Plasma lipidome mediates the causal effect of chronic hepatitis B on cirrhosis: A Mendelian randomization study.

This study aimed to investigate the role of the plasma lipid species in the progression from chronic hepatitis B (CHB) infection to cirrhosis. We conducted 2-sample Mendelian randomization (MR) and genome-wide mediation analyses to explore the causal relationship between CHB and cirrhosis, focusing on lipid species as potential mediators. The inverse variance weighted method was used as the primary analytical approach, supported by complementary estimators. Sensitivity analyses - including Cochran's Q test, MR-Egger intercept test, of Mendelian randomization pleiotropy residual sum and outlier

Genetic Evidence Supporting a Causal Role of Snoring in Keratoconus: A Bidirectional Mendelian Randomization Study.

PURPOSE: To clarify the controversial causal association between snoring and keratoconus (KCN), which is crucial in clinical prevention and treatment. METHODS: This is a 2-sample bidirectional mendelian randomization (MR) case-control study. MR is an innovative method that uses genetic variation as a natural experiment to investigate the causal relationships between potentially modifiable risk factors and health outcomes in observational data. The single nucleotide polymorphisms associated with snoring were retrieved from the UK biobank cohort with 218,346 participants (61,792 cases and 156,5

Causal relationship between schizophrenia and tachyarrhythmia: A Mendelian randomization study based on a European population.

Given the emerging evidence suggesting a link between schizophrenia (SCZ) and tachyarrhythmias, this study utilized a bidirectional Mendelian randomization (MR) approach to explore the causal relationship between SCZ and several common tachyarrhythmias, including tachycardia, atrial flutter (AFL), atrial fibrillation (AF), and supraventricular tachycardia (SVT). Stringent selection criteria were applied to identify instrumental variables, followed by 5 MR analyses grounded in different hypotheses to evaluate the causal relationship between SCZ and tachycardia, SVT, and "AFL and AF." Inverse va

H-ABC syndrome and DYT4: Variable expressivity or pleiotropy of TUBB4 mutations?

Recently, mutations in the TUBB4A gene have been found to underlie hypomyelination with atrophy of the basal ganglia and cerebellum (H-ABC) syndrome, a rare neurodegenerative disorder of infancy and childhood. TUBB4A mutations also have been described as causative of DYT4 ("hereditary whispering dysphonia"). However, in DYT4, brain imaging has been reported to be normal and, therefore, H-ABC syndrome and DYT4 have been construed to be different disorders, despite some phenotypic overlap. Hence, the question of whether these disorders reflect variable expressivity or pleiotropy of TUBB4A mutati

Genetically Predicted Gut Microbiota and Bladder Cancer Risk: A Mendelian Randomisation Analysis.

BACKGROUND: Gut microbiota (GM) has been increasingly implicated in cancer development through immune modulation, metabolic regulation, and systemic inflammatory pathways. Although observational studies have suggested a potential link between GM dysbiosis and bladder cancer (BC), these findings remain susceptible to confounding and reverse causation. To our knowledge, few studies have applied a Mendelian randomisation (MR) framework to systematically evaluate the gut-bladder axis from a genetic perspective. METHODS: We performed a two-sample MR analysis to examine associations between genetic

Concordance of genetic variation that increases risk for tourette syndrome and that influences its underlying neurocircuitry.

There have been considerable recent advances in understanding the genetic architecture of Tourette syndrome (TS) as well as its underlying n … al disinhibition and symmetry behaviour. SNP effect concordance analysis (SECA) was used to examine genetic pleiotropy-the same SNP affecting two traits-and concordance-the agreement in single nucelotide polymorphism (SNP) effect directions across these two traits. In addition, a conditional false discovery rate (FDR) analysis was performed, conditioning the TS risk …

Causal Relationship Between Chronic Obstructive Pulmonary Disease and Respiratory Tuberculosis Susceptibility: A Two-Sample Bayesian Weighted Mendelian Randomization Study.

The causal relationship between chronic obstructive pulmonary disease (COPD) and respiratory tuberculosis (TB) remains unclear due to limited evidence from prospective studies. Therefore, this study aimed to assess the causal relationship between COPD and respiratory TB using a two-sample Bayesian weighted Mendelian randomization (BWMR) study. The genetic instrumental variants (IVs) for COPD and respiratory TB were obtained from the IEU Open GWAS project in 2021. The inverse variance weighted (IVW) method was used as the main statistical analysis method, and the results supplemented with weigh

A Mendelian Randomization Study of the Association Between Polycystic Ovary Syndrome and Serum Urea Levels.

BACKGROUND: Polycystic Ovary Syndrome (PCOS) is a common endocrine and metabolic disorder affecting the reproductive function of women of reproductive age. Its clinical features include irregular menstruation, obesity, insulin resistance, hyperandrogenism, and polycystic ovarian morphology. Elevated serum urea levels may be related to the pathogenesis of PCOS. However, existing observational studies have not been able to clarify the causal relationship between serum urea levels and PCOS. METHODS: This study employed a two-sample Mendelian randomization (MR) approach to evaluate the genetic ca

Causal analysis of gastroesophageal reflux disease and esophageal cancer.

