In vivo bone formation in silk fibroin and chitosan blend scaffolds via ectopically grafted periosteum as a cell source: a pilot study.

Reconstruction of a critical size bone defect in the head and neck after trauma or tumor resection remains challenging. While certain defects, such as isolated orbital floor fractures, may be reconstructed with alloplastic biomaterials, larger defects or those involving load bearing bones usually require autologous tissue reconstruction. Vascularized bone free flaps remain the gold standard for large bone defects of the head and neck. These are generally lengthy, complicated, multi-step procedures that require subspecialty expertise to assure optimal outcomes.1 Invariably any procedure where a

Preclinical model of Echinacea-functionalized silk fibroin-chitosan transparent membranes with enhanced hair follicle regeneration and antibacterial properties for wound healing.

INTRODUCTION: Skin serves as a crucial barrier that separates the human body from the external environment. Its complex structure can make it susceptible to microbial infection and bleeding, leading to poor wound healing. Therefore, it is of important clinical significance to develop a wound dressing that can form scarless tissues with hair follicles, inhibit bacterial infection, and promote hemostasis. METHODS: In this study, we constructed composite dressings based on Echinacea-loaded silk fibroin/chitosan (ESC) that had excellent mechanical properties and air permeability for the promotion

Synergistic effects of berberine and adipose stem cells in a silk fibroin-PEG hydrogel for diabetic wound healing via macrophage polarization modulation.

Diabetic wound repair remains a considerable clinical challenge, largely due to the limited efficacy of current therapies. Herein, we engineered a novel silk fibroin (SF)-polyethylene glycol (PEG) hydrogel (SPB) incorporated with berberine (BBR) and adipose-derived stem cells (ASCs) to synergistically promote diabetic wound healing. By optimizing the physicochemical properties of SPB, we found that a 7:3 SF:PEG ratio provided an ideal balance of pore size (25.83 ± 3.94 μm), mechanical strength (32.33 ± 4.07 kPa) and sustained BBR release. In vitro, SPB significantly enhanced ASC paracrine func

Silk Fibroin Peptides Promote Extracellular Matrix Homeostasis in Photoaging via Modulation of the ITGB1/FAK-TGF-β/Smad Signaling Axis.

Excessive ultraviolet A (UVA) irradiation disrupts extracellular matrix (ECM) homeostasis in skin photoaging by impairing the balance between synthesis and degradation, yet whether silk fibroin peptide (SF), a small bioactive peptide from Bombyx mori, can restore this balance through mechanotransduction pathways remains unknown. Herein, we demonstrate that SF dose-dependently rescues human dermal fibroblasts (HDFs) from UVA-induced oxidative stress, senescence, and ECM disintegration. Notably, SF not only suppresses reactive oxygen species (ROS) and restores activities of antioxidant enzymes,

Transdermal Delivery of Silk-AgNPs through Sucrose/Silk-AgNPs Antibacterial Fibroin Scaffold Packed in PEGDA Microneedles for Enhanced Wound Healing.

Microenvironmental imbalance and bacterial infection are core causes of intractable chronic wounds by hindering nutrient transport and tissue repair. Herein, we innovatively fabricated a hydrophilic antibacterial scaffold based on silk fibroin (SF) and uniformly integrated it into PEGDA microneedle (MN) matrices. The composite MN system can efficiently load hydrophilic model drugs including lidocaine and rhodamine B, achieving uniform drug incorporation and improved drug-loading versatility with controllable, long-term sustained drug release behavior. Biological tests verified that the develop

A printable blue-light photocurable gelatin methacryloyl/silk fibroin hydrogel loaded with salvianolic acid B for enhanced spinal cord repair.

Spinal cord injury (SCI) leads to severe neurological impairment, highlighting the critical need for effective regenerative treatments. While existing therapeutic strategies show limited capacity to promote axonal regeneration, we developed an innovative 3D-printable, blue-light photo-crosslinkable hydrogel system combining silk fibroin with gelatin methacryloyl (GelMA) and incorporating salvianolic acid B (Sal B). The engineered composite was fabricated via a denaturation-refolding protocol in which a denaturant unfolded silk β-sheets to promote rapid ion exchange and high-efficiency Sal B in

Bioorthogonal Click Chemistry Enables Stable Cell Attachment to Silk Fibroin Scaffolds for Tissue Engineering Applications: Proof of Concept in Neural Regeneration.

