The role of H2S in low temperature-induced cucurbitacin C increases in cucumber.

In this study, we first linked the signal molecule H2S with cucurbitacin C, which can cause the bitter taste of cucumber leaves and fruit, and specifically discuss its molecular mechanism. Cucurbitacin C (CuC), a triterpenoid secondary metabolite, enhances the resistance of cucumber plants to pathogenic bacteria and insect herbivores, but results in bitter-tasting fruits. CuC can be induced in some varieties of cucumber on exposure to plant stressors. The gasotransmitter hydrogen sulfide (H2S) participates in multiple physiological processes relating to plant stress resistance. This study focu

Internalization of Listeria monocytogenes in cantaloupes during dump tank washing and hydrocooling.

Recent listeriosis outbreaks and recalls associated with cantaloupes urge for studies to understand the mechanisms of cantaloupe contamination by Listeria monocytogenes. Postharvest practices such as washing and hydrocooling were suggested to facilitate the contamination of fresh fruits by human pathogens. This study assessed the potential of L. monocytogenes internalization into cantaloupes during dump tank washing and immersion-type hydrocooling in water contaminated with L. monocytogenes. The effect of cantaloupe cultivar, water temperature, and harvesting technique on L. monocytogenes inte

Antifungal activity of plant extracts against Colletotrichum lagenarium, the causal agent of anthracnose in cucumber.

BACKGROUND: Colletotrichum lagenarium is an important plant-pathogenic fungus that causes anthracnose of cucumber, a disease that is widespread under both greenhouse and field cultivation. To find a promising method for the control of this disease, extracts of eight plants from China were screened in the study presented here. RESULTS: The results showed that the extract of Cinnamomum camphora (L.) Presl had significantly greater inhibitory activity against C. lagenarium than the other seven plant extracts tested in vitro. At 5 days after inoculation in a potted seedling experiment, C. camphor

Outbreak of Salmonella Newport infections linked to cucumbers--United States, 2014.

In August 2014, PulseNet, the national molecular subtyping network for foodborne disease surveillance, detected a multistate cluster of Salmonella enterica serotype Newport infections with an indistinguishable pulse-field gel electrophoresis (PFGE) pattern (XbaI PFGE pattern JJPX01.0061). Outbreaks of illnesses associated with this PFGE pattern have previously been linked to consumption of tomatoes harvested from Virginia's Eastern Shore in the Delmarva region and have not been linked to cucumbers or other produce items. To identify the contaminated food and find the source of the contaminatio

Bacterial-Mediated Induced Resistance in Cucumber: Beneficial Effect of the Endophytic Bacterium Serratia plymuthica on the Protection Against Infection by Pythium ultimum.

ABSTRACT The potential of the endophytic bacterium Serratia plymuthica strain R1GC4 in stimulating defense reactions in cucumber (Cucumis sativus) seedlings inoculated with the soilborne pathogen Pythium ultimum was explored at the cellular level. Bacterial treatment prior to Pythium inoculation resulted in less seedling disease development as compared with that in nontreated control plants, in which typical root symptoms were visible by 3 days after inoculation with the pathogen. Histological investigations of root samples revealed striking differences in the extent of plant defense reactions…

Abscisic acid confers antibiotic stress tolerance in cucumber by alleviating antibiotic phytotoxicity and decreasing the levels of antibiotic and antibiotic resistance genes.

Antibiotic pollution in the environment causes serious threats to crop growth and human health. However, effective strategies for alleviating antibiotic stress based on endogenous signaling response to antibiotics in plants remain unclear. Here, we found that the toxic effects of ciprofloxacin (CIP) and tetracycline (TC) at 1 mg L-1 on cucumber seedlings were stronger than lincomycin hydrochloride, sulfacetamide, thiamphenicol and tylosin. Importantly, both CIP and TC induced abscisic acid (ABA) accumulation and the transcripts of ABA-related genes. In turn, spraying exogenous ABA alleviated a

CsANU10 maintains chloroplast structural integrity and transcriptional resilience during heat stress in cucumber.

