With the Global Elimination of Trachoma by 2020 program underway, it has become increasingly important to identify the prevalence of ocular chlamydia infection in communities. DNA amplification tests are the gold standard, but are prohibitively expensive. In the present paper, we investigate whether pooling multiple specimens into a single test is feasible. The conjunctivae of 170 children in western Nepal were examined and swabbed. The prevalence of chlamydial infection was estimated in two ways using the ligase chain reaction: by testing all 170 specimens individually, and by testing 34 pool

A comparative study between Chlamydia Testpack and Chlamydiazyme for the detection of chlamydial antigen was performed. In 61 cases suspected of being chlamydial infection, Chlamydia Testpack had a specificity of 92% and a sensitivity of 88% when compared with Chlamydiazyme. On the result of study using Chlamydia trachomatis serotype B, the sensitivity of Chlamydia Testpack which had a lower limit of 6 x 10(4)/ml of EB was less than that of Chlamydiazyme which had a lower limit of 0.7 x 10(4). However, the large number of EB presented in almost all positive specimen, indicated that Chlamydia T…

This chapter describes a lambda Red-based recombineering approach for targeted genetic manipulation in Chlamydia species. The method uses a non-replicative plasmid encoding lambda Red recombination proteins together with locus-specific homology arms to generate precise gene deletions in the Chlamydia chromosome. Detailed protocols are provided for construct design, host cell-based selection, and genetic validation of recombinant strains, and noteworthy considerations for transforming different Chlamydia species and strains are given.

INTRODUCTION: Prior to 2021, the US Centers for Disease Control and Prevention (CDC) recommended either doxycycline or azithromycin as first-line treatments for Chlamydia trachomatis (CT). By 2016, there was evidence showing that doxycycline was more effective than azithromycin for the treatment of rectal CT. This motivated the Public Health-Seattle & King County Sexual Health Clinic to update their clinic's protocol to treat rectal CT with exclusively doxycycline in mid-2016. The CDC later recommended this in July 2021.
METHODS: Leveraging health records from the clinic, we performed a contr…

Due to previously reported associations of ovarian cancer with pelvic inflammatory disease, tubal infertility, and positive Chlamydia trachomatis titers, women undergoing follow-up for ovarian cancer were screened with a C. trachomatis IgG titer. Positive titers were seen in 15 (79%) of 19 women with ovarian cancer, 9 (90%) of 10 age-matched controls, and 14 (67%) of 21 patients with infertility and pain. When analyzed by age, 4 (40%) of 10 women under 30 years of age and 34 (85%) of 40, 30 years of age or older had positive titers (p = 0.007). Of 21 women with positive Chlamydia pneumoniae ti…

We have investigated the ability of both species of chlamydiae (C. trachomatis and C. psittaci), two major biovars of C. trachomatis (lymphogranuloma venereum and trachoma), and the two developmental forms of chlamydia (reticulate and elementary bodies) to stimulate murine spleen lymphocytes. All of these forms of the bacteria induce potent proliferation and differentiation to plaque-forming cells by B lymphocytes in vitro. Chlamydiae induce a broad antibody response, suggesting that stimulation is polyclonal in nature. Although all chlamydiae possess a lipopolysaccharide (LPS) genus-specific

Chlamydia trachomatis (Ct) is a sexually transmitted infection (STI) linked to ectopic pregnancy, infertility, invasive lymphogranuloma venereum (LGV), and the blinding disease trachoma. STI, LGV, and trachoma are associated with disease-specific Ct genovars. Serology is an important surveillance tool for estimating exposure, but the impact of genome variation on the antigenicity of Ct proteins is unclear. Two-dimensional antigenicity profiling of Pgp3 antigens representing LGV, STI, and trachoma demonstrated differential antigenicity using antigen-specific murine sera, but geometric distances…

Male infertility accounts for 20-30% cases of infertility, and may be due to hormonal, genetic, toxic or infectious factors. Male urogenital infections caused by Chlamydia trachomatis (CT) are potential causes of male infertility. This study determined the prevalence of Chlamydia trachomatis infection among infertile men in Cotonou. The descriptive cross-sectional study used Real time PCR (RT PCR) to detect Chlamydia trachomatis infection on the first urine stream from 90 participants who did sperm analysis for infertility problems between March 2023 and December 2023. The mean age of the pati…

