NTP Toxicology and Carcinogenesis Studies of 1,3-Butadiene (CAS No. 106-99-0) in B6C3F1 Mice (Inhalation Studies).

1,3-Butadiene is produced in large volumes for use in the manufacture of synthetic rubber and of thermoplastic resins. In previous inhalation studies conducted by the NTP (NTP, 1984) there was clear evidence of multiple organ carcinogenicity in male and female mice exposed to 625 or 1,250 ppm 1,3-butadiene for 60 or 61 weeks. To better characterize exposure-response relationships for neoplasms and nonneoplastic lesions, toxicology and carcinogenesis studies were conducted by exposing groups of male and female B6C3F1 mice to air containing 1,3-butadiene (greater than 99% pure) for up to 2 years

Pulmonary hypertension is associated with poor cardiovascular and hematologic outcomes in patients with myeloproliferative neoplasms and cardiovascular disease.

Cardiovascular events and hematologic progression to myelofibrosis or leukemia are leading causes of morbidity and mortality among patients with myeloproliferative neoplasms (MPN). Pulmonary hypertension (PH) is also associated with MPN and cardiovascular disease (CVD), though its prognostic significance in MPN is not well characterized. Our primary objective was to investigate the effect of PH, defined as right-ventricular systolic pressure (RVSP) ≥ 50 mmHg on echocardiogram or mean pulmonary artery pressure (mPAP) ≥ 20 on right heart catheterization, on cardiovascular and all-cause mortality

Carcinoid Heart Disease: How to Diagnose and Treat in 2020?

Neuroendocrine tumors (NETs, originally termed "carcinoids") create a relatively rare group of neoplasms with an approximate incidence rate of 2.5 to 5 cases per 100 000 persons. Roughly 30% to 40% of subjects with NETs develop carcinoid syndrome (CS), and 20% to 50% of subjects with CS are diagnosed with carcinoid heart disease (CaHD). The long-standing exposure to high serum serotonin concentration is one of the crucial factors in CaHD development. White plaque-like deposits on the endocardial surface of heart structures with valve leaflets and subvalvular apparatus thickening (fused and sho

The Positive Effects of Exercise in Chemotherapy-Related Cardiomyopathy.

Anthracyclines such as doxorubicin, daunorubicin, epirubicin, mitoxantrone and idarubicin, are powerful chemotherapeutic drugs used both in children and adult populations. Their properties made them particularly suitable for a large variety of neoplasms including breast adenocarcinoma, small cell lung cancer and acute leukemia. Early and late anthracycline-induced cardiotoxicity is a well-known phenomenon, and the incidence of heart failure in patients receiving doxorubicin is 2.2%, with a mortality rate over 60% at 2 years. Prognosis can be improved by prevention, early detection and treatmen

Cardiac hypertrophy associated with myeloproliferative neoplasms in JAK2V617F transgenic mice.

BACKGROUND: Myeloproliferative neoplasms (MPNs) are blood malignancies manifested in increased production of red blood cells, white blood cells, and/or platelets. A major molecular lesion associated with the diseases is JAK2V617F, an activation mutation form of tyrosine kinase JAK2. Cardiovascular events represent the leading cause of morbidity and mortality associated MPNs, but the underlying mechanism is not well understood. METHODS: Previously, we generated JAK2V617F transgenic mice which displayed MPN-like phenotypes. In the present study, we further characterized these mice by analyzing

Presumptive primary mammary gland squamous cell carcinoma with widespread metastases in a young ewe.

Mammary tumors in sheep are exceedingly rare, with all reported cases being benign to noninvasive and slow growing. Here, we describe a squamous cell carcinoma (SCC) with widespread metastases presumptively arising from the mammary parenchyma in a 3.5-y-old ewe. Following a history of chronic mastitis and severe swelling of the udder, the udder was removed surgically and submitted for examination. Three weeks after surgery, because of poor prognosis, the animal was euthanized and autopsied. The udder had no cutaneous lesions but had parenchymal disorganization and extensive areas of necrosis d

Implications of Clonal Hematopoiesis in Hematological and Non-Hematological Disorders.

Clonal hematopoiesis (CH) is associated with an increased risk of developing myeloid neoplasms (MNs) such as myelodysplastic neoplasm (MDS) and acute myeloid leukemia (AML). In general, CH comprises clonal hematopoiesis of indeterminate potential (CHIP) and clonal cytopenia of undetermined significance (CCUS). It is an age-related phenomenon characterized by the presence of somatic mutations in hematopoietic stem cells (HSCs) and hematopoietic stem and progenitor cells (HSPCs) that acquire a fitness advantage under selection pressure. Individuals with CHIP have an absolute risk of 0.5-1.0% per

Identification of curaxin as a potential new therapeutic for JAK2 V617F mutant patients.

Myelofibrosis is a myeloproliferative neoplasm (MPN) which typically results in reduced length and quality of life due to systemic symptoms and blood count changes arising from fibrotic changes in the bone marrow. While the JAK2 inhibitor ruxolitinib provides some clinical benefit, there remains a substantial unmet need for novel targeted therapies to better modify the disease process or eradicate the cells at the heart of myelofibrosis pathology. Repurposing drugs bypasses many of the hurdles present in drug development, such as toxicity and pharmacodynamic profiling. To this end we undertook

Pulmonary tumor thrombotic microangiopathy in patients with low-grade ovarian serous neoplasm: a clinicopathologic review of 2 cases of a previously unknown association.

