Skin advanced glycation end-products as indicators of the metabolic profile in diabetes mellitus: correlations with glycemic control, liver phenotypes and metabolic biomarkers.

INTRODUCTION: The production of advanced glycation end-products (AGEs) is a key pathomechanism related to the complications of diabetes mellitus. The measurement of HbA1c as one of the AGEs is widely used in the clinic, but also other proteins undergo glycation in the course of diabetes. Here, we measure skin AGEs (SAGEs) in patients with diabetes type 1 (DM1) and type 2 (DM2) and correlate them with metabolic markers as well as non-invasively measured liver fibrosis and steatosis. PATIENTS AND METHODS: In this cross-sectional study, a total of 64 patients with either DM1 or DM2 and 28 health

Development of novel pyrazole-1,2,4-triazole derivatives as tyrosinase inhibitors: Design, preparation, mechanism of action and anti-browning application.

Tyrosinase (Polyphenol oxidase), a key enzyme in enzymatic browning, is an attractive target for developing new anti-browning agents in the food industry. In this work, twenty pyrazole-1,2,4-triazole derivatives (3a-3n, 4a-4f) were synthesized and tested in vitro, most of compounds showed potent anti-tyrosinase activity. Of these, 3c (IC50 = 1.02 ± 0.08 μM) was found to be 14 folds stronger than kojic acid (IC50 = 14.74 ± 1.23 μM) and behaved as a mixed type inhibitor. Besides, the disappeared peak of dopaquinone in the HPLC assay intuitively validated the inhibitory effect of 3c. Copper ions

An investigation on the effects of in vitro induced advanced glycation end-products on cortical bone fracture mechanics at fall-related loading rates.

It has been suggested that adverse changes in bone quality due to the accumulation of advanced glycation end-products (AGEs) may play a role in the increased skeletal fragility. These non-enzymatic glycation mediated crosslinks are caused due to the presence of sugars in the extracellular space and can be induced in-vitro. AGEs exist naturally in bone, but with diseases such as type-2 diabetes, they are found at higher levels. While previous studies have examined the relationships between AGE accumulation and some mechanical properties, there is a lack of understanding of how AGE accumulation

Factors influencing the physical and chemical stability of whey permeate powders: A correlational study based on industry samples.

Whey permeate is an important co-product of cheese manufacturing, commonly converted into a powder (WPP) through evaporation, lactose crystallization, and spray drying. The composition of whey permeate powders (WPPs) varies and is influenced by the properties of the cheese produced and the processing parameters used during manufacturing. WPPs may undergo physical and chemical changes, including the Maillard reaction and caking, during storage and transportation, thereby impacting the functional and physical properties of the final product. Therefore, this work aims to establish correlations be…

Ultrasound-induced protein-polysaccharide complexes: Mechanisms, functionalities and applications in food systems.

Understanding the interaction between proteins and polysaccharides is fundamental for designing food structures, yet the potential of ultrasound to precisely modulate these interactions has not been systematically reviewed. This review critically examines the mechanistic role of acoustic cavitation and its associated physical and chemical effects in driving the formation of protein-polysaccharide complexes. We highlight how ultrasound-induced shear forces, micro-turbulence, and free radical generation not only accelerate covalent conjugation via the Maillard reaction but also reinforce non-cov…

Birds and bees do not express RAGE: comparative physiology and non-conventional model organisms hold the key to hyperglycemia tolerance.

Much of the diversity of life is fueled by sugar. The Maillard reaction and subsequent formation of advanced glycated end products (AGEs) are key contributors to hyperglycemia-associated pathologies. Interestingly, birds maintain glucose levels higher than mammals yet avoid diabetic complications. This resistance is attributed to adaptations such as reduced glycation susceptibility of serum proteins, efficient glucose transport mechanisms, and metabolic routing of excess glucose into fat rather than glycogen. Birds possess physiological traits including insulin resistance, constitutive glucose…

Dietary melanoidins as emerging functional components: interactions with gut microbiota and implications for nutritional modulation of intestinal health.

Melanoidins, complex brown polymers formed during the Maillard reaction in thermally processed or fermented foods, are increasingly recognized for their nutritional relevance beyond sensory contributions. Emerging evidence suggests that they may act as prebiotic-like compounds that resist human digestion and undergo microbial fermentation in the colon, producing metabolites such as short-chain fatty acids (SCFAs). These metabolites are proposed to support intestinal barrier function, inflammation, and host metabolism. This review summarizes current knowledge on the gastrointestinal fate, micro…

Elucidating the pattern of flavor evolution during the steaming process of fermented sausages in two dimensions: Strain fermentation and steaming time.

This study elucidated the pattern of change in flavor during the steaming of fermented sausages, with a particular focus on the influence of two key factors: Strain fermentation and steaming time. The results indicated that the steaming procedure resulted in a notable loss of water, as well as protein and fat oxidation, in fermented sausages. Additionally, flavor compounds, predominantly aldehydes, esters, and Maillard reaction products, were produced in considerable quantities during the steaming of sausages. Notably, fermented sausages inoculated with LS starter (Lactiplantibacillus plantaru…

Exploring correlations between soy sauce components and the formation of thermal contaminants during low-salt solid-state fermentation.

Soy sauce is a traditional seasoning in Asia and provides a unique flavor to food. However, some harmful Maillard reaction products (MRPs) were inevitably formed during the manufacturing process. Fermentation is a critical step of soy sauce manufacturing and has a significant impact on MRPs formation. Therefore, this study investigated the formation of some characteristic MRPs (e.g., furan, carboxymethyl lysine (CML), 5-hydroxymethylfurfural (5-HMF), α-dicarbonyl compounds) and their correlation with major quality indicators (e.g., free amino acids, reducing sugar, total acid, ammonia nitrogen…

Glycosylation of oyster peptides generates supramolecular assembly of glycopeptides to relieve DSS-induced colitis.

The incidence of colitis is increasing annually, however, effective treatment options remain limited. Oyster peptides are a food source that has shown promising nutritional strategy for colitis alleviation. However, their clinical translation is hindered by poor stability and rapid in vivo metabolism. To address these limitations, we employed glycosylation modification to enhance the stability and functional properties of oyster peptides. Using a Maillard reaction-driven supramolecular assembly approach, we synthesized glycopeptides with a glycosylation degree of 42.5%. In a murine model of de…

Malondialdehyde interferes with the formation and detection of primary carbonyls in oxidized proteins.

Carbonylation is one of the most remarkable expressions of the oxidative damage to proteins and the DNPH method the most common procedure to assess protein oxidation in biological samples. The present study was elicited by two hypotheses: i) is malondialdehyde, as a reactive dicarbonyl, able to induce the formation of allysine through a Maillard-type reaction? and ii) to which extent does the attachment of MDA to proteins interfere in the assessment of protein carbonyls using the DNPH method? Human serum albumin (HSA), human hemoglobin (HEM) and β-lactoglobulin (LAC) (5 mg/mL) were incubated w
