[SPECIFIC IMMUNE RESPONSE AND PROTECTIVE FACTORS OF NEUTROPHIL GRANULOCYTES IN PULMONARY AND EXTRAPULMONARY TUBERCULOSIS].

An immune response and protective factors of neutrophil granulocytes were comparatively studied in 61 patients with pulmonary tuberculosis, 70 with nephrotuberculosis, and 45 with tuberculous spondylitis. It was shown that there were elevated serum levels of neutrophil cationic proteins in both pulmonary and extrapulmonary tuberculosis. Lower content of intracellular cationic proteins along with suppressed cellular immunity was observed in pulmonary tuberculosis patients with disseminated actively progressive changes. This combination may be regarded as a poor predictor. Higher values of a spe

Interaction of intact porcine spermatozoa with epithelial cells and neutrophilic granulocytes during uterine passage.

New insemination techniques allow a tremendous sperm reduction for successful artificial insemination (AI) if highly diluted semen is deposited in the tip of the uterine horn and close to the utero-tubal junction. High sperm losses are known to occur during uterine passage and it was the general question whether specific binding mechanisms are involved. Upon arrival in the uterus, spermatozoa are confronted with mainly two different cell types: uterine epithelial cells (UEC) and neutrophilic granulocytes (polymorphonuclear neutrophil, PMN). As cell-sperm interactions can hardly be observed in

Mitochondrial DNA spectra of single human CD34+ cells, T cells, B cells, and granulocytes.

Previously, we described the age-dependent accumulation of mitochondrial DNA (mtDNA) mutations, leading to a high degree of mtDNA heterogeneity among normal marrow and blood CD34+ clones and in granulocytes. We established a method for sequence analysis of single cells. We show marked, distinct mtDNA heterogeneity from corresponding aggregate sequences in isolated cells of 5 healthy adult donors-37.9% +/- 3.6% heterogeneity in circulating CD34+ cells, 36.4% +/- 14.1% in T cells, 36.0% +/- 10.7% in B cells, and 47.7% +/- 7.4% in granulocytes. Most heterogeneity was caused by poly-C tract variab

Hepatocyte transplantation--in vitro cytotoxic reaction of autologous granulocytes and mononuclears to isolated hepatocytes.

UNLABELLED: Hepatocyte (HC) transplantation (tx) may be useful for bridging patients to whole organ transplantation and for providing metabolic support during liver failure and for replacing whole organ transplantation in certain liver metabolic diseases. In specific situations where the death rate of host hepatocytes is high, the transplanted cells can repopulate the native liver. Successful transplantation of hepatocytes is hampered by lack of proper cellular (stromal) and humoral (cytokines) environment at the site of implantation. We have found that another factor responsible for low in vi

Interaction of Helicobacter pylori (strain 151) and Campylobacter coli with human peripheral polymorphonuclear granulocytes.

Helicobacter pylori associated gastritis is characterized by dense mucosal inflammatory infiltrations with predominantly neutrophilic granulocytes, together with a local and systemic immune response. Nevertheless, the natural course of the infection is chronic in nature, and active phagocytosis of H. pylori by mucosal granulocytes was only rarely observed. The aim of the present study was to investigate with electronmicroscopic methods the interaction of H. pylori with freshly harvested human peripheral granulocytes, with Campylobacter coli as control organism. Bacteria, either untreated or op

Influence of plasma components on luminol-enhanced chemiluminescence from peripheral granulocytes in juvenile periodontitis.

The generation rate of free oxygen radicals as measured by maximal light intensity of luminol-enhanced chemiluminescence from peripheral blood granulocytes (PMN) stimulated with differently opsonized Staphylococcus aureus was studied in 13 patients with juvenile periodontitis (JP) and pair-matched, healthy controls. Plasma proteins related to inflammation were also assayed. When stimulated with bacteria opsonized with autologous serum, the PMN from the JP patients showed a more intensive chemiluminescence than did their pair-matched controls (p less than or equal to 0.0005). The difference was

Passive material behavior of granulocytes based on large deformation and recovery after deformation tests.

In order to better understand the in vivo rheologic behavior of white cells, we have studied the time-dependent deformability, recovery, and mechanical activation of blood granulocytes. We have used micropipette aspiration methods to measure the large deformation response and recovery after deformation characteristics of neutrophils as functions of time, temperature, and collecting media. The cell response in the pipette experiment was characterized by three time domains: the first phase was the passive deformation response to the fixed suction pressure; the second phase was an obvious transit

Clozapine-Associated Agranulocytosis Treatment With Granulocyte Colony-Stimulating Factor/Granulocyte-Macrophage Colony-Stimulating Factor: A Systematic Review.

PURPOSE/BACKGROUND: Clozapine is associated with hematological abnormalities, notably neutropenia, which may progress to agranulocytosis. Granulocyte colony-stimulating factor (G-CSF) and granulocyte-macrophage colony-stimulating factor (GM-CSF) have been used to reduce the frequency and duration of clozapine-associated neutropenia. This review aims to explore the use, efficacy, and tolerability of these cytokines in the treatment of clozapine-associated agranulocytosis. METHODS/PROCEDURES: We conducted a systematic review of published interventional and observational studies, case series, an

Granulocyte colony-stimulating factor (G-CSF) and granulocyte macrophage colony-stimulating factor (GM-CSF) in Behçet's disease.

