We compared the ocular tissue and systemic blood distribution patterns of clonidine, an alpha 2-adrenergic agonist with ocular hypotensive activity, after topical application with ophthalmic rods or ophthalmic solution (eyedrops) in rabbits. We measured tissue concentrations at 0-240 minutes after administration with rods containing 5 micrograms, 10 micrograms, or 20 micrograms clonidine, or a 0.125% (62.5 micrograms) solution. Administration of 3-isobutyl-1-methylxanthine (IBMX) to rats performing a FR30 operant for food reinforcement produces a dose-dependent suppression of behavior. The authors have used Clonidine for withdrawal from opiates of prisoners. About detoxification of 36 men entering into prison and published studies, they show that Clonidine is an efficacy and safe treatment. Clonidine induced hypotensia is not considered problematic.

Twenty-one patients continued a double-blind crossover study to compare the prophylactic effect on migraine of propranolol and clonidine. The daily dosage of propranolol and clonidine was 160 mg and 100 microgram, respectively. Statistical analysis did not show any significant difference between the two drugs in respect to headache or nausea. The number of sickleave days and the use of symptomatic drugs were both less on propranolol treatment than on clonidine, but there was no statistically conclusive difference.

BACKGROUND: Partial sciatic nerve ligation (PSNL) produces axonal damage, a local inflammatory response, and wallerian degeneration. Cytokines secreted near the site of nerve injury are thought to play important roles in development and maintenance of central sensitization and neuropathic pain. Injection of clonidine at the site and time of nerve injury slows the development of PSNL-induced hypersensitivity and reduces local cytokine expression by actions on alpha2 adrenoceptors. A 70 year old hypertensive patient who had undergone a gastrectomy experienced withdrawal symptoms 10 h after a second epidural injection of morphine (150 micrograms X kg-1) for postoperative analgesia.
