How a Nutritional Deficiency Became Treated with Fluoride.

Ignoring evidence on causes of disease such as smoking can harm public health. This report explores how public health experts started to ignore evidence that pediatric vitamin D deficiencies are associated with dental caries. Historical analyses show that an organization of clinical specialists, the American Dental Association (ADA), initiated this view. The ADA was a world-leading organization and its governing bodies worked through political channels to make fluoride a global standard of care for a disease which at the time was viewed as an indicator of vitamin D deficiencies. The ADA scient

Effects of fluoridated milk on root dentin remineralization.

BACKGROUND: The prevalence of root caries is increasing with greater life expectancy and number of retained teeth. Therefore, new preventive strategies should be developed to reduce the prevalence of root caries. The aim of this study was to investigate the effects of fluoridated milk on the remineralization of root dentin and to compare these effects to those of sodium fluoride (NaF) application without milk. METHODS: Thirty extracted human molars were divided into 6 groups, and the root cementum was removed from each tooth. The dentin surface was demineralized and then incubated with one of

The impact of fluoride on ameloblasts and the mechanisms of enamel fluorosis.

Intake of excess amounts of fluoride during tooth development cause enamel fluorosis, a developmental disturbance that makes enamel more porous. In mild fluorosis, there are white opaque striations across the enamel surface, whereas in more severe cases, the porous regions increase in size, with enamel pitting, and secondary discoloration of the enamel surface. The effects of fluoride on enamel formation suggest that fluoride affects the enamel-forming cells, the ameloblasts. Studies investigating the effects of fluoride on ameloblasts and the mechanisms of fluorosis are based on in vitro cult

Review of fluoride removal from drinking water.

Fluoride in drinking water has a profound effect on teeth and bones. Up to a small level (1-1.5mg/L) this strengthens the enamel. Concentrations in the range of 1.5-4 mg/L result in dental fluorosis whereas with prolonged exposure at still higher fluoride concentrations (4-10mg/L) dental fluorosis progresses to skeletal fluorosis. High fluoride concentrations in groundwater, up to more than 30 mg/L, occur widely, in many parts of the world. This review article is aimed at providing precise information on efforts made by various researchers in the field of fluoride removal for drinking water. T

A health risk assessment for fluoride in Central Europe.

Like many elements, fluorine (which generally occurs in nature as fluoride) is beneficial to human health in trace amounts, but can be toxic in excess. The links between low intakes of fluoride and dental protection are well known; however, fluoride is a powerful calcium-seeking element and can interfere with the calcified structure of bones and teeth in the human body at higher concentrations causing dental or skeletal fluorosis. One of the main exposure routes is via drinking water and the World Health Organisation currently sets water quality guidelines for the element. In Central Europe, g

Assessment of teeth as biomarkers for skeletal fluoride exposure.

Skeletal fluorosis and dental fluorosis are diseases related to fluoride (F) ingestion. Bone is the largest storage site of F in our body. T … mice of three different strains (SWR/J, A/J and 129P3/J) were exposed to different levels of water fluoridation (0, 25, 50 and 100 ppm). Bone (femora and vertebrae) and teeth from these mice were then analyzed for F concentration using Instrumental Neutron Activation Analysis (INAA). In part 2 of the study, human teeth (enamel and dentin) and bone from 30 stu …

Analogues of ethylenediaminetetraacetic acid and sodium fluoride as anticoagulants.

Three analogues of ethylenediaminetetraacetic acid (EDTA), ethyleneglycoltetraacetic acid (EGTA), ethylendiaminedipropionic acid (EDDP), and hexamethylenediaminetetraacetic acid (HDTA), and sodium fluoride (NaF) were evaluated as anticoagulants used for rapid and multi-parametric analysis with minimal amounts of blood. EDDP and HDTA induced rapid blood coagulation (< 30 min), while NaF was not useful for coagulation tests because no coagulation was observed by addition of PT- or APTT-reagents. EGTA could be used for hematology and coagulation tests, although platelet count was significantly de

Mechanism and timing of fluoride effects on developing enamel.

Fluoride appears to specifically interact with mineralizing tissues, causing an alteration of the mineralization process. In enamel, fluorosis results in a subsurface hypomineralization. This hypomineralized enamel appears to be directly related to a delay in the removal of amelogenins at the early-maturation stage of enamel formation. The specific cause for this delay is not known, although existing evidence points to reduced proteolytic activity of proteinases that hydrolyze amelogenin. This delay in hydrolysis of amelogenins could be due to a direct effect of fluoride on proteinase secretio

Fluoride use in periodontal therapy: a review of the literature.

