Genetics of resistance to macrolide antibiotics and lincomycin in natural isolates of Streptococcus pyogenes.

Of 5 clinically isolated strains of Streptococcus pyogenes, 3 showed high-level resistance to erythromycin and lincomycin that was inducible by subinhibitory concentrations of these drugs (IR strains) while 2 strains exhibited constitutive erythromycin and lincomycin resistance (CR strains) which was expressed without prior exposure to low drug concentrations. The CR strain 15346 showed spontaneous loss of resistance whereas resistance in the other strains was quite stable even under curing conditions. The IR strain 13234 was found to be polylysogenic for at least 4 different phages designated

Probiotics in IBD: Evidence and Perspectives on Patient Health and Disease Management.

Inflammatory bowel disease (IBD) includes two distinct diseases: ulcerative colitis (UC) and Crohn's disease, affecting people worldwide regardless of age and gender. It appears that a combination of many factors, primarily genetic background, environmental, host immune response, and a state of reduced microbial diversity are associated with IBD. Gut modulation by probiotics application represents one of the potential strategies for the prevention or treatment of IBD. The gut microbiota has the ability to influence host physiology either directly or through microbial metabolites. This review s

Endogenous bacterial flora in pregnant women and the influence of maternal genetic variation.

Preterm labour, premature prelabour rupture of membranes and low birth weight have all been associated with either specific maternal genital tract infections or an altered vaginal microflora during pregnancy. Factors that influence the variation in microbial-host interaction play an important role in individual susceptibility to adverse pregnancy outcomes. The innate immune responses at mucosal surfaces play a crucial role against microbial invasion. Multiple genes are responsible for the regulation of the innate immune system. Genetic polymorphisms that disrupt innate immune recognition or th

A genotoxin associated with colorectal cancer linked to gut dysbiosis in children with cystic fibrosis.

Cystic fibrosis (CF) alters the gastrointestinal microbiome from an early age, leading to significant changes in microbial composition and functionality. This study explores the physiological and microbiological factors contributing to dysbiosis in children with cystic fibrosis (cwCF), characterized by an increase in potentially pathogenic Escherichia coli and a decrease in beneficial anaerobes, such as Bacteroides. We employed an in vitro medium representative of the nutritional environment of the CF colon to test the role of factors, including mucin, fat, bile, pH, antibiotics, and features

Metagenomics reveals the effects of potassium fertilizer gradients on soil phosphorus-cycling microbial communities and functional genes, and their relationship with yield.

The rational application of potassium (K) is essential for soil nutrient transformation in orchards. Microbial communities drive phosphorus (P) transformation and activation, yet their responses to potassium (K) gradients and the underlying regulation of functional potential remain unclear. Accordingly, a field trial was conducted on 7- to 8-year-old Korla pear trees to evaluate changes in microbial community structure and functional potential across topsoil (0-20 cm) and subsoil (40-60 cm) layers under different K gradients (0, 75, 150, and 225 kg/ha K2O). Results indicated that microbial com

Effects of disinfectant type and dosage on biofilm's activity, viability, microbiome and antibiotic resistome in bench-scale drinking water distribution systems.

Drinking water distribution systems (DWDSs) are important for supplying high-quality water to consumers and disinfectant is widely used to control microbial regrowth in DWDSs. However, the disinfectant's influences on microbial community and antibiotic resistome in DWDS biofilms and the underlying mechanisms driving their dynamics remain elusive. The study investigated the effects of chlorine and chloramine disinfection on the microbiome and antibiotic resistome of biofilms in bench-scale DWDSs using metagenomics assembly. Additionally, the biofilm activity and viability were monitored based o

Gut Microbial Metabolism Inactivates Trifluridine and Drives Loss of Anticancer Efficacy.

Human-associated gut bacteria harbor diverse genetic and metabolic potential. Orally administered therapeutic drugs can interact with these bacteria in the gastrointestinal tract. Recent evidence shows modifications of multiple drugs via gut bacteria, influencing therapeutic outcomes. Trifluridine, a fluoropyrimidine drug used in colorectal cancer treatment, is susceptible to microbial metabolism and premature inactivation, yet the breadth and functional consequences of this metabolism remain incompletely defined. In this work, we further explore the potential of additional gut bacterial strai

Metabolic crosstalk between oral microbiota and the host in OSCC: emerging roles of microbial metabolites in tumor initiation and progression.

