Electroconvulsive therapy (ECT) is a well-established intervention for treatment-resistant major depressive disorder (MDD). Although the neurobiological ramifications of ECT, particularly in relation to the mesolimbic dopaminergic system, have been correlated with the modulation of reward and reinforcement pathways, the capacity of ECT to influence substance-related behaviors remains inadequately elucidated. We report the case of a 42-year-old Iranian woman with a 14-year history of treatment-resistant MDD and no prior history of tobacco or other substance use. Following a second session of in

Drug addiction is a serious brain disorder with widespread somatic, psychological, psychiatric, and legal consequences, characterized by compulsive drug seeking, craving, withdrawal, and relapse, particularly with psychostimulants. A key feature of drugs commonly abused by humans is their ability to enhance dopamine (DA) neurotransmission in mesolimbic circuits, thereby activating DA receptors and altering emotional and motivational behaviors. Recent studies have demonstrated that dopamine D2 receptors (D2Rs), existing both as monomers and as D1/D2 receptor heteromers with dopamine D1 receptor

Evolutionarily conserved neural circuits evolved that mediate switching between feeding and foraging for food, depending on environmental co … anorexia. We highlight how circuits interconnecting the hypothalamus, prefrontal cortex, amygdala, mesolimbic system, and monoaminergic nuclei, interact to modulate animals' decision to feed or forage in the activity-based anorexia paradigm. We then highlight how these recent findings have aided in identifying pathophysiological mechanisms underlying neuropsychiat …

The mesolimbic dopamine system is crucial for drug reinforcement and reward learning, leading to addiction. We previously demonstrated that Arvcf was associated significantly with nicotine and alcohol addiction through genome-wide association studies. However, the role and mechanisms of Arvcf in dopamine-mediated drug reward processes were largely unknown. In this study, we first showed that Arvcf mediates nicotine-induced reward behavior by using conditioned place preference (CPP) model on Arvcf-knockout (Arvcf-KO) animal model. Then, we revealed that Arvcf was mainly expressed in VTA dopamin

Chronic pain attenuates midbrain dopamine (DA) transmission, as evidenced by a decrease in opioid-evoked DA release in the ventral striatum, suggesting that the occurrence of chronic pain impairs reward-related behaviors. However, mechanisms by which pain modifies DA transmission remain elusive. Using in vivo microdialysis and microinjection of drugs into the mesolimbic DA system, we demonstrate in mice and rats that microglial activation in the VTA compromises not only opioid-evoked release of DA, but also other DA-stimulating drugs, such as cocaine. Our data show that loss of stimulated extr

Drugs of abuse activate the mesolimbic dopaminergic pathway. Genetic variations in the dopaminergic system may contribute to drug addiction. Several processes are shared between cocaine and heroin addictions but some neurobiological mechanisms may be specific. This study examined the association of 98 single nucleotide polymorphisms in 13 dopamine-related genes with heroin addiction (OD) and/or cocaine addiction (CD) in a sample of 801 African Americans (315 subjects with OD ± CD, 279 subjects with CD, and 207 controls). Single-marker analyses provided nominally significant evidence for associ

Neurobiological investigation of heroin revealed that abusers of this highly addictive substance show dysregulation in brain circuits for reward processing and cognitive control. Psychologically, personality traits related to reward processing and cognitive control differed between heroin abusers and non-abusers. Yet, there is no direct evidence on the relationship between these neurobiological and psychological findings on heroin abusers, and whether such relationship is altered in these abusers. The present study filled this research gap by integrating findings obtained via magnetic resonanc

Functional neuroimaging studies have implicated an involvement of the prefrontal cortex and mesolimbic reward system (i.e., ventral striatum) in pathological gambling (PG). However, there is a lack of studies focusing on structural changes in frontostriatal brain regions in adult subjects with PG. In order to study differences in local grey matter volume, 20 male subjects with PG and 21 matched controls underwent structural magnetic resonance imaging. Structural brain data were analysed via voxel-based morphometry with a focus on prefrontal areas and ventral striatum. By comparing grey matter

BACKGROUND: Recent studies have raised concerns about subtle long-lasting neurobiological changes that might be triggered by exposure to Cannabis derivatives, especially in a critical phase of brain maturation, such as puberty. The mesolimbic dopamine (DA) system, involved in the processing of drug-induced reward, is a locus of action of cannabinoids and endocannabinoids. Thus, we compared the effects of repeated cannabinoid administration in adolescent and adult rats on DA neuronal functions and responses to drugs of abuse.