Tissue engineering seeks effective strategies to integrate cells into scaffolds while preserving spatial organization and stability. Conventional approaches based on extracellular matrix molecules or adhesion peptides often provide weak and nonspecific attachments, limiting robust assembly of complex cellular architectures. Here, we investigate biorthogonal click chemistry to address these limitations. By enabling covalent cell-biomaterial bonding through strain-promoted azide-alkyne cycloaddition (SPAAC), we achieved stable cell-scaffold integration. As a proof of concept, we engineered artif

Engineered Exosome-Loaded Silk Fibroin Composite Hydrogels Promote Tissue Repair in Spinal Cord Injury Via Immune Checkpoint Blockade.

Spinal cord injury (SCI) is a severe central nervous system disorder characterized by a high rate of disability and limited axonal regeneration. Excessive post-injury inflammation often leads to further neuronal damage. Immune checkpoint (IC) genes, which regulate immune cell activity, play a critical role in modulating post-injury inflammation and thus influence neural repair and functional recovery. In this study, analysis of the GEO database reveals that the IC gene T cell immunoglobulin and mucin domain-containing protein 3 (Tim3) is highly expressed in microglia following SCI, contributin

Peripheral nerve regeneration using a bioresorbable silk fibroin-based artificial nerve conduit fabricated via a novel freeze-thaw process.

While silk fibroin (SF) obtained from silkworm cocoons is expected to become a next-generation natural polymer, a fabrication method for SF-based artificial nerve conduits (SFCs) has not yet been established. Here, we report a bioresorbable SFC, fabricated using a novel freeze-thaw process, which ensures biosafety by avoiding any harmful chemical additives. The SFC demonstrated favorable biocompatibility (high hydrophilicity and porosity with a water content of > 90%), structural stability (stiffness, toughness, and elasticity), and biodegradability, making it an ideal candidate for nerve rege

Construction of multifunctional tracheal substitute based on silk fibroin methacryloyl and hyaluronic acid methacryloyl with decellularized cartilaginous matrix for tracheal defect repair.

The regeneration and functional recovery of tracheal tissue are of paramount importance in the research of tissue-engineered trachea. Current constructs still face some limitations in simulating the complex natural microenvironment and achieving better regenerative capacity and functional recovery. To address these challenges, the application of hydrogels with three-dimensional (3D) network structure and extracellular matrix derived from decellularized tissues and cells has become a more promising strategy. This study aims to introduce a novel bilayer multifunctional tissue-engineered tracheal

Machine learning-assisted high-performance immunoSERS platform using silk fibroin as a natural etching mask for early diagnosis of Alzheimer's disease.

Early and accurate diagnosis of Alzheimer's disease (AD) is a major stride toward pharmacological interventions to delay the onset or progression of the disease in patients with mild symptoms. In this study, we developed a silk fibroin-templated surface-enhanced Raman spectroscopy (SERS)-activated double-sandwich immunoassay (immunoSERS) platform that enhances plasmonic hotspot formation for the ultrasensitive detection of biomarkers. Silk fibroin, acting as a natural etching mask, facilitates the direct fabrication of Au nanocavity (AuNC) substrates and enables the immunoSERS platform to achi

Stabilization of CXCL12 (SDF-1α) via silk fibroin films enhances stem cell migration/retention and functional recovery after stroke.

Brain pathologies such as ischemic stroke or traumatic brain injury (TBI) are among the most impactful diseases worldwide. In ischemic stroke, we currently lack truly effective treatments capable of delaying infarct progression, limiting lesion size or stimulating endogenous brain repair mechanisms to promote neurovascular remodeling and functional recovery. Two main barriers continue to limit the clinical translation of therapeutic molecules: the highly restrictive nature of the blood-brain barrier and that many bioactive molecules exhibit low stability at the target site, with half-lives sho

Anti-Inflammatory Peptide-Conjugated Silk Fibroin/Cryogel Hybrid Dual Fiber Scaffold with Hierarchical Structure Promotes Healing of Chronic Wounds.

Chronic wounds resulting from diabetes, pressure, radiation therapy, and other factors continue to pose significant challenges in wound healing. To address this, this study introduces a novel hybrid fibroin fibrous scaffold (FFS) comprising randomly arranged fibroin fibers and vertically aligned cryogel fibers (CFs). The fibroin scaffold is efficiently degummed at room temperature and simultaneously formed a porous structure. The aligned CFs are produced via directional freeze-drying, achieved by controlling solution concentration and freezing polymerization temperature. The incorporation of a

Slightly photo-crosslinked chitosan/silk fibroin hydrogel adhesives with hemostasis and anti-inflammation for pro-healing cyclophosphamide-induced hemorrhagic cystitis.

