Molecular weapons against agricultural vulnerability and the war on terror.

The multiple reports in this issue of the Journal from the Agenda for Action conference, coupled with the analysis by the National Academy of Sciences, the National Research Council, and the Auditor General (UK) on bioterror preparedness and homeland security, highlight the immediate need for rapid disease detection and advanced diagnostic capabilities to protect the public health, animal agriculture, and the numerous associated economies in the United States. In response to the potentially devastating consequences that could arise, there is an acute need for rapid detection of a variety of th

Animal health risks associated with the transportation and utilisation of wildlife products.

The animal health risks associated with the movement of wildlife products are infinitely less than those associated with the movement of live animals. Very few pathogens are sufficiently robust to survive the significant changes in temperature, pH, moisture content and osmolality which occur post mortem, or which are associated with preservation processes such as pickling, smoking or drying. Certain pathogens, however, (e.g. foot and mouth disease, classical swine fever [hog cholera] and African swine fever viruses and the anthrax bacillus) are hardy and resistant to these environmental change

Foodborne Zoonoses Common in Hunted Wild Boars.

The northern European wild boar population has increased during the last decade. Highest wild boar numbers in Finland have been reported in the southeastern part near the Russian border. Wild boars may be infected with several human and animal pathogens. In this study, we investigated the presence of important foodborne pathogens in wild boars hunted in 2016 in Finland using serology, PCR and culturing. Seroprevalence of Salmonella (38%) and Yersinia (56%) infections was high in wild boars. Antibodies to hepatitis E virus, Toxoplasma gondii and Brucella were found in 18%, 9% and 9% of the wild

Modulation of T cell and monocyte function in the spleen following infection of pigs with African swine fever virus.

Infection of pigs with many strains of African Swine Fever Virus (ASFV) has been shown to cause a loss or marked decrease in the ability of splenocytes to respond to mitogens. These observations have been extended by cell fractionation and reconstitution experiments to show that the mitogen stimulated proliferative capacity of both the CD4+ and CD8+ T cells is affected. Similarly, monocytes which are directly infectable by virus, are functionally defective as antigen presenting cells when added to mitogen stimulated normal T cells. Interestingly, the same T cells which respond poorly in mitoge

Annexin A2 orchestrates CSFV entry route selection via the annexin A2/S100A10 heterotetrameric complex.

Classical swine fever virus (CSFV) is a highly contagious pathogen that causes substantial economic losses in swine production, yet the mechanisms governing its cellular entry remain poorly defined. Here, through a combination of pharmacological inhibition, genetic knockout/overexpression, and co-immunoprecipitation, we identify annexin A2 (ANXA2) as a pivotal host determinant of CSFV entry into PK-15 cells. Confocal microscopy revealed that CSFV infection rapidly induced the translocation of ANXA2 to the cell surface, and functional assays confirmed that its plasma membrane localization was e…

Genetic variation of Border disease virus species strains.

The 5'-untranslated region of Pestivirus strains isolated from domestic and wild animals were analysed to determine their taxonomic status according to nucleotide changes in the secondary genomic structure using the palindromic nucleotide substitutions (PNS) method. A total of 131 isolates out of 536 Pestivirus strains evaluated, were clustered as Border disease virus (BDV) species. The BDV strains were further divided into at least 8 genotypes or subspecies. Thirty-two isolates from small ruminants suffering from clinical symptoms of Border disease were clustered into bovine viral diarrhoea v

Activation and modulation of antiviral and apoptotic genes in pigs infected with classical swine fever viruses of high, moderate or low virulence.

The immune response to CSFV and the strategies of this virus to evade and suppress the pigs' immune system are still poorly understood. Therefore, we investigated the transcriptional response in the tonsils, median retropharyngeal lymph node (MRLN), and spleen of pigs infected with CSFV strains of similar origin with high, moderate, and low virulence. Using a porcine spleen/intestinal cDNA microarray, expression levels in RNA pools prepared from infected tissue at 3 dpi (three pigs per virus strain) were compared to levels in pools prepared from uninfected homologue tissues (nine pigs). Promotion of immunity of mice to Pasteurella multocida and hog cholera vaccine by pig interleukin-6 gene and CpG motifs.

