Highly efficient removal of patulin using magnetic Fe3O4 nanoparticle-immobilized Papiliotrema flavescens.

Patulin is a mycotoxin mainly produced by Penicillium expansum and commonly contaminates fruits, vegetables, and their derivative products. Patulin degradation by microorganisms or enzymes is currently the most extensively studied method; however, the broader application of these organisms and enzymes is limited by the isolation from products and the requirement of expensive cofactors. In this study, a strain of Papiliotrema flavescens LY3 with excellent patulin degradation capability was isolated. It was further immobilized with magnetic nano-Fe3O4 particles by non-covalent electrostatic inte

Smart dual-ligand-functionalized magnetic nanocomposites for rapid disruption of EpCAM-positive extracellular vesicles.

BACKGROUND: Epithelial Cell Adhesion Molecule positive (EpCAM+) extracellular vesicles (EVs) can promote cancer progression and metastasis. Thus, it is necessary to develop targeted strategies to disable EpCAM+ EVs. RESULTS: In this work, inspired by the ability of antiviral curvature-sensing peptides to disrupt highly curved lipid membranes (<300 nm diameter), we develop magnetic nanoparticles co-functionalized with curvature-sensing peptide AH-D and EpCAM aptamer (denoted AH-D/Apt@MNP) for synergistic targeting and disruption of EpCAM+ EVs. A rapid, sensitive, and straightforward bacterial capture tool -boronic acid-functionalized Fe3O4 nanoparticle- for antimicrobial susceptibility testing.

Pathogenic multi-resistant bacterial infections represent a significant and persistent threat to public health. The development of a rapid, accurate, and convenient method for antimicrobial susceptibility testing (AST) is crucial for the timely diagnosis of pathogenic bacteria in clinical settings and is essential for the rational use of antibiotics. However, the low abundance of pathogenic multi-resistant bacteria in patient samples typically necessitates a waiting period of 24 h to 3 days to obtain positive samples for AST. To address this challenge, a novel, rapid, and straightforward bacte

Non-invasive methods of monitoring Fe3O4 magnetic nanoparticle toxicity in human liver HepaRG cells using impedance biosensing and Coherent anti-Stokes Raman spectroscopic (CARS) microscopy.

Functionalized nanoparticles have been developed for use in nanomedicines for treating life threatening diseases including various cancers. To ensure safe use of these new nanoscale reagents, various assays for biocompatibility or cytotoxicity in vitro using cell lines often serve as preliminary assessments prior to in vivo animal testing. However, many of these assays were designed for soluble, colourless materials and may not be suitable for coloured, non-transparent nanoparticles. Moreover, cell lines are not always representative of mammalian organs in vivo. In this work, we use non-invasi

A magnetic nanoparticle-based microfluidic device fabricated using a 3D-printed mould for separation of Escherichia coli from blood.

Herein, A microfluidic device is described, produced with a 3D-printed master mould that rapidly separates and concentrates Escherichia coli directly from whole blood samples, enabling a reduction in the turnaround time of bloodstream infections (BSIs) diagnosis. Moreover, it promotes the cleansing of the blood samples whose complexity frequently hampers bacterial detection. The device comprises a serpentine mixing channel with two inlets, one for blood samples (spiked with bacteria) and the other for magnetic nanoparticles (MNPs) functionalized with a (bacterio)phage receptor-binding protein

From Bench to Cell: A Roadmap for Assessing the Bioorthogonal "Click" Reactivity of Magnetic Nanoparticles for Cell Surface Engineering.

In this work, we report the use of bioorthogonal chemistry, specifically the strain-promoted click azide-alkyne cycloaddition (SPAAC) for the covalent attachment of magnetic nanoparticles (MNPs) on living cell membranes. Four types of MNPs were prepared, functionalized with two different stabilizing/passivation agents (a polyethylene glycol derivative and a glucopyranoside derivative, respectively) and two types of strained alkynes with different reactivities: a cyclooctyne (CO) derivative and a dibenzocyclooctyne (DBCO) derivative. The MNPs were extensively characterized in terms of physicoch

Magnetic Particle Imaging (MPI): Experimental Quantification of Vascular Stenosis Using Stationary Stenosis Phantoms.

