Coordinated neural progenitor adaptations contribute to primate neocortical downscaling.

Most primates have a large, highly folded (gyrencephalic) neocortex, a feature present in the primate common ancestor. In contrast, several New World monkey species display a comparatively small and largely smooth, unfolded (lissencephalic) neocortex. One prominent example is the common marmoset, an increasingly popular model in neuroscience. This phenotype likely reflects an evolutionary reduction from the ancestral primate condition, implying modifications in neurodevelopmental programs. One essential basis for neocortical development is the activity and behavior of neural progenitor cells (

Morphological Investigation of the Female Internal Genital Organs of Marca's Marmoset (Mico marcai).

This study provides the first description of the anatomy and histology of the female internal genital organs of Mico marcai. Descriptions were based on samples collected from two free-ranging specimens. As in other species of Platyrrhini, the ovaries were oval-shaped. Histological findings suggest both females were at the beginning of the luteal phase, as shown by the presence of luteal tissue in the ovaries and the high incidence of follicular atresia. As described in other primates, branching longitudinal folds and a tunica muscularis were seen in uterine tubes in M. marcai. The tunica muscu

Comprehensive DNA methylation analysis of brain and peripheral tissues following chronic risperidone treatment in common marmosets.

Antipsychotic drugs are increasingly recognized to exert therapeutic effects not only through neurotransmitter modulation but also, in part, through epigenetic mechanisms. Risperidone is an atypical antipsychotic drug widely used to treat schizophrenia, yet its in vivo epigenomic effects remain poorly understood. We investigated genome-wide DNA methylation changes induced by chronic risperidone administration in the common marmoset, a non-human primate with high translational relevance. Adult males were treated orally with risperidone for 28 days. DNA methylation was analyzed in four brain reg

Early parental deprivation during primate infancy has a lifelong impact on gene expression in the male marmoset brain.

Adverse early life experiences are well-established risk factors for neurological disorders later in life. However, the molecular mechanisms underlying the impact of adverse experiences on neurophysiological systems throughout life remain incompletely understood. Previous studies suggest that social attachment to parents in early development are indispensable for infants to grow into healthy adults. In situations where multiple offspring are born in a single birth in common marmosets, human hand-rearing is employed to ensure the survival of the offspring in captivity. However, hand-reared marm

Distribution of [11C]-JNJ-42491293 in the marmoset brain: a positron emission tomography study.

JNJ-42491293 is a metabotropic glutamate 2 (mGlu2) positive allosteric modulator (PAM) that was radiolabelled with [11C]- to serve as a positron emission tomography (PET) ligand. Indeed, in vitro, the molecule displays high selectivity at mGlu2 receptors. However, PET experiments performed in rats, macaques and humans, have suggested that [11C]-JNJ-42491293 could interact with an unidentified, non-mGlu2 receptor binding site. The brain distribution of [11C]-JNJ-42491293 has not been determined in the brain of the common marmoset, a small non-human primate increasingly used in neuroscience rese

Frontal cortex activity during the production of diverse social communication calls in marmoset monkeys.

Vocal communication is essential for social behaviors in humans and non-human primates. While the frontal cortex is crucial to human speech production, its role in vocal production in non-human primates has long been questioned. It is unclear whether activities in the frontal cortex represent diverse vocal signals used in non-human primate communication. Here we studied single neuron activities and local field potentials (LFP) in the frontal cortex of male marmoset monkeys while the animal engaged in vocal exchanges with conspecifics in a social environment. We found that both single neuron ac

Diversity of Ganglion Cell Responses to Saccade-Like Image Shifts in the Primate Retina.

Saccades are a fundamental part of natural vision. They interrupt fixations of the visual gaze and rapidly shift the image that falls onto the retina. These stimulus dynamics can cause activation or suppression of different retinal ganglion cells, but how they affect the encoding of visual information in different types of ganglion cells is largely unknown. Here, we recorded spiking responses to saccade-like shifts of luminance gratings from ganglion cells in isolated marmoset retinas and investigated how the activity depended on the combination of presaccadic and postsaccadic images. All iden

Natural SARS-CoV-2 Infection in a Free-Ranging Black-Tailed Marmoset (Mico melanurus) from an Urban Area in Mid-West Brazil.

The emergence of spillover pathogens such as SARS-CoV-2 is a risk to vulnerable human populations. We report natural SARS-CoV-2 infection in a free-ranging adult female black-tailed marmoset (Mico melanurus) from an urban area of Cuiabá, Mato Grosso State, Brazil. The animal was found after a motor vehicle collision without previous clinical history. Necropsy confirmed polytrauma. Severe multifocal to coalescent haemorrhage and mild multifocal peribronchial lymphocytic hyperplasia were seen in lung sections. The alveolar septa were multifocally expanded by a few lymphocytes. Mild lymphocytic p

Reduced differentiation of intestinal epithelial cells in wasting marmoset syndrome.

Wasting marmoset syndrome (WMS) is a serious disease in captive common marmoset (Callithrix jacchus) colonies. Because of the high mortality rates, elucidation of the underlying mechanisms is essential. In this study, we compared the histopathology, the number of each epithelial cell in the jejunum and colon, and the expression patterns of some molecular markers between healthy and WMS-affected marmosets. Atrophy of villi in the jejunum and mononuclear cell infiltration in the lamina propria were observed in the intestinal tract of WMS-affected marmosets. Although the numbers of transient ampl…

Marmosets: a promising model for probing the neural mechanisms underlying complex visual networks such as the frontal-parietal network.

