Calmodulin infused intracerebroventricularly enhances food intake in the cat.

In the fasted cat, calmodulin (CaM) infused into the cerebral ventricle produces an increase in the normal intake of food in a dose-dependent manner. The enhancement of feeding by CaM seems to be functionally specific since the response was: (1) abolished by the simultaneous intraventricular infusion of calcineurin, a specific CaM antagonist; (2) not mimicked by another calcium binding protein, troponin C; and (3) independent of the CaM's lack of effect on body temperature and water intake. This finding opens up the dual possibility that this Ca2+ binding protein may affect receptors other tha

[Morphine antagonizes the central hypotensive effect of clonidine in the anesthetized cat].

A dose of 1 microgram/kg of clonidine injected intracerebroventricularly (I.C.V.) in the normotensive anaesthetized Cat elicits a significant and long lasting drop in the mean arterial blood pressure (23 +/- 5 mmHg) and the heart rate (34 +/- 5 beats/min.). A pretreatment with morphine (100 micrograms/kg, I.C.V.) completely prevents the central hypotensive effect of clonidine, whereas the bradycardia is only weakly influenced. Administered after clonidine, morphine reverses the hypotensive effect of clonidine whereas morphine does not affect its bradycardic action. These data demonstrate that

Structural and biochemical changes in rat cerebral cortex after neonatal 6-hydroxydopamine administration.

Newborn rats received an intracisternal injection of 6-hydroxydopamine (100 micrograms) within 16 h after birth. Treatment effects upon noradrenaline uptake (with or without desmethylimipramine pre-incubation), endogenous noradrenaline, dopamine, and serotonin were biochemically assayed. Noradrenaline uptake and endogenous noradrenaline content were permanently reduced to less than 5% of control values. Reduction of endogenous dopamine content was less marked: at day 60, values were about 40% of controls. Serotonin content remained unaffected. Cell density countings in postnatal day 15 tempora

Cerebromedullospinal disconnection after intraventricular methotrexate.

A case of cerebromedullospinal disconnection or "locked-in" syndrome after intraventricular methotrexate is described. This is an unusual complication of intraventricular methotrexate instillation. Ventral pontine infarction is postulated to be responsible for the syndrome.

Impact of Intraventricular Human Adipose-Derived Stem Cells Transplantation with Pregnenolone Treatment on Remyelination of Corpus Callosum in A Rat Model of Multiple Sclerosis.

OBJECTIVE: Multiple sclerosis (MS) is known as a nerve tissue disorder, which causes demyelination of central nervous system (CNS) fibers. Cell-based treatment is a novel strategy for the treatment of demyelinating diseases such as MS. Adipose-derived stem cells (ADSCs) have neuroprotective and neuroregenerative effects and pregnenolone as a neurosteroid has remarkable roles in neurogenesis. We intend to examine the impact of intraventricular transplantation of human ADSCs and systemic injection of pregnenolone on the remyelination of a rat model cuprizone-induced demyelination. Efficacy and safety of intraventricular nimodipine in patients with aneurysmal subarachnoid hemorrhage compared to oral nimodipine: a systematic review and meta-analysis.

Aneurysmal subarachnoid hemorrhage (aSAH) is associated with high morbidity and mortality. Nimodipine is the only drug approved and administered orally as well as intravenously to improve the outcomes of patients with vasospasm post-SAH. EG-1962 is a sustained-release formulation of nimodipine administered into the subarachnoid space in patients with aSAH. We hypothesize that this may improve the efficacy of nimodipine and minimize its adverse effects. We searched PubMed, Cochrane, Scopus, and Web of Science on August 2, 2024, using relevant keywords. Studies were screened for eligibility. We

Central ventilatory and cardiovascular actions of serotonin in trout.