Cyclophosphamide is commonly used in the treatment of various cancers and autoimmune diseases, while concurrently imposing substantial toxicity on the bladder, frequently manifesting hemorrhagic cystitis. Intravesical interventions, such as hyaluronic acid supplementation, present a therapeutic strategy to reinstate bladder barrier function and alleviate the effects of metabolic toxicants. However, it remains a great challenge to achieve efficient cyclophosphamide-induced hemorrhagic cystitis (CHC) management with accelerated tissue repair owing to the low wet-adhesion, poor hemostasis, and ac

A 3D-printed acinar-mimetic silk fibroin-collagen-astragalus polysaccharide scaffold for tissue reconstruction and functional repair of damaged parotid glands.

Salivary glands are the principal organs responsible for secreting saliva in the oral cavity. Tumors, trauma, inflammation, and other factors can cause functional or structural damage to the glands, leading to reduced saliva secretion. In this study, we innovatively prepared a acinar-mimetic silk fibroin-collagen-astragalus polysaccharide (SCA) scaffold using low-temperature three-dimensional (3D) printing and freeze-drying techniques. We evaluated the material properties and cell compatibility of the scaffold in vitro and implanted it into the damaged parotid glands (PG) of rats to assess its

Silk Fibroin-Based Bioengineered Scaffold for Enabling Hemostasis and Skin Regeneration of Critical-Size Full-Thickness Heat-Induced Burn Wounds.

Millions of people around the globe are affected by full-thickness skin injuries. A delay in the healing of such injuries can lead to the formation of chronic wounds, posing several clinical and economic challenges. Current strategies for wound care aim for skin regeneration and not merely skin repair or faster wound closure. The present study aimed to develop a bioactive wound-healing matrix comprising natural biomaterial silk fibroin (SF), clinical-grade human fibrin (FIB), and human hyaluronic acid (HA), resulting in SFFIBHA for regeneration of full-thickness burn wounds. A porous, hemostat

A New Preparation Method for Anisotropic Silk Fibroin Nerve Guidance Conduits and Its Evaluation In Vitro and in a Rat Sciatic Nerve Defect Model.

Over the past decade, silk fibroin (SF) has been emergently used in peripheral nerve tissue engineering. Current approaches aiming at producing SF-based nerve guidance conduits (SF-NGCs) used dissolved silk based on either aqueous solutions or organic solvents. In this study, we describe a novel procedure to produce SF-NGCs: A braided tubular structure of raw Bombyx mori silk is subsequently processed with the ternary solvent CaCl2/H2O/ethanol, formic acid, and methanol to improve its mechanical and topographical characteristics. Topographically, the combination of the treatments results in a

A polycaprolactone/silk-fibroin nanofibrous composite combined with human umbilical cord serum for subacute tympanic membrane perforation; an in vitro and in vivo study.

Due to its good mechanical reinforcement and large number of basic amino acids, silk fibroin (SF) has been widely used in tissue regenerative applications as a bioactive dispersed phase of a biocomposite system. In this work, we fabricated an electrospun polycaprolactone (PCL)/SF composite and combined the composite scaffold with human umbilical cord serum (UCS) to facilitate cell growth. Various in vitro cellular activities (cell-viability, MTT assay, and fluorescence analysis) for the electrospun pure PCL, pure SF, PCL/SF, and PCL/SF/UCS scaffolds were compared. The PCL/SF/UCS based scaffold

Silk Fibroin Coated Lactobacillus Plantarum Lac16 Ameliorates Alcoholic Liver Disease by Modulating Bile Acid Metabolism and Gut-Liver Axis.

Alcoholic liver disease (ALD) is a major global health burden with limited therapeutic methods. Emerging evidence highlights the gut-liver axis as a critical role of ALD pathogenesis, involving gut microbiota dysbiosis, intestinal barrier dysfunction, and disrupted bile acid metabolism. Probiotics, particularly Lactobacillus plantarum, have shown promise in mitigating ALD, but their efficacy is limited by poor gastrointestinal survival. Here, we developed silk fibroin coated Lactobacillus plantarum Lac16 (SLac16) to enhance probiotic viability and evaluated its protective effects in a murine A

Silk Fibroin-Modified Metal Organic Framework Acid-Controlled Release Nanocarrier for Polyetheretherketone to Combat Implant-Associated Infections and Promote Angiogenesis.

Polyetheretherketone (PEEK) has been regarded as a prospective alternative orthopedic implant, because of its elastic modulus approximately equal to human cortical bone. However, the poor antibacterial and angiogenic properties of bioinert PEEK have become a limitation to its clinical application. Therefore, a biomimetic metal organic framework acid-controlled release nanocarrier consisting of Zeolitic Imidazolate Framework-8 (ZIF-8) nanoparticles loaded with gentamicin and modified with silk fibroin nanospheres for PEEK implants is prepared. In this study, acidity-responsive PEEK implant can
