Oligodendrocyte-axon metabolic coupling is mediated by extracellular K+ and maintains axonal health.

The integrity of myelinated axons relies on homeostatic support from oligodendrocytes (OLs). To determine how OLs detect axonal spiking and how rapid axon-OL metabolic coupling is regulated in the white matter, we studied activity-dependent calcium (Ca2+) and metabolite fluxes in the mouse optic nerve. We show that fast axonal spiking triggers Ca2+ signaling and glycolysis in OLs. OLs detect axonal activity through increases in extracellular potassium (K+) concentrations and activation of Kir4.1 channels, thereby regulating metabolite supply to axons. Both pharmacological inhibition and OL-spe

Early-activated extracellular matrix proteins shape the metabolic and spatial dynamics of the kidney fibrotic microenvironment.

The fibrotic kidney microenvironment is shaped by cellular crosstalk, extracellular matrix (ECM) remodelling, metabolic reprogramming and spatial heterogeneity. While late-stage ECM changes dominate fibrosis, the role of early-activated matrix proteins remains unclear. Here we show that ECM1 is an early regulator of kidney remodelling. Global Ecm1 knockout mice develop spontaneous fibrosis and early death, whereas ECM1 levels markedly increase in biofluids during chronic kidney disease. Targeting Ecm1 through AAV9-mediated knockdown or fibroblast-specific deletion substantially reduces renal f

Obesity Disrupts CtBP2-Mediated Maintenance of Transcriptional Equilibrium in Hypothalamic Feeding Circuitry.

Feeding behavior, which is crucial for all mammals, is regulated by a delicate equilibrium between the orexigenic and anorexigenic activities of hypothalamic neurons. This exquisite control of the neuropeptides that govern the feeding behavior could be a focal point in the pathogenesis of obesity. We reported that inactivation of C-terminal binding protein 2 (CtBP2), a transcriptional corepressor with metabolite-sensing capabilities, contributes to the pathogenesis of obesity in liver tissues and pancreatic β-cells. Here, we describe a transcriptional system regulated by CtBP2 in the hypothala

[Mek1 and Mek2 functions in the formation of the blood placental barrier].

The ERK/MAPK signaling pathway is involved in several cellular functions. Inactivation in mice of genes encoding members of this pathway is often associated with embryonic death resulting from abnormal placental development. The placenta is essential for nutritional and gaseous exchanges between maternal and embryonic circulations, as well as for the removal of metabolic wastes. These exchanges take place without direct contact between the two circulations. In mice, the hematoplacental barrier consists in a triple layer of trophoblast cells and endothelial cells of the embryo. MEK1 and MEK2 ar

Control of Kir channel gating by cytoplasmic domain interface interactions.

Inward rectifier potassium (Kir) channels are expressed in almost all mammalian tissues and play critical roles in the control of excitability. Pancreatic ATP-sensitive K (KATP) channels are key regulators of insulin secretion and comprise Kir6.2 subunits coupled to sulfonylurea receptors. Because these channels are reversibly inhibited by cytoplasmic ATP, they link cellular metabolism with membrane excitability. Loss-of-function mutations in the pore-forming Kir6.2 subunit cause congenital hyperinsulinism as a result of diminished channel activity. Here, we show that several disease mutations

Toxicological significance of mechanism-based inactivation of cytochrome p450 enzymes by drugs.

Cytochrome P450 (P450) enzymes oxidize xenobiotics into chemically reactive metabolites or intermediates as well as into stable metabolites. If the reactivity of the product is very high, it binds to a catalytic site or sites of the enzyme itself and inactivates it. This phenomenon is referred to as mechanism-based inactivation. Many clinically important drugs are mechanism-based inactivators that include macrolide antibiotics, calcium channel blockers, and selective serotonin uptake inhibitors, but are not always structurally and pharmacologically related. The inactivation of P450s during dru

Cytochrome P450-mediated bioactivation of the epidermal growth factor receptor inhibitor erlotinib to a reactive electrophile.

