Regional metal concentrations in Parkinson's disease, other chronic neurological diseases, and control brains.

Metal deficiency or toxicity states have been recognized as a cause of several neurological disorders and are suspected in others. We analyzed four brain regions (frontal cortex, caudate nucleus, substantia nigra, and cerebellum) in 36 human brains for concentrations of 24 metals (Ag, Al, As, B, Be, Ca, Cd, Co, Cr, Cu, Fe, K, Pb, Mg, Mn, Mo, Na, Ni, P, Se, Ti, V, W, Zn). Regional metal concentrations, measured using atomic absorption and atomic emission spectroscopy, were compared between 9 Parkinson's disease (PD) brains, 15 brains from patients with other chronic neurological diseases, and 1…

Connectome-based predictive modelling predicts frailty levels in older adults.

Frailty is characterized by a persistent and progressive decline in physiological reserves, leading to increased vulnerability to stressors … ividual differences in grip strength, with the Subcortical and Cerebellum network, particularly the caudate nucleus, functional connectivity being the strongest predictor. These findings demonstrate that task-based functional connectomes can serve as personalized markers that can predict individual behavioral measures, including handgrip strength, and point to in …

Rhesus monkeys exhibiting spontaneous ritualistic behaviors resembling obsessive-compulsive disorder.

Obsessive-compulsive disorder (OCD) is a chronic and debilitating psychiatric disorder that affects ∼2%-3% of the population globally. Study … humans. Brain imaging showed that monkeys with SMBs had larger gray matter (GM) volumes in the left caudate nucleus and lower fractional anisotropy of the corpus callosum. The GM volume of the left caudate nucleus correlated positively with the daily duration of SMBs. Notably, exposure to a stressor (human presence) significantly increased SMBs. In addition, fluo …

The pathological anatomy of posthemiplegic athetosis.

Disorders of movement after hemiplegia have been described for more than a century, but their pathological anatomy and physiology have remained poorly understood because of ambiguous terminology and incomplete studies. We examined the brains of 5 patients which had been serially sectioned where there had been well documented pure motor hemiplegia acquired in childhood. In 4 patients handicapped by hemiathetosis the main lesion was partial destruction of the caudate nucleus and putamen. In the fifth case, where non-disabling involuntary movements only appeared in later life, there was gliosis o…

Putamen FDOPA uptake and its relationship tot cognitive functioning in PD.

Impaired executive functioning is frequently found in PD patients. As a result, impairments in the executive components of other cognitive functions such as memory, visuo-spatial functions and fluency are also present. From a neuroanatomical point of view, different loops are mediating cognitive, emotional and motor functions. The putamen is part of the motor loop, while the caudate nucleus is part of the motor loop but also of the cognitive loop. In PD, multiple neurotransmitter systems are involved in cognitive functioning, including the dopaminergic system. Cognitive functioning in PD is re…

Sulfonylurea binding sites in normal human brain and in Parkinson's disease, progressive supranuclear palsy and Huntington's disease.

In human brain, [3H]glibenclamide binds with high affinity (KD about 3.5 nM) to sulfonylurea binding sites which are associated with ATP-sensitive potassium (KATP) channels. Regarding to the important neuromodulatory action of KATP channels in some neuronal populations, sulfonylurea binding sites were measured in several cortical areas (frontal and temporal cortex, hippocampus) and striatum (caudate nucleus and putamen) in controls and patients with Parkinson's disease or progressive supranuclear palsy. There was no modification of [3H]glibenclamide specific binding in the cerebral regions stu…

Calcineurin in human brain and its relation to extrapyramidal system. Immunohistochemical study on postmortem human brains.

