Inhibition of mitochondrial dynamics in Dictyostelium discoideum through the overexpression of FszA and FszB proteins.

Mitochondria play a central role in cells through energy production, calcium regulation, and cell death regulation. Dysfunction of mitochondria can impair energy production causing cellular damage which could be detrimental to an organism. Mitochondrial dynamics such as fission, fusion, and motility determine the organelle's structure and can indicate the overall health of the cell. Dictyostelium discoideum, a well-established model for mitochondrial dynamics, contains two GTPase proteins that are predicted to mediate mitochondrial dynamics, FszA and FszB. In this study, we overexpressed GFP t…

Diurnal dynamics and psychobiological regulation of cell-free mitochondrial and nuclear DNA in human saliva.

Cell-free mitochondrial DNA (cf-mtDNA) has emerged as a dynamic molecular signal responsive to psychological stress and a potential biomarker of stress-related physiological adaptations. While previous work has established the acute stress reactivity of cf-mtDNA in saliva, little is known about its diurnal regulation. In this intensive-sampling study, we characterized the diurnal dynamics of saliva cf-mtDNA across one weekday and one weekend day in healthy adults (N = 25, 52% female, 826 samples). Saliva was collected at awakening, during the first hour post-awakening, and at hourly intervals

Delay Differential Equation Approach to Oligomerization Reveals the Delay Dynamics Underlying Oscillations in Mitochondrial Fission.

Disruptions in the balance of mitochondrial fission and fusion are implicated in a host of diseases including cardiovascular, metabolic, and neurodegenerative, as well as cancer. Leinheiser et al. proposed a mechanistic model for mitochondrial fission which relies on the oligomerization of Dynamin-related protein 1 (Drp1). In this work, we propose an alternative state-dependent delay-differential equation (sdDDE) framework for mitochondrial fission, which reveals that the intrinsic delay dynamics in Drp1 oligomerization can drive oscillations in the rate of mitochondrial fission. To develop th

Cyclic tensile loading regulates nucleus pulposus cell autophagy through mitochondrial dynamics: molecular mechanisms and implications.

BACKGROUND: Chronic low back pain is predominantly driven by intervertebral disc degeneration (IVDD), a process rooted in the dysregulation of extracellular matrix (ECM) homeostasis within nucleus pulposus (NP) cells. These cells reside in a harsh microenvironment characterized by hypoxia, nutrient scarcity, and mechanical stress, making the regulatory pathways of autophagy and mitochondrial dynamics critical for their survival and function. MAIN BODY: This review synthesizes current evidence demonstrating that cyclic tensile loading (CTL) is a decisive factor directing NP cell fate through th…

Metformin Attenuates Osteoarthritis Progression by Modulating Mitochondrial Dynamics via Activation of the AMPK/Drp1 Pathway.

BACKGROUND: Osteoarthritis (OA) is a chronic degenerative disease primarily characterized by articular cartilage degradation and chondrocyte dysfunction. Mitochondrial impairment and oxidative stress in chondrocytes are pivotal contributors to OA pathogenesis. Emerging evidence suggests that metformin, beyond its role in glucose regulation, exhibits antioxidative and anti-inflammatory properties via activation of AMP-activated protein kinase (AMPK). Nonetheless, how metformin regulates mitochondrial dynamics and autophagy in OA remains to be fully elucidated. METHODS: A mouse anterior cruciat…

Hyperbaric Oxygen Attenuates Cerebral Ischemia-Reperfusion Injury Through ROS-Dependent Remodeling of Microglial Mitochondrial Dynamics.

Hyperbaric oxygen (HBO) shows neuroprotective potential in cerebral ischemia-reperfusion (CIR) injury, but its variable efficacy suggests that the underlying cellular mechanisms remain incompletely defined. We previously showed that HBO suppresses microglial NLRP3 inflammasome activation after CIR injury in a reactive oxygen species (ROS)-dependent manner; yet, how ROS couples to this effect remains unclear. Since mitochondria regulate ROS and inflammasome signaling, we investigated whether HBO modulates microglial mitochondrial dynamics in CIR injury. In adult male ICR mice (n = 71, 8-12 week…

Co-mutation Based Genetic Networks to Infer Temporal Mutation Dynamics in Ancient Human Mitochondrial Genomes.