Oral squamous cell carcinoma (OSCC) is an aggressive malignancy characterized by profound metabolic reprogramming and a persistently poor clinical outcome. Beyond genetic and environmental risk factors, growing evidence indicates that dysbiosis of the oral microbiome is associated with metabolic perturbations observed in OSCC and may contribute to tumor initiation and progression. Microbiome-derived metabolites represent a previously underappreciated layer of cancer metabolism, linking microbial activity to host metabolic states, epigenetic regulation, and immune dysfunction within the tumor m

An ancient Chinese wisdom for metabolic engineering: Yin-Yang.

In ancient Chinese philosophy, Yin-Yang describes two contrary forces that are interconnected and interdependent. This concept also holds true in microbial cell factories, where Yin represents energy metabolism in the form of ATP, and Yang represents carbon metabolism. Current biotechnology can effectively edit the microbial genome or introduce novel enzymes to redirect carbon fluxes. On the other hand, microbial metabolism loses significant free energy as heat when converting sugar into ATP; while maintenance energy expenditures further aggravate ATP shortage. The limitation of cell "powerhou

Gut microbiota-metabolite axis in gastric cancer: Genetic causality and Ruminococcus subgroup shifts.

Observational studies have long suggested an association between gut microbiota dysbiosis and gastric cancer, yet the causal relevance and underlying biological pathways remain poorly defined. Here, we applied a comprehensive Mendelian randomization (MR) framework to gut microbiota and gastric cancer GWAS summary statistics to identify 14 microbial taxa significantly associated with gastric cancer risk. Sensitivity analyses, including tests for horizontal pleiotropy, heterogeneity, and genetic co-localization, revealed no strong evidence of shared causal variants (H4 < 2%); however, convergent

Small Intestinal Bacterial Overgrowth: Microbiome Dysregulation, Gut-Brain Axis Disruption, and Systemic Consequences.

Small intestinal bacterial overgrowth (SIBO) is a gastrointestinal disorder characterized by excessive bacterial colonization in the small intestine, leading to impaired digestion and nutrient malabsorption. Increasing evidence indicates that SIBO exerts systemic effects beyond the gut, contributing to metabolic, neurological, cardiovascular, dermatological, and autoimmune conditions. Current management strategies include nonsystemic antibiotics such as rifaximin, dietary interventions (low-FODMAP and biphasic diets), and nutraceuticals including berberine, oregano oil, peppermint oil, garlic

Establishing and sustaining mosquito colonies: insights into morphology, bionomics, and advances in the rearing of Anopheles, Culex, and Aedes.

Mosquitoes of the genera Anopheles, Culex, and Aedes are of major medical importance as vectors of malaria, filariasis, Japanese encephalitis, dengue, chikungunya, and Zika. Understanding their morphology and bionomics is fundamental for vector biology, ecological studies, and the design of effective control strategies. Laboratory colonies of mosquitoes serve as an indispensable resource for investigating mosquito genetics, physiology, and vector-pathogen interactions, while also enabling the evaluation of new interventions such as Wolbachia-based control and the sterile insect technique. New vectors and optimal conditions for allelic exchange in hypervirulent Klebsiella pneumoniae.

The emergence of antibiotic-resistant Klebsiella pneumoniae is a significant global health threat that has led to increased morbidity and mortality. This resistance also hinders basic research, as many strains are no longer susceptible to antibiotics commonly used in microbial genetics. Addressing this requires the development of new genetic tools with alternative selective markers. In this report, we introduce new allelic exchange vectors for use in drug-resistant strains. These vectors feature a conditional R6K origin of replication, an origin of transfer, SacB counter-selection, and alterna

A gut bacterial pathway metabolizes aromatic amino acids into nine circulating metabolites.

