Chemical modification of polymer brushes via nitroxide photoclick trapping.

The preparation of polymer brushes (PBs) bearing α-hydroxyalkylphenylketone (2-hydroxy-2-methyl-1-phenylpropan-1-one) moieties as photoreactive polymer backbone substituents is presented. Photoreactive polymer brushes with defined thicknesses (up to 60 nm) and high grafting densities are readily prepared by surface initiated nitroxide mediated radical polymerization (SINMP). The photoactive moieties can be transformed via Norrish-type I photoreaction to surface-bound acyl radicals. Photolysis in the presence of a persistent nitroxide leads to chemically modified PBs bearing acylalkoxyamine moi

Synthesis and characterization of a new red-emitting Ca2+ indicator, calcium ruby.

Calcium Ruby m-Cl (X = H, Y = Cl) is a visible-light excited red-emitting calcium concentration ([Ca2+]) indicator dye (579/598 nm peak excitation/emission) with a side arm for conjugation via EDC or click chemistry. Its large molar extinction and high quantum yield rank it among the brightest long-wavelength Ca2+ indicators. Calcium Ruby is a promising alternative to existing dyes for imaging [Ca2+] in multicolor fluorescence applications or in the presence of yellow-green cellular autofluorescence.

Modular in vivo antibody-ADC click to reverse drug resistance in tumours.

Antibody-drug conjugates (ADCs) have significantly advanced cancer therapy by enabling the selective delivery of cytotoxic agents to tumour cells. However, ADC efficacy remains constrained by its dependence on a single target antigen, which limits tumour targeting and promotes resistance in heterogeneous tumours with variable and low antigen expression1-6. Here we introduce an in vivo bioorthogonal ligation strategy that generates functional antibody-ADC click constructs following systemic administration. This platform provides a modular and translatable approach for enhanced targeted drug del

Chemoenzymatic method for site-selective fluorophore conjugation of a native IgG Fab fragment.

Antibody-derived bioconjugates have emerged as practical biomolecules for applications in diagnostics and therapeutics. A key challenge is achieving site-selective bioconjugation to preserve the native function of the labeled biomolecule. Methods that target native amino acid residues can broaden the applicability of bioconjugates; however, most existing methods rely on genetically engineered antibodies to ensure site-specific labeling. In this report, a chemoenzymatic strategy for modifying a native Fab fragment derived from trastuzumab was investigated using the EzMTG-pG fusion protein, whic

Accelerated Blood Clearance of Antibodies by Nanosized Click Antidotes.

Long blood half-life is one of the advantages of antibodies over small molecule drugs. At the same time, prolonged half-life is a problem for imaging applications or in the case of antibody-induced toxicities. There is a substantial need for antidotes that can quickly clear antibodies from systemic circulation and peripheral tissues. Engineered nanoparticles exhibit intrinsic affinity for clearance organs (mainly liver and spleen). trans-Cyclooctene (TCO) and methyltetrazine (MTZ) are versatile copper-free click chemistry components that are extensively being used for in vivo bioorthogonal cou

Artificial Nucleosides as Diagnostic Probes to Measure Translesion DNA Synthesis.

The misreplication of damaged DNA, a biological process termed translesion DNA synthesis (TLS), produces a large number of adverse effects on human health. This chapter describes the application of an artificial nucleoside/nucleotide system that functions as a biochemical probe to quantify TLS activity under in vitro and in vivo conditions. For in vitro studies, the artificial nucleotide, 3-ethynyl-5-nitroindolyl-2'-deoxyriboside triphosphate (3-Eth-5-NITP), is used as it is efficiently inserted opposite an abasic site, a highly pro-mutagenic DNA lesion produced by several types of DNA-damagin

Preparation of Metalloporphyrin-Bound Superparamagnetic Silica Particles via "Click" Reaction.

A facile approach using click chemistry is demonstrated for immobilization of metalloporphyrins onto the surface of silica-coated iron oxide particles. Oleic-acid stabilized iron oxide nanocrystals were prepared by thermal decomposition of iron(III) acetylacetonate. Their crystallinity, morphology, and superparamagnetism were determined using X-ray diffraction, transmission electron microscopy, and a superconducting quantum interference device. Monodisperse core-shell particles were produced in the silica-coating of iron oxide via microemulsion synthesis. Surface modification of these particle

Modular Synthesis of PEG-Dendritic Block Copolymers by Thermal Azide-Alkyne Cycloaddition with Internal Alkynes and Evaluation of their Self-Assembly for Drug Delivery Applications.

