The incidence of cytogenetically abnormal rogue cells in peripheral blood.

PURPOSE: To compare the occurrence of cytogenetically abnormal rogue cells, characterized by a high frequency of chromosome-type aberrations, in people exposed to ionizing radiation and in non-exposed subjects. MATERIALS AND METHODS: Data on rogue cells from a total of nine cytogenetic studies on radiation-exposed populations and controls were collected from three laboratories in the United Kingdom, France and Finland. The studies were conducted on first-division metaphases of peripheral blood lymphocytes. Solid Giemsa-stained, G- or R-banded and FISH chromosome-painted material was included.

Autografting Ph-negative blood precursor cells in chronic myeloid leukaemia.

The study was devised to evaluate whether it was possible to collect Philadelphia-negative precursor cells in patients with chronic myeloid leukaemia. The approach was based on previous experience showing that complete remission (Ph-negative bone marrow cells) is rarely achieved after chemotherapy and is very short-lasting. We decided to explore whether it was possible to collect Ph-negative precursor cells in peripheral blood during the early phase of haemopoietic recovery. These data show that: the collection of Ph-negative precursor cells occurred in 12/16 (75%) patients mobilized within on

Cytokine production by peripheral blood mononuclear cells in human coccidioidomycosis.

The production and mRNA expression of the cytokines interferon-gamma (IFN-gamma) and interleukin (IL)-4, -10, and -12 by peripheral blood mononuclear cells (PBMC) after incubation with the coccidioidal antigen toluene spherule lysate (TSL) from various subjects were measured. The IFN-gamma concentration in PBMC supernatants incubated for 72 h from 8 subjects with disseminated coccidioidomycosis was significantly less than that from 7 healthy, coccidioidal-immune subjects (P = .015). No differences were seen among the subject groups in the concentrations of IL-4, -10, or -12. By use of competit

[Nitrergic reactivity in endothelial cells of human uveal blood vessels].

BACKGROUND: We investigated the presence of Nicotinamid-adenin-dinucleotid-phosphat (NADPH)-diaphorase activity in endothelial cells of the human uveal vessels, using a histochemical procedure which presumably localizes the reaction product nitric oxide. MATERIAL AND METHODS: 11 donor eyes destined for corneal transplantation (male: 6; female: 5; age: 56 +/- 8 years) were used within 14 +/- 7 h postmorten. After removal of the cornea and retinal pigment epithelium, flat mounts of the choroid were made. Serial sections, 12-16 microns in thickness were cut from the ciliary body and iris using a

Natural killer (NK) cells inhibit human umbilical cord blood erythropoiesis.

To examine the role of NK cells on in vitro human umbilical cord (HUC) blood erythropoietic progenitor growth, 25 normal HUC blood samples were depleted of CD56+ cells by using immunomagnetic beads coated with CD56 monoclonal antibodies (mAb). When stimulated by erythropoietin (Epo) to form colonies in plasma clot medium, the CD(56+)-depleted preparations demonstrated a two-fold increase in the number of early erythropoietic progenitors (BFU-E) over nondepleted preparations. This stimulatory effect of CD56+ depletion on BFU-E growth was not due to artifactual stimulations of other accessory ce

[Harvesting of mononuclear cells from peripheral blood in malignant hemopathies].

After demonstrating through animal models that progenitor cells with haemopoietic regenerative capability are present in peripheral blood, such precursor cells were also found in the mononucleated cell (MNC) fraction of human peripheral blood. It was shown that such cells increase in the recuperative period following chemotherapy. A number of studies have demonstrated that autologous infusion of such MNC achieved by cytapheresis may lead to a quick restoration of haemopoiesis. The initial results of peripheral blood MNC achievement in 18 patients with different malignancies of the blood are pr

Detection of IgM autoantibodies in eluates from red blood cells.