The evolutionary history of Homo sapiens is marked by complex interactions between environmental, cultural, and genetic factors. To investigate the molecular signatures of these processes, we analyzed ancient mitochondrial DNA (mtDNA) across temporal and geographic contexts using principles of co-occurrence of minor alleles defined as co-mutation, through spatiotemporal co-mutation networks of variable sites. Haplogroup-based assessments of variable sites revealed a major transition from foraging to agrarian lifestyles during the Copper-Bronze Age. Genetic network analyses demonstrated that CO

Early Life Stress Induces Brain Mitochondrial Dynamics Changes and Sex-Specific Adverse Effects in Adulthood.

Early life stress exposure exerts detrimental effects in adulthood and is a risk factor for psychiatric disorders. Studies addressing the molecular mechanisms of early life stress have primarily focused on hormones and stress circuits. However, little is known on how mitochondria and mitochondrial dynamics (i.e., the orchestration of mitochondrial fission, fusion, mitophagy, and biogenesis) modulate early life stress responses. Here, we used a maternal separation with early weaning (MSEW) paradigm to investigate the behavioral and molecular early life stress-elicited effects in male and female…

Genomic-Encoded Mitovirus RdRp Is Required for Embryo Development and Maintaining Mitochondrial Dynamics in Arabidopsis.

Mitoviral-derived sequences are frequently detected in plant genomes, encoding an RNA-dependent RNA polymerase (RdRp). These sequences share many similarities with mitoviruses that are known to commonly infect plant mitochondria. However, the functional characterization of nuclear-encoded mitoviral-RdRp remains unclear. This study elucidates the critical role of mRdRp (AT2G07749) in maintaining mitochondrial homeostasis and embryo viability, highlighting the dual role of viral-derived genes in plant development and stress response. Phylogenetic analysis reveals that mRdRp shares 96.8% identity

BDH1 regulates cardiomyocyte apoptosis and diabetic cardiomyopathy by modulating mitochondrial dynamics and attenuating oxidative stress.

Diabetic cardiomyopathy (DCM) is a significant cardiovascular consequence of diabetes, marked by increasing myocardial dysfunction that culm … ion by inhibiting apoptosis through two primary mechanisms: (1) interaction with KIF1Bβ to modulate mitochondrial dynamics, and (2) suppression of ROS-mediated apoptosome formation (Cytc/Apaf-1 complex). These findings designate BDH1 as an innovative therapeutic target for DCM by illustrating its dual function in metabolic regulation and apoptosis inhibition, while reco …

Copper deficiency disrupts OXPHOS and mitochondrial dynamics through MTCH2-dependent copper trafficking in skeletal muscle.

Copper is an essential trace element required for mitochondrial respiration and cellular metabolism, yet its role in skeletal muscle remains incompletely understood. Here, we show that skeletal muscle-specific deletion of the high-affinity copper importer Ctr1 (SMKO) in mice leads to copper deficiency, resulting in exercise intolerance, metabolic dysfunction, and hallmarks of mitochondrial myopathy, including ragged-red fibers, lactic acidosis, and aberrant mitochondrial morphology. Copper deficiency disrupted electron transport chain proteome and induced mitochondrial hyperfusion. We identifi

SENP1 prevents high fat diet-induced non-alcoholic fatty liver diseases by regulating mitochondrial dynamics.

Mitochondrial dynamics plays a crucial role in the occurrence and development of non-alcoholic fatty liver diseases (NAFLD). SENP1, a SUMO-specific protease, catalyzes protein de-SUMOylation and involves in various physiological and pathological processes. However, the exact role of SENP1 in NAFLD remains unclear. Therefore, we investigated the regulatory role of SENP1 in mitochondrial dynamics during the progression of NAFLD. In the study, the NAFLD in vivo model induced by high fat diet (HFD) and in vitro model induced by free fatty acids (FFA) were established to investigate the role and un…

Effects of Dietary Vitamin D Deficiency on Markers of Skeletal Muscle Mitochondrial Biogenesis and Dynamics.

We examined the effects of dietary vitamin D deficiency on markers of mitochondrial biogenesis and dynamics in rat soleus muscle. Male Wistar rats were fed a chow with no vitamin D (No-D; 0 IU/kg) or a moderate dose (Mod-D; 2,000 IU/kg) of vitamin D chow for 8 wk. Compared to the Mod-D group, at 8 wk the No-D group showed significantly lower serum 25(OH)D levels. Although vitamin D deficiency had no effect on body composition, the No-D rats showed significantly decreased levels of PGC-1α, a marker of skeletal muscle mitochondrial biogenesis, and DRP1, a marker of skeletal muscle mitochondrial
