Mitochondrial genomes of two subspecies of Citellophilus tesquorum: Insights into the molecular evolution and phylogenetic analysis within Siphonaptera.

BACKGROUND: Citellophilus tesquorum is a major flea vector in natural plague foci across the Palaearctic region of China, representing a significant threat to veterinary and public health. Precise identification and sound taxonomic delimitation of its subspecies are critical for effective vector surveillance and evolutionary research. METHODS: We first performed morphological identification to guarantee specimen authenticity and accuracy, then sequenced and annotated the complete mitochondrial genomes of C. t. dzetysuensis and C. t. mongolicus using the Illumina NovaSeq platform. Comparative

A single-cell atlas of the bobtail squid visual and nervous system highlights molecular principles of convergent evolution.

The cephalopod and vertebrate visual systems are a textbook example of convergent evolution with unknown molecular underpinnings. Here we characterize 98,537 single-cell transcriptomes in the bobtail squid Euprymna berryi to understand how the cephalopod retina and optic lobes relate to the vertebrate retina. We confirm the overall relative simplicity of the cephalopod retina but identify two related photoreceptor cell subtypes expressing distinct r-opsins. By contrast, the adult optic lobe contains a diverse repertoire of neuronal and glial cell types, with a predominance of dopaminergic neur

Molecular evolution of coxsackievirus A6 associated with atypical hand, foot, and mouth disease in Northern Brazil in 2019.

Globally, coxsackievirus A6 (CVA6) is a major cause of hand, foot, and mouth disease (HFMD) outbreaks, although studies on its evolutionary and transmission dynamics are limited. Here, we analyzed samples from atypical HFMD cases in Brazil. Enteroviruses were detected in 49.2% (156/317) of the specimens, with CVA6 being the primary pathogen. Phylogenetic analysis based on VP1 gene sequences revealed that Brazilian CVA6 isolates belonged to the lineage D3 and were divided into two distinct clades from different regions. Temporal analysis indicated that the most recent common ancestor dates back

Tracing the Origin, Spread, and Molecular Evolution of Dengue Type 1 Cases That Occurred in Northern Italy in 2023.

The dengue virus (DENV) is a mosquito-borne flavivirus endemic to many tropical and subtropical regions. Over the past few decades, the global incidence of dengue has risen dramatically, with the virus now present in over 100 countries, putting nearly half of the world's population at risk. This increase is attributed to several factors, including urbanization, climate change, and global travel, which facilitate the spread of both the virus and its mosquito vectors. While dengue is primarily associated with tropical regions, outbreaks in temperate areas are becoming increasingly common due to

Molecular Evolution of Apolipoprotein Multigene Family and the Original Functional Properties of Serum Apolipoprotein (LAL2) in Lampetra japonica.

Apolipoprotein (APO) genes represent a large family of genes encoding various binding proteins associated with plasma lipid transport. Due to the long divergence history, it remains to be confirmed whether these genes evolved from a common ancestor through gene duplication and original function, and how this evolution occurred. In this study, based on the phylogenetic tree, sequence alignment, motifs, and evolutionary analysis of gene synteny and collinearity, APOA, APOC, and APOE in higher vertebrates may have a common ancestor, lamprey serum apolipoprotein LAL1 or LAL2, which traces back to

The dynamics of molecular evolution of emerging avian reoviruses through accumulation of point mutations and genetic re-assortment.

In the last decade, the emergence of variant strains of avian reovirus (ARV) has caused enormous economic impact in the poultry industry across Canada and USA. ARVs are non-enveloped viruses with ten segments of double-stranded RNA genome. So far, only six genotyping cluster groups are identified worldwide based on sequence analysis of the σC protein encoded by the S1 segment. In this study, we performed deep next generation whole-genome sequencing and analysis of twelve purified ARVs isolated from Saskatchewan, Canada. The viruses represent different genotyping cluster. A genome-wide sequence

Role of accelerated segment switch in exons to alter targeting (ASSET) in the molecular evolution of snake venom proteins.

BACKGROUND: Snake venom toxins evolve more rapidly than other proteins through accelerated changes in the protein coding regions. Previously we have shown that accelerated segment switch in exons to alter targeting (ASSET) might play an important role in its functional evolution of viperid three-finger toxins. In this phenomenon, short sequences in exons are radically changed to unrelated sequences and hence affect the folding and functional properties of the toxins. RESULTS: Here we analyzed other snake venom protein families to elucidate the role of ASSET in their functional evolution. ASSE

Molecular cloning of Echis ocellatus disintegrins reveals non-venom-secreted proteins and a pathway for the evolution of ocellatusin.

We report the cloning and sequence analysis of Echis ocellatus cDNAs coding for dimeric disintegrin subunits and for the short disintegrin ocellatusin. All the dimeric disintegrin subunit messengers belong to the short-coding class, indicating that short messengers may be more widely distributed than previously thought. Mass spectrometric analysis of the HPLC-separated venom proteins was performed to characterize the dimeric disintegrins expressed in the venom proteome. In addition to previously reported EO4 and EO5 heterodimers, a novel dimeric disintegrin containing RGD- and KGD-bearing subu

Lactate dehydrogenase genes of caiman and Chinese soft-shelled turtle, with emphasis on the molecular phylogenetics and evolution of reptiles.

L-Lactate dehydrogenase (LDH) cDNAs encoding for LDH-A(4) (muscle) and LDH-B(4) (heart) isozymes from caiman (Caiman crocodilus apaporiensis) belonging to the order Crocodilia and Chinese soft-shelled turtle (Pelodiscus sinensis) belonging to the order Chelonia were sequenced. The phylogenetic relationships of the newly determined cDNA and their deduced protein sequences, as well as the previously published sequences of vertebrate LDH isozymes, were analyzed by various phylogenetic tree construction methods. These results indicated that Chelonia is indeed more closely related to Crocodilia. Th

Sequence diversity and molecular evolution of the leukotoxin (lktA) gene in bovine and ovine strains of Mannheimia (Pasteurella) haemolytica.

