Developmental effects of fumonisin B1-containing Fusarium moniliforme culture extract in CD1 mice.

Pregnant Charles River CD1 mice were treated with a semipurified extract of Fusarium moniliforme culture containing 0, 12.5, 25, 50 or 100 mg FB1/kg each day orally (diluted in distilled water) between gestational days (GD) 7 and 15 to evaluate the developmental toxicity of FB1. Following sacrifice of dams on GD 18, litters were examined for gross abnormalities and divided equally for skeletal or visceral examination by routine techniques. Significant maternal mortality was observed at doses of 50 and 100 mg FB1/kg. Dose-dependent decreases in maternal body weight gains, number of live offspri…

Risk and Molecular Mechanism of Metconazole Resistance in the Rice Bakanae Pathogen Fusarium fujikuroi.

Rice bakanae disease, caused by Fusarium fujikuroi, is a destructive seed-borne disease. Metconazole has excellent control potential, but its resistance risk and mechanism remain unreported. This study confirmed metconazole has strong antifungal activity, with EC50 values ranging from 0.0073 to 0.0468 μg/mL. After fungicide taming, we obtained seven metconazole-resistant mutants with low-level, genetically stable resistance. Biological fitness showed mycelial growth and virulence did not decrease significantly, but sporulation increased significantly. Cross-resistance found the mutants had cro…

A banana susceptibility gene underlying fusarium wilt: validation as a target for disease resistance.

BACKGROUND: Fusarium wilt, caused by the soil-borne fungus Fusarium oxysporum f. sp. cubense, remains the principal constraint on banana production, particularly for the widely cultivated cultivar 'Prata-Ana' (AAB) in Brazil. Given the limited efficacy of conventional control strategies, susceptibility (S) genes have emerged as promising targets for developing resistant cultivars. This study investigated the expression of the banana DMR6 gene during the interaction with Foc. DMR6 was selected because it is a conserved plant susceptibility gene that negatively regulates salicylic acid-mediated …

Establishment of an Isolation System for Extracellular Vesicles of Fusarium oxysporum and Its Proteomic Analysis.

Extracellular vesicles (EVs) secreted by Fusarium oxysporum play an important role in the process of its infestation of the host, but the in vitro research system for EVs of F. oxysporum (Fo-EVs) has not yet been improved, and the mechanism of its action remains unclear. In this study, particle size distribution, particle concentration, number of particles per unit of protein, number of particles per unit of mycelial biomass, and concentration of contaminated proteins were used as indicators to evaluate the yield and purity of Fo-EVs. The optimal method for Fo-EV preparation and extraction was…

Synergism between nisin and citronellal against Fusarium graminearum and their application in maize preservation.

Fusarium graminearum (F. graminearum), responsible for Fusarium head blight in cereals, causes significant yield losses and produces mycotoxins contaminating food and feed. The combined effects of nisin and citronellal against F. graminearum were investigated in this study. The combination of nisin and citronellal not only exhibited a synergistic effect against F. graminearum spores (FICI <0.3125), but also significantly inhibited mycelial growth, with inhibition rates of (76.93 ± 6.74) % and (95.24 ± 4.33) % for mycelial diameter and weight, respectively. The nisin/citronellal combination mai…

Screening for Susceptibility of Macadamia to Euwallacea fornicatus and its Fungal Symbiont Fusarium euwallaceae.

The polyphagous shothole borer (Euwallacea fornicatus, PSHB), an ambrosia beetle, with its fungal symbiont, Fusarium euwallaceae, is responsible for Fusarium dieback (FD) in a wide range of woody hosts. In 2019, the first suspected case of E. fornicatus was reported in macadamia in South Africa. The aims of this study were to confirm the E. fornicatus report and thereafter to assess the susceptibility of commercially planted macadamia cultivars to FD caused by F. euwallaceae. The identities of the beetle and associated fungal symbionts were confirmed by means of DNA sequence analysis of the 28…

Fusarium-Induced Cellulitis in an Immunocompetent Patient With Sickle Cell Disease: A Case Report.

Fungal infections due to Fusarium species are mostly present in immunocompromised and patients with poorly controlled diabetes mellitus. We report a case of lower extremity skin infection caused by Fusarium species in a 61-year-old woman diagnosed with sickle cell disease. Single skin ulceration caused by Fusarium species can result from fungal inoculation into damaged tissue, so any condition that damages the skin can be considered as a risk factor for inoculation. Long-standing sickle cell disease may develop vaso-occlusion in the skin that can produce lower extremity ulcers and myofascial s…

Chlorine gas is an effective alternative to sterilize carnation leaves for Fusarium spp. identification.