Pulmonary tumor thrombotic microangiopathy (PTTM) is a rare complication occurring during tumor dissemination that can lead to severe, commonly unrecognized pulmonary hypertension, right-sided heart failure, and sudden death. Histologically, it is characterized by tumor microthrombi within small arteries and arterioles and associated fibrocellular and fibromuscular intimal proliferation. Gastric adenocarcinoma is the most common tumor type with this association. Of gynecologic malignancies, a single case of ovarian clear cell carcinoma has been linked to PTTM. We report 2 patients who underwen

Electrochemotherapy Treatment for Squamous Cell Carcinoma of the Lip: A Case Report.

BACKGROUND: Squamous cell carcinoma is a malignant tumor that affects the skin and mucous membranes. It may present as painless superficial lesions, or as rapidly growing, exophytic masses, sometimes ulcerated, which can invade and damage surrounding tissues. It is the second most common malignant neoplasm of the skin, characterized by high clinical variability, which makes early diagnosis and timely treatment essential to improve prognosis. The aim of this case report is to evaluate the clinical efficacy, tolerability, and durability of response of electrochemotherapy in the treatment of cuta

Fissural pleural effusion mimicking a lung mass: a diagnostic pitfall in the Emergency Department.

BACKGROUND: Phantom tumour, also known as a vanishing tumour or fissural pseudotumor, is an uncommon condition that represents a localized, transudative, loculated interlobar pleural effusion. It occurs predominantly in patients with heart failure or renal dysfunction and may mimic a pulmonary neoplasm on chest radiography. Rapid resolution following diuretic therapy is characteristic and typically occurs in the setting of acute decompensated heart failure. CASE PRESENTATION: We report the case of a 79-year-old male with a known history of ischemic heart disease who presented to the Emergency

Does Telotristat Have a Role in Preventing Carcinoid Heart Disease?

Carcinoid heart disease (CHD) is a frequent and life-threatening complication in patients with carcinoid tumors. Its clinical management is challenging is some cases since serotonin-induced valve fibrosis leads to heart failure. Telotristat is an inhibitor of tryptophan-hydroxylase (TPH), a key enzyme in serotonin production. Telotristat use in patients with carcinoid syndrome and uncontrollable diarrhea under somatostatin analogs is approved, but its specific role in patients with CHD is still not clear. IN this context, we aimed to explore the effect of telotristat in heart fibrosis using a

Cardiac Myxoma With Gamna-Gandy Bodies in a Nicaraguan Patient: Leveraging Advanced Imaging to Reduce Mortality Risk.

Myxomas are the most frequent primary cardiac neoplasms, presenting with diverse clinical manifestations depending on their size and location due to obstructive effects on blood flow. In patients with pre-existing structural heart disease, their nonspecific symptoms can closely mimic acute heart failure, valvular disease, or even pulmonary thromboembolism, leading to significant diagnostic challenges. We describe the case of a 59-year-old Hispanic male patient with hypertensive heart disease with preserved ejection fraction and paroxysmal atrial fibrillation, poorly adherent to treatment. He i

Primary prevention of cardiovascular disease with pravastatin in Japan (MEGA Study): a prospective randomised controlled trial.

BACKGROUND: Evidence-based treatment for hypercholesterolaemia in Japan has been hindered by the lack of direct evidence in this population. Our aim was to assess whether evidence for treatment with statins derived from western populations can be extrapolated to the Japanese population. METHODS: In this prospective, randomised, open-labelled, blinded study, patients with hypercholesterolaemia (total cholesterol 5.69-6.98 mmol/L) and no history of coronary heart disease or stroke were randomly assigned diet or diet plus 10-20 mg pravastatin daily. The primary endpoint was the first occurrence

Ovarian carcinoid tumor with carcinoid heart syndrome: A case report and literature review.

BACKGROUND: Primary ovarian carcinoid tumors are rare neuroendocrine neoplasms comprising <0.1% of ovarian tumors and 1% of all carcinoid tumors. Up to 30% present with carcinoid syndrome, and approximately one-quarter of these develop carcinoid heart syndrome. Unlike gastrointestinal carcinoid tumors, ovarian lesions can cause carcinoid syndrome without hepatic metastases due to direct venous drainage into the systemic circulation. Diagnostic Dilemma of a Neuroendocrine Tumour Complicated by Simultaneous Retroperitoneal Fibrosis and Carcinoid Heart Disease in a Perimenopausal Woman.

Carcinoid tumours are rare, slow-growing neuroendocrine neoplasms that often remain asymptomatic until metastatic spread or the development of carcinoid syndrome. Carcinoid syndrome is characterised by flushing, diarrhoea, and bronchospasm due to the secretion of vasoactive hormones. These tumours commonly arise in the gastrointestinal tract but can also occur in other organs, namely, the lungs, genitourinary tract, and pancreas. Retroperitoneal fibrosis (RPF), a rare inflammatory disease, involves chronic inflammation leading to fibrous scarring and compression of surrounding structures like

Successful one-stage resection of intracardiac intravenous leiomyomatosis: A case report.

This case report is about a 47-year-old patient, who was diagnosed with intracardiac intravenous leiomyomatosis and received treatment at our institution. Intravenous leiomyomatosis is a rare, histologically benign, uterine neoplasm, which is characterized by non-invasive intravascular proliferation of smooth muscle cells. Intravenous leiomyomatosis arises from the myometrium and, in its most extensive form, can reach the heart via the pelvic veins and the inferior vena cava, causing hemodynamic complications. Treatment of choice is the complete resection of the tumor, even though there is no
