Prenatal ultrasound charts of orbital total axial length measurement (TAL): a valuable data for correct fetal eye malformation assessment.

OBJECTIVE: To construct prenatal age-specific reference intervals using ultrasound measurement of total axial length (TAL) in normal fetuses for assessing microphthalmia. METHOD: Prospective cross-sectional study of fetuses assessed at a prenatal ultrasound unit between 2011 and 2014. The study cohort comprised 309 pregnant women attending for routine fetal biometry, viability, or anomaly scan between 14 and 41 weeks of gestation. Only singleton viable fetus with normal anatomy, adequate amniotic fluid, accurate gestational age, and no maternal medical complications of pregnancy were enrolled

The effects of IgG purified from women with SLE and associated pregnancy loss on rat embryos in culture.

PROBLEM: Recurrent fetal loss occurs in approximately 1% of women. Autoimmune causes have been suggested as a factor in some of these cases. High rates of intrauterine fetal growth retardation and increased incidence of prematurity is associated with systemic lupus erythematosus (SLE) and the anti-phospholipid syndrome (APS). Autoantibodies from sera of SLE/APS patients affect reproductive outcome in pregnant mice, as was studied in vivo, where injection of immunoglobulin (Ig)G purified from patients with APS to mice caused fetal resorptions and growth retardation. METHODS: In order to invest

When Simplicity Matters: Simplified Skin Tissue Cryopreservation Media for Wildlife Conservation.

INTRODUCTION: The conservation of threatened species requires effective strategies to preserve genetic diversity, with genome resource banks representing a valuable tool in this context. However, many cryopreservation protocols rely on complex media and laboratory infrastructure, limiting their applicability under field conditions. OBJECTIVES: This study evaluated whether simplified cryopreservation media composed of widely available reagents could effectively preserve skin tissue from Neotropical deer using slow freezing and needle-immersed vitrification. METHODS: Skin samples from red broc

Development of oyster protein-enhanced scaffolds with seven-band grouper muscle satellite cells for cultured seafood production.

Cell-based aquaculture holds significant promise for revolutionizing the seafood industry by addressing the limitations of traditional fishing and aquaculture practices. In this study muscle cells from Seven-band grouper (SBG) Epinephelus septemfasciatus were isolated using an enzymatic digestion method. Initial cell attachment and growth were monitored, showing robust proliferation when cultured in L-15 medium supplemented with 10% fetal bovine serum (FBS) and basic fibroblast growth factor (bFGF). We assessed the biocompatibility and cytotoxicity of two protein sources, oyster protein (OP) a

Advances in Management for Preterm Fetuses at Risk of Delivery.

Preterm birth accounts for only 11% of live births but contributes to up to 75% of neonatal mortality and more than half of long-term morbidity. Targeted interventions to reduce the most common causes of perinatal morbidity and mortality include intrapartum group B Streptococcus prophylaxis, magnesium sulfate for fetal neuroprotection, antenatal corticosteroids for fetal lung maturity, latency antibiotics for preterm premature rupture of membranes, and tocolysis to allow corticosteroid administration and transfer to a tertiary care center. This article reviews the evidence for interventions to

Effects of isotretinoin (13-cis-retinoic acid) on the development of mouse limbs in vivo and in vitro.

Isotretinoin (13-cis-RA) is known to be teratogenic in humans and laboratory animals. The relatively low potency of 13-cis-RA in NRMI mice in comparison to the all-trans isomer has been proposed to be due to minimal transfer across the placenta (Creech-Kraft et al., '87). To further delineate the teratogenic potential of 13-cis-RA, a dose-response, temporal study was conducted in vivo and in vitro using submerged limb culture and image analysis evaluation of development. Dose-dependent embryotoxicity was produced by treatment on GD 7, while later treatments produced inconsistent effects on res

Placental Pathophysiology in Maternal Psychoactive Substance Use: Biological, Clinical, and Forensic Perspectives.

Maternal psychoactive substance use during pregnancy represents a major threat to placental integrity and fetal development. As the central interface for maternal-fetal exchange, the placenta is highly susceptible to psychoactive substances, including alcohol, tobacco, cannabis, cocaine, opioids, and synthetic drugs, which can cross the placental barrier and induce structural and functional alterations. This review synthesizes current evidence on the biological mechanisms, diagnostic approaches, and forensic relevance of psychoactive substances-induced placental pathology. We summarize how dif

Anti-Toxoplasma gondii activity of two novel synthetic compounds based on pyridine, benzimidazole and benzoxazole scaffolds in human maternal-fetal interface models.

Congenital toxoplasmosis remains a major public health concern, and current therapy with sulfadiazine and pyrimethamine is limited by toxicity and efficacy, highlighting the need for safer alternatives. This study evaluated the anti-Toxoplasma gondii activity of two novel synthetic compounds, RVI 30 and RVI 33, containing pyridine, benzimidazole, and benzoxazole scaffolds, using BeWo cells and human third-trimester villous explants as models of the maternal-fetal interface. Cell and tissue viability were assessed by MTT; parasite proliferation and invasion by β-galactosidase assay; ultrastruct

Mimicking partial to total placental insufficiency in a rabbit model of cerebral palsy.