The epidermal growth factor receptor tyrosine kinase inhibitor erlotinib (ERL) is approved for treatment of non-small-cell lung cancer. Numerous reports of ERL-associated toxicities are consistent with immune-mediated toxicity, including drug-induced hepatitis, interstitial lung disease, Stevens-Johnson syndrome, and toxic epidermal necrolysis. Although the mechanism of toxicity has not been established, we present evidence that reactive intermediates are formed during the metabolism of ERL, which can covalently conjugate to the cysteine group of the peptide-mimetic GSH. Seven ERL-GSH conjugat

A framework for optimized reaction phenotyping of the seven major hepatic P450 isoforms in human liver microsomes using six mechanism-based inactivators and one reversible inhibitor.

Cytochrome P450 (CYP) enzymes play a central role in drug metabolism. Understanding the contribution of individual CYP isoforms, known as CYP reaction phenotyping, is essential for elucidating metabolic pathways and predicting drug-drug interactions. Reversible inhibitors are commonly used in human liver microsome (HLM)-based CYP reaction phenotyping. However, selecting appropriate inhibitor concentrations is challenging as experimental results will vary depending on the HLM concentration, substrate concentration, and incubation duration selected. These factors collectively influence the obser

Introduction to epigenetic mechanisms in the brain.

This chapter provides an introductory overview of epigenetic mechanisms, focusing on the molecular processes that regulate gene expression without altering the DNA sequence. It outlines the foundational concepts of DNA methylation, histone modifications, chromatin remodeling, and noncoding RNAs, with emphasis on their roles in cell identity and transcriptional regulation. The interactions between these epigenetic mechanisms, such as the crosstalk of histone acetylation and DNA methylation, are also discussed. Key mechanisms of long-term gene inactivation, like X-chromosome inactivation and gen

Harnessing wine yeasts: Unveiling their potential as lipid supplements.

In industry, clarifying white grape juice may lead to the partial loss of lipids, which yeasts use to alter their plasma membrane composition to overcome the stresses associated with wine fermentation. In this work, a selection of yeasts isolated from the broader wine environment was screened for their lipid composition using flow cytometry, gravimetric assays and GCMS. Inactivated dry yeasts were produced from six of the screened yeast strains and were tested as lipid supplements in over-clarified Chenin Blanc grape juice, alongside pure lipid mixtures matching the lipid concentrations occurr

[Intestinal enzymopathies of children (author's transl)].

In recent years it has been successfully demonstrated that, in relation to congenital metabolic abnormalities, every enzyme of the gastro-intestinal tract may be absent or inactive.. In serious diseases of the intestinal tract, secondary inactivation of numerous enzymes may result. In this comprehensive presentation the pathology of the disaccharidases, the peptidases and lypolysis is described, the physiology being gone into briefly in each case. Furthermore, all known disturbances of absorption of sugar, aminoacids and fats are briefly dealt with. Finally, congenital chloridorrhea is describ

The effect of chronic exposure to tobacco smoke on the antibacterial defenses of the lung.

To evaluate the effects of cigarette smoking on the host defenses of the lung, male CD rats were exposed to fresh whole smoke for up to 60 consecutive days. Intrapulmonary deposition of smoke and animal exposure levels, quantified with decachlorobiphenyl and other smoke tracers, indicated a daily cigarette exposure equivalent to approximately a pack and a half per day in man. Pulmonary alveolar macrophage function in situ was quantified by the inactivation of an aerosolized challenge of Staphylococcus aureus six hours after inoculation. Controls (n=120) inactivated 88.8+/-0.64% of the staphylo

Rational design of a fluorescent probe for selective sensing of human cytochrome P450 2C19 and its application in exploration of enzymatic inactivation.

Human cytochrome P450 2C19 (CYP2C19) is a drug-metabolizing enzyme frequently implicated in adverse drug reactions. However, the identification of inhibitors, especially irreversible inactivators, that underlie CYP2C19-mediated drug interactions remains challenging, chiefly owing to the paucity of robust molecular probes capable of quantification of enzymatic inactivation profiles in biospecimens. In the present study, we propose a molecular modification strategy that fine-tunes unfavorable substrate-aromatic interactions within the CYP2C19 pocket by rationally modifying a key junction domain

Inability of Ogataea parapolymorpha pho91-Δ mutant to produce active methanol oxidase can be compensated by inactivation of the PHO87 gene.