The molecular evolution of the leukotoxin structural gene (lktA) of Mannheimia (Pasteurella) haemolytica was investigated by nucleotide sequence comparison of lktA in 31 bovine and ovine strains representing the various evolutionary lineages and serotypes of the species. Eight major allelic variants (1.4 to 15.7% nucleotide divergence) were identified; these have mosaic structures of varying degrees of complexity reflecting a history of horizontal gene transfer and extensive intragenic recombination. The presence of identical alleles in strains of different genetic backgrounds suggests that as

Functional consistency and molecular evolution: a comprehensive characterization of primary endometrial cancer-associated fibroblasts during in vitro expansion.

BACKGROUND: Cancer-associated fibroblasts (CAFs) are critical components of the tumor microenvironment in endometrial cancer (EC). Primary isolation and culture of CAFs serve as the standard method for investigating their biological roles. However, it remains unclear whether prolonged in vitro culture alters the intrinsic properties of CAFs, potentially affecting experimental reproducibility. This study aims to evaluate the phenotypic and transcriptomic evolution of primary EC-derived CAFs across different passages. METHODS: CAFs were isolated from human EC tissues using enzymatic digestion a

Research note: Molecular characteristics and genetic evolution of H1N1 avian influenza virus from wild birds in Shanghai, China.

The H1N1 influenza virus is a major pandemic and seasonal pathogen with a broad host range, posing a substantial threat to human health and underscoring the need for continuous surveillance. Wild birds, as natural reservoirs of avian influenza viruses (AIVs), carry H1N1 strains capable of reassorting with other influenza viruses, which can drive pandemic emergence. The global migration of wild birds facilitates the spread of these viruses, and their interactions with poultry increase the risk of cross-species transmission, further amplifying the public health threat. However, knowledge of H1N1

Molecular epidemiological surveillance of the feline coronavirus reveals novel recombinant FCoV strains with evidence of ongoing viral evolution.

Feline Infectious Peritonitis Virus (FIPV) is a virulent biotype of feline coronavirus (FCoV) that causes fatal systemic disease in cats and continues to emerge through mutation and recombination. We conducted demographic data collection, molecular surveillance and genomic characterization of FCoV circulating in feral and shelter cats. Among 139 cats screened, 22.3% tested positive for FCoV. Lower weight and Body Condition Scores indicate higher likelihood for FCoV. Whole-genome sequencing identified three novel FCoV (FIPV-170, FIPV-193, and FIPV-246) belonging to FCoV type I. The viral genome

Spatial Profiling Reveals Distinct Molecular and Immune Evolution of Mouse Lung Adenocarcinoma Precancers with or Without Carcinogen Exposure.

Tumor evolution involves genetic, transcriptional, and phenotypic alterations that shape cancer cell behavior and interactions with the microenvironment. While single-cell technologies have advanced our understanding of this process, spatial dynamics remain incompletely characterized. Here, whole-exome sequencing (WES), imaging mass cytometry (IMC), and spatial transcriptomics (ST) were integrated to study molecular evolution and immune responses in two lung adenocarcinoma (LUAD) mouse models: a genetically engineered model (129S4/Sv-KrasLSL-G12D, termed 129S4 K) and a carcinogen-induced preca

Molecular phylogenetics provides insights into biogeography and character evolution in the assassin bug tribe Saicini (Hemiptera: Reduviidae: Emesinae).

Reduviidae (assassin bugs) are a species-rich group of terrestrial predatory insects, exhibiting remarkable diversity in foreleg morphology. The tribe Saicini (Hemiptera: Reduviidae: Emesinae) displays various morphological modifications of the foreleg. Although the definition and classification of Saicini have undergone considerable changes in recent years, the higher-level phylogeny and evolutionary history of the tribe remain poorly understood. Here, we generated a molecular dataset for Saicini to investigate its phylogeny, historical biogeography and foreleg character evolution. Our phylog

Longitudinal Molecular Characterization of a Rare TPM3::PDGFRB-Rearranged Myeloid/Lymphoid Neoplasm With Late CNL-Like Neutrophilic Evolution.

OBJECTIVES: Myeloid/lymphoid neoplasms with tyrosine kinase fusions are rare but important because many are highly sensitive to targeted therapy. PDGFRB rearrangements are well-established drivers, whereas TPM3 is an exceptionally rare fusion partner. METHODS: We performed longitudinal analysis of an elderly patient with hypereosinophilia, integrating morphology, cytogenetics, PDGFRB break-apart FISH, whole-transcriptome RNA sequencing, optical genome mapping, targeted myeloid DNA sequencing and exploratory fusion-specific RT-qPCR. RESULTS: A balanced t(1;5)(q21;q33) with PDGFRB rearrangemen

The Tolerated functional decline Model: Relaxed constraint in the molecular evolution of mammalian master Sex-Determining gene SRY.

While most genes are subject to strong purifying selection to maintain essential functions, some master sex-determining genes (MSDGs) appear to evolve under unexpectedly relaxed constraints. To understand the molecular mechanisms and evolutionary rationale behind this phenomenon, we investigated the eutherian Sry gene, a top-level regulator of testis development that originated from Sox3. We focused on the high mobility group (HMG) box DNA-binding domain, analyzing its sequences across ten representative species. Comparative analyses revealed that Sry/SRY has a dN/dS ratio approximately 14 tim
