The critical role of miRNA in bacterial zoonosis.

The public's health and the financial sustainability of international societies remain threatened by bacterial zoonoses, with limited reliable diagnostic and therapeutic options available for bacterial diseases. Bacterial infections influence mammalian miRNA expression in host-pathogen interactions. In order to counteract bacterial infections, miRNAs participate in gene-specific expression and play important regulatory roles that rely on translational inhibition and target gene degradation by binding to the 3' non-coding region of target genes. Intriguingly, according to current studies, that

Dissecting the essential role of luxS-relevant small RNAs in biofilm formation of Streptococcus mutans.

Streptococcus mutans is considered the key contributor to human dental caries. The LuxS/AI-2 quorum‑sensing (QS) system in S. mutans plays a crucial role in the development of cariogenic oral biofilms. Many QS systems rely on regulation carried out by small RNAs (sRNAs) to achieve optimal performance. However, the identities and functions of sRNAs in S. mutans QS system are not well elucidated. Here, we identify two novel sRNAs named SmqsR2 and SmqsR4 (Streptococcus mutans quorum-sensing-related small RNA 2 and 4), which respond to luxS expression and support biofilm formation by mediating exo

Transcriptional regulation drives heteroresistance in Pseudomonas aeruginosa: evidence from genomic and targeted gene expression analysis.

Bacterial heteroresistance, characterized by the presence of antibiotic-resistant subpopulations within phenotypically susceptible bacterial colonies, poses significant challenges to clinical antimicrobial susceptibility testing and treatment outcomes. While heteroresistance has been increasingly recognized across diverse bacterial species, the underlying mechanisms in Pseudomonas aeruginosa remain incompletely understood, particularly regarding carbapenem and monobactam resistance. In this study, we employed a multi-omics approach that integrates phenotypic screening, whole-genome sequencing,

Food-grade expression of Helicobacter pylori ureB subunit in Lactococcus lactis and its immunoreactivity.

Helicobacter pylori is the principal cause of chronic active gastritis, peptic ulcer, and gastric cancer. To develop an oral vaccine against H. pylori infection, we had expressed the H. pylori ureB gene (Genbank accession no. FJ436980) in nisin-controlled expression vectors using Lactococcus lactis NZ3900 as host. The ureB gene was amplified by PCR from a H.pylori strain MEL-Hp27. Then the ureB gene was fused translationally downstream of the nisin-inducible promoter nisA in a L. lactis plasmid pNZ8149. Lactose utilization based on the complementation of the lacF gene was used as a dominant se

Inactivation of the GacA response regulator in Pseudomonas fluorescens Pf-5 has far-reaching transcriptomic consequences.

The GacS/GacA signal transduction system is a central regulator in Pseudomonas spp., including the biological control strain P. fluorescens Pf-5, in which GacS/GacA controls the production of secondary metabolites and exoenzymes that suppress plant pathogens. A whole genome oligonucleotide microarray was developed for Pf-5 and used to assess the global transcriptomic consequences of a gacA mutation in P. fluorescens Pf-5. In cultures at the transition from exponential to stationary growth phase, GacA significantly influenced transcript levels of 635 genes, representing more than 10% of the 614

Global gene expression profile of Yersinia pestis induced by streptomycin.

Plague, caused by Yersinia pestis, is one of the most dangerous diseases that impressed a horror onto human consciousness that persists to this day. Cases of plague can be normally controlled by timely antibiotic administration. Streptomycin is the first-line antibiotic for plague treatment. In this study, a DNA microarray was used to investigate the changes in the gene expression profile of Y. pestis upon exposure to streptomycin. A total of 345 genes were identified to be differentially regulated, 144 of which were up-regulated, and 201 down-regulated. Streptomycin-induced transcriptional ch

RNA-Seq-based monitoring of infection-linked changes in Vibrio cholerae gene expression.

