Diallyl trisulfide reduced the reproductive capacity of male Sitotroga cerealella via the regulation of juvenile and ecdysone hormones.

Environmental pollution and resistance in animals are major concerns for the application of synthetic pesticides. Diallyl trisulfide (DAT), an active compound in garlic essential oil, is a novel tool for active and safe control of agricultural insect pests. In this study, we analysed the effects of DAT (0.01 μL/L) on the protein content in male reproductive tissues (accessory glands, ejaculatory ducts, and testis), and juvenile hormone (JH) and ecdysone titres in a highly detrimental pest of stored products, Sitotroga cerealella. Evaluation of the expression profile of JH and ecdysone pathway-

Antibodies Against the Clock Proteins Period and Cryptochrome Reveal the Neuronal Organization of the Circadian Clock in the Pea Aphid.

Circadian clocks prepare the organism to cyclic environmental changes in light, temperature, or food availability. Here, we characterized the master clock in the brain of a strongly photoperiodic insect, the aphid Acyrthosiphon pisum, immunohistochemically with antibodies against A. pisum Period (PER), Drosophila melanogaster Cryptochrome (CRY1), and crab Pigment-Dispersing Hormone (PDH). The latter antibody detects all so far known PDHs and PDFs (Pigment-Dispersing Factors), which play a dominant role in the circadian system of many arthropods. We found that, under long days, PER and CRY are

Targeting the Type 1 Tyramine Receptor LsTAR1 Inhibits Reproduction, Feeding and Survival in the Small Brown Planthopper Laodelphax striatellus.

Laodelphax striatellus is one of the most destructive rice pests. However, the functions of TARs in rice pests remain largely unknown. Here, we cloned LsTAR1 from L. striatellus. LsTAR1 shares considerable sequence identity with its orthologous receptors, and clusters closely with its corresponding receptor groups. LsTAR1 was most highly expressed in the egg stage and brain of L. striatellus. Knockdown of LsTAR1 by RNA interference (RNAi) prolonged the preoviposition and oviposition period, and reduced the fecundity. Furthermore, LsTAR1 knockdown significantly decreased the mRNA levels of vite

Chemosensory protein BmCSP4 enhances silkworm adaptation to mulberry by suppressing plant defense responses.

The silkworm (Bombyx mori) feeds exclusively on mulberry leaves; however, the genetic basis underlying its adaptation to mulberry defenses remains unclear. In this study, we identified a silkworm chemosensory protein (CSP), BmCSP4, which is highly expressed in the salivary glands of larvae and secreted onto mulberry leaf wounds during feeding. Biochemical and molecular docking assays revealed that BmCSP4 binds to various mulberry volatiles, particularly cis-3-hexenyl acetate, cis-jasmone, and trans-3-hexen-1-ol. Mulberry leaves chewed by silkworms with exogenous BmCSP4 application or transgeni

Identification of a sex-determining region potentially involved in resolving genetic conflicts over sex ratio.

Sex-determining genes remain largely uncharacterized outside classical models in vertebrates and insects, leaving a gap in our understanding of their evolutionary emergence. The terrestrial isopod Armadillidium vulgare provides an excellent model for investigating this question, as it presents multiple genetic sex determinants. Some lineages possess a masculinizing dominant allele at a locus called the 'M gene', which is analogous to an XY system. This allele is hypothesized to have been selected due to the deficit of males caused by a non-Mendelian feminizing factor previously described. The

Sexual Mosaicism and Teratogenesis in Simulium guianense s.l. and S. incrustatum s.l. (Diptera: Simuliidae), Onchocerciasis vectors, from Environmentally Impacted Areas in the Yanomami Indigenous Land, Brazil.

Sexual mosaics in Simuliidae members have been detected at low frequencies in various locations worldwide. These phenotypic expressions are expected in very small amounts in any natural insect population and may result from inherent individual formation factors, such as hormonal or external elements, i.e., environmental pollution and parasitism, both of which can disrupt sexual differentiation development processes. This study assessed a Simuliidae collection sampled as a result of the Brazilian focus on endemic onchocerciasis in the Yanomami Indigenous Land in 2013, 2014, 2017, and 2021. Some

Ecdysteroid UDP-Glucosyltransferase Expression in Beauveria bassiana Increases Its Pathogenicity against Early Instar Silkworm Larvae.