Sterile carnation leaves are required for proper morphological identification of Fusarium spp. but the gamma irradiation equipment required for leaf sterilization is not available to everyone. This study evaluated three different methods for sterilizing carnation leaves: microwave radiation, ultraviolet, and chlorine gas (CG) sterilization. Both microwave and ultraviolet treatments did not sufficiently sterilize leaf tissue, however, exposure to CG for 2 h resulted in no growth of either fungi or bacteria. Exposure times of carnation leaves to CG were also evaluated for spore production, spore…

Identification and Characterization of Fusarium proliferatum, a New Species of Fungi that Cause Fungal Keratitis.

Fusarium proliferatum (F. proliferatum) is known as a pathogen of corn and other crops, but its role in fungal keratitis has not been well investigated. Among 877 Fusarium isolates, we identified 155 (17.7%) stains as F. proliferatum according to their morphological features and partial DNA sequencing of translation elongation factor-[Formula: see text] (EF-[Formula: see text]) in this study. In vitro antifungal susceptibility tests showed that the F. proliferatum strains were sensitive to natamycin and vorionazole but resistant to amphotericin B, fluconazol, ketoconazole and itaconazole. Most…

Modified Fusarium Mycotoxins in Cereals and Their Products-Metabolism, Occurrence, and Toxicity: An Updated Review.

Mycotoxins are secondary fungal metabolites, toxic to humans, animals and plants. Under the influence of various factors, mycotoxins may undergo modifications of their chemical structure. One of the methods of mycotoxin modification is a transformation occurring in plant cells or under the influence of fungal enzymes. This paper reviews the current knowledge on the natural occurrence of the most important trichothecenes and zearalenone in cereals/cereal products, their metabolism, and the potential toxicity of the metabolites. Only very limited data are available for the majority of the identi

A tomatinase-like enzyme acts as a virulence factor in the wheat pathogen Fusarium graminearum.

During their interactions with plants, fungal pathogens employ large numbers of pathogenesis-associated molecules including secreted effectors and enzymes that can degrade various defence compounds. However, in many cases, in planta targets of pathogen-produced enzymes remain unknown. We identified a gene in the wheat pathogen Fusarium graminearum, encoding a putative enzyme that shows 84% sequence identity to FoTom1, a tomatinase produced by the tomato pathogen Fusarium oxysporum f. sp. lycopersici. In F. oxysporum f. sp. lycopersici, FoTom1 is a virulence factor involved in the degradation o…

Effects of Selenium on Fusarium Growth and Associated Fermentation Products and the Relationship with Chondrocyte Viability.

This study determined the effects of selenium on the growth of Fusarium strains and the effects of products extracted from the fungal cultures on relevant indicators of chondrocytes injury. The results showed that selenium supplementation resulted in differential effects on the mycelial growth of the strains. Levels of the chondrocyte injury indicators, including cell viability, proteoglycan and type II collagen contents and their mRNA expressions, were all reduced to varying degrees when the chondrocytes were incubated with fermentation extracts, the inhibitory effect varied depending on sele…

Genotyping and In Vitro Antifungal Susceptibility Testing of Fusarium Isolates from Onychomycosis in India.

Onychomycosis refers to fungal infection of the nail and is commonly caused by dermatophytes, while yeasts and non-dermatophytic molds (NDM) are increasingly recognized as pathogens in nail infections. The present study was done to delineate molecular epidemiology of Fusarium onychomycosis in India. Five hundred nail samples of Indian patients clinically suspected of onychomycosis were subjected to direct microscopy and fungal culture. Representative Fusarium isolates were further identified to species level by multi-locus sequencing for internal transcribed spacer, translation elongation fact…

Identification of ABC transporter genes of Fusarium graminearum with roles in azole tolerance and/or virulence.

Fusarium graminearum is a plant pathogen infecting several important cereals, resulting in substantial yield losses and mycotoxin contamination of the grain. Triazole fungicides are used to control diseases caused by this fungus on a worldwide scale. Our previous microarray study indicated that 15 ABC transporter genes were transcriptionally upregulated in response to tebuconazole treatment. Here, we deleted four ABC transporter genes in two genetic backgrounds of F. graminearum representing the DON (deoxynivalenol) and the NIV (nivalenol) trichothecene chemotypes. Deletion of FgABC3 and FgABC…