Heat stress destabilizes chloroplast function and frequently leads to leaf chlorosis in crops, yet the genetic mechanisms linking chloroplast integrity to nuclear stress responses remain poorly defined. Here, we identify CsANU10, encoding a chloroplast-localized homolog of Arabidopsis ANGULATA10, as a key regulator of thermosensitive chloroplast stability in cucumber. A recessive missense mutation in CsANU10 causes temperature-dependent and largely irreversible chlorosis accompanied by enlarged and structurally disorganized chloroplasts. Under heat stress, CsANU10 deficiency accelerates chloro

Evaluation of commercially available produce antimicrobial washes to improve the quality and microbial safety of fresh produce.

Despite ongoing efforts to mitigate foodborne pathogen contamination, outbreaks associated with the consumption of fresh produce persist. This research evaluated the efficacy of three sanitizers in reducing various microorganisms on the surfaces of fresh cucumbers, broccoli, and lettuce under retail crisping conditions. Specifically, this study examined the ability of peracetic acid (PAA; 80 mg/L), hypochlorous acid (FAC; 50 mg/L free available chlorine), and accelerated hydrogen peroxide (AHP; 500 mg/L) to reduce pathogens (0.79-2.41 log CFU/g) and natural microbiota, including aerobic bacter

Involvement of jasmonic acid, ethylene and salicylic acid signaling pathways behind the systemic resistance induced by Trichoderma longibrachiatum H9 in cucumber.

BACKGROUND: Trichoderma spp. are effective biocontrol agents for many plant pathogens, thus the mechanism of Trichoderma-induced plant resistance is not fully understood. In this study, a novel Trichoderma strain was identified, which could promote plant growth and reduce the disease index of gray mold caused by Botrytis cinerea in cucumber. To assess the impact of Trichoderma inoculation on the plant response, a multi-omics approach was performed in the Trichoderma-inoculated cucumber plants through the analyses of the plant transcriptome, proteome, and phytohormone content. RESULTS: A novel

The chlorophyll-deficient golden leaf mutation in cucumber is due to a single nucleotide substitution in CsChlI for magnesium chelatase I subunit.

The cucumber chlorophyll-deficient golden leaf mutation is due to a single nucleotide substitution in the CsChlI gene for magnesium chelatase I subunit which plays important roles in the chlorophyll biosynthesis pathway. The Mg-chelatase catalyzes the insertion of Mg(2+) into the protoporphyrin IX in the chlorophyll biosynthesis pathway, which is a protein complex encompassing three subunits CHLI, CHLD, and CHLH. Chlorophyll-deficient mutations in genes encoding the three subunits have played important roles in understanding the structure, function and regulation of this important enzyme. In a

Design, synthesis and structure-activity relationship of novel diphenylamine derivatives.

Diphenylamine derivatives have been reported with good fungicidal, insecticidal, acaricidal, rodenticidal and/or herbicidal activities. To find new lead compound of this kind, a series of novel diphenylamine derivatives were designed and synthesized by the approach of Intermediate Derivatization Methods. All compounds were identified by (1)H NMR and elemental analysis. Bioassays demonstrated that some compounds substituted at 2,4,6-positions or 2,4,5-positions of phenyl ring B exhibited excellent fungicidal activities. The optimal compounds P30 and P33 showed 80% and 85% control respectively a

Influence of nutrient amendments on the phytoextraction of weathered 2,2-bis(p-chlorophenyl)-1,1-dichloroethylene by cucurbits.

Field experiments were conducted to determine the impact of nutrient amendments on the phytoextraction of weathered 2,2-bis(p-chlorophenyl)-1,1-dichloroethylene (p,p '-DDE) by eight cultivars of cucurbits over a single growing season. Four cultivars of Cucurbita pepo ssp pepo are accumulators and extract percent level quantities of persistent organic pollutants (POPs), whereas C. pepo ssp ovifera and Cucumis sativus are nonaccumulators. The nonamended accumulators phytoextracted 1.0% of the p,p'-DDE and had a translocation factor of 0.44; however, the nonaccumulators removed 0.16% of the conta…
