Once-weekly D-cycloserine effects on negative symptoms and cognition in schizophrenia: an exploratory study.

BACKGROUND: Daily dosing with d-cycloserine has inconsistently improved negative symptoms in schizophrenia patients, whereas intermittent dosing significantly facilitated exposure-based therapy in two studies of patients with phobic anxiety. In animal models, single-dose administration enhances memory consolidation, but tachyphylaxis develops with repeated dosing. The objective of this exploratory study was to assess whether once-weekly dosing with d-cycloserine will produce persistent improvements in negative symptoms and cognition. METHODS: Fifty stable adult schizophrenia outpatients treat…

Forgetting, reminding, and remembering: the retrieval of lost spatial memory.

Retrograde amnesia can occur after brain damage because this disrupts sites of storage, interrupts memory consolidation, or interferes with memory retrieval. While the retrieval failure account has been considered in several animal studies, recent work has focused mainly on memory consolidation, and the neural mechanisms responsible for reactivating memory from stored traces remain poorly understood. We now describe a new retrieval phenomenon in which rats' memory for a spatial location in a watermaze was first weakened by partial lesions of the hippocampus to a level at which it could not be …

The Orbitofrontal Cortex Underlies Consolidation of Pavlovian Anticipatory Arousal Responses in Macaque Monkeys.

Altered arousal is characteristic of many mental health disorders, including major depressive disorder (MDD). Several studies link neural activity in the orbitofrontal cortex (OFC) with anticipation of reward, including anticipatory sympathetic arousal, which is blunted in MDD. We therefore studied acquisition and consolidation of appetitive pavlovian memories in two groups of adult male rhesus monkeys: unoperated controls (N = 4) and those with selective neurotoxic lesions of OFC (N = 4). The dependent measure was conditioned sympathetic arousal as indexed by differential pupil dilation, and

Functional Evidence for Memory Stabilization in Sensorimotor Adaptation: A 24-h Resting-State fMRI Study.

Adaptation learning is crucial to maintain precise motor control in face of environmental perturbations. Although much progress has been made in understanding the psychophysics and neurophysiology of sensorimotor adaptation (SA), the time course of memory consolidation remains elusive. The lack of a reproducible gradient of memory resistance using protocols of retrograde interference has even led to the proposal that memories produced through SA do not consolidate. Here, we pursued an alternative approach using resting-state fMRI to track changes in functional connectivity (FC) induced by lear…

Pharmacological manipulation of memory reconsolidation: towards a novel treatment of pathogenic memories.

Well-consolidated memories, when retrieved, may return to a transiently fragile state, and need to be consolidated again in order to be maintained. This process has been referred to as memory reconsolidation and presumably serves to modify or strengthen memory traces. In recent years, our understanding of the neurobiological mechanisms underlying this phenomenon has increased rapidly. Here, we will briefly review some of the pharmacological evidence, stressing a crucial role for the brain's major neurotransmitter systems, such as glutamate and noradrenaline, in memory reconsolidation. Pharmaco

Can sleep affect destination memory? A prospective narrative review.

The destination memory is the ability of individuals to remember to whom the information has been delivered. The memory system is an essential and critical piece of social communication and human social cognition. Previously, research has been done on the relationship between source memory and sleep which are critical and fundamental processes influencing our daily lives. However, this present prospective narrative review sheds light on the idea of beginning new research to understand the relationship between destination memory and sleep. Although no such literature exists that can explain thi

Transforming experiences: Neurobiology of memory updating/editing.

Long-term memory is achieved through a consolidation process where structural and molecular changes integrate information into a stable memory. However, environmental conditions constantly change, and organisms must adapt their behavior by updating their memories, providing dynamic flexibility for adaptive responses. Consequently, novel stimulation/experiences can be integrated during memory retrieval; where consolidated memories are updated by a dynamic process after the appearance of a prediction error or by the exposure to new information, generating edited memories. This review will discus

Schema-dependent gene activation and memory encoding in neocortex.

When new learning occurs against the background of established prior knowledge, relevant new information can be assimilated into a schema and thereby expand the knowledge base. An animal model of this important component of memory consolidation reveals that systems memory consolidation can be very fast. In experiments with rats, we found that the hippocampal-dependent learning of new paired associates is associated with a striking up-regulation of immediate early genes in the prelimbic region of the medial prefrontal cortex, and that pharmacological interventions targeted at that area can prev…

The role of telencephalic NO and cGMP in avoidance conditioning in goldfish (Carassius auratus).

Our previous study with N-methyl-D-aspartate (NMDA) receptor antagonist D-AP5 suggested that NMDA receptors were involved in learning but not memory consolidation of avoidance conditioning. The present study investigated whether nitric oxide (NO) and cyclic guanosine monophosphate (cGMP) were involved in memory consolidation but not learning of avoidance conditioning in goldfish. Experiments 1 to 3 investigated amnestic and performance effects of NO inhibitor L-NAME and cGMP inhibitor LY-83583. Experiment 4 investigated whether posttraining intratelencephalic injection of NO donor SNAP amelior…

Stimulating the Stimulated Cortex-Frontocortical Anodal Electric Stimulation Combined With Closed-Loop Acoustic Stimulation During Sleep Impairs Memory in Subjects With High Cognitive Ability.

