[Ependymal cyst--case report].

An eleven-month-old male infant with an ependymal cyst is reported. He was born after full-term pregnancy and vacuum extraction. His growth and development was somewhat retarded and he could control his head at the age of five months. He was admitted to our hospital because of the enlargement of his head and the paretic right upper limb. On admission he was found to be a well-nourished infant with the head circumference of 54.5 cm and the height of 75.5 cm. Physical examination disclosed slight weakness in the right upper extremity but no signs of increased intracranial pressure. Computed tomo…

Cephalothoracopagus janiceps malformation. A contribution to the pathogenesis of cerebral malformation.

We report the first histologic findings of the CNS in cephalothoracopagus janiceps monosymmetros. The findings are compared with the other ten reported cases of this type of conjoined twin. The faces and CNSs showed asymmetry of development manifested chiefly in the prosencephalon, specifically the cortical gyri, corpus callosum, and subependymal mantle layer. It is difficult to explain discordance for congenital malformations in monovular or conjoined twins by either genetic or environmental mechanisms. It appears related to the vascular supply of the brain and a state of chronic ischemia rat…

Neurobrucellosis (Brucella ceti) in striped dolphins (Stenella coeruleoalba): Immunohistochemical studies on immune response and neuroinflammation.

Neurobrucellosis is a shared condition of cetaceans and humans. However, the pathogenesis and immune response in cetacean neurobrucellosis h … interleukin [IL]-1β, IL-2, and IL-6) markers were investigated. Inflammation of the leptomeninges, ependyma, and/or choroid plexus was lymphohistiocytic, containing macrophages/microglia (IBA-1+), T-cells (CD3+), and B-cells (CD20+) in equal proportion. B-cells occasionally formed tertiary follicles. GFAP expression showed astrocytosis in most cases. Expr …

Overcoming Limitations Inherent in Sulfamidase to Improve Mucopolysaccharidosis IIIA Gene Therapy.

Sulfamidase (SGSH) deficiency causes mucopolysaccharidosis type IIIA (MPS IIIA), a lysosomal storage disease (LSD) that affects the CNS. In earlier work in LSD mice and dog models, we exploited the utility of adeno-associated viruses (AAVs) to transduce brain ventricular lining cells (ependyma) for secretion of lysosomal hydrolases into the cerebrospinal fluid (CSF), with subsequent distribution of enzyme throughout the brain resulting in improved cognition and extending lifespan. A critical feature of this approach is efficient secretion of the expressed enzyme from transduced cells, for deli…

Automated segmentation of neurons and spinal cord structures in immunofluorescence images using SpineDL.

In this study, we present SpineDL, an open-source deep learning (DL) approach for neuron and anatomical structure segmentation of the spinal cord in fluorescence images immunostained with NeuN and DAPI, within the context of murine models of spinal cord injury (SCI). SpineDL comprises two main modules: SpineDL-Neuron, for instance-level identification of neuronal somas; and SpineDL-Structure, for semantic segmentation of key spinal cord structures including gray matter, white matter, ependyma, and damaged tissue. To train the models, we developed the SpineDL dataset, a curated collection of 16…

Tissue distribution and cell tropism of Brucella canis in naturally infected canine foetuses and neonates.

Brucella canis infection is an underdiagnosed zoonotic disease. Knowledge about perinatal brucellosis in dogs is extremely limited, although … epithelial cells of gastrointestinal mucosa, renal tubules, epidermis, adipocytes, choroid plexus, ependyma, neuroblasts, blood vessels endothelium, muscle cells, and in the intestinal lumen. These results largely expand our knowledge about perinatal brucellosis in the dog, clearly demonstrating a pantropic distribution of B. canis in naturally infected f …

Systematic Analysis of CCNO Variants in a Defined Population: Implications for Clinical Phenotype and Differential Diagnosis.

Reduced generation of multiple motile cilia (RGMC) is a novel chronic destructive airway disease within the group of mucociliary clearance d … ity (22%). Consistent with these findings, we demonstrate that CCNO expression is present in murine ependyma and fallopian tubes. CCNO is mutated more frequently than expected from the rare previous clinical case reports, leads to severe clinical manifestations, and should therefore be considered an important differential diagnosis of mucociliary clearance …

Characterisation and localisation of [3H]2-(2-benzofuranyl)-2-imidazoline binding in rat brain: a selective ligand for imidazoline I2 receptors.