The diminished capacity to experience pleasure or reward ("anhedonia") has its biological underpinnings in the mesolimbic dopamine system and is strongly implicated in risk for a variety of addictive behaviors. The present study tested the prediction that patients with anorexia nervosa (AN) would be more anhedonic than those with bulimia nervosa (BN)-a factor that could contribute to their respective avoidance and approach relationship to food. We also tested the idea that anhedonia would be correlated with high-level exercising from the viewpoint that the latter serves as a compensatory behav

BACKGROUND: Perimenopause marks a high-risk period for psychiatric instability, often precipitating or exacerbating disorders like schizophrenia. While traditional antipsychotics can be effective, they often worsen vasomotor symptoms and carry side effects that drive high discontinuation rates. We hypothesized that hippocampal hyperactivity, resulting from a decrease in GABAergic transmission, drives mesolimbic dopamine dysregulation in this state. Thus, we proposed that this circuit-level dysfunction can be reversed by targeting α5-containing GABAA receptors (α5-GABAAR).

The neurodegenerative and neurodevelopmental hypotheses represent the basic etiological framework for the origin of schizophrenia. Additionally, the dopamine hypothesis, adopted more than two decades ago, has repeatedly asserted the position of dopamine as a pathobiochemical substrate through the action of psychostimulants and neuroleptics on the mesolimbic and mesocortical systems, giving insight into the origin of positive and negative schizophrenic symptoms. Meanwhile, cognitive impairments in schizophrenia remain incompletely understood but are thought to be present during all stages of th

The dopaminergic mesolimbic system plays a key role in the mechanisms of reinforcement elicited by alcohol (ethanol) and other drugs of abuse. Numerous lines of evidence indicate that ethanol reinforcement mechanisms involve, at least partially, the ethanol-induced activation of the endogenous opioid system. Ethanol may alter opioidergic transmission at different levels, including the biosynthesis, release, and degradation of opioid peptides, as well as binding of endogenous ligands to opioid receptors. Several studies suggest that mu and delta opioid receptors play a major role in ethanol rei…

Visual hallucinations (VH) in Parkinson's disease (PD) are a chronic complication in 30 to 60% of treated patients and have a multifaceted p … instem filtering capacities through fluctuating vigilance, and medication-related overactivation of mesolimbic systems. Different etiologies likely produce different phenomenologies and the prognosis may not be uniform. This new conceptual framework permits an anatomical view of VH and suggests new, testable hypotheses regarding their pathophysiology and therapy.

Prolonged periods of opioid use have been shown to cause neuroadaptations in the brain's reward circuitry, contributing to addictive behaviors and drug dependence. Recently, considerable focus has been placed on the role of the endocannabinoid system (ECS) and its CB receptors in opioid-driven behaviors. However, opioid-induced neuroadaptations to the ECS remain understudied. In this study, we systematically assessed CB1 receptor (CB1R) protein expression within the cortico-mesolimbic-basal ganglia circuit in rats following chronic fentanyl exposure. Male and female Long Evans rats were admini

Cocaine use disorder (CUD) is a major public health challenge with a high relapse rate and lack of effective pharmacotherapies; therefore, there is a substantial need to identify novel medications to treat this epidemic. Since the advent of glucagon-like peptide-1 (GLP-1) receptors (GLP-1Rs) agonists (GLP-1RAs), their potential has been extensively explored and expanded. In this review, we first summarized the biological effects of GLP-1, GLP-1Rs, and GLP-1RAs. Subsequently, the recent literature examining the behavioral effects and the possible pharmacological mechanisms of GLP-1RAs on CUD wa
