Muscarinic cholinergic receptors in area postrema and brainstem areas regulating emesis.

Central cholinergic pathways modulate both the perception of excessive motion stimuli and the expression of motion sickness symptoms, such as nausea and vomiting. Specific brainstem areas which mediate motion-induced emesis include the area postrema (AP), vagal nuclear complex (VNC), reticular formation (RF) at the site of the vomiting center, and the vestibular complex (VC). In this report, histological studies indicated the cellular organization of brainstem structures mediating emesis was similar in bovine and squirrel monkey brain. The objective of this study was to characterize biochemica

Activation of acetylcholine receptors and microglia in hypoxic-ischemic brain damage in newborn rats.

OBJECTIVE: We previously showed that acetylcholine receptor (AChR) agonist reduced hypoxic-ischemic brain damage in the newborn rats. To further investigated the interaction between hypoxia and chorinergic anti-inflammatory pathway, we examined the effect of AChR antagonist on brain damage and to see the relation between microglial activation and protective effect of AChR agonist. STUDY DESIGN: Seven-day-old Wistar rats were divided into 2 groups, one receiving AChR antagonists to see if they have deleterious effects on hypoxic-ischemic brain damage, and the other receiving AChR agonist, carb

Case of cholinergic urticaria accompanied by anaphylaxis.

Cholinergic urticaria occasionally occurs in combination with anaphylactic symptoms. However, this has not been widely reported. Herein, we report the case of a 14-year-old Japanese male who was diagnosed with cholinergic urticaria accompanied by anaphylaxis. The patient, who was suffering from atopic dermatitis and bronchial asthma, had developed wheals after exercising or bathing, which would have increased his core body temperature, since summer 2014. He experienced two episodes of severe systemic symptoms and wheal development when he took a bath after eating in December 2014 and the follo

Muscarinic effects and foetal cardiovascular and hormonal responses in utero.

INTRODUCTION: Cholinergic mechanisms play an important role in the control of hormonal and vascular regulation. However, in utero development of cholinergic regulation in the foetal hormonal systems is not clearly understood. This study investigated foetal hormonal and cardiovascular responses following application of the muscarinic antagonist atropine. MATERIALS AND METHODS: Chronically prepared near-term ovine foetuses (control and experimental: n=5, each group) were used. After 4-5 days' surgical recovery, conscious ewes and their foetuses were tested in vivo. RESULTS: In response to intra

Interactions between scopolamine and muscarinic cholinergic agonists or cholinesterase inhibitors on spatial alternation performance in rats.

The effects on working memory of the muscarinic cholinergic agonists oxotremorine, arecoline, RS86 and pilocarpine, and the cholinesterase inhibitors physostigmine and tetrahydroaminoacadine were investigated in male F344 rats. Working memory was assessed by behavior maintained under a spatial alternation schedule of food presentation in which the interval between trials was varied from 2 to 32 sec. Under control conditions the percentage of correct responses decreased as the retention interval was varied from 2 to 32 sec. Administered alone the cholinergic agonists oxotremorine (0.01-0.1 mg/k

Central muscarinic receptor subtypes and carrageenin-induced paw oedema in rats.

In an earlier report from this laboratory, it was demonstrated that the central cholinergic system exerted a modulatory pro-inflammatory effect on carrageenin-induced paw inflammation in rats. In this study, the role of the central muscarinic receptors in cholinergic modulation of peripheral inflammation was investigated by using several M1 and M2 receptor agonists and antagonists. The M1 receptor agonists aceclidine and arecholine and the nonspecific muscarinic receptor agonist oxotremorine augmented carrageenin oedema, an effect attenuated by the M1 receptor antagonist scopolamine. Physostig

Characterization and localization of the muscarinic cholinergic receptor in human prostatic tissue.

Radioligand receptor binding and autoradiography were used to characterize and localize the muscarinic cholinergic receptor in human benign prostatic hyperplastic tissue. These methods have not been used previously to investigate the autonomic innervation of the human prostate. The binding of [3H]N-methylscopolamine ([3H]NMS), a muscarinic cholinergic antagonist, to homogenates of human prostate was saturable and of high affinity. The equilibrium dissociation constant, (Kd), for [3H]NMS binding to human prostate homogenates was 0.10 +/- 0.03 nM (mean +/- SEM). The values of the Kd's for [3H]NM

Regulation of enteric nervous system in the proximal and distal parts of the normal human pyloric sphincter--in vitro study.

BACKGROUND/AIMS: The structure of the pyloric sphincter (PS) muscle has recently been shown to divide into two parts (proximal and distal parts). To clarify the functional differences in the human PS between proximal and distal parts, we investigated the enteric nerve responses in normal proximal and distal PS specimens. METHODOLOGY: Normal PS specimens derived from 20 patients with early gastric cancer (13 men and 7 women aged from 50 to 64 years, average 58.2 years) were used. These PS muscles were divided into 2 parts [1/2 oral site of PS; proximal part (PPS; n=26), 1/2 anal site of PS; di

Oxybutynin-associated Cognitive Impairment: Evidence and Implications for Overactive Bladder Treatment.