Beauveria bassiana (B. bassiana) is a broad-spectrum entomopathogenic fungus that can control pests in agriculture and forestry. In this study, encoding ecdysteroid uridine diphosphate glucosyltransferase gene (egt) was successfully screened in B. bassiana on the medium containing 500μg/mL G418 sulfate solution through the protoplast transformation method. This enzyme has the function of 20E (20-hydroxyecdysone) inactivation, thus increasing the mortality of the early instar larvae infected with B. bassiana. In this study, we transformed B. bassiana with the egt gene, which deactivates 20-hydr

Targeted mutagenesis and functional analysis of adipokinetic hormone-encoding gene in Drosophila.

Adipokinetic hormones (Akhs) are small peptides (8-10 amino acid [aa] residues long) found in insects that regulate metabolic responses to stress by stimulating catabolic reactions and mobilizing energy stores. We employed Transcription activator-like effector nuclease (TALEN) mutagenesis and isolated an Akh(1) mutant carrying a small deletion in the gene that resulted in a truncated peptide; the second aa (Leu) was missing from the functional octapeptide. This null Dmel/Akh mutant is suitable to study Akh function without any effect on the C-terminal associated peptide encoded by the same gen

Neuropeptide receptors NPR-1 and NPR-2 regulate Caenorhabditis elegans avoidance response to the plant stress hormone methyl salicylate.

Methyl salicylate (MeSa) is a stress hormone released by plants under attack by pathogens or herbivores . MeSa has been shown to attract predatory insects of herbivores and repel pests. The molecules and neurons underlying animal response to MeSa are not known. Here we found that the nematode Caenorhabditis elegans exhibits a strong avoidance response to MeSa, which requires the activities of two closely related neuropeptide receptors NPR-1 and NPR-2. Molecular analyses suggest that NPR-1 expressed in the RMG inter/motor neurons is required for MeSa avoidance. An NPR-1 ligand FLP-18 is also re

dissatisfaction, a gene involved in sex-specific behavior and neural development of Drosophila melanogaster.

Few mutations link well defined behaviors with individual neurons and the activity of specific genes. In Drosophila, recent evidence indicates the presence of a doublesex-independent pathway controlling sexual behavior and neuronal differentiation. We have identified a gene, dissatisfaction (dsf), that affects sex-specific courtship behaviors and neural differentiation in both sexes without an associated general behavioral debilitation. Male and female mutant animals exhibit abnormalities in courtship behaviors, suggesting a requirement for dsf in the brain. Virgin dsf females resist males dur

In vivo interactions of the Drosophila Hairy and Runt transcriptional repressors with target promoters.

The Hairy and Runt pair-rule proteins regulate Drosophila segmentation by repressing transcription. To explore the ability of these proteins to function as promoter-bound regulators in vivo, we examined the effects of Hairy and Runt derivatives containing heterologous transcriptional activation domains (HairyAct and RunAct). Using this approach, we find that Hairy and Runt efficiently target such activation domains to specific segmentation gene promoters, leading to rapid induction of transcription. Our results strongly suggest that Hairy normally acts as a promoter-bound repressor of fushi ta

Identification of regions interacting with ovoD mutations: potential new genes involved in germline sex determination or differentiation in Drosophila melanogaster.

Only a few Drosophila melanogaster germline sex determination genes are known, and there have been no systematic screens to identify new genes involved in this important biological process. The ovarian phenotypes produced by females mutant for dominant alleles of the ovo gene are modified in flies with altered doses of other loci involved in germline sex determination in Drosophila (Sex-lethal+, sans fille+ and ovarian tumor+). This observation constitutes the basis for a screen to identify additional genes required for proper establishment of germline sexual identity. We tested 300 deletions,

Impact of mediterranean fruit fly rearing residues and biological supplementation on performance of gimmizah chicks.

