Hypoxanthine incorporation was studied in growing HPRT mutant cells by preincubating them with extracts from normal cells, HPRT mutant cells, and extracts of their lyophilized cell sediment. HPRT mutant cells showed no increase of hypoxanthine uptake after preincubation with extracts of mutant cells, whereas after preincubation with extracts from normal cells and lyophilized sediment of HPRT mutant cells the incorporation rate was increased. This effect could not be observed when normal cells were preincubated with extracts of lyophilized sediments of normal cell lines.

Hypoxanthine--guanine phosphoribosyltransferase (HPRT) is a purine salvage enzyme that catalyzes the conversion of hypoxanthine to inosine monophosphate and guanine to guanosine monophosphate. The pharmacokinetics of vidarabine were studied in 8 patients with polyarteritis nodosa related to hepatitis B virus infection. The drug was administered by continuous infusion for three weeks at doses of 15 (1 week) and 7.5 (2 weeks) mg/kg per day, during which time 15 plasma exchanges were performed. Two brothers with hereditary xanthinuria (xanthine oxidase deficiency) and several members of their family were studied. A novel point mutation (I137T) was identified in the hypoxanthine-guanine phosphoribosyltransferase (HPRT) encoding gene, in a patient with partial deficiency of the enzyme.