The technology, methodology and models used by visual neuroscientists have provided great insights into the structure and function of individual brain areas. However, complex cognitive functions arise in the brain due to networks comprising multiple interacting cortical areas that are wired together with precise anatomical connections. A prime example of this phenomenon is the frontal-parietal network and two key regions within it: the frontal eye fields (FEF) and lateral intraparietal area (area LIP). Activity in these cortical areas has independently been tied to oculomotor control, motor pr

Novel gastrointestinal disease in common marmosets characterised by duodenal dilation: a clinical and pathological study.

Common marmosets (Callithrix jacchus) are frequently used for biomedical research but gastrointestinal diseases have been major health problems to maintain captive marmosets. We have diagnosed a novel gastrointestinal disease in marmosets, as which we propose to call 'marmoset duodenal dilation syndrome'; this disease is characterised by proximal duodenal obstruction and dilation. This study aimed to reveal the clinical and pathological findings of this syndrome and establish appropriate diagnostic imaging methods. Animals with the syndrome comprised 21.9% of the necropsy cases at the Central …

Functional Localization of the Frontal Eye Fields in the Common Marmoset Using Microstimulation.

The frontal eye field (FEF) is a critical region for the deployment of overt and covert spatial attention. Although investigations in the macaque continue to provide insight into the neural underpinnings of the FEF, due to its location within a sulcus, the macaque FEF is virtually inaccessible to electrophysiological techniques such as high-density and laminar recordings. With a largely lissencephalic cortex, the common marmoset (Callithrix jacchus) is a promising alternative primate model for studying FEF microcircuitry. Putative homologies have been established with the macaque FEF on the ba…

Rapid modulations of the vocal structure in marmoset monkeys.

Humans and some animal species show flexibility in vocal production either voluntarily or in response to environmental cues. Studies have shown rapid spectrotemporal changes in speech or vocalizations during altered auditory feedback in humans, songbirds and bats. Non-human primates, however, have long been considered lacking the ability to modify spectrotemporal structures of their vocalizations. Here we tested the ability of the common marmoset (Callithrix jacchus), a highly vocal New World primate species to alter spectral and temporal structures of their species-specific vocalizations in t…

Manganese-enhanced MRI visualizes V1 in the non-human primate visual cortex.

MRI at 7 Tesla has been used to investigate the accumulation of manganese in the occipital cortex of common marmoset monkeys (Callithrix jacchus) after administering four fractionated injections of 30 mg/kg MnCl(2) . 4H(2)O in the tail vein. We found a statistically significant decrease in T(1) in the primary (V1) and secondary (V2) areas of the visual cortex caused by an accumulation of manganese. The larger T(1) shortening in V1 (DeltaT(1) = 640 ms) relative to V2 (DeltaT(1) = 490 ms) allowed us to robustly detect the V1/V2 border in vivo using heavily T(1)-weighted MRI. Furthermore, the dor…

Visual areas in lateral and ventral extrastriate cortices of the marmoset monkey.

The representation of the visual field in visual areas of the dorsolateral, lateral, and ventral cortices was studied by means of extracellular recordings and fluorescent tracer injections in anaesthetised marmoset monkeys. Two areas, forming mirror-symmetrical representations of the contralateral visual field, were found rostral to the second visual area (V2). These were termed the ventrolateral posterior (VLP) and the ventrolateral anterior (VLA) areas. In both areas, the representation of the lower quadrant is located dorsally, between the foveal representation of V2 and the middle temporal

Four-week repeated inhalation study of HCFC 225ca and HCFC 225cb in the common marmoset.

Four male and three female marmosets in each group were exposed to air only, 1000 ppm of HCFC 225ca or 5000 ppm of HCFC 225cb, for 6 h per day for 28 consecutive days. HCFC 225ca caused a slight reduction in body weight. HCFC 225cb occasionally caused somnolence during exposure and vomiting on the first day of exposure. Clinical chemistry findings included a mild reduction of triglyceride, cholesterol and phospholipid levels and increased GOT level in the HCFC 225ca exposure group. HCFC 225cb also caused a reduction of triglyceride levels in some animals. HCFC 225ca caused a slight increase of

Effects of small doses of dioxins on the immune system of marmosets and rats.

There is no doubt that TCDD is capable of inducing effects on a variety of components and functions of the immune system in a variety of species. In fact, such changes seem to belong to the most sensitive variables affected by TCDD. Some of the biological effects, induced at rather high doses of TCDD exhibiting general toxicity (> 3 micrograms TCDD/kg body wt), may be considered unspecific or the result of the pronounced thymus involution. However, other effects (such as that on lymphocyte subtype patterns in marmosets or a reduced resistance of mice to influenza viruses) have been reported to

Local production of progesterone by the corpus luteum of the marmoset monkey in response to perfusion with chorionic gonadotrophin and melatonin in vivo.

The effect of human chorionic gonadotrophin (hCG) and melatonin on the local production of progesterone by the marmoset corpus luteum was investigated in vivo using a perfusion cannula system. Progesterone secretion was measured in 10-min fractions of buffer which had been perfused through the corpus luteum at a flow rate of 70 microliters/min for a maximum of 3 h in anaesthetized animals. Two corpora lutea were cannulated in each animal; one for perfusion of test material and the other for perfusion with buffer alone as a control.