Cells of the methylotrophic yeast Ogataea parapolymorpha have two genes encoding low-affinity phosphate transporters: PHO87, encoding the plasma membrane transporter, and PHO91, encoding a protein, which is homologous to the Saccharomyces cerevisiae vacuolar membrane transporter. Earlier, we reported that inactivation of PHO91 in O. parapolymorpha interferes with methanol utilization due to the lack of activity of methanol oxidase encoded by the MOX gene. In this work, we showed that this defect was completely suppressed by inactivating the PHO87 gene or introducing additional copies of the MO

Effects of trivalent chromium on hepatic CYP-linked monooxygenases in laying hens.

Chromium is an essential nutrient required for the metabolism of carbohydrates and lipids in humans and many animal species. We evaluated whether feeding laying hens with high levels of different chemical forms of trivalent chromium may affect hepatic metabolizing cytochrome P-450 (CYP)-linked enzymes. Modulation of CYP-dependent monooxygenases (which play a pivotal role in the endogenous metabolism) by Cr(III) was tested using either specific substrates as probes of different CYPs or testosterone as a multi-bioprobe. The CYP-supported oxidases were studied in liver microsomes from laying hens

Safety of COVID-19 vaccines in children with inborn errors of metabolism in terms of developing metabolic decompensation.

AIM: There are no recommended guidelines or clinical studies on safety of COVID-19 vaccines in patients with inborn errors of metabolism (IEMs). Here, we aimed to examine the relationship between COVID-19 vaccination and metabolic outcome in paediatric IEM patients. METHODS: Patients with IEM between the ages of 12 and 18 were enrolled. Term metabolic decompensation was defined as acute disruption in metabolic homeostasis due to vaccination. Clinical and biochemical markers were compared between pre- and post-vaccination periods. RESULTS: Data from a total of 36 vaccination episodes in 18 pa

Increased susceptibility to 4-HNE-induced toxicity and impaired development in a model of ALDH4A1-deficient pediatric epilepsy carrying the S352L variant.

Certain human mutations in the mitochondrial aldehyde dehydrogenase 4A1 (ALDH4A1) lead to a severe, paediatric form of epilepsy and developmental abnormalities, yet the precise molecular mechanism leading to the clinical phenotypes remains unexplained. ALDH4A1 metabolizes glutamic-γ-semialdehyde (GSA). Mutations in ALDH4A1, which lead to inactive enzyme variants, cause GSA to accumulate and vitamin B6 inactivation. Patients with severe ALDH4A1 deficiency have paediatric epilepsy and are resistant to prescribed therapies. We develop knock-in cell culture and mouse models of the S352L variant to

Defective ring A reduction of cortisol as the major metabolic error in the syndrome of apparent mineralocorticoid excess.

Impaired peripheral metabolism of cortisol in the syndrome of apparent mineralocorticoid excess is currently understood to be causally related to the severe but otherwise unexplained manifestations of mineralocorticoid excess. A normally ambivalent mineralocorticoid receptor responding equally well to glucocorticoids and mineralocorticoids requires prereceptor inactivation of glucocorticoids to elicit a specific mineralocorticoid effect. The failed inactivation step in the form of the syndrome of apparent mineralocorticoid excess first described (type 1) involves the 11 beta-hydroxydehydrogena

Nitric Oxide Inhibition of Glycyl Radical Enzymes and Their Activases.

Innate immune response cells produce high concentrations of the free radical nitric oxide (NO) in response to pathogen infection. The antimicrobial properties of NO include nonspecific damage to essential biomolecules and specific inactivation of enzymes central to aerobic metabolism. However, the molecular targets of NO in anaerobic metabolism are less understood. Here, we demonstrate that the Escherichia coli glycyl radical enzyme pyruvate formate lyase (PFL), which catalyzes the anaerobic metabolism of pyruvate, is irreversibly inhibited by NO. Using electron paramagnetic resonance and site

Mg2+ Supplementation Mitigates Metabolic Deficits Associated With TRPM7 Disruption.

Transient receptor potential channel subfamily M member 7 (TRPM7) regulates cellular and systemic Mg2+ homeostasis through its channel domain and induces protein phosphorylation via its kinase domain. We recently found that mice with selective deletion of Trpm7 in β-cells develop glucose intolerance and declines in insulin secretion, primarily due to the impaired enzymatic activity of this protein. Accumulating evidence suggests that Mg2+ supplementation effectively mitigates the detrimental effects of TRPM7 disruption in various cell types. However, the impact of Mg2+ supplementation on metab