This study was undertaken to investigate the central actions of 5-HT on ventilatory and cardiovascular variables in the unanesthetized trout. Compared to vehicle, intracerebroventricular injection (ICV) of 5-HT elevated the total ventilation. This elevation was due to its stimulatory action on ventilatory amplitude. Moreover, 5-HT produced a dose-dependent increase in mean dorsal aortic blood pressure (PDA) without change in heart rate (fH). Methysergide, a 5-HT1/5-HT2 receptor antagonist, reduced the hyperventilatory and hypertensive actions of 5-HT. 8-OH-2-(di-n-propylamino) tetralin, a 5-HT

A single episode of central glucoprivation reduces the adrenomedullary response to subsequent hypoglycemia in rats.

Hypoglycemia-associated autonomic failure (HAAF) is a complication of recurrent hypoglycemia in patients with diabetes. The present experiments were performed to study the contribution of brain glucose-sensitive structures to HAAF. In conscious rats, brain glucoprivation was elicited by injection of 2 deoxy-D-glucose (2-DG) into the third cerebral ventricle (intracerebroventricular, ICV), while controls received saline ICV. Two days after ICV injection, hormone responses to systemic hypoglycemic challenge were measured. The epinephrine response 60 min after insulin injection was reduced by 46%

The anorectic effect of neurotensin is mediated via a histamine H1 receptor in mice.

Neurotensin (NT), a tridecapeptide found in the mammalian brain and peripheral tissues, induces a decrease in food intake after central administration. In this investigation, we examine whether the histaminergic system is involved in NT-induced suppression of feeding. Intracerebroventricular injection of NT (0.1-1 nmol/mouse) led to dose-dependent inhibition of food intake in fasted ddY mice. The anorectic effect induced by NT (0.1 nmol/mouse) was ameliorated upon co-administration of pyrilamine (3 nmol/mouse), an antagonist for histomine H1 receptor. The NT-induced anorectic effect was partia

Enhancement of natural immunity seen after voluntary exercise in rats. Role of central opioid receptors.

Chronic voluntary exercise in wheels for 5 weeks in spontaneously hypertensive rats (SHR) augments in vivo natural killer (NK) cell cytotoxicity. Endogenous beta-endorphin is increased in cerebrospinal fluid after voluntary exercise in rats and we have recently shown that beta-endorphin administered i.c.v. augments NK cell mediated cytotoxicity in vivo in a similar way as chronic voluntary exercise. We have now further investigated the involvement of central opioid systems in the exercise-induced augmentation in natural immunity. Exercise consisted of voluntary running in wheels for 5 weeks. I

Acute decreases in cerebrospinal fluid glutathione levels after intracerebroventricular morphine for cancer pain.

UNLABELLED: Intracerebroventricular (ICV) morphine administration is effective for the management of refractory cancer pain. Recent preclinical observations of acute depletion of the major endogenous intracellular antioxidant glutathione (GSH) in brain and peripheral organs after ICV morphine in rodents led us to apply microchemical methods to profile the neurochemical effects of ICV morphine in three patients treated for intractable cancer pain. Assessment of morphine, morphine-6-glucuronide, and a panel of endogenous compounds and metabolites in ventricular and cisternal cerebrospinal fluid

The intrinsic antinociceptive effects of oxycodone appear to be kappa-opioid receptor mediated.

Our previous studies in the Sprague-Dawley rat showed that the intrinsic antinociceptive effects of oxycodone are naloxone reversible in a manner analogous to morphine but that in contrast to morphine, oxycodone's antinociceptive effects have a rapid onset of maximum effect (approximately 5-7 min compared to 30-45 min for morphine), comprise one antinociceptive phase (compared to two phases) and are of relatively short duration (approximately 90 min compared to approximately 180 min). In the present study, administration of a range of selective opioid receptor antagonists has shown that the in

Effects of melatonin infused into the III ventricle on prolactin, beta-endorphin and luteotropin secretion in ewes during the different stages of the reproductive cycle.

BACKGROUND: Injuries constitute around 16% of the total disease burden in India with respect to Disability-Adjusted Life Years. More than two third of these injuries are unintentional and occur at household level. Domestic accidents are preventive and can be drastically reduced by effective measures and safety consciousness. This study aims to find the prevalence of domestic accidents, the household safety practices and their association with socio demographic factors in selected urban wards of Puducherry. METHODS: A population-based cross sectional analytical study was conducted in the servi

The effect of norbinaltorphimine, beta-funaltrexamine and naltrindole on NPY-induced feeding.