Although the relationship between fluoride and dental caries has been widely studied and definitively determined, the relationship between fluoride and periodontal health and disease is not clear. Conflicting studies in the literature either suggest routine fluoride usage as an adjunct to conventional periodontal therapy or argue against topical fluoride use on periodontally involved teeth. This review summarizes the literature that addresses the utility of fluoride in patients with periodontal disease and aims to advance a rational criterion for the use of fluorides in the management of perio

The effect of topical fluoride agents on saliva secretion.

The aim of this study was to assess the possible influence of various topical fluoride agents on the rate of saliva secretion. Fifteen healthy volunteers, both males and females, aged 20-60 years, participated in this study. Aminfluorides and monofluorophosphate products in gel, toothpaste and rinse states were used. Each subject brushed or rinsed the teeth and mouth for 2 minutes. Secreted saliva was collected for 5 minutes. Salivary secretion was significantly greater after toothbrushing with aminfluoride compounds as compared to toothbrushing with monofluorophosphate. There was a tendency f

Animal caries models for evaluating fluoride dentifrices.

To determine the ability of animal caries models to evaluate the cariostatic potential of fluoride dentifrices, we examined four different coronal caries models. These currently used rat caries models were: Francis' hypomineralized area model, Gaffar's CARA rat model, the Connecticut model, and the Indiana model. Available data indicated that all of these models were capable of detecting significant cariostatic benefits from clinically proven fluoride dentifrices. For the most part, these animal models were also able to demonstrate a response to increasing concentrations of fluoride, particula

Cancer incidence and mortality in workers exposed to fluoride.

BACKGROUND: Although a recent bioassay showed increased frequency of bone cancer in rats with high oral intake of fluoride, the data are reported as equivocal evidence of carcinogenicity. In humans, occupational fluoride exposure may cause skeletal fluorosis, and our earlier follow-up of fluoride-exposed workers showed increased incidence of respiratory cancers. PURPOSE: To further evaluate occupational fluoride exposure as a carcinogenic risk factor, we extended by approximately one decade the follow-up of a cohort of 425 men and 97 women employed for at least 6 months in the period 1924-196

In vitro studies on the effects of fluoride on de- and remineralization.

The recent literature extensively describes the role of ambient fluoride in the de- and remineralization of dental enamel. Fluoride in sub-ppm concentrations is effective in promoting mineral deposition and inhibiting mineral dissolution. The latter phenomenon is most likely attributable to the concomitant precipitation of a fluoride-rich mineral phase which inhibits further dissolution. These fundamental processes result in an inhibition of enamel demineralization and an enhancement of enamel lesion remineralization. Alternatively, fluoride may also induce the 'arrestment' of enamel lesions.

Fluoride-induced fractures: relation to osteogenic effect.

The possible effects of fluoride in inducing fractures were studied in 61 patients treated with sodium fluoride (NaF), 40-60 mg daily in combination with calcium and vitamin D. Nine patients developed the fluoride-(F) related lower extremity pain syndrome. Four other patients had stress fractures associated with trauma. Seven of the 61 patients had 10 upper femur fractures of which 5 were stress fractures. The bone mineral mass of the central skeleton including the hips was measured by neutron activation and the results expressed as a calcium bone index (CaBI) which normalizes the results to t

[Fluoride level in human nails and its significance].

Bone tissue investigation provides the best indicator for fluorine saturation of the organism. However, the intravital bone biopsy--as an invasive method, is not applicable in prophylactic studies. Thus, attention was focussed on the nails that constitute an easily accessible material being able to reflect the chronic accumulation of fluorine in the organism. In the actual paper it has been endeavoured to find out the routes, on which fluorine is deposited in the nails, if there is any dependence between the fluorine content in the nails and the age of the studied subjects, and whether the con

Fluoride, aluminum, and phosphate kinetics in cryolite workers.

Exposure to cryolite (Na3AlF6) dust may result in skeletal fluorosis. Eight male workers at a cryolite concentrator participated in a 4-day study after 5 days of vacation. Dust exposures were 0.16 to 21.2 mg/m3. Urine was collected before work began and during two 4-hour periods. Preshift urine fluoride concentrations increased during the week. Fluoride concentrations in postshift urine and serum both correlated with the dust exposures. Serum fluoride concentrations decreased with a half-life of 3.3 to 6.9 hours after work. Fluoride clearance was 40.5 to 76.5 mL/min at urinary flow rates of 0.
