Histiocytic sarcoma - a case with evenly distributed multinucleated giant cells.

Histiocytic sarcoma is an uncommon neoplasm of mature histiocytes with a poor clinical outcome. We report a case of a true histiocytic sarcoma with prominent and evenly distributed multinucleated giant cells that mimics a giant cell tumor of soft tissue. The tumor was located between the appendix, right ovary, and the terminal ileum with severe adhesion. The liver and spleen were not enlarged. Grossly, the tumor appeared grayish white, solid, and soft. Microscopically, polygonal mononuclear tumor cells aggregated to form somewhat epithelioid nests, which occasionally showed coagulative necrosi…

Giant cells: contradiction to two-hit model of tuber formation?

1. Tuberous sclerosis (TSC) is an autosomal dominant disease characterized by the formation of hamartomatous lesions in many organs, includi … process of tuberous sclerosis, but, interestingly, not in brain lesions, containing characteristic giant cells. 3. In the present paper we review literature covering origination of giant cells and present several hypotheses explaining why in spite of the presence of hamartin and tuberin, brain lesions form in TSC patients.

Evidence of abnormal differentiation in giant cells of tuberous sclerosis.

To characterize the giant cells in tuberous sclerosis, we examined immunoreactivity for nestin, vimentin, microtubule-associated protein 1B (MAP1B), MAP2, neurofilament, and glial fibrillary acidic protein (GFAP) in cortical tubers detected in brain specimens from 6 patients with tuberous sclerosis who had undergone surgical resection for treatment of intractable epilepsy. Giant cells with a neuronal appearance, "neuron-like giant cells," had a round centrally-placed nucleus with a single, prominent nucleolus, and Nissl substance was commonly present in cortex. These neuron-like giant cells de…

Polyploid giant cancer cells: Survival, progeny formation, and therapeutic targeting.

Polyploid giant cancer cells (PGCCs) are enlarged tumor cells with markedly increased genomic content that can arise or become enriched afte … ting progeny vary in proliferative capacity, genomic organization, and phenotypic plasticity, while giant cells and their progeny can influence stromal and immune responses. Human studies associate PGCC-related phenotypes with adverse pathology or outcomes, but their contribution to recurrent tumor lineages remains unresolved. Preclinical interventions act at …

Undifferentiated pancreatic cancer with osteoclast-like giant cells: imaging features.

AIM: The 2019 WHO classification of pancreatic ductal adenocarcinoma (PDAC) recognizes undifferentiated carcinoma with osteoclast-like giant cells (UCOG) as a rare subset. There is sparse literature on the imaging appearances of this type of cancer. Pathologists also increasingly recognize that poorly-differentiated PDAC may occasionally contain osteoclast-like giant cells (PDO). This study aims to compare the imaging features and natural history of these two groups. MATERIALS AND METHODS: In this retrospective case-controlled study, we searched the Institutional pathology and imaging databas…

DRESS syndrome-associated acute necrotizing eosinophilic myocarditis with giant cells.

Drug reaction with eosinophilia and systemic symptoms (DRESS) syndrome is an uncommon drug hypersensitivity reaction caused by a wide variety of agents. It has a characteristic latent period between 2 and 8 weeks from the onset of drug ingestion followed by a slow resolution with the potential for relapse. Despite being a potentially fatal disease, little is understood about its variable clinical presentation and why it can present long after removal of the offending drug. Visceral organ involvement typically occurs, but rarely results in clinically manifested cardiac injury. In its most aggre

Tanycytic ependymoma of the filum terminale with pleomorphic giant cells.

A tanycytic ependymoma measuring 1.5 cm in maximal dimension, which involved the filum terminale and conus medullaris of a 55-year-old woman, is reported. The tumor consisted of a compact fascicular proliferation of spindle cells having long bipolar cytoplasmic processes, and immunohistochemical and ultrastructural studies demonstrated the ependymal features of neoplastic cells. The most prominent finding was an appearance of many atypical and pleomorphic, often monstrous, giant cells, which was not associated with an increase in proliferative activity. Remarkable nuclear atypism and pleomorph…

Participation of ezrin in bacterial uptake by trophoblast giant cells.

BACKGROUND: Trophoblast giant (TG) cells are involved in systematic removal of bacterial pathogens from the maternal-fetal interface of the placenta. In particular, TG cells have the ability to take up extracellular antigens by active phagocytosis induced by interferon-gamma (IFN-gamma). We previously reported that heat shock cognate protein 70 (Hsc70) present on the surface of TG cells mediated the uptake of Brucella abortus. However, the mechanism of bacterial uptake by TG cells is not completely understood. Here we identified ezrin, a member of ezrin-radixin-moesin (ERM) protein family, as

Immunohistochemical and microscopic studies on giant cells in tuberous sclerosis.

Tuberous sclerosis (TSC) is an autosomal dominant disease, caused by mutations in TSC1 or TSC2 genes, encoding hamartin and tuberin, respectively. The clinical picture of the disease is connected with the formation of hamartomas, mainly in the heart, kidneys and the brain. In three types of brain lesions: cortical tubers, subependymal nodules and subependymal giant-cell astrocytoma (SEGA) characteristic, so-called "giant cells" are found. In the present review we summarise immunohistochemical findings of two types of studies performed on giant cells aiming at establishing the expression of ham…

Dysgerminoma with syncytiotrophoblastic giant cells presenting as a hydatidiform mole.

There have been a total of 11 cases of pure dysgerminoma with syncytiotrophoblastic giant cells (SGC) previously reported in the literature as of June 1981. All were Stage IA, all were treated with unilateral salpingo-oophorectomy, and the preoperative serum human chorionic beta-gonadotropin (beta-hCG) titers ranged from 2000 to 3160 mlU/ml. A recent case at William Beaumont Army Medical Center involved a 20-year-old Hispanic female who presented with a 20 weeks' size pelvic-abdominal mass which was sonographically consistent with a hydatidiform mole. An initial preoperative serum beta subunit…

Seminoma with syncytiotrophoblastic giant cells. A special form of seminoma.

Testicular seminomas may occur in various forms, of which the classical and spermatocytic are distinct, the anaplastic or atypical seminomas, however, less clearly defined. Lately, a separate group of particular clinical significance, comprising seminomas with syncytiotrophoblastic giant cells (STGC), has been specified. Although this type of seminoma had been recognized morphologically long ago, recent investigations have shown its ability to secret HCG, a fact that raises serious difficulties in its differential diagnosis with combined seminomas and choriocarcinomas. Two cases of seminomas w…

H3F3A-mutated giant cell tumour of bone without giant cells-clinical presentation, radiology and histology of three cases.

AIMS: Giant cell tumour of bone (GCTB) is histologically defined as a lesion containing reactive giant cells and a neoplastic mononuclear cell population; in up to 92% of cases, GCTB is characterised by a specific mutation of the histone gene H3F3A. The cellular composition ranges from giant-cell-rich to giant-cell-poor. The diagnosis of GCTB can be challenging, and several other lesions need to be excluded, e.g. aneurysmal bone cysts, non-ossifying fibromas, chondroblastomas, brown tumours, and giant-cell-rich osteosarcomas. Our aim was to analyse the clinical history, imaging, molecular path…