Pathogens adapt to the host environment by altering their patterns of gene expression. Microarray-based and genetic techniques used to characterize bacterial gene expression during infection are limited in their ability to comprehensively and simultaneously monitor genome-wide transcription. We used massively parallel cDNA sequencing (RNA-seq) techniques to quantitatively catalog the transcriptome of the cholera pathogen, Vibrio cholerae, derived from two animal models of infection. Transcripts elevated in infected rabbits and mice relative to laboratory media derive from the major known V. ch

Genomic traits, transcriptional responses, and metabolic activities determine functional divergence in mineral dissolution between Priestia megaterium PMA2-2 and Priestia flexa PMD1-1.

Priestia strains are gram-positive bacteria and globally distributed in mineral-rich environments. However, the functional divergence in mineral dissolution and its molecular drivers among Priestia strains remains poorly understood. Here, we employ comparative phenotypic and multi-omics analyses to decipher the molecular mechanisms for divergent mineral dissolution phenotypes between the efficient mineral-dissolving Priestia megaterium PMA2-2 and the inefficient mineral-dissolving Priestia flexa PMD1-1. Compared to PMD1-1, PMA2-2 exhibited significantly enhanced biotite dissolution, medium aci

Exploring the Protein Landscape of Dormant Mycobacterium tuberculosis Through In Vitro Functional Studies.

The investigation of functional proteins during dormancy in Mycobacterium tuberculosis (Mtb) through in vitro experiments provides crucial insights into the bacterium's survival mechanisms and pathogenicity. Dormancy constitutes a pivotal phase in the Mtb life cycle, enabling the pathogen to persist within hostile host environments, evade immune defenses, and establish latent tuberculosis infections. Although dormancy is often conceptually divided into early, mid, and late stages, these phases frequently overlap, representing a continuum of physiological and molecular adaptations rather than d

The MutRS quorum-sensing system controls lantibiotic mutacin production in the human pathogen Streptococcus mutans.

Microbes use quorum-sensing systems to respond to ecological and environmental changes. In the oral microbiome, the pathogenic bacterium Streptococcus mutans uses quorum-sensing to control the production of bacteriocins. These antimicrobial peptides kill off ecological competitors and allow S. mutans to dominate the microenvironment of dental plaques and form dental caries. One class of bacteriocins produced by S. mutans, the lantibiotic mutacins, are particularly effective at killing due to their broad spectrum of activity. Despite years of study, the regulatory mechanisms governing productio

Hacking heterocysts: advances in the genetic regulation of heterocyst differentiation.

Heterocyst differentiation in certain filamentous cyanobacteria is a multifaceted process essential for nitrogen fixation, orchestrated by a sophisticated regulatory network that encompasses several key stages. These include induction, pattern differentiation, commitment, extracellular layer formation, cell-cell communication, and ultimately, nitrogen fixation and metabolism. Key regulators like NtcA and HetR control heterocyst development, while proteins such as PatS, HetN, and PatA modulate pattern formation. Certain non-coding RNAs, such as NsiR1, Yfr1, and NsiR4, also regulate gene express

Proteomic Profiling Reveals RisR as a Novel Regulatory Element in the Virulence Network of Bordetella pertussis.

Bordetella pertussis, the causative agent of whooping cough, regulates its virulence through the BvgAS two-component system, which controls the transition between virulent (Bvg+) and avirulent (Bvg-) phases. In the virulent phase, virulence-activated genes (vags) are expressed, while in the avirulent phase, these are repressed. Virulence-repressed genes (vrgs) are induced under the control of the response regulator RisA. Phosphorylation of RisA by the nonoperonic kinase RisK is essential for vrgs expression. Adjacent to risK lies risR, a gene encoding a putative response regulator, whose funct

Revisiting old friends: updates on the role of two-component signaling systems in Listeria monocytogenes survival and pathogenesis.

Listeria monocytogenes is well recognized for both its broad resistance to stress conditions and its ability to transition from a soil bacterium to an intracellular pathogen of mammalian hosts. The bacterium's impressive ability to adapt to changing environments and conditions requires the rapid sensing of environmental cues and the coordinated response of gene products that enable bacterial growth and survival. Two-component signaling systems (TCSs) have been long recognized for their ability to detect environmental stimuli and transmit those signals into transcriptional responses; however, o

Unveiling the regulatory mechanisms of salicylate degradation gene cluster cehGHIR4 in Rhizobium sp. strain X9.