This study investigates the impact of simultaneous anodal transcranial direct-current stimulation (tDCS) and closed-loop acoustic stimulation (CLAS) during slow-wave sleep on memory consolidation and neural oscillations. In this experiment, anodal tDCS was used to modulate cortical excitability, aiming to alter the brain state and investigate the resultant impact of CLAS-induced effects on sleep electroencephalography and overnight memory consolidation. Twenty participants (aged 18-30) completed two experimental nights involving either CLAS alone or tDCS combined with CLAS (AmodCLAS). Offline …

Maternal Deprivation Induces Memory Deficits That Are Reduced by One Aerobic Exercise Shot Performed after the Learning Session.

During the neonatal period, the brain is susceptible to external influences. Exposure to stressful events during this phase of life influences brain development and impacts adult life. In animals, the maternal deprivation (MD) model is effective in mimicking stress in the early stages of development. In contrast, physical exercise seems to be able to prevent deficits in memory consolidation. Although the effects of chronic exercise in cognition are already well established, little is known about the effects of acute aerobic exercise. Here, male Wistar rats divided into deprived (MD) and nondep…

A Lyapunov theory demonstrating a fundamental limit on the speed of systems consolidation.

The nervous system reorganizes memories from an early site to a late site, a commonly observed feature of learning and memory systems known as systems consolidation. Previous work has suggested learning rules by which consolidation may occur. Here, we provide conditions under which such rules are guaranteed to lead to stable convergence of learning and consolidation. We use the theory of Lyapunov functions, which enforces stability by requiring learning rules to decrease an energy-like (Lyapunov) function. We present the theory in the context of a simple circuit architecture motivated by class

Danger Changes the Way the Brain Consolidates Neutral Information; and Does So by Interacting with Processes Involved in the Encoding of That Information.

This study examined the effect of danger on consolidation of neutral information in two regions of the rat (male and female) medial temporal lobe: the perirhinal cortex (PRh) and basolateral amygdala complex (BLA). The neutral information was the association that forms between an auditory stimulus and a visual stimulus (labeled S2 and S1) across their pairings in sensory preconditioning. We show that, when the sensory preconditioning session is followed by a shocked context exposure, the danger shifts consolidation of the S2-S1 association from the PRh to the BLA; and does so by interacting wi

Post-training, intrahippocampal HDAC inhibition differentially impacts neural circuits underlying spatial memory in adult and aged mice.

Converging evidence indicates that pharmacologically elevating histone acetylation using post-training, systemic or intrahippocampal, administration of histone deacetylase inhibitor (HDACi) can enhance memory consolidation processes in young rodents but it is not yet clear, whether such treatment is sufficient to prevent memory impairments associated with aging. To address this question, we used a 1-day massed spatial learning task in the water maze to investigate the effects of immediate post-training injection of the HDACi trichostatin A (TSA) into the dorsal hippocampus on long-term memory …

Sleep deprivation and memory: A neurobiological perspective.

Memory consolidation and the preservation of cognitive function are fundamentally influenced by sleep. Lack of sleep affects synaptic plasticity and neurogenesis, as well as oxidative stress and neuroinflammation, all of which impair memory accuracy, cognitive flexibility, and attention. The behavioural repercussions include decreased learning ability, attentional problems, emotional dysregulation, and heightened vulnerability to false memories. This review examines the neurobiological impact of sleep deprivation on memory and cognition, emphasising how the brain systems performing these funct…

Memory Engrams in the Neocortex.

While in the past much of our knowledge about memory representations in the brain has relied on loss-of-function studies in which whole brain regions were temporarily inactivated or permanently lesioned, the recent development of new methods has ushered in a new era of downright "engram excitement." Animal research is now able to specifically label, track, and manipulate engram cells in the brain. While early studies have mostly focused on single brain regions like the hippocampus, recently more and more evidence for brain-wide distributed engram networks is emerging. Memory research in humans

Memory facilitation by naloxone is due to release of dopaminergic and beta-adrenergic systems from tonic inhibition.

The post-training IP administration of naloxone (0.8 mg/kg) facilitates memory consolidation of the habituation of a rearing response to a tone in rats. Amphetamine (1.0 - 2.5 mg/kg or nicotine (0.2 - 0.5 mg/kg), and amphetamine (2.5 mg/kg) plus nicotine (0.5 mg/kg) have no effect. The higher doses of amphetamine or nicotine, however, when given together with a dose of naloxone which is ineffective alone (0.2 mg/kg), markedly enhance consolidation. Haloperidol (0.5 mg/kg), propranolol (0.5 mg/kg), and phenoxybenzamine (2.0 mg/kg) have no effect on their own; whereas tolazoline (2.0 mg/kg) impa…

Hippocampus supports long-term maintenance of language representations: Evidence of impaired collocation knowledge in amnesia.

Traditional systems consolidation theories of memory suggest that the role of the hippocampus in maintaining memory representations diminishes over time, with learned information eventually becoming fully independent of the hippocampus. Knowledge of collocations in one's native (L1) language are acquired during development and are solidly acquired by adulthood. Remote semantic knowledge of collocations might therefore be expected to be resistant to hippocampal pathology. Patients with hippocampal damage and severe anterograde amnesia completed two tasks testing English collocation knowledge or

Status epilepticus induces chronic silencing of burster and dominance of regular firing neurons during sharp wave-ripples in the mouse subiculum.

Sharp wave-ripples (SWRs) are hippocampal oscillations associated with memory consolidation. The subiculum, as the hippocampal output structure, ensures that hippocampal memory representations are transferred correctly to the consolidating neocortical regions. Because patients with temporal lobe epilepsy often develop memory deficits, we hypothesized that epileptic networks may disrupt subicular SWRs. We therefore investigated the impact of experimentally induced status epilepticus (SE) on subicular SWRs and contributing pyramidal neurons using electrophysiological recordings in mouse hippocam…
