Comprehensive genomic analysis of ESBL- and carbapenemase-producing Escherichia coli ST131 in Japan: Genetic characteristics of pandemic clade C.

INTRODUCTION: The global dissemination of extended-spectrum β-lactamase (ESBL)- and carbapenemase-producing Escherichia coli sequence type 131 (ST131), particularly clade C, represents a major public health concern. However, the clade-specific genomic features underlying the long-term persistence and international spread of this lineage remain incompletely understood. METHODS: We performed whole-genome sequencing and comparative genomic analyses of 121 clinical E. coli isolates, including 83 ST131 and 38 non-ST131 strains, collected from 24 healthcare-associated facilities in Japan between 20

A broadly applicable split-luciferase biosensor approach for rapid antibody detection in emerging infectious diseases.

Conventional immunoassays such as ELISA are widely used in serological testing, yet their reliance on multi-step workflows and labeled reagents limits diagnostic scalability and speed. Bioluminescence-based biosensors are attractive alternatives that offer high sensitivity, operational simplicity, and cost efficiency for detecting diverse analytes. Here, we present a broadly compatible bioluminescent biosensor for antibody detection based on analyte-mediated reconstitution of split-Nanoluciferase (NanoLuc). The platform utilizes engineered bifunctional probes, comprising the protein G C2 domai

Integrative Multi-Omics and Molecular Simulations Identify PRKCD as a Biological Modulator in Colon Adenocarcinoma: Validation With Phoenix dactylifera Phytochemicals and Comparison With Rottlerin.

Colorectal cancer (CRC) is the leading cause of cancer-related deaths worldwide. Colon adenocarcinoma (COAD), the most common histological subtype, is characterized by a poor prognosis in advanced stages due to significant molecular heterogeneity. Protein kinase C delta (PKCδ), encoded by PRKCD, is a context-dependent regulatory kinase in tumor biology. Its expression, genomic alterations, and binding interactions with phytochemicals from Phoenix dactylifera were studied using a comprehensive in silico approach. PRKCD was found to be downregulated in all TCGA-COAD tumor subgroups when compared

A COLQ Missense Mutation in Sphynx and Devon Rex Cats with Congenital Myasthenic Syndrome.

An autosomal recessive neuromuscular disorder characterized by skeletal muscle weakness, fatigability and variable electromyographic or muscular histopathological features has been described in the two related Sphynx and Devon Rex cat breeds (Felis catus). Collection of data from two affected Sphynx cats and their relatives pointed out a single disease candidate region on feline chromosome C2, identified following a genome-wide SNP-based homozygosity mapping strategy. In that region, we further identified COLQ (collagen-like tail subunit of asymmetric acetylcholinesterase) as a good candidate

Initial presentation with elevated transaminases and subsequent hematuria in limb-girdle muscular dystrophy type 2B: A case report.

RATIONALE: Limb-girdle muscular dystrophy type 2B (LGMD2B) is a degenerative muscle disorder induced by mutations in the dysferlin gene. Dysferlin is involved in membrane repair and vesicle fusion through its 7 C2 calcium-binding domains, which mediate these calcium-dependent processes. It is currently considered an untreatable neuromuscular condition with a poor prognosis. The estimated incidence of this disease is 1 to 6.5 per 100,000 individuals. The primary clinical features of LGMD2B include proximal muscle weakness and elevated serum creatine kinase (CK) levels; occasionally, patients ma

Collectins and collectin receptors in innate immunity.

This thesis is based on nine papers and a review on the collectins and collectin receptors in innate immunity. The collectins are a family of proteins in which the individual chains consist of a C-type lectin domain attached to a collagen domain via an alpha-coiled neck region. The chains are organized into a triple collagen helix and oligomerized through N-terminally located cysteines. The collectins have a dual function: one is to bind specifically to carbohydrate structures on the surface of a pathogen; the other is subsequently to recruit other cells and molecules to destroy the pathogen.

Continuous gait cycle index estimation for electrical stimulation assisted foot drop correction.

BACKGROUND: Walking impairment after stroke can be addressed with the use of drop foot stimulators (DFS). Many studies have demonstrated that DFS improves walking speed, reduces spasticity and reduces the physiologic effort of walking. Current DFS, through activation of the common peroneal nerve, elicit ankle dorsiflexion during swing phase of gait. DFS are generally piloted by force sensing resistor placed in the shoe of the affected side with stimulation triggered ON by heel rise and triggered OFF by heel strike. A tilt sensor can also be used with stimulation triggered by the tilt of the sh

How Does the maternal immune system contribute to the development of pre-eclampsia?

