Swine production is vital for global food security; however, its sustainability is threatened by the overuse of antibiotics, which contributes to the emergence and dissemination of multidrug-resistant pathogens. Probiotics and bacteriocin-producing bacteria are promising strategies to support animal health and reduce antibiotic dependence. In this study, Enterococcus lactis LBM-M3D, isolated from the porcine gastrointestinal tract, was evaluated for in vitro antimicrobial activity, tolerance to simulated gastrointestinal conditions, genomic safety features, and bacteriocinogenic potential. OBJECTIVES: Oral health's inextricable links to systemic health are highlighted by the emerging oral-gut-brain axis and other well-known axes. There is growing evidence of a complex oral-gut-brain axis linking mouth and gut microbiomes with the central nervous system. Axis disruptions, characterized as oral and gut dysbiosis or microbial imbalances, can trigger oral and systemic inflammation and neuroinflammation, contributing to diseases such as Alzheimer's disease and Parkinson's disease.
MATERIALS AND METHODS: We summarize the oral-gut-brain axis mechanistic pathways, key evidence from hum

Antimicrobial resistance is a natural and inevitable phenomenon that constitutes a severe threat to global public health and economy. Innovative products, active against new targets and with no cross- or co-resistance with existing antibiotic classes, novel mechanisms of action, or multiple therapeutic targets are urgently required. For these reasons, antimicrobial peptides such as bacteriocins constitute a promising class of new antimicrobial drugs under investigation for clinical development. Here, we review the potential therapeutic use of AS-48, a head-to-tail cyclized cationic bacteriocin…

Ready-to-eat (RTE) sliced emulsion type sausages are sensitive to recontamination with Listeria (L.) monocytogenes during processing and packaging steps. Since Listeria spp. are able to grow on those products under cold storage conditions, taking steps to reduce the recontamination risk and implementing antibacterial hurdles contribute to consumer safety and increase the product quality. With this study data about the suitability of culture broth, cell-free supernatant (CFS) or concentrated bacteriocin preparations (CFSconc) of bacteriocin-producing lactic acid bacteria (LAB) obtained from fer

Two NE strains (JGS4143 and JGS4064) produced bacteriocins, which inhibited each other and normal flora strains (n=17), while normal flora strains inhibited neither NE strains nor each other. Thus, it appears that naturally occurring dominance of the gut by NE strains can be reproduced experimentally. Bacte …

OBJECTIVE: This study explores the potential of postbiotic metabolites from lactic acid bacteria (LAB) isolated from human milk and tempeh against Porphyromonas gingivalis, a key pathogen in periodontitis.
DESIGN: Selected LAB strains and their treated cell-free supernatants (TCFS), lyophilized TCFS (L-TCFS), and bacteriocin-like inhibitory substances (BLIS) were tested individually and in combination using agar well diffusion, MIC, MBC, autoaggregation, coaggregation, and biofilm inhibition assays. LAB identification was performed using the API 50 CHL kit and 16S rDNA sequencing.
RESULTS: S

Biofilm-forming pathogens are a major cause of persistent infections, showing limited response to antibiotic treatment. The search for alternative strategies has therefore driven extensive research into the antimicrobial potential of beneficial microorganisms. In the present study, the antibacterial and antibiofilm activity of the commercial probiotic strain Lacticaseibacillus casei ATCC 393 (Lc393) was examined against clinical isolates of Staphylococcus aureus, Salmonella enterica subsp. enterica serovar Enteritidis and Escherichia coli. Lc393 reduced pathogen viability and attachment to the

The current study was carried out to investigate metabolic activities and main probiotic characteristics of two Latilactobacillus sakei strains (8.P1 and 28.P2) isolated from pastırma, a highly seasoned, air-dried cured beef. Both strains showed antimicrobial activity against important foodborne pathogens like Listeria monocytogenes, Staphylococcus aureus, Pseudomonas aeruginosa and so forth. For the characterization of antimicrobial activity, the effect of various enzymes, temperature and pH were tested. The results of the tests demonstrated that the antimicrobial activity of strains was base

BACKGROUND: The development and utilization of probiotics had many environmental benefits for replacing antibiotics in animal production. Bacteria in the intestinal mucosa have better adhesion to the host intestinal epithelial cells compared to bacteria in the intestinal contents. In this study, lactic acid bacteria were isolated from the intestinal mucosa of broiler chickens and investigated as the substitution to antibiotic in broiler production.
RESULTS: In addition to acid resistance, high temperature resistance, antimicrobial sensitivity tests, and intestinal epithelial cell adhesion, En

In this review, the existing research regarding antimicrobial molecules from microorganisms is summarized. The potential antimicrobial compounds from actinomycetes, particularly Streptomyces spp.; archaea; fungi including endophytic, filamentous, and marine-derived fungi, mushroom; and microalgae are briefly described. Furthermore, this review briefly summarizes bacteriocins, halocins, sulfolobicin, etc., that target multiple-drug resistant pathogens an…

Microcin C7 (McC) as a viable immunomodulator peptide can be a potential solution for pathogenic microbial infection in the post-antibiotic era and has gained substantial attention. This study was designed to evaluate the immunomodulatory activity of Microcin C7 in a cyclophosphamide (CTX)-induced immunodeficient mouse model. We show that Microcin C7 treatment significantly alleviated the CTX-caused body weight loss, improved the feed and water consumption to improve the state of the mice, and elevated the absolute number and proportion of peripheral blood lymphocytes as well as the level of h

A mycobacteriocin (M12) produced by Mycobacterium smegmatic ATCC 25855 was partially purified by ammonium sulphate precipitation followed by DEAE-cellulose chromatography and Sephadex G100 chromatography. Production of M12 was maximal when bacteria were harvested after 3 d cultivation in liquid medium and disrupted by sonication. The molecular weight of M12, estimated by Sephadex G100 chromatograpy, was about 85000. M12 was sensitive to proteolytic enzymes but resistant to DNAase and RNAase, and was relatively stable to heat treatment, sonication, ultraviolet irradiation and pH over the range

Microcin J25 (MccJ25) is of special interest due to its "lariat" topology. The lasso architecture is thought to be responsible for its remarkable thermal stability (100°C for 30 min incubation) and potency against Gram-negative bacteria (i.e., MBC of ~4.75 µM against E. coli strain DH5α). This naturally occurring lasso peptide is formed via post-translational modifications (PTMs), including leader peptide removal and macrolactam cyclization between the N-terminus and 8-Glu side chain. Since the discovery of MccJ25 in 1992, synthetic endeavors toward the total chemical synthesis of MccJ25 have