Anticholinergic medications have long been a mainstay of overactive bladder (OAB) treatment. Oxybutynin, a first-generation anticholinergic, still accounts for more than half of all OAB medication prescriptions, despite associations with impaired memory and cognition, as well as mounting evidence that it may increase the risk of incident dementia. This review details the current literature regarding oxybutynin and cognition, including evidence from preclinical, clinical, and real-world studies that show that oxybutynin binds nonspecifically to muscarinic receptors in the brain and is associate

Ureteral Stent Discomfort and Its Management.

PURPOSE OF REVIEW: Ureteral stent placement is a very common urologic procedure and a necessary component of many minimally invasive surgeries including ureteroscopy. Despite its widespread use, stent placement results in bothersome urinary symptoms and pain in approximately 80% of patients. This review discusses the proposed mechanisms underlying ureteral stent-related symptoms and current approaches to symptom management and the evidence to support them. RECENT FINDINGS: Though it has been extensively studied, the exact cause of stent-related symptoms remains unknown but is likely related t

Chemogenetic activation of cholinergic intrinsic cardiac ganglia improves border zone oxygenation and reduces arrhythmias during acute local ischemia.

BACKGROUND: Current therapeutic strategies for acute myocardial infarction rely on reperfusion and pharmacological management, which are typically administered hours after an event. Activation of the cardiac cholinergic efferents superior to the heart soon after a coronary occlusion has shown promise as a potential therapy to reduce arrhythmias and improve ventricular function. We tested whether selective activation of cholinergic neurons within the intrinsic cardiac ganglia (ICG) would also reduce arrhythmias and improve oxygenation of ischemic border zone tissue after an acute coronary occlu

[Current prospects for improving the quality of life of patients with internal stents].

Internal drainage of the upper urinary tract by stenting is a common urological procedure. The use of stenting in modern urological practice is on the rise due to widespread adoption of modern endourological, laparoscopic, percutaneous and high-tech robotic interventions. The presence of a stent in the patients body may cause stent-related symptoms and negatively affect the quality of life, resulting in additional outpatient visits and hospitalizations. The review provides an analysis of the causes, diagnostic evaluation, prevention and treatment of stent-related symptoms. The authors describe

Pharmacological agents for the treatment of urinary incontinence due to overactive bladder.

Although the exact aetiology of overactive bladder is unknown to date, pharmacological therapy has been targeted to both the central and peripheral nervous systems. Potential CNS targets include GABA, opioid, serotonin (5-HT), dopamine and glutaminergic receptors as well as the alpha-adrenoceptors. Potential PNS targets include muscarinic receptors, calcium and potassium channels and alpha- and beta-adrenergic receptors. Since acetylcholine is the primary excitatory neurotransmitter involved in bladder (detrusor) contraction and emptying, anticholinergic agents are the primary compounds used c

Management of overactive bladder--looking to the future.

OBJECTIVES: To offer a perspective on potential future advances in the understanding and management of the overactive bladder. METHODS: Consideration is given to potential developments in the areas of pathophysiology, prevention, early intervention, and new pharmacologic targets for the treatment of overactive bladder. RESULTS: The future will likely reveal that motor urgency is usually a premature micturition reflex with a limited and variable degree of penetrance. Overactivity of all types in the lower urinary tract (LUT) will be regarded as the result of a partially uncontrolled on-off me

Natural Polyphenols in the Treatment of Alzheimer's Disease.

Neurodegenerative disease is an incurable disease which involves the degeneration or death of the nerve cells. Alzheimer's Disease (AD) is a neurodegenerative disease discovered in 1906 by Alois Alzheimer, a German clinical psychiatrist and neuroanatomist. The main pathological hallmarks of this disease are the formation of extracellular amyloid β (Aβ) plaques and intracellular neurofibrillary tangle (NFT). The accumulation of the amyloid protein aggregates in the brain of AD patients leads to oxidative stress and inflammation. Other postulated reasons for the development of this disease are c

Altered neurogenic and mechanical responses to acetylcholine, ATP and substance P in detrusor from rat with outlet obstruction.

The well-known side effects of anticholinergic compounds used to treat urinary incontinence caused by detrusor overactivity have addressed the interest on other pharmacological intervention. The purpose of the present work was to investigate the possible changes in purinergic and cholinergic components of parasympathetic neurotransmission in obstructed rat bladders with detrusor overactivity, and to examine the effect of the association of suramin, atropine and indomethacin on nerve-mediated responses to electrical field stimulation (EFS). Mechanical responses to exogenous acetylcholine, ATP a

Stimulation by interleukin-1 (IL-1) of the release of rat corticotropin-releasing factor (CRF), which is independent of the cholinergic mechanism, from superfused rat hypothalamo-neurohypophysial complexes.

The effects of interleukin-1 (IL-1) and interferon-gamma (Ifn-gamma) on the release of corticotropin-releasing factor (CRF) from superfused hypothalamo-neurohypophysial complexes (HNC) of rats were examined in the present study. In this in vitro system, the release of CRF from HNC was not affected by any dose of human recombinant Ifn-gamma tested (0.1, 1 and 10 nM). In contrast, a rapid increase of CRF from HNC was elicited in a dose-dependent manner by human recombinant IL-1 alpha and -1 beta in concentrations of 0.1-10 nM. The involvement of the cholinergic system in the mediation of the sti
