cAMP response element-binding protein is required for dopamine-dependent gene expression in the intact but not the dopamine-denervated striatum.

The cAMP response element-binding protein (CREB) is believed to play a pivotal role in dopamine (DA) receptor-mediated nuclear signaling and neuroplasticity. Here we demonstrate that the significance of CREB for gene expression depends on the experimental paradigm. We compared the role of CREB in two different but related models: l-DOPA administration to unilaterally 6-hydroxydopamine lesioned rats, and cocaine administration to neurologically intact animals. Antisense technology was used to produce a local knockdown of CREB in the lateral caudate-putamen, a region that mediates the dyskinetic…

Dopamine and endogenous opioid regulation of picrotoxin-induced locomotion in the ventral pallidum after dopamine depletion in the nucleus accumbens.

Microinjection of picrotoxin or the mu-opioid agonist, Tyr-D-Ala-Gly-NmePhe-Gly-OH (DAMGO), into the ventral pallidum (VP) produces an increase in locomotor activity that is antagonized by dopamine receptor blockade. To investigate the regulation of VP-induced locomotion by the dopaminergic innervation of the nucleus accumbens (NA) and the role of opioid receptors in this regulation, dopamine innervation of the NA was bilaterally lesioned with 6-hydroxydopamine (6-OHDA). The lesions resulted in an 89-97% depletion of tissue dopamine levels in the nucleus accumbens compared with sham-lesioned r…

Divergent Dopamine Dynamics within the Accumbens Core.

Dopamine in the nucleus accumbens Core powerfully influences motivation and learning, yet the information encoded by Core dopamine remains unresolved. Competing theories typically assume a spatially uniform signal, and standard recording methods average dopamine over substantial tissue volumes. Using two-photon imaging through microprisms in behaving mice, we reveal a pronounced spatial organization of Core dopamine dynamics. Posterior Core showed phasic dopamine increases to both rewards and aversive stimuli, with transfer to a predictive cue after learning. Anterior Core showed phasic decrea…

Optimizing cognitive enhancement through dopamine transporter modulation.

Psychostimulants act on monoamine systems including the dopamine transporter (DAT), a key regulator of dopamine reuptake following synaptic release. While these compounds can enhance energy, cognition, and sociability, their clinical utility is often limited by their abuse potential and peripheral side effects. Research has traditionally focused more on their addictive properties rather than their therapeutic potential. However, certain psychostimulants, such as R-modafinil, enhance cognitive performance without inducing significant euphoria or addiction, making them promising lead candidates …

Clinical trial highlights: Dopamine cell-replacement therapies.

Parkinson's disease is a common neurodegenerative disorder, which is characterised by motor features, many of which relate to the loss of the dopaminergic nigrostriatal pathway. The use of grafted cells to replace the lost dopaminergic neurons as a therapy for Parkinson's has been explored since the 1980s, with mixed clinical outcomes. Much of the heterogeneity in outcomes has been related to the major problems with the cell source for these trials, being derived from human fetal brain tissue. It is, however, now possible to derive authentic midbrain dopamine cells from human pluripotent stem …

Dopamine receptor sensitivity and Pavlovian conditioned approach.

Understanding the determinants of individual differences in cue-reactivity and drug sensitivity is critical to identifying neurobiological mechanisms underlying vulnerability to addiction. In this study, we examined the relationship between dopamine D1 and D2 receptor sensitivity and the attribution of incentive salience to reward cues and sensitivity to cocaine. Male Sprague Dawley rats were classified as having high or low sensitivity to the D2 receptor agonist quinpirole, and a subset was tested with the D1 receptor agonist SKF 82958. Cue-reactivity was assessed using a Pavlovian conditione…

Dopamine Modulates Effective Connectivity in Frontal Cortex.

There is increasing evidence that the left lateral frontal cortex is hierarchically organized such that higher-order regions have an asymmetric top-down influence over lower order regions. However, questions remain about the underlying neuroarchitecture of this hierarchical control organization. Within the frontal cortex, dopamine plays an important role in cognitive control functions, and we hypothesized that dopamine may preferentially influence top-down connections within the lateral frontal hierarchy. Using a randomized, double-blind, within-subject design, we analyzed resting-state fMRI d…

Mesial temporal dopamine: From biology to behaviour.

While colloquially recognized for its role in pleasure, reward, and affect, dopamine is also necessary for proficient action control. Many motor studies focus on dopaminergic transmission along the nigrostriatal pathway, using Parkinson's disease as a model of a dorsal striatal lesion. Less attention to the mesolimbic pathway and its role in motor control has led to an important question related to the limbic-motor network. Indeed, secondary targets of the mesolimbic pathway include the hippocampus and amygdala, and these are linked to the motor cortex through the substantia nigra and thalamus…

Midbrain dopamine neurons arbiter OCD-like behavior.

The neurobiological understanding of obsessive-compulsive disorder (OCD) includes dysregulated frontostriatal circuitry and altered monoamine transmission. Repetitive stereotyped behavior (e.g., grooming), a featured symptom in OCD, has been proposed to be associated with perturbed dopamine (DA) signaling. However, the precise brain circuits participating in DA's control over this behavioral phenotype remain elusive. Here, we identified that DA neurons in substantia nigra pars compacta (SNc) orchestrate ventromedial striatum (VMS) microcircuits as well as lateral orbitofrontal cortex (lOFC) du…

Dopamine agonists for preventing ovarian hyperstimulation syndrome.