Calcineurin immunoreactivity has been successfully detected in formalin-fixed paraffin-embedded postmortem human brain tissue using the peroxidase-antiperoxidase method. We have examined two autopsy cases with Huntington's disease (HD), three cases with Parkinson's disease, and two senile patients as controls. In the controls, calcineurin immunoreactivity was present in neuronal cells only and highly concentrated in the caudate nucleus, putamen, globus pallidus (striato-pallidal pathway), substantia nigra (striato-nigral pathway) and hippocampal formation. These localizations were similar to t…

In autism, abnormalities of the basal ganglia, in particular the caudate nucleus, are the best replicated findings in neuroimaging studies. Interestingly, the DRD3 gene is highly expressed in the basal ganglia, most notably the caudate nucleus. The rs167771 SNP was recently also found to be related to risperidone-induced extra-p …

Alterations in dopaminergic function in Rett syndrome.

Rett syndrome is a neurological disorder associated with cortical atrophy, stereotyped hand movements, dementia, and extrapyramidal dysfunction. Endogenous levels of dopamine and its metabolites are decreased throughout the neocortex and basal ganglia and the number of dopamine type 2 receptors are decreased in the putamen. The present study investigated changes in dopamine uptake sites and dopamine type-1 receptors in the brains of eleven Rett syndrome patients (4-30 yrs) and ten normal female controls (2.5-20 yrs). The number of dopamine type 1 receptors within the caudate nucleus were uncha…

Do forebrain structures compete for behavioral expression? Evidence from amphetamine-induced behavior, microdialysis, and caudate-accumbens lesions in medial frontal cortex damaged rats.

The neurochemical basis of behavioral changes following medial frontal cortex damage were investigated. Experiment 1 examined locomotion in response to D-amphetamine (1.5 and 5 mg/kg) in rats that had received bilateral aspirative lesions of the medial frontal cortex alone or in combination with 6-hydroxydopamine (6-OHDA) lesions of the nucleus accumbens or caudate-putamen. Relative to controls, medial frontal cortex rats were initially hypoactive (day 1 postoperative) but rapidly became hyperactive (days 5-15 postoperative). Locomotor-time profiles and stereotypy ratings showed that amphetami

Functional brain networks and neuroanatomy underpinning nausea severity can predict nausea susceptibility using machine learning.

KEY POINTS: Nausea is an adverse experience characterised by alterations in autonomic and cerebral function. Susceptibility to nausea is difficult to predict, but machine learning has yet to be applied to this field of study. The severity of nausea that individuals experience is related to the underlying morphology (shape) of the subcortex, namely of the amygdala, caudate and putamen; a functional brain network related to nausea severity was identified, which included the thalamus, cingulate cortices (anterior, mid- and posterior), caudate nucleus and nucleus accumbens. Sympathetic nervous sys…

Association of Iron and Myelin Alterations in the Contralesional Dentate Nucleus and Thalamus With Functional Outcome in Acute Ischemic Stroke: A Susceptibility Source Separation Study.

BACKGROUND: Susceptibility alterations in deep gray matter (DGM) nuclei after acute ischemic stroke (AIS) may relate to impaired functional independence. However, conventional quantitative susceptibility mapping (QSM) cannot separate paramagnetic iron-related from diamagnetic myelin-related sources, potentially obscuring relevant pathophysiological alterations. PURPOSE: To apply χ-separation to disentangle paramagnetic susceptibility (χpara) and diamagnetic susceptibility (χdia) in DGM nuclei after AIS and assess associations with 3-month functional independence. STUDY TYPE: Prospective. PO

Differential reward coding in the subdivisions of the primate caudate during an oculomotor task.

The basal ganglia play a pivotal role in reward-oriented behavior. The striatum, an input channel of the basal ganglia, is composed of subdivisions that are topographically connected with different cortical and subcortical areas. To test whether reward information is differentially processed in the different parts of the striatum, we compared reward-related neuronal activity along the dorsolateral-ventromedial axis in the caudate nucleus of monkeys performing an asymmetrically rewarded oculomotor task. In a given block, a target in one position was associated with a large reward, whereas the o…