In a previous study, the novel gene cluster cehGHI was found to be involved in salicylate degradation through the CoA-mediated pathway in Rhizobium sp. strain X9 (Mol Microbiol 116:783-793, 2021). In this study, an IclR family transcriptional regulator CehR4 was identified. In contrast to other regulators involved in salicylate degradation, cehR4 forms one operon with the gentisyl-CoA thioesterase gene cehI, while cehG and cehH (encoding salicylyl-CoA ligase and salicylyl-CoA hydroxylase, respectively) form another operon. cehGH and cehIR4 are divergently transcribed, and their promoters overl

A Streptomyces tendae Specialized Metabolite Inhibits Quorum Sensing in Group A Streptococcus.

Quorum sensing (QS) is a means of bacterial communication accomplished by microbe-produced signals and sensory systems. QS systems regulate important population-wide behaviors in bacteria, including secondary metabolite production, swarming motility, and bioluminescence. The human pathogen Streptococcus pyogenes (group A Streptococcus [GAS]) utilizes Rgg-SHP QS systems to regulate biofilm formation, protease production, and activation of cryptic competence pathways. Given their reliance on small-molecule signals, QS systems are attractive targets for small-molecule modulators that would then a

BolA promotes the generation of multicellular behavior in S. Typhimurium by regulating the c-di-GMP pathway genes yeaJ and yhjH.

The generation of multicellular behavior enhances the stress adaptability, antibiotic resistance, and pathogenic potential of Salmonella enterica serovar Typhimurium (S. Typhimurium), which is challenging for its prevention and control. Therefore, determination of the mechanism of multicellular behavior development is urgently required. Accordingly, this study investigated BolA, a transcription factor that promotes bacterial survival under different stresses. We found that BolA promoted the generation of multicellular behavior. Furthermore, transcriptome analysis revealed that BolA affected th

The Global Regulator MftR Controls Virulence and Siderophore Production in Burkholderia thailandensis.

Burkholderia thailandensis is a member of the Burkholderia pseudomallei complex. It encodes the transcription factor MftR, which is conserved among the more pathogenic Burkholderia spp. and previously shown to be a global regulator of gene expression. We report here that a B. thailandensis strain in which the mftR gene is disrupted is more virulent in both Caenorhabditis elegans and onion. The ΔmftR strain exhibits a number of phenotypes associated with virulence. It is more proficient at forming biofilm, and the arcDABC gene cluster, which has been linked to anaerobic survival and fitness wit

Role of the fsr Quorum-Sensing System in Enterococcus faecalis Bloodstream Infection.

Enterococcus faecalis is an important intestinal colonizing bacteria and can cause various tissue infections, including invasive blood infection (BI). The annual incidence of E. faecalis BI has been estimated to be ~4.5 per 100,000, with a fatality rate that can reach 20%. However, whether bacterial colonization or invasive infections are tissue based has not been thoroughly studied. In this study, we analyzed 537 clinical isolates from 7 different tissues to identify the key genomic elements that facilitate the colonization and invasive infection of E. faecalis. Comparative genomic analysis r

GlnR-mediated regulation of KstR controls cholesterol catabolism in Mycobacterium smegmatis.

Tuberculosis, caused by mycobacteria, continues to pose a substantial public health threat. Mycobacteria typically use cholesterol from the membranes of host macrophages as a carbon and energy source. Most genes that control cholesterol degradation are regulated by KstR, which is highly conserved in Mycobacterium tuberculosis and Mycobacterium smegmatis. Through bioinformatic analysis, we found a typical global nitrogen regulator (GlnR)-binding motif (CCGAC-AACAGT-GACAC) in the promoter region of kstR of M. smegmatis, and we determined its binding activity in vitro using electrophoretic mobili