Increases in the number and activity of peripheral polymorphonuclear neutrophils (PMNs) is often found in Behçet's disease (BD), indicating that PMN may play an important role in the pathogenesis of this disorder. It has recently been reported that G-CSF and GM-CSF, a family of hematopoietic growth factors, enhance PMN activity. To explore the role of these two CSFs in BD, we first examined the chemotactic response of PMNs to these CSFs by performing a polarization assay. PMN response to G-CSF in BD patients was lower than that in controls, while PMN response to GM-CSF was similar in patients

Effects of stem cell factor and granulocyte colony-stimulating factor on granulocyte recovery and Candida albicans infection in granulocytopenic mice.

The combined action of stem cell factor (SCF) and granulocyte colony-stimulating factor (G-CSF) on granulocyte recovery and infection with Candida albicans was studied. In mice rendered granulocytopenic with 200 mg/kg cyclophosphamide, daily coinjection of SCF (100 micrograms/kg) and G-CSF (10 micrograms/kg) for 5 days synergistically stimulated granulocyte recovery compared with either factor alone. Growth of C. albicans in the kidneys of granulocytopenic mice with experimental C. albicans infection was reduced by the combined regimen to 25% of that in vehicle-treated controls (P = .018), whe

Emergency department evaluation of immature granulocytes for complicated acute appendicitis: a retrospective cohort study.

BACKGROUND: Preoperative identification of complicated acute appendicitis (CAA) is important for reducing morbidity. We assessed the diagnostic value of immature granulocyte (IG) count and percentage (IG%) for predicting CAA and histopathologically confirmed perforation in adults presenting to the emergency department. METHODS: We retrospectively reviewed consecutive adults (≥18 years) with histopathologically confirmed appendicitis admitted to a tertiary care center between January 1, 2025 and January 1, 2026. Cases were classified as CAA or non-complicated acute appendicitis (NAA). Perforat

Granulocytes and their Involvement in the Foreign Body Response to Biomaterials and Tissue Repair.

Granulocytes, long considered short-lived effector cells, are increasingly recognized as key modulators of the foreign body response (FBR) to biomaterials and determinants of regenerative outcomes. This review summarizes current evidence on the roles of neutrophils, basophils, and eosinophils in biomaterial-associated inflammation and tissue remodeling. Particular focus is placed on protein adsorption, cytokine release, and downstream effects across diverse biomaterial classes, including bone substitutes, collagen scaffolds, titanium, magnesium, and synthetic polymers. Neutrophils dominate the

CD177 Deficiency Defines a Stable Subtype of Human Neutrophil Granulocytes with Tumor Promoting Activity.

The surface protein CD177 on human neutrophil granulocytes is linked to tissue infiltration. In most individuals, circulating neutrophils comprise distinct fractions of CD177-expressing (CD177+) and non-expressing (CD177-) cells, which differ functionally and influence disease outcomes. Currently, CD177 is considered a dynamic activation marker ultimately expressed by most neutrophils. Here, we show that, instead, CD177--neutrophils represent a stable subpopulation that never acquires CD177. CD177+- to CD177--neutrophil ratios persist over time in individuals, regardless of circadian rhythms o

Adaptation of the living therapeutic materials concept to the immune sensing of neutrophil granulocytes.

Neutrophils are innate immune cells that perpetually patrol the circulation and tissues. They sense and migrate toward invading microbes to initiate and orchestrate a robust immune response. Their highly reactive nature, driven by multiple and redundant receptor families recognizing bacterial components, makes them particularly sensitive to contaminants or nonsterile implants. This often leads to a neutrophil-driven foreign body reaction that shields the implant and triggers inflammation, collateral tissue damage, or even sepsis. This presents a significant challenge for living therapeutic mat

Transcriptional pathways of terminal differentiation in high- and low-density blood granulocytes in sepsis.

BACKGROUND: Trauma and infection induce emergency granulopoiesis. Counts of immature granulocytes and transcriptional pathways of terminal granulocytic differentiation in blood are elevated in sepsis but correlate with disease severity. This limits their performance as sepsis biomarkers in critically ill patients. We hypothesized that activation of these pathways in sepsis is attributable to immature low-density (LD) rather than mature high-density (HD) granulocytes. METHODS: We included patients with sepsis and systemic inflammatory response syndrome (SIRS) of comparable disease severity, an

Indicator of Inflammation and NETosis-Low-Density Granulocytes as a Biomarker of Autoimmune Hepatitis.

Introduction. Interest in the potential role of low-density granulocytes (LDGs) in the development of autoimmune diseases has been renewed recently. Due to their pro-inflammatory action, more and more attention is paid to the role of LDGs, including those expressing the enzyme myeloperoxidase (MPO), in the development of autoimmune hepatitis (AIH). LDGs are actively involved in the formation of neutrophil extracellular traps (NETs). This phenomenon may favour the externalization of the autoantigen and lead to damage to internal organs, including the liver. Aim. The main aim of the study was to

A new and early marker in the diagnosis of acute complicated appendicitis: immature granulocytes.

BACKGROUND: The aim of this study was to investigate the effectiveness of the immature granulocyte (IG) count (IGC) and percentage (IG%) in both diagnosing acute appendicitis (AA) and discriminating between simple appendicitis (SA) and complicated appendicitis (CA). METHODS: This study was carried out using the data of 438 adult patients who underwent an appendectomy. Demographic details, the preoperative white blood cell (WBC) count, neutrophil/lymphocyte ratio (NLR), IGC and IG%, operation findings, and pathology results were assessed retrospectively. The patients were grouped as AA and nor