Linear-dendritic block copolymers assemble in solution due to differences in the solubility or charge properties of the blocks. The monodispersity and multivalency of the dendritic block provide unparalleled control for the design of drug delivery systems when incorporating poly(ethylene glycol) (PEG) as a linear block. An accelerated synthesis of PEG-dendritic block copolymers based on the click and green chemistry pillars is described. The tandem composed of the thermal azide-alkyne cycloaddition with internal alkynes and azide substitution is revealed as a flexible, reliable, atom-economica

Replenishable drug depot to combat post-resection cancer recurrence.

Local drug presentation made possible by drug-eluting depots has demonstrated benefits in a vast array of diseases, including in cancer, microbial infection and in wound healing. However, locally-eluting depots are single-use systems that cannot be refilled or reused after implantation at inaccessible sites, limiting their clinical utility. New strategies to noninvasively refill drug-eluting depots could dramatically enhance their clinical use. In this report we present a refillable hydrogel depot system based on bioorthogonal click chemistry. The click-modified hydrogel depots capture prodrug

Enhanced osteogenic differentiation of MC3T3-E1 on rhBMP-2-immobilized titanium via click reaction.

In the present study, we report about the efficacy of titanium surface-immobilized with bone morphogenetic protein-2 (BMP-2) via click reaction on enhanced osteogenic differentiation of MC3T3-E1 cells. The surface was characterized by static contact angles and XPS measurements, which indicated that pristine titanium (Ti-1) was successfully surface-modified via click chemistry (aminated titanium, Ti-4). By quantitative analysis of heparin immobilized on aminated titanium (Ti-4), we found that the Ti-4 can be used as a good candidate to immobilize biomolecules such as heparin. BMP-2 from titaniu

Synthesis of polyvinylethylene glycols (PVEGs) via polyetherification of vinylethylene carbonate by synergistic catalysis.

An efficient and controllable polyetherification of vinylethylene carbonate (VEC) using diols as initiators is developed. By using a synergistic catalysis with palladium and boron reagents under mild conditions, the polymerization process enables the regioselective production of a series of polyvinylethylene glycols (PVEGs) bearing pendent vinyl groups in high yields with accurate molecular weight control and narrow molecular weight distribution. The utility of PVEGs is demonstrated by the production of functional polyurethanes and post-polymerization modification via thiol-ene photo-click che

A functionalized hydroxydopamine quinone links thiol modification to neuronal cell death.

Recent findings suggest that dopamine oxidation contributes to the development of Parkinson's disease (PD); however, the mechanistic details remain elusive. Here, we compare 6-hydroxydopamine (6-OHDA), a product of dopamine oxidation that commonly induces dopaminergic neurodegeneration in laboratory animals, with a synthetic alkyne-functionalized 6-OHDA variant. This synthetic molecule provides insights into the reactivity of quinone and neuromelanin formation. Employing Huisgen cycloaddition chemistry (or "click chemistry") and fluorescence imaging, we found that reactive 6-OHDA p-quinones ca

Can Biomimetic Zinc Compounds Assist a (3 + 2) Cycloaddition Reaction? A Theoretical Perspective.

Compounds bearing tetrazole rings are valuable facets in numerous branches of chemistry including metabolic imaging and the production of drugs and high-energy materials. Synthesis of tetrazoles with the help of organocatalysts has intensively been explored, while enzyme-directed click chemistry has attracted less attention and was only used in inhibitor design so far. Herein, we have investigated the possibility of a biomimetic catalyst based on a Lewis acid approach by exploring the anionic cycloaddition of azide and acetonitrile catalyzed by ZnBr2 in comparison with different biomimetic Zn(

Catalytic and Gating Nanoreactors: Cu(I)/Cu(II)-MOF@HMS for Size-Selective DNA-Templated Click Ligation Chain Reaction and Detection of Nucleic Acids.

Distinguishing short nucleic acid targets from their highly homologous precursors remains a formidable challenge in precision diagnostics, primarily due to the inherent sequence embedding. Herein, we report a spatially confined nanoreactor strategy that synergizes a rigorous size-selective kinetic filter with DNA-templated click ligation chain reaction (DT-CLCR) to achieve highly specific exponential amplification. We engineered a core-shell architecture comprising a mixed-valence Cu(I)/Cu(II)-MOF core encapsulated within a hollow mesoporous silica (HMS) shell. By leveraging the HMS shell as a

A catalyst-free bioorthogonal reaction for malononitrile addition to azodicarboxylates.

Developing bioorthogonal reactions that enable fundamental exploration of biological processes remains a key pursuit of chemical biology. We report here malononitrile addition to azodicarboxylate as a distinct class of catalyst-free bioorthogonal reactions. Our findings demonstrate that malononitriles react rapidly with azodicarboxylates in both organic and aqueous environments at ambient temperature, without requiring catalysts, bases, or additives. The facile modification of malononitriles facilitates their incorporation into biomolecules such as RNA and proteins. This malononitrile addition