Forty-two patients with autoimmune haemolysis, whose red cells were sensitized with IgM, were studied in order to determine suitable techniques for investigating the form of erythrocyte-bound IgM, particular emphasis being placed on identifying forms of low molecular weight. Four elution techniques were assessed for their efficiency in removing cell-bound IgM; these were the heat elution, acid stromal, organic solvent and freeze-thaw methods. Eluted IgM was detected by polyacrylamide gel electrophoresis followed by immunoblotting; the heat elution method proved to be the best. Although oligome

Human lupus-type inclusions in umbilical cord blood mononuclear cells.

Mononuclear cells purified from umbilical cord blood activated oligoadenylate synthetase and formed human lupus-type inclusions (LI) when cultured with the purified recombinant human leukocyte interferon, IFLrA. LI frequencies increased from 0% to a low of 0.75% and a high of 6.25% in 4-day cultures with IFLrA (100 units/ml). These interferon-induced responses in the mononuclear cells of neonates indicate that LI are solely an intrinsic product of normal cells, and not an exogenous virus or some other environmental agent.

Deformability and intrinsic material properties of neonatal red blood cells.

Whole cell and membrane deformability are essential for red blood cell (RBC) survival and for effective blood flow. Neonatal RBCs display several specific properties (eg, large size, high hemoglobin F) that could influence their deformation characteristics and contribute to their shortened life span. The present study was designed to compare selected rheologic properties (cellular deformability, pressure required to aspirate RBC into micropipettes, static and dynamic viscoelastic material properties) of neonatal and adult RBCs. RBC deformability, as studied by a rheoscope, was similar for neon

Are the occasional aneuploid cells in peripheral blood cultures significant?

Cytogenetic results of 1,500 consecutive clinical cases from a young population were analyzed for rare cells with hypermodality (greater than or equal to 47 chromosomes) or hypomodality (less than or equal to 45 chromosomes). Such instances of non-modal chromosome gains or losses were random relative to referral diagnosis or modal karyotype. However, chromosome loss was correlated with size, smaller chromosomes being lost more frequently (correlation coefficient = 0.794). Sex chromosome gain or loss in vitro was of particular interest since mosaicism in vivo is frequently found in patients pre

Vascular resistance and transit time of sickle red blood cells.

Changes in vascular resistance and mean transit time caused by the introduction either normal or sickled red blood cells (SSRBC) into the circulation were measured in the coronary and mesenteric vascular beds. The cat mesentery was perfused at constant flow through the superior mesenteric artery with a Ringer solution containing albumin. A rise in input pressure was observed after introducing RBC's. (10% hematocrit) at a constant rate, into the perfusate. The pressure rise caused by SSRBC's relative to that caused by normal RBC's represents the relative change in vascular resistance caused by …

Cells containing Epstein--Barr nuclear antigen (EBNA) in peripheral blood.

The anti-complement immunofluorescence (ACIF) method was used to demonstrate EBNA in B lymphocytes separated from the peripheral blood of patients with infectious mononucleosis (IM) or other diseases. In the acute phase of IM of Epstein-Barr virus (EBV) origin, 0.5--1% of the B lymphocytes proved to be positive in 6 of the 8 patients tested. In two of the positive cases the test was repeated 20 and 26 days, respectively, after clinical symptoms had appeared; the result was negative in both cases. No EBNA-positive cells were found in the peripheral blood of 17 patients with Hodgkin disease, 3 w

Blood Cell Disorders and the Nervous System.

PURPOSE OF REVIEW: This article discusses the epidemiology, diagnosis, treatment, and prevention of neurologic complications of common and rare blood cell disorders. RECENT FINDINGS: A growing number of preventive treatment options are available for stroke in sickle cell disease. Paroxysmal nocturnal hemoglobinuria and immune thrombocytopenia can lead to stroke. Thrombotic thrombocytopenic purpura frequently causes neurologic symptoms and should be considered in the differential diagnosis of a patient with neurologic symptoms, thrombocytopenia, and hemolytic anemia. Polycythemia vera and esse
