We examined 212 sera from patients with respiratory diseases (n = 55), from women regularly attending a gynaecology clinic (n = 155) and from patients with cat-scratch disease (n = 2), using the complement fixation test against Chlamydia psittaci and IgG indirect immunofluorescence tests against 3 different strains of C. psittaci, C. trachomatis (serotype L2) and the TWAR strain. Competitive and standard enzyme-linked immunosorbent assays (ELISAs), rose bengal (RB), complement fixation, and agar gel immunoprecipitation with native hapten (AGID-NH) were compared by using sera from Brucella-free, Brucella melitensis-infected, and B. melitensis Rev1-vaccinated sheep. The most sensitive tests were indirect ELISA and RB, and the most specific tests were AGID-NH and competitive ELISA. We show that RB followed by AGID-NH is a simple and effective system for diagnosing sheep brucellosis.

Five mares and a stallion were studied from three to 30 months after experimental infection with Brucella abortus strain 544. The mares bred normally. No organisms were recovered from horses or from pregnant Friesian heifer contacts. Titres of serum antibody in the antiglobulin (Coombs) and complement fixation tests fell more slowly than those assessed by other tests. The serum of one foal yielded maternal antibody. An intradermal test was positive in infected adults only, and negative in all foals.

A method is described by which a soluble antigen was prepared from the yeast phase of Histoplasma capsulatum. This soluble preparation had a specificity greater than that of whole-cell yeast-phase antigens. In complement fixation tests with sera from human cases of histoplasmosis, blastomycosis, and coccidioidomycosis, the soluble antigen reacted in 12.1% of 141 tests with heterologous sera, whereas conventional whole-cell yeast antigens reacted in 47.3% of 91 tests with heterologous sera. The reactivities of the two types of antigens with homologous sera were essentially the same.

A cryopreserved stabilate was prepared from Babesia bovis-infected Boophilus microplus ticks. The stabilate was used to infect a group of mature Bos taurus-Bos indicus cows. 1,600 random serum samples from inhabitants of Hesse were investigated for rickettsial antibodies by complement fixation test. Antibodies were found in 19 samples (1,2%) against RMSF-group antigen with titers between 1:10 and 1:640. These results confirm recent reports on natural foci of such rickettsial infections in the Federal Republic of Germany. Antibodies against rickettsia of the Typhus-group or Coxiella burnetii could not be detected by the complement fixation reaction. None of the sera reacted with significant titer in the Weil-Felix-Test.

Quantitative precipitation and complement fixation tests were performed on sera from a group of patients with hypersensitivity pneumonitis (pigeon breeders' disease). Their antibody levels were compared to a selected group of similarly exposed but asymptomatic subjects. The enzyme-linked immunosorbent assay (ELISA) was used to detect antibodies to yellow fever virus in 110 sera from patients living in an epidemic yellow fever area. The results were then compared with those obtained with the hemagglutination-inhibition (HI), complement-fixation (CF), neutralization (NT), and indirect immunofluorescence (IFA) tests. This ELISA, which used a type-specific antigen, showed the same results as the NT test and was found to be more sensitive and more specific than the HI and CF tests.

Five cases of cerebral paragonimiasis presenting with hemianopsia, convulsion, and gait disturbance are discussed. The cases were all in the chronic stage. The intradermal paragonimiasis reaction, complement fixation, and Ouchterlony tests were not useful for diagnosis. Computed tomography demonstrated calcifications in all cases in sites consistent with the foci of symptoms. Surgical treatment in two cases failed to improve symptoms.

During a 4-year (2007-2010) survey, the presence of Brucella suis infection in domestic pigs in Sardinia was investigated. Serum samples were collected from breeding pigs located on 108 commercial farms with documented reproductive problems and analysed using the Rose Bengal (RBT) and complement fixation (CFT) tests for screening and confirmation of Brucella, respectively.
