Bioinformatics analysis and clinical validation identify FBXW9 as a biomarker in bladder urothelial carcinoma.

BACKGROUND: F-box and WD repeat domain-containing protein 9 (FBXW9), a member of the F-box protein family, is dysregulated and involved in the progression of human malignancies. However, its functional mechanisms and clinical significance in bladder urothelial carcinoma (BLCA) remain poorly understood. This study aimed to systematically investigate the diagnostic and prognostic value of FBXW9 in BLCA. METHODS: Data obtained from The Cancer Genome Atlas (TCGA) and the Gene Expression Omnibus (GEO) databases were utilized to examine the differential expression of FBXW9 in BLCA tissues compared

Establishing Molecular Mechanism of Carnosine Against Hypobaric Hypoxia Induced Muscle Protein Loss via In-Silico and In-Vivo Approach.

Skeletal muscle is highly susceptible to hypobaric hypoxia linked muscle protein loss due to disturbed proteostasis and redox homeostasis. Carnosine, an endogenous dipeptide possesses antioxidant, anti-inflammatory, anti-glycation and pH buffering properties. The present study investigated the beneficial efficacy of carnosine in amelioration of muscle protein loss during chronic hypobaric hypoxia exposure in rats. Further, probable molecular mechanism for it's protective efficacy was also established via molecular docking study along with support of in-vivo study. Male Sprague-Dawley rats were

The negative regulated immune mechanism of Skp1-Cullin1-F-box (SCF) E3 ubiquitin ligase-mediated HOS15-dependent responses in banana.

Under Foc TR4 stress, MaSKP1-1, MaCULI1, and MaHOS15 form SCFHOS15-type E3 ubiquitin ligase, which inhibits the expression level of SA signal-related factors to regulate the immune process of bananas. Banana wilt disease, caused by Fusarium oxysporum f. sp. cubense, tropical race 4 (Foc TR4), poses a significant threat to global banana production. In Musa spp., HOS15 is believed to play a role in enhancing the response to Foc TR4 stress by the salicylic acid signaling pathway. Previous studies have indicated that HOS15 functions as an F-box protein within the SCF ubiquitin ligase complex, cont

CCDC65, a Gene Knockout that leads to Early Death of Mice, acts as a potentially Novel Tumor Suppressor in Lung Adenocarcinoma.

CCDC65 is a member of the coiled-coil domain-containing protein family and was only reported in gastric cancer by our group. We first observed that it is downregulated in lung adenocarcinoma based on the TCGA database. Reduced CCDC65 protein was shown as an unfavorable factor promoting the clinical progression in lung adenocarcinoma. Subsequently, CCDC65-/- mice were found possibly dead of hydrocephalus. Compared with the CCDC65+/+ mice, the downregulation of CCDC65 in CCDC65+/- mice significantly increased the formation ability of lung cancer induced by urethane. In the subsequent investigati

Screening for Rho GTPase Modulators in Actin-Dependent Processes Exemplified by Phagocytosis.

Rho GTPases, a family of molecular switches, are essential for the assembly and rearrangement of the cellular actin network. Actin remodeling is a central component of many important biological phenomena including chemotaxis, immunological synapse formation, and phagocytosis. Proper execution of these processes requires careful modulation of Rho GTPase activity in space and time. This is accomplished by delicate coordination of Rho GTPase activation and inactivation by Rho guanine nucleotide exchange factors (RhoGEFs) and Rho GTPase-activating proteins (RhoGAPs), respectively. Elucidating the

Clonal biases dictate availability of colonic cancer driver mutations for transformation.

Aged normal tissues harbour cancer mutations predisposing to transformation. However, how different pro-oncogenic events in the human colon compare in frequency, behaviour and subsequent transformation risk remains unclear. Here, we analyse mutation hotspot regions in five colorectal cancer genes (APC, KRAS, TP53, FBXW7 and CTNNB1) using targeted sequencing of 76,800 normal colonic glands from 56 patients. We show that cancer-driving mutations are present in all genes in histologically normal tissue. Reconstruction of clone dynamics reveals that FBXW7 R465C mutations preferentially become fixe

6-Shogaol Attenuates Doxorubicin-Induced Cardiac and Skeletal Muscle Atrophy by Inhibiting E3 Ubiquitin Ligases and Necroptosis.

Doxorubicin (DOX) is an effective anticancer agent, but it not only induces dose-dependent cardiotoxicity but also causes severe cardiac and skeletal muscle atrophy. Recent studies have indicated that cardiac atrophy may play an important role in DOX-induced cardiotoxicity. However, the mechanisms responsible for DOX-induced cardiac and skeletal muscle atrophy remain unclear. 6-Shogaol (6-SH) is a bioactive component from ginger, which exhibits various bioactive effects and can alleviate cisplatin-induced cachexia. In this work, we systematically assessed the effects of DOX and/or 6-SH on hear

VCP controls KCC2 degradation through FAF1 recruitment and accelerates emergence from anesthesia.

Ubiquitin-proteasomal degradation of K+/Cl- cotransporter 2 (KCC2) in the ventral posteromedial nucleus (VPM) has been demonstrated to serve as a common mechanism by which the brain emerges from anesthesia and regains consciousness. Ubiquitin-proteasomal degradation of KCC2 during anesthesia is driven by E3 ligase Fbxl4. However, the mechanism by which ubiquitinated KCC2 is targeted to the proteasome has not been elucidated. We report in cultured neuro-2a cells that the valosin-containing protein (VCP) transported ubiquitinated KCC2 to the proteasome and in mice in vivo experiments that inhibi

Selective deletion of E3 ubiquitin ligase FBW7 in VE-cadherin-positive cells instigates diffuse large B-cell lymphoma in mice in vivo.