The human gut microbiota produces dozens of metabolites that accumulate in the bloodstream, where they can have systemic effects on the host. Although these small molecules commonly reach concentrations similar to those achieved by pharmaceutical agents, remarkably little is known about the microbial metabolic pathways that produce them. Here we use a combination of genetics and metabolic profiling to characterize a pathway from the gut symbiont Clostridium sporogenes that generates aromatic amino acid metabolites. Our results reveal that this pathway produces twelve compounds, nine of which a

Transfer of a Catabolic Pathway for Chloromethane in Methylobacterium Strains Highlights Different Limitations for Growth with Chloromethane or with Dichloromethane.

Chloromethane (CM) is an ozone-depleting gas, produced predominantly from natural sources, that provides an important carbon source for microbes capable of consuming it. CM catabolism has been difficult to study owing to the challenging genetics of its native microbial hosts. Since the pathways for CM catabolism show evidence of horizontal gene transfer, we reproduced this transfer process in the laboratory to generate new CM-catabolizing strains in tractable hosts. We demonstrate that six putative accessory genes improve CM catabolism, though heterologous expression of only one of the six is

Chikungunya virus-vector interactions.

Chikungunya virus (CHIKV) is a mosquito-borne alphavirus that causes chikungunya fever, a severe, debilitating disease that often produces chronic arthralgia. Since 2004, CHIKV has emerged in Africa, Indian Ocean islands, Asia, Europe, and the Americas, causing millions of human infections. Central to understanding CHIKV emergence is knowledge of the natural ecology of transmission and vector infection dynamics. This review presents current understanding of CHIKV infection dynamics in mosquito vectors and its relationship to human disease emergence. The following topics are reviewed: CHIKV inf

Chromosomal complementation using Tn7 transposon vectors in Enterobacteriaceae.

Genetic complementation in many bacteria is commonly achieved by reintroducing functional copies of the mutated or deleted genes on a recombinant plasmid. Chromosomal integration systems using the Tn7 transposon have the advantage of providing a stable single-copy integration that does not require selective pressure. Previous Tn7 systems have been developed, although none have been shown to work effectively in a variety of enterobacteria. We have developed several mini-Tn7 and transposase vectors to provide a more versatile system. Transposition of Tn7 at the chromosomal attTn7 site was achiev

MALT lymphoma : recent advances in aetiology and molecular genetics.

Mucosa-associated lymphoid tissue (MALT) lymphoma is a common low grade B-cell lymphoma arising from a background of chronic inflammatory disease at a number of mucosal sites. Those originating in the stomach are causatively linked to Helicobacter pylori infection and eradication of the bacterium with antibiotics leads to long-term complete regression of the lymphoma in aproximately 70% of cases. Now, there is further evidence of linking Campylobacter jejuni, Borrelia burgdorferi and Chlamydia psittaci infection with immunoproliferative small intestine disease, MALT lymphoma of the skin and oc

Polysaccharide hydrogel integrated microbial consortia for reducing relative abundance of antibiotic resistance genes in soil: physical adsorption and biological regulation.

The occurrence of antibiotics and antibiotic resistance genes (ARGs) in soil ecosystems poses a significant risk to human health. However, the techniques for removing ARGs in soil remain limited. Here we developed a natural polysaccharide hydrogel (low-acyl gellan gum/chitosan, GGC) to immobilize the microbial consortia for mitigating the propagation of ARGs. The introduction of microbial consortia in GGC demonstrated high degradation efficiencies of antibiotics, which attributed to the activation of the metabolic pathway network through microbial consortia, further reducing selective pressure

Intestinal Microbiota in Diabetes-Does the Pathomechanism and Diversity Depend on the Type of Diabetes and Coexisting Metabolic Disorders?

Emerging evidence strongly suggests that gut microbiota dysbiosis plays a significant role in the development and progression of both type 1 and type 2 diabetes mellitus. Quantitative and qualitative changes in the intestinal microbiota's composition are linked to these distinct pathomechanisms. In type 1 diabetes mellitus, dysbiosis is thought to initiate or accelerate the autoimmune destruction of pancreatic beta cells. This may occur through increased intestinal permeability, which allows microbial components and endotoxins to enter the systemic circulation. This exposure triggers inflammat