Background: Chlamydia trachomatis (CT) infections are among the most prevalent sexually transmitted infections (STIs), particularly affecting young adults. In women, CT can cause pelvic inflammatory disease, infertility, and ectopic pregnancy, justifying routine vaginal swab screening. However, due to diverse sexual practices, CT can colonize multiple anatomical sites, including the pharynx. Pharyngeal CT carriage in women is underdiagnosed and may contribute to ongoing transmission. Objectives and Methods: This study aimed to assess the rate of pharyngeal CT carriage, describe the rate of iso…

The cervicovaginal microbiome (CVM) is increasingly being considered as an important aspect of women's health, particularly in relation to t … with better or neutral reproductive health, including reduced coincident detection of STIs such as Chlamydia trachomatis. In contrast, CST IV is composed of diverse, predominantly anaerobic, microbial species and is associated with CVM dysbiosis, bacterial vaginosis, and a heightened risk of STI acquisition. This review examines the complex interplay between the CVM, …

Chlamydia trachomatis, is a kind of obligate intracellular pathogen. The removal of C. trachomatis relies primarily on specific cellular immunity. It is currently considered that CD4+ Th1 cytokine responses are the major protective immunity against C. trachomatis infection and reinfection rather than CD8+ T cells. The non-specific immunity (innate immunity) also plays an important role in the infection process. To survive inside the cells, the first process that C. trachomatis faces is the innate immune response. As the "sentry" of the body, mast cells attempt to engulf and remove C. trachomat…

In recent years, substantial progress has been made in the understanding of the intracellular lifestyle of Chlamydia trachomatis and how the bacteria establish themselves in the human host. As an obligate intracellular pathogenic bacterium with a strongly reduced coding capacity, C. trachomatis depends on the provision of nutrients from the host cell. In this Review, we summarize the current understanding of how C. trachomatis establishes its intracellular replication niche, how its metabolism functions in the host cell, how it can defend itself against the cell autonomous and innate immune re…

BACKGROUND: In Cuba, little is known regarding the prevalence of Chlamydia trachomatis (CT) infection in adolescents and young people. We study the frequency of CT infection in these populations, and its association with clinical-epidemiological variables.
METHODS: A total of 496 individuals aged 12 to 24 were recruited from November 2018 to November 2019. Of them, 302 were patients attending at sexually transmitted infections (STI) services and 194 were young volunteers. CT detections were carried out by real-time PCR and IgG serology.
RESULTS: The prevalence of CT using PCR was 9.1% (45/49…

Currently, Chlamydia trachomatis still possesses a significant impact on public health, with more than 130 million new cases each year, alongside a high prevalence of asymptomatic infections (approximately 80% in women and 50% in men). C. trachomatis infection involves a wide range of different cell types, from cervical epithelial cells, testicular Sertoli cells to Synovial cells, leading to a broad spectrum of pathologies of varying severity both in women and in men. Several two-dimensional in vitro cellular models have been employed for investigating C. trachomatis host-cell interaction, alt…

Sexually transmitted infections (STIs), such as Chlamydia trachomatis (Ct) infection, have serious consequences for sexual and reproductive health worldwide. Ct is one of the most common sexually transmitted bacterial infections in the world, with approximately 129 million new cases per year. C. trachomatis is an obligate intracellular Gram-negative bacterium. The infection is usually asymptomatic, notwithstanding, it could also be associated with severe sequels and complications, such as chronic pain, infertility, and gynecologic cancers, and thus there is an urgent need to adequately treat t…

Sexual contact is the most common route of HIV transmission, and the concurrent presence of sexually transmitted infections (STIs) such as Chlamydia trachomatis (CT) and Neisseria gonorrhoeae (gonococcus, GC) is known to increase the HIV risk. Antibiotic treatment decreases the incidence of STIs but not HIV. CT and GC activate Toll-like receptors (TLRs) 2 and 4, which act as sensors of microbial infection are critical for initiating immune responses to control infection. We have previously shown that GC enhances HIV infection of primary resting CD4+ T cells through activation of TLR2 but not T…
