N-Acetylserotonin (NAS) inhibits the peroxidation of linoleic acid induced by a water-soluble initiator, 2,2'-azobis(2-amidinopropane) (ABAP). Lipid peroxidation was detected by the formation of conjugated dienes, monitored spectrophotometrically at 236 nm. N-Acetylserotonin, at concentrations ranging from 200 nM to 20 microM, reduced the rate of ABAP-initiated lipid peroxidation in a concentration-dependent manner. Under in vitro conditions, the pyridoindole stobadine inhibited alloxan-induced lipid peroxidation in a model biological membrane with the efficacy comparable with that of the standard Trolox. The present study was designed to determine whether the supplementation of vitamin E in the copper-deficient diet would ameliorate the severity of copper deficiency in fructose-fed rats. Ten trauma patients and 13 burns patients were studied intensively for the first 36 h and subsequent 6 days post injury in order to investigate the mechanism of trauma and burn associated proteinuria. Burns patient's initial maximum proteinuria occurred between 4 and 8 h post injury, whilst trauma patients showed greatest proteinuria within 4 h. In this study the role of free radicals and lipid peroxidation as mediators of chemically induced mucosal damage was investigated. Two enzymatic antioxidants, superoxide dismutase or catalase injected intravenously, reduced mucosal damage either by ethanol or aspirin. Lipid peroxidation is increased by ferrous or ferric ions in rat lung microsomes both from rats pretreated with 3-methylcholanthrene and from untreated controls. This increase was dependent on the concentration of these ions in the reaction mixture. Crocidolite alone increased peroxidation in microsomal fractions. However, addition of ferric or ferrous ions with the crocidolite did not give a greater increase in the amount of peroxidation in microsomes. The effects of the interactions between dietary carbohydrates and copper deficiency on superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) activities and their roles in peroxidative pathways were investigated. Weanling rats were fed diets deficient in copper and containing either 62% starch, fructose, or glucose.