All placental abruptions begin as partial abruptions, which sometimes manifest as fetal bradycardia. The progression from partial to total abruption was mimicked by a new rabbit model of placental insufficiency, and we compared it, with sufficient statistical power, with the previous model mimicking total placental abruption. The previous model uses total uterine ischemia at E22 or E25 (70% or 79% term, respectively), in pregnant New Zealand white rabbits for 40 min (Full H-I). The new model, Partial+Full H-I, added a 30-min partial ischemia before the 40-min total ischemia. Fetuses were deliv

Developmental exposure to ethanol increases the neuronal vulnerability to oxygen-glucose deprivation in cerebellar granule cell cultures.

Prenatal alcohol exposure is associated with microencephaly, cognitive and behavioral deficits, and growth retardation. Some of the mechanisms of ethanol-induced injury, such as high level oxidative stress and overexpression of pro-apoptotic genes, can increase the sensitivity of fetal neurons towards hypoxic/ischemic stress associated with normal labor. Thus, alcohol-induced sequelae may be the cumulative result of direct ethanol toxicity and increased neuronal vulnerability towards metabolic stressors, including hypoxia. We examined the effects of ethanol exposure on the fetal cerebellar gra

Intrastriatal transplantation of rat adrenal chromaffin cells seeded on microcarrier beads promote long-term functional recovery in hemiparkinsonian rats.

Possible biologic treatments for Parkinson's disease, a disorder caused by the deterioration of dopaminergic neurons bridging the nigrostriatal system, have recently focused on fetal cell transplantation. Because of ethical and tissue availability issues concerning fetal cell transplantation, alternative cell sources are being developed. The adrenal medulla has been used as a cell transplant source because of the capacity of the cells to provide catecholamines and to transform into a neuronal phenotype. However, adrenal tissue transplants have shown limited success, primarily because of their

Social, ethical and practical aspects of perinatal substance use.

Substance use in pregnancy has garnered increasing attention over the last decade as a particularly concerning facet of the larger national drug problem. This concern stems from the unique circumstance presented by pregnancy, in which the fetus may suffer harm as a result of maternal behavior. Furthermore, organizing a response to this problem is complicated by the ethically and legally challenging nature of the maternal-fetal relationship. The medical implications of perinatal substance use are profound. A discussion of these associated medical and obstetrical complications lies outside the f

What we have learned about antenatal prediction of neonatal morbidity and mortality.

When extremely preterm birth is anticipated, a reliable estimate of neonatal outcome is essential for the parents and health care providers who face difficult management decisions. Estimates of birth weight and gestational age are most commonly used for this purpose. The National Institute of Child Health and Human Development Maternal-Fetal Medicine Units (NICHD MFMU) Network performed an observational study of data available before delivery of infants with birth weights < 1000 g. Ultrasonographic variables (estimated fetal weight, obstetrically estimated gestational age, femur length, and bi

Free radicals and ethanol-induced cytotoxicity in neural crest cells.

Associations between ethanol-induced cranial neural crest cell (NCC) damage in mammalian embryos and subsequent malformations as observed in human fetal alcohol syndrome have previously been illustrated. The vulnerability of NCCs to this teratogen may result, at least in part, from their sensitivity to free radical damage. To examine relationships between free radical generation and NCC cytotoxicity, primary culture of mouse NCCs was used. NCC viability was determined in both dose- and time-response studies involving ethanol exposure. After 48 hr of culture, cell viability was significantly di

The relationship between plasma oestrone sulphate concentrations in pregnant dairy cattle and calf birth weight, calf viability, placental weight and placental expulsion.

A total of 54 Holstein-Friesian cows (13 primiparous and 41 multiparous) was used to study maternal plasma oestrone sulphate (E1S) during pregnancy and its relationship to birth weight and viability of calves and time required for placental expulsion after calving. Plasma samples were obtained from the tail vein of cows once every month from days 90 to 180, every 2 weeks from days 181 to 270, and every day from day 270 of gestation to parturition. The E1S concentrations were measured by radioimmunoassay, and birth weight, placental measurements, neonatal viability and the period from calving t

Periparturient metabolic disease in small ruminants: interactions among maternal adaptation, nutrient partitioning, and placental-mammary function.

Periparturient metabolic diseases remain a major challenge to the productivity, health, and welfare of small ruminants. Traditionally, disorders including pregnancy toxemia, early lactation ketotic disorders, hypocalcemia, and hypomagnesemia have been considered distinct disease entities arising primarily from nutritional deficiencies during late gestation and early lactation. However, increasing evidence suggests these conditions represent interconnected manifestations of disrupted physiological adaptation during the transition from pregnancy to lactation. During this period, maternal tissues

Fetal Age Assessment From Primary Teeth and Long Bones.

This study evaluated traditional and expedited methods for assessing the age of fetal remains. Because of their rare occurrence, the discovery of fresh, decomposing, disfigured, or skeletal fetuses engenders heightened awareness by forensic pathologists primarily tasked with age estimation in relation to viability. With decomposed complete or isolated fetal remains, dentists focus on primary molar mineralization, whereas anthropologists perform long bone measurements along with discernment of other indicators of skeletal maturity to obtain an age estimation. The results of this study are 4-fold

Comparison of prostate cancer cell line growth in fetal bovine serum and animal free supplements.

Fetal bovine serum (FBS) is widely used as a growth supplement in cell culture due to its richness in embryonic growth-promoting factors, but presents ethical concerns, batch-to-batch variability, high cost, and biosafety issues. Alternative supplements are needed to support consistent and reliable cell growth in vitro. Human platelet lysate (hPL) and chemically defined supplements offer potential substitutes, but they require evaluation across different cell types. This study aimed to evaluate the effectiveness of two commercial serum supplements, PLTGold (an hPL product) and FastGro (a chemi
