[Globalization and infectious diseases].

We live in an ever more connected global village linked through international travel, politics, economics, culture and human-human and human-animal interactions. In public health a similar combination of factors can be seen. Today the concept of globalization including global exposure to infectious diseases is becoming more apparent. In 2003 outbreaks included monkeypox , SARS and avian influenza. This article examines some basic problems to infectious diseases emergence and control.

Advances in the development of organoid-based models of the trachea and esophagus.

During organogenesis, the anterior foregut of the mammalian embryo undergoes morphological changes that give rise to the ventral trachea and the dorsal esophagus. Subsequently, the trachea on the ventral side functions as an air passage between the larynx and the bronchi, whereas the esophagus on the dorsal side develops a lumen to transport food from the pharynx to the stomach. Although mouse genetics has provided insights into various cell-cell interaction signals involved in the development of the trachea and esophagus, the exact mechanism remains elusive. Additionally, differences in tissu

Associations between Cryptococcus Genotypes, Phenotypes, and Clinical Parameters of Human Disease: A Review.

The genus Cryptococcus contains two primary species complexes that are significant opportunistic human fungal pathogens: C. neoformans and C. gattii. In humans, cryptococcosis can manifest in many ways, but most often results in either pulmonary or central nervous system disease. Patients with cryptococcosis can display a variety of symptoms on a spectrum of severity because of the interaction between yeast and host. The bulk of our knowledge regarding Cryptococcus and the mechanisms of disease stem from in vitro experiments and in vivo animal models that make a fair attempt, but do not recapi

Screening and identification of a human domain antibody against Brucella abortus VirB5.

Brucellosis is caused by the genus Brucella. Brucella is widely distributed in cattle, swine, sheep, goat and other mammals including human. Animal brucellosis causes severe economic losses and affects related international transportation and trade. Human brucellosis causes both acute and chronic symptoms of multi-organ dysfunction. Brucella type IV secretion system (T4SS) VirB5 was required for macrophages infection and essential for virulence in mice. VirB5 is located on the cell surface and serves as a specific adhesin targeting host cell receptors. The aim of this study was to isolate and

Human lung ex vivo infection models.

Pneumonia is counted among the leading causes of death worldwide. Viruses, bacteria and pathogen-related molecules interact with cells present in the human alveolus by numerous, yet poorly understood ways. Traditional cell culture models little reflect the cellular composition, matrix complexity and three-dimensional architecture of the human lung. Integrative animal models suffer from species differences, which are of particular importance for the investigation of zoonotic lung diseases. The use of cultured ex vivo infected human lung tissue may overcome some of these limitations and compleme

The role of stem cells in the pharmacological assessment of nanoparticles.

Stem cells aid in the pharmacological assessment of nanoparticles by serving as a human-relevant, dynamic, and scalable model. Their integration into nanoparticle research enhances toxicity screening and therapeutic optimization by evaluating absorption, distribution, metabolism, excretion and toxicity (ADMET) parameters across models ranging from simple in vitro cultures to complex organoids. They provide insights into how nanoparticles interact with biological systems, guiding the rational design of safer, more effective nanomedicines. Conventional in vitro and animal models often fall short

Listeria monocytogenes behaviour in presence of non-UV-irradiated titanium dioxide nanoparticles.

Listeria monocytogenes is the agent of listeriosis, a food-borne disease. It represents a serious problem for the food industry because of its environmental persistence mainly due to its ability to form biofilm on a variety of surfaces. Microrganisms attached on the surfaces are a potential source of contamination for environment and animals and humans. Titanium dioxide nanoparticles (TiO2 NPs) are used in food industry in a variety of products and it was reported that daily exposure to these nanomaterials is very high. Anti-listerial activity of TiO2 NPs was investigated only with UV-irradiat

Review: Creutzfeldt-Jakob disease: prion protein type, disease phenotype and agent strain.

The human transmissible spongiform encephalopathies or human prion diseases are one of the most intensively investigated groups of rare human neurodegenerative conditions. They are generally held to be unique in terms of their complex epidemiology and phenotypic variability, but they may also serve as a paradigm with which other more common protein misfolding disorders might be compared and contrasted. The clinico-pathological phenotype of human prion diseases appears to depend on a complex interaction between the prion protein genotype of the affected individual and the physico-chemical prope

The antiviral immunity of ticks against transmitted viral pathogens.

Ticks, being obligate hematophagous arthropods, are exposed to various blood-borne pathogens, including arboviruses. Consequently, their feeding behavior can readily transmit economically important viral pathogens to humans and animals. With this tightly knit vector and pathogen interaction, the replication and transmission of tick-borne viruses (TBVs) must be highly regulated by their respective tick vectors to avoid any adverse effect on the ticks' biological development and viability. Knowledge about the tick-virus interface, although gaining relevant advances in recent years, is advancing

Experiments on Analysing Voice Production: Excised (Human, Animal) and In Vivo (Animal) Approaches.

UNLABELLED: Experiments on human and on animal excised specimens as well as in vivo animal preparations are so far the most realistic approaches to simulate the in vivo process of human phonation. These experiments do not have the disadvantage of limited space within the neck and enable studies of the actual organ necessary for phonation, i.e., the larynx. The studies additionally allow the analysis of flow, vocal fold dynamics, and resulting acoustics in relation to well-defined laryngeal alterations. PURPOSE OF REVIEW: This paper provides an overview of the applications and usefulness of ex

From rodents to chips: preclinical models of inflammatory bowel disease with emphasis on host-microbiome interactions.