During the maturation of hematopoietic stem/progenitor cells (HSPCs) to fully differentiated mature B lymphocytes, developing lymphocytes may undergo malignant transformation and produce B-cell lymphomas. Emerging evidence shows that through the endothelial-hematopoietic transition, specialized endothelial cells called the hemogenic endothelium can differentiate into HSPCs. However, the contribution of genetic defects in hemogenic endothelial cells to B-cell lymphomagenesis has not yet been investigated. Here, we report that mice with endothelial cell-specific deletion of Fbw7 spontaneously de

FBXL5 promotes lipid accumulation in alcoholic fatty liver disease by promoting the ubiquitination and degradation of TFEB.

BACKGROUND: Alcoholic fatty liver disease (AFLD) is characterized by abnormal lipid droplet accumulation in liver. Epigenetic regulation plays an important role in the pathogenesis of AFLD. Comprehensive bioinformatics analysis revealed that an E3 ubiquitin ligase, F-box and leucine-rich repeats protein 5 (FBXL5), was significantly upregulated in AFLD mice. METHODS: The mouse model of AFLD was established by feeding Lieber-DeCarli liquid diet containing ethanol. An in vitro model of AFLD was established by treating HepG2 cells with ethanol (EtOH). The FBXL5 expression was assessed by quantita

Human Papillomavirus 16-Transgenic Mice as a Model to Study Cancer-Associated Cachexia.

Cancer cachexia is a multifactorial syndrome characterized by general inflammation, weight loss and muscle wasting, partly mediated by ubiquitin ligases such as atrogin-1, encoded by Fbxo32. Cancers induced by high-risk human papillomavirus (HPV) include anogenital cancers and some head-and-neck cancers and are often associated with cachexia. The aim of this study was to assess the presence of cancer cachexia in HPV16-transgenic mice with or without exposure to the chemical carcinogen 7,12-dimethylbenz(a)anthracene (DMBA). Male mice expressing the HPV16 early region under the control of the cy

KL-Biome (Postbiotic Formulation of Lactiplantibacillus plantarum KM2) Improves Dexamethasone-Induced Muscle Atrophy in Mice.

Sarcopenia refers to an age-related decrease in muscle mass and strength. The gut-muscle axis has been proposed as a promising target to alleviate muscle atrophy. The effect of KL-Biome-a postbiotic preparation comprising heat-killed Lactiplantibacillus plantarum KM-2, its metabolites, and an excipient (soybean powder)-on muscle atrophy was evaluated using dexamethasone (DEX)-induced atrophic C2C12 myoblasts and C57BL/6J mice. KL-Biome significantly downregulated the expression of genes (Atrogin-1 and MuRF1) associated with skeletal muscle degradation but increased the anabolic phosphorylation

Membrane ATG8ylation in secretory autophagy.

Mammalian Atg8-family (ATG8) proteins are crucial for macroautophagic/autophagic degradation in the lysosome and facilitate non-degradative processes including multiple distinct forms of unconventional protein secretion. These secretion pathways, collectively termed secretory autophagy, depend upon ATG8 conjugated to membranes to both specify and traffic molecules for extracellular release. Here, we review the current understanding of how membrane ATG8ylation supports secretory autophagy, and propose a cell biological framework for classifying the growing repertoire of secretory autophagy path

Topical MTH1 Inhibition Suppresses SKP2-WNT5a-Driven Psoriatic Hyperproliferation.

Topically applied TH1579 alleviated the psoriatic phenotype in the imiquimod-induced psoriasis mouse model by decreasing CD45+, Ly6b+, and CD3+ cell infiltration and downregulating the expression of the proliferation marker PCNA. Moreover, TH1579 strongly suppressed IL-17 expression in mouse skin, accompanied by reduced infiltration of IL-17-producing γδ-T cells. Furthermore, TH1579 decreased keratinocyte viability and proliferation. Mass spectrometry data analysis revealed the enrichment of proteins associated with nucleotide excision repair and cell cycle regulation. The key cell cycle regul

Mutation Severity of FBXO43 Determines Spermatogenic Outcome and Informs Precision ART Strategies.

OBJECTIVE: FBXO43 is a known inhibitor of the anaphase-promoting complex/cyclosome (APC/C), a key E3 ubiquitin ligase that controls meiotic cell cycle progression. However, how different FBXO43 mutations affect APC/C inhibition and lead to divergent clinical phenotypes remains unclear. This study is aimed at clarifying how different FBXO43 mutations produce divergent clinical phenotypes in male infertility and to investigate their preliminary molecular mechanisms, thereby providing evidence-based guidance for precision-assisted reproduction in affected individuals. METHODS: Two infertile pati

Wnt/β-catenin activation by mutually exclusive FBXW11 and CTNNB1 hotspot mutations drives salivary basal cell adenoma.

Basal cell adenoma (BCA) and basal cell adenocarcinoma (BCAC) of the salivary gland are rare tumours that can be difficult to distinguish from each other and other salivary gland tumour subtypes. Using next-generation sequencing, we identify a recurrent FBXW11 missense mutation (p.F517S) in BCA that is mutually exclusive with the previously reported CTNNB1 p.I35T gain-of-function (GoF) mutation with these mutations collectively accounting for 94% of BCAs. In vitro, mutant FBXW11 is characterised by defective binding to β-catenin and higher protein levels within the nucleus. This is consistent