A novel oligodeoxynuleotides containing 11 CpG motifs was synthesized and inserted into the VR1020 plasmid containing pig interleukin-6 (IL-6) gene (VPIL6) to construct recombinant plasmid, VPIL6C. The chitosan nanoparticles (CNP) were prepared by ionic cross linkage to entrap the VPIL6C (VPIL6C-CNP), VPIL6 (VPIL6-CNP) and CpG (CpG-CNP). 42-Day old female mice were divided into four groups and intramuscularly injected respectively with 6 pmol VPIL6C-CNP, VPIL6-CNP, CpG-CNP and VR1020-CNP along with the bivalent vaccines against the Pasteurellosis and hog cholera. A real-time PCR to detect and analyze virulent EMCV loads in sows and piglets.

A real-time polymerase chain reaction with SYBR Green was developed for the detection and quantification of encephalomyocarditis virus (EMCV) in porcine tissues; the method uses two primers specific for the 3D gene. The detection limit of this assay was 22 gene copies/reaction, equivalent to 0.001 TCID(50)/ml. The assay was linear over a 10(7) dilution range of template concentrations and was specific for EMCV; it did not amplify other porcine pathogens (porcine circovirus 2, porcine reproductive and respiratory virus, classical swine fever virus, pseudorabies virus, or porcine teschovirus). Wild boars as sources for infectious diseases in livestock and humans.

Wild boars (Sus scrofa) are indigenous in many countries in the world. These free-living swine are known reservoirs for a number of viruses, bacteria and parasites that are transmissible to domestic animals and humans. Changes of human habitation to suburban areas, increased use of lands for agricultural purposes, increased hunting activities and consumption of wild boar meat have increased the chances of exposure of wild boars to domestic animals and humans. Wild boars can act as reservoirs for many important infectious diseases in domestic animals, such as classical swine fever, brucellosis

Classical swine fever (hog cholera): review of aspects relevant to control.

Classical swine fever (CSF) has the ability to spread over large distances when human intervention such as illegal swill feeding facilitates its movement. This was apparent during 2005 when CSF appeared in South Africa (SA) after an absence of 87 years. In this review, various newly published developments in terms of the diagnosis of the disease and vaccination are described and applied to situations similar to SA. The role of wildlife such as feral pigs and European wild boar in the dissemination and maintenance of CSF virus are discussed, and the dearth of knowledge on the potential of other

Seroprevalence and associated risk factors of important pig viral diseases in Bhutan.

A cross-sectional serological study was conducted in Bhutan between October 2011 and February 2012 to determine the prevalence of antibodies to classical swine fever virus (CSFV), porcine reproductive and respiratory syndrome virus (PRRSV), porcine circovirus type 2 (PCV2), swine influenza virus (SIV) subtype H1N1 and Aujeszky's disease virus (ADV). Furthermore, risk factors for the seropositive status were investigated. Antibodies to SIV, subtype H1N1 (likely pandemic H1N1 2009) were detected in 49% of the pigs in the government farms, and 8% of the village backyard pigs. For PCV2, these perc…

Prevalence of Antibodies to Japanese Encephalitis Virus and Severe Fever with Thrombocytopenia Syndrome Virus in Wild Boars Captured Across Different Locations in Toyama Prefecture, Japan.

Wild boars inhabit fields, hills, and farms across Japan, where they are fed on by numerous arthropods, including mosquitoes and ticks. Consequently, they are frequently exposed to arthropod-borne pathogens. In Toyama Prefecture, blood samples from captured wild boars have long been collected for classical swine fever virus antibody testing, with detailed records kept on the capture locations. In this study, we investigated the prevalence of antibodies against Japanese encephalitis virus (JEV) and severe fever with thrombocytopenia syndrome virus (SFTSV) using 3059 serum samples collected from…