Magnetic Particle Imaging (MPI) is able to provide high temporal and good spatial resolution, high signal-to-noise ratio and sensitivity. Furthermore, it is a truly quantitative method as its signal strength is proportional to the concentration of its tracer, superparamagnetic iron oxide nanoparticles (SPIOs). Because of that, MPI is proposed to be a promising future method for cardiovascular imaging. Here, an interesting application may be the quantification of vascular pathologies like stenosis by utilizing the proportionality of the SPIO concentration and the MPI signal strength. In this st

Recent patents and advances on applications of magnetic nanoparticles and thin films in cell manipulation.

Cell manipulation is instrumental in most biological applications. One of the most promising methods in handling cells and other biological particles is the magnetic manipulation technique. In this technique, magnetic nanoparticles are employed to magnetize cells. Such cells then can be manipulated, sorted, or separated by applying an external magnetic field. In this work, first recent works and patents on the synthesis methods used for producing magnetic nanoparticles are investigated. These methods include co-precipitation, solvothermal, electrical wire explosion, microemulsion, laser pyroly

Magnetic boronate-Ti(IV) dual-affinity nanoparticles for simultaneous enrichment and sequential elution of glycopeptides and phosphopeptides.

The simultaneous enrichment of glycopeptides and phosphopeptides from complex peptide mixtures remains challenging because boronate affinity and immobilized metal ion affinity chromatography (IMAC) typically require different pH conditions for efficient binding. We evaluated the previously reported dual boronate-Ti(IV) magnetic nanomaterial, Mag@DBA-Ti(IV), as a sample-preparation platform for the single-sorbent, single-loading-condition enrichment and sequential elution of glycopeptides and phosphopeptides. The material, prepared by co-immobilizing a low-pKₐ boronic acid ligand and alendronat

Unveiling the Physicochemical Properties of Magnetic Nanoparticles as Solid Carriers for Laccase Immobilization Toward Different Reducing Substrates.

Laccases are multicopper oxidases capable of oxidizing a wide range of substrates using molecular oxygen as the terminal electron acceptor, producing water as the sole by-product. High-redox potential fungal laccases, such as those from Trametes versicolor, are particularly attractive for industrial and environmental applications, although their use is often limited by sensitivity to operational conditions. Enzyme immobilization represents an effective strategy to enhance laccase stability and reusability. In this work, magnetic nanoparticles (MNPs) were investigated as support for laccase imm

Intra-Mesopore Immunoassay Based on Core-Shell Structured Magnetic Hierarchically Porous ZIFs.

It is crucial yet challenging to sensitively quantify low-abundance biomarkers in blood for early screening and diagnosis of various diseases. Herein, an analytical model of intra-mesopore immunoassay (IMIA) was proposed, which was competent to examine various biomarkers at the femtomolar level. The success is rooted in the design of an innovative superparamagnetic core-shell structure with Fe3O4 nanoparticles (NPs) at the core and hierarchically porous zeolitic imidazolate frameworks as a shell (Fe3O4@HPZIF-8), achieved through a soft-template directed self-assembly coupled with confinement g

Binding Capacity and Adsorption Stability of Uremic Metabolites to Albumin-Modified Magnetic Nanoparticles.

Kidney disease causes the retention of uremic metabolites in blood, which is associated with many comorbidities. Hemodialysis does not properly clear many metabolites, including large, middle-sized, and small protein-bound uremic toxins (PBUTs). Adsorption strategies for metabolite removal require the development of engineered adsorbents with tailored surfaces to increase the binding of desired metabolites. Albumin is uniquely positioned for modifying blood-contacting surfaces to absorb uremic metabolites, as it (i) minimizes non-specific protein adsorption and (ii) binds a range of molecules
