Emerging Nano Drug Delivery Systems Targeting Cancer-Associated Fibroblasts for Improved Antitumor Effect and Tumor Drug Penetration.

Currently, clinically used antitumor nanomedicine is usually insufficient to eradicate malignancies, due to the tumor stroma exerting therapeutic resistance and physical barriers for proper drug delivery. As the most abundant cells in the tumor stroma, cancer-associated fibroblasts (CAFs) produce a critical tumor-promoting effect and barriers preventing the physical delivery of nanomedicines through secreting pro-tumorigenic cytokines, increasing solid tumor pressure and interstitial fluid pressure (IFP), and nonspecific internalization. Therefore, beyond treatment centered on cancer cells, re

Targeted treatment for osteoarthritis: drugs and delivery system.

The management of osteoarthritis (OA) is a clinical challenge due to the particular avascular, dense, and occluded tissue structure. Despite numerous clinical reports and animal studies, the pathogenesis and progression of OA are still not fully understood. On the basis of traditional drugs, a large number of new drugs have been continuously developed. Intra-articular (IA) administration for OA hastens the development of targeted drug delivery systems (DDS). OA drugs modification and the synthesis of bioadaptive carriers contribute to a qualitative leap in the efficacy of IA treatment. Nanopar…

An experimental platform for systemic drug delivery to the retina.

Degenerative retinopathies, including age-related macular degeneration, diabetic retinopathy, and hereditary retinal disorders--major causes of world blindness--are potentially treatable by using low-molecular weight neuroprotective, antiapoptotic, or antineovascular drugs. These agents are, however, not in current systemic use owing to, among other factors, their inability to passively diffuse across the microvasculature of the retina because of the presence of the inner blood-retina barrier (iBRB). Moreover, preclinical assessment of the efficacies of new formulations in the treatment of suc

Smart drug delivery systems for immunotherapeutic agents: focus on biological/biogenic platform-based intelligent delivery in cancer immunotherapy.

Cancer immunotherapy marks a major advance in oncology, yet its effectiveness is constrained by challenges such as inefficient tumor targeting, immune-related toxicities, low response rates, and an immunosuppressive tumor microenvironment (TME). To overcome these obstacles, smart drug delivery systems (SDDSs) have been developed to deliver immunotherapeutic agents in a targeted, controlled manner in response to specific stimuli. This review summarizes recent advances in SDDSs for cancer immunotherapy, focusing on biological and biogenic platforms. These systems respond to endogenous and exogen…

Multi-Functional Applications of Hydrogel Delivery Systems in Inflammatory Bowel Disease: Drug Delivery, Anti-Inflammation, and Intestinal Repair.

Inflammatory bowel disease (IBD) represents a chronic inflammatory disorder of the gastrointestinal tract with a multifactorial etiology that remains incompletely elucidated. Accumulating evidence implicates dysregulation of the intestinal micro-ecosystem, aberrant neuroimmune interactions, and compromised epithelial barrier integrity as key contributors to IBD pathogenesis. While oral administration remains the predominant therapeutic approach, the acidic gastric milieu and enzymatic catabolism markedly compromise drug efficacy. Consequently, selecting an optimal drug delivery method has beco

PURPOSE: Propofol is a relatively short-acting potent anesthetic lipophilic drug used during short surgical procedures. Despite the success of propofol intravenous emulsions, drawbacks to such formulations include inherent emulsion instability, the lack of a safe vehicle to prevent sepsis, and concern regarding hyperlipidemia-related side effects. The aim of the current investigation was to develop a novel, lipid-based self-nanoemulsifying drug delivery system (SNEDDS) for propofol with improved stability and anesthetic activity for human use. METHODS: A series of SNEDDS formulations were dev

Formulation and in vitro evaluation of liposomal and self-microemulsifying drug delivery systems for oral delivery of cannabidiol.

Cannabidiol (CBD) is a highly lipophilic molecule characterized by low oral bioavailability, primarily due to its limited solubility and extensive first-pass metabolism. This study aims to improve apparent solubility of cannabidiol through the development of liposomal and self-microemulsifying drug delivery systems (SMEDDS). Liposomes incorporating ω-3 fatty acids, and SMEDDS formulations were prepared and assessed regarding their hydrodynamic diameter, polydispersity index, and z-potential. Furthermore, tablets were developed from lyophilized powders and encapsulated into enteric-coated capsu…

Phase Behavior of Potential Drug Delivery Systems, PolycaprolactoneNonsteroidal Anti-Inflammatory Drug in a Pressurized Carbon Dioxide Medium.

Although high-pressure technologies are widely used, for example, in the polymer and pharmaceutical industries, measuring phase equilibria in high-pressure media is still challenging, especially for multicomponent systems. Although the most reliable methods in solid-liquid-(-gas) equilibrium measurements, also providing additional information, are high-pressure differential scanning calorimetry (DSC) and transitiometry, the most common techniques for detecting solid-liquid phase transitions in pressurized media are visual observation-based methods. In this research, the melting behavior of pot