In rat whole brain homogenates, saturation binding analysis revealed that both [3H]2-BFI (2-(2-benzofuranyl)-2-imidazoline) and [3H]idazoxan … n nuclei, notably the area postrema, interpeduncular nucleus, arcuate nucleus, mammillary peduncle, ependyma and pineal gland. These data indicate that [3H]2-BFI recognises imidazoline I2 receptors in rat brain with higher affinity and selectivity than [3H]idazoxan and thus represents a superior radioligand to [3H]idazoxan for the study of imidazoline I2 r …

Intraventricular infusion of basic fibroblast growth factor (bFGF) in the MPTP-treated common marmoset.

Basic fibroblast growth factor (bFGF) prevents damage to the nigrostriatal system in rodents. We now report the effects of bFGF administered … r animals receiving bFGF. Histological examination revealed proliferation of the choroid plexus and ependyma which was most marked in the high dose animals. Adverse effects, in the form of hydrocephalus and neurological deterioration, were presumably secondary to an ependymal and choroid plexus reaction induced by bFGF.

TMEM106B amyloid filaments in the Biondi bodies of ependymal cells.

Biondi bodies are filamentous amyloid inclusions of unknown composition in ependymal cells of the choroid plexuses, ependymal cells lining cerebral ventricles and ependymal cells of the central canal of the spinal cord. Their formation is age-dependent and they are commonly associated with a variety of neurodegenerative conditions, including Alzheimer's disease and Lewy body disorders. Here, we show that Biondi bodies are strongly immunoreactive with TMEM239, an antibody specific for inclusions of transmembrane protein 106B (TMEM106B). Biondi bodies were labelled by both this antibody and the …

Spinal intramedullary ependymal cyst and tethered cord in an adult. Case report.

Spinal intramedullary ependymal cysts are extremely rare. Fourteen cases have been reported in the literature, and only one was associated with another congenital lesion. The authors describe the case of a 33-year-old man in whom an intramedullary ependymal cyst and filar lipoma were present. These lesions were diagnosed using magnetic resonance imaging and were treated surgically. Pathological examination confirmed the diagnosis. At 6 months postsurgery, there was no evidence of recurrence.

Subependymal pseudocysts in the fetal brain: prenatal diagnosis of two cases and review of the literature.

Subependymal pseudocysts are cerebral cysts found in 5% of all neonates. When they are isolated and typical, they result from persistence of the germinal matrix, have a good prognosis and regress spontaneously within a few months. However, associated anomalies are frequent and in such cases the prognosis is poor. They can be of infectious, vascular, metabolic or chromosomal origin. Subependymal pseudocysts have rarely been described in the antenatal period. We report the prenatal diagnosis of two cases of isolated pseudocysts at 32 and 23 weeks. Both ultrasonography and magnetic resonance imag…

The efficacy of shunting the hydrocephalic edema.

The efficacy of shunting the hydrocephalic edema was evaluated by means of transmission electromicroscopical observation (TEM) comparing ultrastructural alterations seen in either valid or invalid shunted feline hydrocephalus. Owning to shunt placement, deteriorated clinical symptoms recovered and one of the observed morphological alterations such as gliosis developed chronologically after the initiation of hydrocephalus. On the other hand, so called hydrocephalic edema observed in the region of periependymal tissue after shunt placement was improved not only in both valid and invalid shunted …

Pain reaction after topical NA and lesions of the obex region in the alert guinea pig.

A study was carried out on the effect of topical application of 3.0 micrograms/microliters noradrenalin (NA) to the obex of awake guinea pigs bearing electrolyte lesions and submitted to noxious peripheral stimulation (electric pulses). The animals with extensive lesions covering the area postrema (AP) and the solitary tract nucleus (STN) showed a marked increase in vocalization (V) and defense motor response (M) to the noxious peripheral stimulus after NA application. In the group bearing lesions restricted to the AP, the analgesic effect of NA was potentiated, with a marked decrease in V and