An immunological aura has hovered over the study of pre-eclampsia for many years but there has still been little progress in explaining the various 'immune' phenomena associated with this elusive disease. When considering the primary defect of placentation that leads to pre-eclampsia the focus should be on the intermingling of the invasive placental trophoblast cells with maternal leukocytes in the uterine wall. The MHC status of trophoblast cells is a crucial factor to be considered, as these molecules can act as ligands for uterine immune cells, including T cells, NK cells and myelomonocytic

A new Mel-CAM (CD146)-specific monoclonal antibody, MN-4, on paraffin-embedded tissue.

Mel-CAM (also termed CD146 or MUC18) is a cell-adhesion molecule belonging to the immunoglobulin gene superfamily. It mediates cell-cell interaction through heterophilic Mel-CAM/ligand adhesion. Previous studies showed Mel-CAM immunoreactivity in normal and neoplastic human tissues, but an extensive assessment of Mel-CAM distribution was not performed because of the lack of a Mel-CAM-specific monoclonal antibody that could be used in routinely processed, formalin-fixed, paraffin-embedded tissues. Therefore, we developed a mouse monoclonal antibody, MN-4, that specifically recognized a fixation

Metabolic shifts induced by fatty acid synthase inhibitor orlistat in non-small cell lung carcinoma cells provide novel pharmacodynamic biomarkers for positron emission tomography and magnetic resonance spectroscopy.

PURPOSE: Abnormal fatty acid (FA) synthesis is one of the common features of cancer. Fatty acid synthase (FASN), a multifunctional enzyme playing a key role in biosynthesis of FA, is up-regulated in prostate, breast, and lung carcinomas. Orlistat is a FDA-approved anti-obesity drug that inhibits the thioesterase domain of FASN, interferes with cellular FA synthesis, can arrest tumor cell proliferation, and induces tumor cell apoptosis. The current study was aimed to investigate the metabolic changes associated with FASN inhibition by orlistat and to understand the molecular mechanisms behind t

A new class of potent matrix metalloproteinase 13 inhibitors for potential treatment of osteoarthritis: Evidence of histologic and clinical efficacy without musculoskeletal toxicity in rat models.

OBJECTIVE: Matrix metalloproteinases (MMPs) have long been considered excellent targets for osteoarthritis (OA) treatment. However, clinical utility of broad-spectrum MMP inhibitors developed for this purpose has been restricted by dose-limiting musculoskeletal side effects observed in humans. This study was undertaken to identify a new class of potent and selective MMP-13 inhibitors that would provide histologic and clinical efficacy without musculoskeletal toxicity. METHODS: Selectivity assays were developed using catalytic domains of human MMPs. Freshly isolated bovine articular cartilage

Computational analysis of congenital heart disease associated SNPs: unveiling their impact on the gene regulatory system.

Congenital heart disease (CHD) is a prevalent condition characterized by defective heart development, causing premature death and stillbirths among infants. Genome-wide association studies (GWASs) have provided insights into the role of genetic variants in CHD pathogenesis through the identification of a comprehensive set of single-nucleotide polymorphisms (SNPs). Notably, 90-95% of these variants reside in the noncoding genome, complicating the understanding of their underlying mechanisms. Here, we developed a systematic computational pipeline for the identification and analysis of CHD-associ

A Member of the Ferlin Calcium Sensor Family Is Essential for Toxoplasma gondii Rhoptry Secretion.

Invasion of host cells by apicomplexan parasites such as Toxoplasma gondii is critical for their infectivity and pathogenesis. In Toxoplasma, secretion of essential egress, motility, and invasion-related proteins from microneme organelles is regulated by oscillations of intracellular Ca2+ Later stages of invasion are considered Ca2+ independent, including the secretion of proteins required for host cell entry and remodeling from the parasite's rhoptries. We identified a family of three Toxoplasma proteins with homology to the ferlin family of double C2 domain-containing Ca2+ sensors. In humans

Novel phospholipase C zeta 1 mutations associated with fertilization failures after ICSI.

STUDY QUESTION: Are phospholipase C zeta 1 (PLCZ1) mutations associated with fertilization failure (FF) after ICSI? SUMMARY ANSWER: New mutations in the PLCZ1 sequence are associated with FFs after ICSI. WHAT IS KNOWN ALREADY: FF occurs in 1-3% of ICSI cycles, mainly due to oocyte activation failure (OAF). The sperm PLCζ/PLCZ1 protein hydrolyzes phosphatidylinositol (4, 5)-bisphosphate in the oocyte, leading to intracellular calcium release and oocyte activation. To date, few PLCZ1 point mutations causing decreased protein levels or activity have been linked to FF. However, functional altera