BACKGROUND: Ovarian hyperstimulation syndrome (OHSS) is a potentially serious complication of ovarian stimulation in assisted reproduction technology (ART). It is characterised by enlarged ovaries and an acute fluid shift from the intravascular space to the third space, resulting in bloating, increased risk of venous thromboembolism, and decreased organ perfusion. Most cases are mild, but forms of moderate or severe OHSS appear in 3% to 8% of in vitro fertilisation (IVF) cycles. Dopamine agonists were introduced as a secondary prevention intervention for OHSS in women at high risk of OHSS unde…

Dysfunctional striatal dopamine signaling in Huntington's disease.

Dopamine signaling in the striatum is critical for a variety of behaviors including movement, behavioral flexibility, response to reward and many forms of learning. Alterations to dopamine transmission contribute to pathological features of many neurological diseases, including Huntington's disease (HD). HD is an autosomal dominant genetic disorder caused by a CAG repeat expansion in the Huntingtin gene. The striatum is preferentially degenerated in HD, and this region receives dopaminergic input from the substantia nigra. Studies of HD patients and genetic rodent models have shown changes to …

Cavity Carbon-Nanopipette Electrodes for Dopamine Detection.

Microelectrodes are typically used for neurotransmitter detection, but nanoelectrodes are not because there is a trade-off between spatial resolution and sensitivity that is dependent on surface area. Cavity carbon-nanopipette electrodes (CNPEs), with tip diameters of a few hundred nanometers, have been developed for nanoscale electrochemistry. Here, we characterize the electrochemical performance of CNPEs with fast-scan cyclic voltammetry (FSCV) for the first time. Dopamine detection using cavity CNPEs, with a depth equivalent to a few radii, is compared with that using open-tube CNPEs, an es…

Cariprazine:New dopamine biased agonist for neuropsychiatric disorders.

Cariprazine (RGH-188, MP-214, Vraylar[TM]) is a new dopamine receptor ligand developed for the treatment of several neuropsychiatric diseases including schizophrenia and bipolar disorders. Cariprazine displays higher affinity at dopamine D3 receptors and a similar affinity at D2 and 5-HT2B receptors. At variance with some atypical antipsychotics, its affinity at 5-HT1A, 5-HT2A and histamine H1 receptors is modest compared with its three main targets. Cariprazine could correspond to a biased agonist at dopamine receptors, displaying either antagonist or partial agonist properties depending on t…

[Scans without Evidence of Dopamine Deficit (SWEDDs)].

Dopamine transporter (DaT) single-photon emission computed tomography (SPECT) and [18F]fluoro-L-DOPA ([18F]DOPA) positron emission tomography (PET) facilitate the investigation of dopaminergic hypofunction in neurodegenerative diseases. DaT SPECT and [18F]DOPA PET have been adopted as survey tools in clinical trials. In a large study on Parkinson's disease, 4-15% of subjects clinically diagnosed with early-stage Parkinson's disease had normal dopaminergic functional imaging scans. These are called Scans without Evidence of Dopamine Deficit (SWEDDs), and are considered to represent a state diff…

Aberrant mesolimbic dopamine-opiate interaction in obesity.

Dopamine and opioid neurotransmitter systems share many functions such as regulation of reward and pleasure. μ-Opioid receptors (MOR) modulate the mesolimbic dopamine system in ventral tegmental area and striatum, key areas implicated in reward. We hypothesized that dopamine and opioid receptor availabilities correlate in vivo and that this correlation is altered in obesity, a disease with altered reward processing. Twenty lean females (mean BMI 22) and 25 non-binge eating morbidly obese females (mean BMI 41) underwent two positron emission tomography scans with [(11)C]carfentanil and [(11)C]r…

Imaging dopamine transmission parameters in cannabis dependence.

Low striatal dopamine D2/3 receptor (D2/3) availability and low ventrostriatal dopamine release have been observed in alcoholism, cocaine and heroin dependence. Multiple studies to date have examined D2 availability in cannabis dependence and have consistently failed to demonstrate alterations. In addition, the response of the dopamine system to an amphetamine challenge and to a stress challenge has also been examined, and did not show alterations. We review these studies here and conclude that cannabis dependence is an exception among commonly abused drugs in that it is not associated with bl…

Dopamine-induced nonmotor symptoms of Parkinson's disease.

Nonmotor symptoms of Parkinson's disease (PD) may emerge secondary to the underlying pathogenesis of the disease, while others are recognized side effects of treatment. Inevitably, there is an overlap as the disease advances and patients require higher dosages and more complex medical regimens. The non-motor symptoms that emerge secondary to dopaminergic therapy encompass several domains, including neuropsychiatric, autonomic, and sleep. These are detailed in the paper. Neuropsychiatric complications include hallucinations and psychosis. In addition, compulsive behaviors, such as pathological …

Neural reward processing under dopamine agonists: imaging.

Impulse control disorders (ICDs) are increasingly reported as a considerable side-effect of treatment with dopaminergic medication (both levodopa and dopamine agonists (DA)). ICDs together with punding are described within the entity of dopamine dysregulation syndrome along with immediate reward seeking and addictive behaviors. The brain functions involved in reward processing in general and their modulation by medication can be characterized by neuropsychological assessments and underlying neurobiology can be investigated by functional neuroimaging techniques such as functional magnetic reson…
