In vivo imaging to trace the dissemination of Aeromonas hydrophila in common carp (Cyprinus carpio) after intestinal infection.

Aeromonas hydrophila (A. hydrophila) is a pathogenic bacterium that often causes serious economic losses to aquaculture and fisheries. Its infection and pathogenesis of fish are currently not fully understood. In this study, A. hydrophila was labeled with Fluorescein Isothiocyanate (FITC) and administered to common carp via gavage to trace its dissemination in common carp (Cyprinus carpio) after intestinal infection by in vivo imaging system. The optimal FITC concentration for labeling was determined to be 30 μg/mL, with a labeling time of 3 h at a bacterial concentration of 108 CFU/mL, with n

Grass carp (Ctenopharyngodon idella) resist heat stress by maintaining mitochondrial quality and enhancing energy production.

This study aims to explore the resistance mechanism of grass carp (Ctenopharyngodon idella) to heat stress (HS). Grass carp were cultured at 28°C, and then exposed to 30°C, 32°C or 34°C with a heating rate of 1°C/d, respectively. The results show that serum aspartate aminotransferase (AST), alanine aminotransferase (ALT) levels and liver malondialdehyde (MDA) content significantly increased after HS. In contrast, liver total antioxidant capacity (T-AOC) was significantly reduced after HS. Additionally, serum triglyceride (TG) and free fatty acid (FFA) levels increased after HS. Notably, the he

Growth-promoting potential and immunostimulatory of poly-β-hydroxybutyrate in common carp (Cyprinus carpio) fingerlings culture.

The natural polymer poly-β-hydroxybutyrate (PHB) is converted to β-hydroxybutyric acid, which is similar to short-chain fatty acids, via microbial fermentation and host enzyme breakdown. This study investigated the impact of different PHB concentrations (Control, 1.00, 3.00, and 5.00% substitution) on growth performances and fish welfare in common carp fingerlings. After a 60-day trial, fish fed on diet containing 1.00% PHB exhibited significantly higher weight gain and improved feed conversion efficiency compared to the control group. Furthermore, analysis of enzymatic activity showed elevate

Spring viraemia of carp virus modulates the time-dependent unfolded protein response to facilitate viral replication.

INTRODUCTION: The spring viraemia of carp virus (SVCV) poses a significant threat to global aquaculture, yet effective antiviral drugs and vaccines remain unavailable. Understanding the interplay between host-pathogen interactions and SVCV replication is crucial for devising preventive strategies. METHODS: ZF4 cells were exposed to UV-inactivated SVCV or live SVCV at different multiplicities of infection, and the modulation of the unfolded protein response (UPR) was assayed by qPCR at different times. Moreover, ZF4 cells were treated with several UPR modulators to investigate their effect on

Genome-Resolved Co-Infection by Aeromonas veronii and Shewanella sp. in Koi Carp: A Zoonotic Risk for Aquarists.

Co-infections are increasingly recognized as drivers of disease in ornamental fish, yet their genomic underpinnings and zoonotic implications remain underexplored compared to farmed species. Leveraging a One Health perspective, we investigated an acute mortality event in koi carp and characterized a co-infection by opportunistic aquatic bacteria that are also implicated in human disease. We isolated Aeromonas veronii and Shewanella sp. from a moribund koi using culture, biochemical assays, and MALDI-TOF MS, then generated draft genomes and performed orthology (OrthoVenn3), pathway annotation (

Identification of an IRF8 gene in common carp (Cyprinus carpio. L) and its regulatory role in immune responses.

BACKGROUND: Interferon (IFN) regulatory factors (IRF) are the crucial transcription factors for IFN expression and leading host cells response to viral infection. IRF8 in mammals plays vital roles in the innate and adaptive immune systems. In this study, we identified and characterized the common carp (Cyprinus carpio. L) IRF8 gene (ccIRF8) to further clarify the function of IRF8 in teleost fish. RESULTS: The complete cDNA sequence of ccIRF8 was 1431 bp and encodes a polypeptide of 431 amino acids. Analysis of the putative amino acid sequence showed that ccIRF8 encodes structures typical of t

Effects of a Synthetic Isoquinoline Derivative Against Ichthyophthirius multifiliis In Vivo and In Vitro in Grass Carp (Ctenopharyngodon idella).

White spot disease, caused by the parasitic Ichthyophthirius multifiliis, induces mortality exceeding 80% in intensive aquaculture systems, resulting in global annual losses of >$1 billion. Current chemical controls (e.g., formaldehyde) face environmental persistence and drug resistance challenges. This study developed a synthetic isoquinoline derivative, BHTCA (s)-2-tert-butoxycarbonyl-7-hydroxy-1,2,3,4-tetrahydroisoquinoline-3-carboxylic acid), as a biodegradable alternative against I. multifiliis in grass carp (Ctenopharyngodon idella). Analysis of the mechanism of lncRNA_89636 in the regulation of asip by targeting miR-200a in koi carp skin color differentiation.

As a globally renowned ornamental fish, the body color and patterns of koi carp play a decisive role in their market value. Although genetic factors drive color diversity, the non-coding regulatory mechanisms underlying koi color variation are poorly understood. This study focused on two color patterns (red and black) in Hi-Utsuri koi. Histological analysis revealed that black skin primarily consists of dendritic melanocytes, whereas red skin contains numerous granular erythrophores. Whole transcriptome and small RNA sequencing identified many differentially expressed non-coding RNAs (lncRNAs,

Ferulic acid alleviates avermectin induced renal injury in carp by inhibiting inflammation, oxidative stress and apoptosis.