Patients with gastroesophageal reflux disease (GERD) are more likely to develop esophageal cancer (EC). However, a causal relationship betwe … nsitivity analysis also demonstrated similar results with the causal explanation, and major bias in genetic pleiotropy was not identified (intercept, 0.001; standard error, 0.001; P = .418). Causal link between thyroid function and schizophrenia: a two-sample Mendelian randomization study.

Schizophrenia is a chronic psychiatric disorder with inconsistent behavioral and cognitive abnormalities with profound effects on the individual and the society. Individuals with schizophrenia have altered thyroid function, but results from observational studies are conflicting. To date, it remains unclear whether and in which direction there is a causal relationship between thyroid function and schizophrenia. To investigate causal paths, a bidirectional two-sample Mendelian randomization (MR) study was conducted using summary statistics from genome-wide association studies including up to 330

Abundant genetic overlap between blood lipids and immune-mediated diseases indicates shared molecular genetic mechanisms.

Epidemiological studies suggest a relationship between blood lipids and immune-mediated diseases, but the nature of these associations is not well understood. We used genome-wide association studies (GWAS) to investigate shared single nucleotide polymorphisms (SNPs) between blood lipids and immune-mediated diseases. We analyzed data from GWAS (n~200,000 individuals), applying new False Discovery Rate (FDR) methods, to investigate genetic overlap between blood lipid levels [triglycerides (TG), low density lipoproteins (LDL), high density lipoproteins (HDL)] and a selection of archetypal immune-

Mutant Huntingtin Drives Development of an Advantageous Brain Early in Life: Evidence in Support of Antagonistic Pleiotropy.

OBJECTIVE: Huntington's disease (HD) is a neurodegenerative disease caused by a triplet repeat expansion within the gene huntingtin (HTT). Antagonistic pleiotropy is a theory of aging that posits that some genes, facilitating individual fitness early in life through adaptive evolutionary changes, also augment detrimental aging-related processes. Antagonistic pleiotropy theory may explain a positive evolutionary pressure toward functionally advantageous brain development that is vulnerable to rapid degeneration. The current study investigated antagonistic pleiotropy in HD using a years-to-onset

A Similar Genetic Architecture Underlies the Convergent Evolution of the Selfing Syndrome in Capsella.

Whether, and to what extent, phenotypic evolution follows predictable genetic paths remains an important question in evolutionary biology. Convergent evolution of similar characters provides a unique opportunity to address this question. The transition to selfing and the associated changes in flower morphology are among the most prominent examples of repeated evolution in plants. In this study, we take advantage of the independent transitions to self-fertilization in the genus Capsella to compare the similarities between parallel modifications of floral traits and test for genetic and developm

Contraception ends the genetic maintenance of human same-sex sexual behavior.

Because human same-sex sexual behavior (SSB) is heritable and leads to fewer offspring, it is puzzling why SSB-associated alleles have not been selectively purged. Current evidence supports the antagonistic pleiotropy hypothesis that SSB-associated alleles benefit individuals exclusively performing opposite-sex sexual behavior by increasing their number of sexual partners and consequently their number of offspring. However, here we show that having more sexual partners no longer predicts more offspring since the availability of oral contraceptives in the 1960s and that SSB is now negatively ge

Gut microbiota and risk of lower respiratory tract infections: a bidirectional two-sample Mendelian randomization study.

INTRODUCTION: Observational studies have reported the association between gut microbiota and the risk of lower respiratory tract infections (LRTIs). However, whether the association reflects a causal relationship remains obscure. METHODS: A bidirectional twosample Mendelian randomization (MR) analysis was conducted by assessing genome-wide association study (GWAS) summary statistics for gut microbiota taxa and five common LRTIs. MR methods including inverse-variance-weighted (IVW), MR-Egger, weighted median, simple mode, and weighted mode were used to analyze the causality. Gene pleiotropy wa

Low cardiac vagal control is associated with genetic liability for elevated triglycerides and risky health behaviors.

Low parasympathetic regulation of cardiac activity, known as cardiac vagal control (CVC), is robustly associated with poor health outcomes. However, the etiological mechanism that undergirds this association remains largely unknown. One explanation is a causal relationship wherein health problems cause low CVC, or vice versa. However, an alternative explanation is that a common set of genetic factors contributes to both increased liability for poor health and low CVC (i.e., pleiotropy). The present study uses polygenic risk scores for a number of health-related phenotypes (physical, mental, be

Morphological and genetic diversity of Pura Raza Español horse with regard to the coat colour.

Gene mutations influencing melanocytes also impact on physiological and behavioural functions. In this study, we investigated their association with four different coat colours in the Pura Raza Español (PRE) horse using morphological traits and molecular datasets. Four different subpopulations were identified according to individual coat colour: grey, bay, chestnut and black. Coat colour significantly associated with morphological measurements. Observed and expected heterozygosity values were low in grey compared with the other three subpopulations, suggesting the presence of unique ancestral

Genetic overlap and causality between blood metabolites and migraine.

The availability of genome-wide association studies (GWASs) for human blood metabolome provides an excellent opportunity for studying metabolism in a heritable disease such as migraine. Utilizing GWAS summary statistics, we conduct comprehensive pairwise genetic analyses to estimate polygenic genetic overlap and causality between 316 unique blood metabolite levels and migraine risk. We find significant genome-wide genetic overlap between migraine and 44 metabolites, mostly lipid and organic acid metabolic traits (FDR < 0.05). We also identify 36 metabolites, mostly related to lipoproteins, tha
