Preparation, isolation, purification and identification of ACE inhibitory fractions from yak casein fermented by Bacillus cereus (XBMU-13).

Food-derived angiotensin-converting enzyme (ACE) inhibitory peptides have been shown to contribute to blood pressure reduction. In this study, a protease-producing bacterial strain was isolated and identified for fermentation of yak milk casein to produce ACE inhibitory peptides. Subsequently, fermentation conditions were optimized, and the resulting peptides were purified, and identified. The results showed that a Bacillus cereus strain, designated XBMU-13, was isolated from naturally fermented bean curd and exhibited a protease activity of 86.27 ± 3.82 U/mL. Using response surface methodolog

Arsenic intervened anarchy in cauda epididymis and spermatozoa of adult rats is amended by casein and pea enriched high protein diet.

Arsenic-mediated male reprotoxicosis has emerged as a serious health concern. Arsenic-induced oxidative stress is known to impair sperm quality, cause erectile dysfunction, and lead to infertility. On the other hand, a formulated high-protein diet (FHPD) enriched with casein and pea is considered to possess significant antioxidant potential. The present study aimed to investigate the protective efficacy of such a protein-rich diet against arsenic-induced epididymal and spermatozoal dysfunctions. Adult male rats were randomly divided into three groups (n = 8 per group). Group I served as contro

Sodium Caseinate Induces Apoptosis in Cytarabine-Resistant AML by Modulating SIRT1 and Chemoresistance Genes, Alone or in Combination with Cytarabine or Daunorubicin.

Resistance to cytarabine (Ara-C) remains a major obstacle to the successful treatment of acute myeloid leukemia (AML). Therefore, modulating Ara-C resistance is indispensable for improving clinical outcomes. We previously demonstrated that sodium caseinate (SC), a salt derived from casein, the principal milk protein, inhibits proliferation and modulates the expression of Ara-C resistance-related genes in chemoresistant cells. However, it remains unclear whether the combination of SC with antineoplastic agents enhances apoptosis, modulates chemoresistance-related genes, and prolongs the surviva

Mucoadhesive and sodium transport properties of some animal (Casein and Gelatin) and plant (Lotus Seed) proteins as a strategy for saltiness enhancement.

Proteins possess advantageous mucoadhesive and mucopenetrative properties when in contact with the oral mucosa, which might be used to modulate saltiness perception. The present study investigated the mucoadhesive properties of lotus seed protein (LP), compared to some animal proteins known for their mucoadhesion ability [casein (CA) and gelatin (G)]. To optimize and maximize the protein binding to mucin and the oral mucosa, we tested the binding at different protein concentrations (2 % - 10 % w/v) and pH levels (1.2 to 6.8). Since the human oral environment is about 37°C, all experiments were

Synergistic effects of ultrasonication and alkaline treatment on curcumin-induced cross-linking and gelation of casein-soy protein isolate hybrids.

The development of hybrid protein gels through the partial replacement of animal proteins with plant-based proteins represents a promising strategy for sustainable food production. This study investigated whether ultrasonication-generated free radicals, alone or combined with alkaline treatment, could induce curcumin to act as a cross-linker between casein and soy protein isolate. Gels were prepared under different pretreatments (alkali, ultrasound, or their combination) and characterized using rheology, spectroscopy, solubility assays, and microscopy. Results demonstrated that only ultrasound

Porphyrinic metal-organic framework PCN-224 supported Prussian blue enables selective detection of casein and phytic acid.

Monitoring casein and phytic acid content is of great significance for food safety and human health. In this paper, porphyrinic metal-organic framework PCN-224 supported Prussian blue (PCN-224@PB) nanoprobe was constructed by in-situ growth of PB on PCN-224 layer. The supported PCN-224 not only reduces the size and improves the dispersity of PB, but also provides specific affinity for casein and phytic acid. Based on the formation of Zr-OP structure, the peroxidase (POD)-like activity of PCN-224@PB is inhibited when encountering casein. In the presence of phytic acid, the deposited PB is decom

Effect of β-Casein Fortification in Milk Protein on Digestion Properties and Release of Bioactive Peptides in a Suckling Rat Pup Model.

β-casein (β-CN) is the predominant casein fraction in breast milk, while current infant milk formula (IMF) contains substantially lower β-CN levels than breast milk. The impact of β-CN fortification on neonatal digestive characteristic and bioactive peptide release remains an understudied area in vivo. This study investigated the effect of β-CN fortification in milk protein on digestion properties and release of bioactive peptides using a suckling rat pup model. Rat pups were, respectively, gavaged with two milk protein solutions: one with ordinary β-CN content (OBCN) and the other with fortif

VTA dopamine neurons are hyperexcitable in 3xTg-AD mice due to casein kinase 2-dependent SK channel dysfunction.