Inflammatory bowel disease (IBD), comprising Crohn's disease and ulcerative colitis, is a chronic relapsing inflammatory disorder with steadily increasing global prevalence, significantly impairing patient quality of life. Preclinical research in IBD has traditionally relied on animal models to investigate disease etiology, immunopathogenesis, and therapeutic responses. A wide range of experimental models primarily rodents, along with limited use of larger animals such as pigs and nonhuman primates, have been developed to reproduce key clinical, histological, and immunological features of huma

Mouse models for actinic keratosis and squamous cell carcinoma.

This manuscript focuses on the use of mice to study the genetics and biology of cutaneous squamous cell carcinoma (SCC). Mice develop actinic keratosis-like lesions and SCC resembling those seen in humans. As an animal model, the mouse provides great experimental flexibility and has been useful in investigating aspects of the genetics and biology of SCC that are difficult to study in humans. We discuss the pros and cons of the various murine models available. How well mouse pathology in general mimics human disease remains an open question due to the vast differences in animal strain backgroun

Frank A. Beach award. Homologies of animal and human sexual behaviors.

Theoretical models of animal and human sexual behavior have evolved from two very different literatures, yet they contain many common behavioral components that may reflect the action of similar neuroendocrine and neurochemical systems. The study of animal sexual behavior has been largely concerned with mechanisms that underlie the pattern of consummatory behaviors observed during copulation, behaviors that tend to be highly stereotyped, sexually differentiated, and species-specific. There are important species differences in the behavioral topography, endocrine control, and neural substrates

Seeded fibrillation as molecular basis of the species barrier in human prion diseases.

Prion diseases are transmissible spongiform encephalopathies in humans and animals, including scrapie in sheep, bovine spongiform encephalopathy (BSE) in cattle, chronic wasting disease (CWD) in deer, and Creutzfeldt-Jakob disease (CJD) in humans. The hallmark of prion diseases is the conversion of the host-encoded prion protein (PrP(C)) to its pathological isoform PrP(Sc), which is accompanied by PrP fibrillation. Transmission is not restricted within one species, but can also occur between species. In some cases a species barrier can be observed that results in limited or unsuccessful transm

Evidence that fibulin family members contribute to the steroid-dependent extravascular sequestration of sex hormone-binding globulin.

Sex hormone-binding globulin (SHBG) binds steroids in the blood but is also present in the extravascular compartments of some tissues. Mice expressing a human SHBG transgene in the liver have human SHBG in their blood. In these animals, human SHBG accumulates within the stromal matrix of the endometrium and epididymis. This is remarkable because these tissues do not express the transgene. Human SHBG administered intravenously to wild-type mice in the presence of estradiol is rapidly sequestered within the endometrial stroma, and this prompted us to search for SHBG interacting proteins. Yeast t

Experimental disease models for the assessment of meningococcal vaccines.

Animal infection models are valuable for the development and preclinical assessment of meningococcal vaccines in the absence of clear in vitro correlates of protection for protein-based serogroup B vaccines. It is only in animal models that interactions of the organism with the innate, humoral and cellular immune systems can be assessed. However, humans are the only natural host for Neisseria meningitidis and there is no ideal disease model using laboratory animals that mimics the course of human disease. The two most widely used models are intraperitoneal (i.p.) infection of adult mice or inf

A direct functional link between the multi-PDZ domain protein GRIP1 and the Fraser syndrome protein Fras1.

Cell adhesion to extracellular matrix (ECM) proteins is crucial for the structural integrity of tissues and epithelial-mesenchymal interactions mediating organ morphogenesis. Here we describe how the loss of a cytoplasmic multi-PDZ scaffolding protein, glutamate receptor interacting protein 1 (GRIP1), leads to the formation of subepidermal hemorrhagic blisters, renal agenesis, syndactyly or polydactyly and permanent fusion of eyelids (cryptophthalmos). Similar malformations are characteristic of individuals with Fraser syndrome and animal models of this human genetic disorder, such as mice car

Gendered knowledge practices and human-animal-ecological health in Papua New Guinea: a qualitative study during a zoonotic tuberculosis investigation.

There have been consistent calls to incorporate gender frameworks into One Health research and policy. Men's and women's roles in relation to animals in many rural and agricultural communities may influence potential pathogen exposure and transmission patterns of zoonotic infectious diseases. However, men's and women's roles related to animals in rural societies are not static but change over time. This article draws on the results of qualitative research about perceptions and practices related to animal species and animal products in five sites located in Eastern Highlands Province and East N

Animal models of human anthrax: the Quest for the Holy Grail.

Anthrax is rare among humans, few data can be collected from infected individuals and they provide a fragmentary view of the dynamics of infection and human host-pathogen interactions. Therefore, the development of animal models is necessary. Anthrax has the particularity of being a toxi-infection, a combination of infection and toxemia. The ideal animal model would explore these two different facets and mimic human disease as much as possible. In the past decades, the main effort has been focused on modelling of inhalational anthrax and the perception of specific aspects of the infection has

The clinical pharmacology of appetite suppressant drugs.

One way of gaining a greater understanding of the central mechanisms underlying hunger and the regulation of feeding behaviour in humans is to examine the actions and interactions on hunger and food intake of drugs with known or presumed pharmacological modes of action. To this end we have undertaken a number of studies which fall into three main categories: the mechanisms by which amphetamine anorexia is induced; the possible role of endogenous opioids in feeding; the action of amino acids thought to be involved in the regulation of feeding. In this field the potential for cross-fertilization