The non-selective opioid receptor antagonist naloxone decreases the robust feeding observed after i.c.v. injection of neuropeptide Y (NPY). In the present study we evaluated the effects of three selective opioid receptor antagonists on NPY-induced feeding. Graded doses of norbinaltorphimine (norBNI), beta-funaltrexamine (beta-FNA) and naltrindole (NTI), antagonists of the kappa, mu and delta receptors respectively, were preinjected (i.c.v.) in male rats prior to injection of 5 micrograms NPY (i.c.v.). Food intake was measured 1, 2 and 4 h post-NPY injection. Injection of beta-FNA and norBNI we

Effect of morphine and morphine-like drugs on carbachol-induced fighting in cats.

In the present experiments, morphine, methadone or pethidine was injected into the cerebral ventricle of the unanesthetized cat after fighting was induced with carbachol injected previously. The fighting evoked by carbachol was sensitive to the depressant action of morphine or pethidine but not to the depressant effect of methadone. The most likely explanation of the depressant effects of the former compounds is that they act on the postsynaptic receptors of central cholinergic neurons.

Effects of clonidine on gastric acid secretion in the rat.

The effect of clonidine on gastric acid secretion was studied in the pylorus ligated stomach (Shay test) and in the perfused stomach of the rat. Clonidine produced biphasic (inhibitory and stimulatory) effects on gastric acid secretion. In the Shay test, clonidine produced only an inhibitory effect on spontaneous gastric acid secretion in a dose-dependent manner (ED50 = 0.042 mg/kg, intraduodenal administration). In the perfused stomach, clonidine in a dose range of 0.625 to 5 mg/kg i.p. slightly increased acid secretion. The secretion, induced by 5 mg/kg of clonidine, was antagonized by cimet

Drug administration into the third ventricle of the brain of anesthetized and conscious cats.

A method is described for the administration of drugs into the third ventricle of the brain of conscious and anesthetized cats. Noradrenaline administered by this route produced cardiovascular stimulant effects as against depressor effects when administered into a lateral cerebral ventricle. The pressor responses evoked by third ventricular administration of noradrenaline were antagonized by spinal section in anesthetized cats and by hexamethonium in conscious animals. Conscious cats were more sensitive to the pressor effects of third ventricular noradrenaline than chloralose-anesthetized anim

Effects of centrally administered pentazocine and ethylketocyclazocine on thermoregulation in the cat.

The mechanisms for the biphasic response of unrestrained cats to centrally administered pentazocine were examined by injecting 1 mg pentazocine into the third cerebral ventricle at various ambient temperatures (TaS). The initial phase was hyperthermia at Ta = 34 degrees C, but at Ta = 22 degrees C hypothermia, which was enhanced at Ta = 0 degrees C, developed. The second phase was an increase in body temperature that occurred at all three TaS. No evidence was seen of compensatory thermoregulatory effector activity during development of either phase. The first phase can be attributed to general

[Effects of intracerebroventricular injections of nalorphine on feeding behaviour and rumination in sheep (author's transl)].

Two sheep were fitted with permanent cannulus in the first cerebral ventricule and permanent electrodes were stitched to the wall of the reticulum. They were housed in individual pens and fed lucern pellets twice a day (total duration 1.30 hour) at fixed hours. Food intake was weighed. Chewing movements were recorded permanently by means of a submandibular balloon. Reticular motility was recorded by electromyography during feeding periods. Feeding behaviour and rumination were previously recorded during two control periods. No treatment was applied during the first control period of 6 days (Ta

Effect of calcium on locomotor activity of mice.

Intracerebroventricular administration of calcium severely depressed the locomotor activity of unanesthetized mice, producing ataxic gait and motor incoordination. Pretreatment with pentobarbital (30 mg/kg, IP) twice daily for 2 and 6 days partially reversed this effect of calcium, suggesting that calcium and pentobarbital may in part exert their depressant effect by a similar mechanism.