For a transformation of the global food system towards sustainability, circular approaches and nutrient-rich side-stream valorization are mandatory. Moreover, affordable and sustainable alternatives to corn, soy, and fish meal are needed in poultry production. Recently, insects and their derived products have gained research interest as alternative sources of conventional feed ingredients in poultry nutrition. The Mediterranean fruit fly (medfly; Ceratitis capitata) production industry using sterile insect technology amasses thousands tonnes of rearing residues annually. This study is the firs

Resistance to Cry14A family Bacillus thuringiensis crystal proteins in Caenornabditis elegans operates via the nhr-31 transcription factor and vacuolar-type ATPase pathway.

Bacillus thuringiensis (Bt) has been successfully used commercially for more than 60 years for biocontrol of insect pests. Since 1996, transgenic plants expressing Bt crystal (Cry) proteins have been used commercially to provide protection against insects that predate on corn and cotton. More recently, Bt Cry proteins that target nematodes have been discovered. One of these, Cry14Ab, has been expressed in transgenic soybean plants and found to provide significant protection against the soybean cyst nematode, Heterodera glycines. However, to date there has been no description of high-level resi

A tyramine receptor gene LsTAR2 is involved in reproduction and feeding in the small brown planthopper Laodelphax striatellus.

Tyramine (TA) is an important biogenic amine present in the central nervous system of insects, and mediates a variety of physiological and behavioral functions via tyramine receptors (TARs). However, TARs have not yet been characterized in planthoppers, and their physiological functions remain poorly understood in rice pests. Here, we cloned a tyramine receptor gene (LsTAR2) from the small brown planthopper Laodelphax striatellus, one of the most destructive rice pests. LsTAR2 shares high sequence identity with its orthologous receptors, and is closely related to its corresponding receptor gro

Evidence for the involvement of 3-oxo-delta 4 intermediates in ecdysteroid biosynthesis.

Although the involvement of 3-oxo-delta 4 compounds as intermediates in arthropod ecdysteroid biosynthesis has been postulated for a long time, it has not yet been directly demonstrated. In the present study, 3-oxo-delta 4-steroids have been synthesized and incubated in vitro with dissociated moulting gland cells from the crab Carcinus maenas. The tritiated compounds were converted into 3-dehydroecdysone, ecdysone and/or 25-deoxyecdysone, i.e. final ecdysteroids. This means that the 3-oxo-delta 4 compounds had undergone a 5 beta-reduction, to give the 5 beta-conformation of ecdysteroids. Our r

Persistent ectopic expression of Drosophila homeotic genes resulting from maternal deficiency of the extra sex combs gene product.

Like other members of the Polycomb group, the extra sex combs gene (esc) is required for the correct repression of loci in the major homeotic gene complexes. We show here that embryos lacking both maternal and zygotic esc+ function display transient, general derepression of both the Ultrabithorax (Ubx) and Antennapedia (Antp) genes during germ band shortening, but Sex combs reduced (Scr) expression is almost normal in the epidermis and lacking in the central nervous system (CNS). In addition, embryos that are maternally esc- but receive two paternal copies of esc+ often are characterized by ec

Cestode infection is linked to transcriptional shifts in neuropeptide signalling and caste-specific ageing pathways in a social insect.

Social insects are key models for phenotypic plasticity, as queens and workers show striking differences in behaviour and lifespan despite sharing the same genome. Neuropeptides are central regulators of behavioural plasticity, and caste-specific ageing is governed by the insulin/insulin-like growth factor 1 signalling-target of rapamycin-juvenile hormone network. Parasites that manipulate host behaviour and lifespan may exploit these regulators by targeting conserved neuropeptides or by altering ageing pathways. Here, we examine a host-parasite system in which the cestode Anomotaenia brevis e

Anti-diuretic hormone ITP signals via a guanylate cyclase receptor to modulate systemic homeostasis in Drosophila.

Insects have evolved a variety of neurohormones that enable them to maintain nutrient and osmotic homeostasis. Here, we characterized the ion transport peptide (ITP) signaling system in Drosophila. The Drosophila ITP gene can generate three different peptide isoforms: ITP amidated (ITPa) and two ITP-like (ITPL1 and ITPL2) isoforms. We comprehensively characterized the expression of all three ITP isoforms in the nervous system and peripheral tissues. Our analyses reveal widespread expression of ITP isoforms. Moreover, we show that ITPa-producing neurons are activated and release ITPa during deh