Alzheimer's disease (AD) patients exhibit neuropsychiatric symptoms that extend beyond classical cognitive deficits, suggesting involvement of subcortical areas. Here, we investigated the role of midbrain dopamine (DA) neurons in AD using the amyloid + tau-driven 3xTg-AD mouse model. We found deficits in reward-based operant learning in AD mice, suggesting possible VTA DA neuron dysregulation. Physiological assessment revealed hyperexcitability and disrupted firing in DA neurons caused by reduced activity of small-conductance calcium-activated potassium (SK) channels. RNA sequencing from conte

Effect of Casein Phosphopeptide-Amorphous Calcium Phosphate Added to Milk, Chewing Gum, and Candy on Dental Caries: A Systematic Review.

Casein is one of the most studied proteins with activity against dental caries. In particular, casein phosphopeptide-amorphous calcium phosphate (CPP-ACP) has shown promising remineralizing properties. In vivo evidence on the anticaries potential of CPP-ACP added to foodstuffs is elusive, nonetheless. Hence, this systematic review aimed at determining whether the use of CPP-ACP added to foodstuffs has a remineralizing or inhibitory action on dental demineralization either in vivo or in situ. The review protocol followed the PRISMA-P criteria and was registered in PROSPERO. PubMed, Scopus, and

Inhibition of casein kinase 2 induces cell death in tyrosine kinase inhibitor resistant chronic myelogenous leukemia cells.

Chronic myelogenous leukemia (CML) is a myeloproliferative disease characterized by the BCR-ABL oncogene. Despite the high performance of treatment with tyrosine kinase inhibitors (TKI), about 30% of patients develop resistance to the therapy. To improve the outcomes, identification of new targets of treatment is needed. Here, we explored the Casein Kinase 2 (CK2) as a potential target for CML therapy. Previously, we detected increased phosphorylation of HSP90β Serine 226 in patients non-responding to TKIs imatinib and dasatinib. This site is known to be phosphorylated by CK2, which was also l

In vitro digestion of milk proteins including intestinal brush border membrane peptidases. Transepithelial transport of resistant casein domains.

The use of enzymes from the brush border membrane (BBM) in simulated gastrointestinal digestion of milk proteins has been evaluated. With this purpose, the resistant sequences from casein and milk whey proteins after INFOGEST in vitro digestion with and without BBM have been analyzed by tandem mass spectrometry. The use of BBM revealed additional cleavages to those found with pancreatic enzymes, although the number of total identified peptides decreased due to the reduction of the peptide size. These new cleavages were mainly attributed to the activity of amino- and carboxy-peptidases, which w

Impact of high-pressure treatment on casein micelles, whey proteins, fat globules and enzymes activity in dairy products: a review.

The quality and safety of food products are the two factors that most influence the demands made by consumers. Contractual food sterilization and preservation methods often result in unfavorable changes in functional properties of foods. High-pressure processing (HPP) (50-1000 MPa) is a non-thermal preservation technique, which can effectively reduce the activity of spoilage and pathogenic microorganisms with minimal impact on the functional and nutritional properties of food. Comprehensive inquires have disclosed the potential profits of HPP as an alternative to heat treatments by affecting t

Structural and molecular bases of angiotensin-converting enzyme inhibition by bovine casein-derived peptides: an in silico molecular dynamics approach.

The angiotensin-converting enzyme (ACE) plays a key role in blood pressure regulation process, and its inhibition is one of the main drug targets for the treatment of hypertension. Though various peptides from milk proteins are well-known for their ACE-inhibitory capacity, research devoted to understand the molecular bases of such property remain scarce, specifically for such peptides. Therefore, in this work, computational molecular docking and molecular dynamics calculations were performed to enlighten the intermolecular interactions involved in ACE inhibition by six different casein-derived

Characterization of casein-derived peptide bioactivity: Differential effects on angiotensin-converting enzyme inhibition and cytokine and nitric oxide production.

Lactic acid bacteria (LAB) are used as starter cultures in the production of fermented dairy products and have the potential to confer bioactivity relevant to cardiovascular health, as they possess extensive proteolytic systems that liberate small bioactive peptides from larger milk proteins. Certain casein-derived peptides released by various LAB strains during fermentation have been shown to reduce hypertension and to modulate the immune system. We investigated the growth and peptide production of 2 LAB strains, Lactobacillus helveticus R0389 and Lactocaseibacillus rhamnosus R0011, their imm