Avamectin (AVM), a macrolide antibiotic, is widely used in fisheries, agriculture, and animal husbandry, however, its irrational use poses a great danger to aquatic organisms. Ferulic acid (FA) is a natural chemical found in the cell walls of plants. It absorbs free radicals from the surrounding environment and acts as an antioxidant. However, the protective effect of FA against kidney injury caused by AVM has not been demonstrated. In this study, 60 carp were divided into the control group, AVM group (2.404 μg/L), FA+AVM group and FA group (400 mg/kg). Pathological examination, quantitative r

Decoding the effect of photoperiodic cues in transducing kisspeptin-melatonin circuit during the pubertal onset in common carp.

This study unravels the intricate interplay between photoperiod, melatonin, and kisspeptin to orchestrate the pubertal onset of Common carp. Female fingerlings exposed to long days (LD) exhibited a hormonal crescendo, with upregulated hypothalamic-pituitary-ovarian (HPO) axis genes (kiss1, kiss1r, kiss2, gnrh2, gnrh3) and their downstream targets (lhr, fshr, ar1, esr1). However, the expression of the melatonin receptor (mtnr1a) diminished in LD, suggesting a potential inhibitory role. This hormonal symphony was further amplified by increased activity of key transcriptional regulators (gata1, g

Ornamental fish mortality reveals an old parasite introduction: A case study of Koi carp and fish louse.

Koi carp are globally known for their colors and cultural significance. The introduction of these fish to new environments poses a threat to local biodiversity, in addition to releasing parasites, such as argulid ectoparasites. This study presents a record of Argulus japonicus infecting carp in an artificial lake in Southern Brazil using morphological and molecular methods, with a 100% prevalence (n = 3) and a mean intensity of 21.6 parasites per host, distributed over the body surface. The invasion history of hosts in the study locality indicates that the introduction of A. japonicus occurred

Surviving without oxygen involves major tissue specific changes in the proteome of crucian carp (Carassius carassius).

The crucian carp (Carassius carassius) can survive complete oxygen depletion (anoxia) for several months at low temperatures, making it an excellent model for studying molecular adaptations to anoxia. Still, little is known about how its global proteome responds to anoxia and reoxygenation. By applying mass spectrometry-based proteome analyses on brain, heart and liver tissue from crucian carp exposed to normoxia, five days anoxia, and reoxygenation, we found major changes in particularly cardiac and hepatic protein levels in response to anoxia and reoxygenation. These included tissue-specific

NMR technique revealed the metabolic interference mechanism of the combined exposure to cadmium and tributyltin in grass carp larvae.

Widespread human activity has resulted in the presence of different pollutants in the aquatic environment that does not exist in isolation. The study of the effects of contamination of aquatic organisms is of great significance. To assess the individual and combined toxicity of cadmium (Cd) and tributyltin (TBT) to aquatic organisms, juvenile grass carp (Ctenopharyngodon idella) were exposed to Cd (2.97 mg/L), TBT (7.5 μg/L), and their mixture MIX. The biological response was evaluated by nuclear magnetic resonance (NMR) analysis of plasma metabolites. Plasma samples at 1, 2, 4, 8, 16, 32, and

Changes of the clock gene expression in central and peripheral organs of common carp exposed to constant lighting conditions.

In both mammals and fish, the circadian system is composed of oscillators that function at the cellular, tissue, and system levels and show the cyclic expression of clock genes. The organization and functioning of the biological clock in fish has not yet been characterized in detail, therefore, in the present study, an extensive analysis of the rhythmic expression of the main components of the biological clock in the central and peripheral organs of common carp was performed. The diurnal changes in clock gene expression were determined with respect to the subjective light cycle in fish exposed

Grass carp (Ctenopharyngodon idellus) Cdc25a down-regulates IFN 1 expression by reducing TBK1 phosphorylation.

In vertebrates, TANK Binding Kinase 1 (TBK1) plays an important role in innate immunity, mainly because it can mediate production of interferon to resist the invasion of pathogens. In mammals, cell division cycle-25a (Cdc25a) is a member of the Cdc25 family of cell division cycle proteins. It is a phosphatase that plays an important role in cell cycle regulation by dephosphorylating its substrate proteins. Currently, many phosphatases are reported to play a role in innate immunity. This is because the phosphatases can shut down or reduce immune signaling pathways by down-regulating phosphoryla

Susceptibility of Goldfish to Cyprinid Herpesvirus 2 (CyHV-2) SH01 Isolated from Cultured Crucian Carp.

Cyprinid herpesvirus 2 (CyHV-2), a member of the Alloherpesviridae family belonging to the genus Cyprinivirus, is a fatal contagious aquatic pathogen that affects goldfish (Carassius auratus) and crucian carp (Carassius carassius). Although crucian carp and goldfish belong to the genus Carassius, it is unclear whether they are susceptible to the same CyHV-2 isolate. In addition, the origin of the crucian carp-derived CyHV-2 virus isolate remains unclear. CyHV-2 SH01 was isolated during herpesviral hematopoietic necrosis disease (HVHN) outbreaks in crucian carp at a local fish farm near Shangha

3nLcn2, a teleost lipocalin 2 that possesses antimicrobial activity and inhibits bacterial infection in triploid crucian carp.

Lipocalin 2 (Lcn2) has been identified in mammals, however, the in vivo function of fish Lcn2 is essentially unknown. Triploid crucian carp (3n = 150) of red crucian carp (female, 2n = 100) and allotetraploid (male, 4n = 200) shows better resistance to pathogenic infections. To elucidate the antimicrobial mechanism of triploid crucian carp, we examined the function of a novel Lcn2 from triploid crucian carp (3nLcn2). 3nLcn2 is 183 residues in length and contains a conserved lipocalin domain. Quantitative real time reverse transcription PCR (qRT-PCR) analysis showed that 3nLcn2 expression occur
