Leguminous plants nodulated by selected strains of Cupriavidus necator grow in heavy metal contaminated soils amended with calcium silicate.

Increasing concern regarding mining area environmental contamination with heavy metals has resulted in an emphasis of current research on ph … f dry matter from the aerial part of the plant, an increase in nodule number and an increase in the nitrogen fixation rate. The results of the present study demonstrate that the combination of rhizobia, leguminous plants and calcium silicate may represent a key factor in the remediation of areas contaminated by heavy metals.

Host-specific symbiotic requirement of BdeAB, a RegR-controlled RND-type efflux system in Bradyrhizobium japonicum.

Multidrug efflux systems not only cause resistance against antibiotics and toxic compounds but also mediate successful host colonization by certain plant-associated bacteria. The genome of the nitrogen-fixing soybean symbiont Bradyrhizobium japonicum encodes 24 members of the family of resistance/nodulation/cell division (RND) multidrug efflux systems, of which BdeAB is genetically controlled by the RegSR two-component regulatory system. Phylogenetic analysis of the membrane components of these 24 RND-type transporters revealed that BdeB is more closely related to functionally characterized or

Identification and functional characterization of NifA variants that are independent of GlnB activation in the photosynthetic bacterium Rhodospirillum rubrum.

The activity of NifA, the transcriptional activator of the nitrogen fixation (nif) gene, is tightly regulated in response to ammonium and oxygen. However, the mechanisms for the regulation of NifA activity are quite different among various nitrogen-fixing bacteria. Unlike the well-studied NifL-NifA regulatory systems in Klebsiella pneumoniae and Azotobacter vinelandii, in Rhodospirillum rubrum NifA is activated by a direct protein-protein interaction with the uridylylated form of GlnB, which in turn causes a conformational change in NifA. We report the identification of several substitutions i…

Synthesis and characterization of In(III) S-thiobenzoylthioglycolate complexes and their catalytic applications in CO2 fixation and multicomponent synthetic reactions.

Indium(III) complexes of S-thiobenzoylthioglycolate (Stbtg) with nitrogen-donor ligands, such as 2,2'-bipyridyl and 1,10-phenanthroline, have been synthesized. The complexes [In(1,10-phen)(Stbtg)3] (1), [In(2,2'-bipy)(Stbtg)3] (2), and a thioglycolate salt, Na[In(2,2'-bipy)(SCH2COO)2]·H2O (3), obtained by the decomposition of thiobenzoylthioglycolate complexes, were fully characterized using NMR and IR spectroscopy and single-crystal X-ray diffraction. The catalytic activities of these complexes were evaluated for two significant types of reactions. Complex 1 demonstrated exceptional catalytic

Long-Term Survival of Synechococcus and Heterotrophic Bacteria without External Nutrient Supply after Changes in Their Relationship from Antagonism to Mutualism.

Marine phytoplankton and heterotrophic bacteria share a very close but usually changeable relationship. However, the ultimate fate of their … nship with Synechococcus. In the final mutualism stage, a self-sufficient nitrogen cycle, including nitrogen fixation, denitrification, and organic nitrogen degradation, contributed to the healthy survival of Synechococcus for 2 years without an exogenous nutrient supply. This natural selective trait of Synechococcus and heterotrophic bacteria toward mutualism unde …

Mineral nutrition of plants: a short history of plant physiology.

The development of the knowledge on the mineral nutrition of plants begins between the 17th and 18th centuries when some European naturalist … ms related to some indispensable mineral salts, while Boussingault and Ville attempted to prove the nitrogen fixation from air without giving any convincing evidence. Liebig was the pioneer of the agricultural chemistry: he epitomised the knowledge of that period by imposing the so-called "law of the minima", already acknowledged by Sprengel, and patronised the use …

Metabolite shift in Medicago truncatula occurs in phosphorus deprivation.

Symbiotic nitrogen (N) fixation entails successful interaction between legume hosts and rhizobia that occur in specialized organs called nodules. N-fixing legumes have a higher demand for phosphorus (P) than legumes grown on mineral N. Medicago truncatula is an important model plant for characterization of effects of P deficiency at the molecular level. Hence, a study was carried out to address the alteration in metabolite levels of M. truncatula grown aeroponically and subjected to 4 weeks of P stress. First, GC-MS-based untargeted metabolomics initially revealed changes in the metabolic prof

Dynamic proteome analysis of Cyanothece sp. ATCC 51142 under constant light.

Understanding the dynamic nature of protein abundances provides insights into protein turnover not readily apparent from conventional, static mass spectrometry measurements. This level of data is particularly informative when surveying protein abundances in biological systems subjected to large perturbations or alterations in environment such as cyanobacteria. Our current analysis expands upon conventional proteomic approaches in cyanobacteria by measuring dynamic changes of the proteome using a (13)C(15)N-l-leucine metabolic labeling in Cyanothece ATCC51142. Metabolically labeled Cyanothece A

[Adenosine-3':5'-monophosphate phosphodiesterase from Rhizobium].

The phosphodiesterase (PDE) activity of adenosine-3':5'-monophosphate was detected in the cells of tubercular bacteria of Rhizobium lupini and Rhizobium japonicum. The specific activity of three Rhizobium forms, e.g. bacteroids from lupine root tubercles, free-nitrogen-fixing culture and vegetative cells grown on a mannitol--yeast agar, were compared. In the bacteroids PDE is represented both by soluble and membrane-bound forms. The optimal enzyme activity is revealed in an alkaline medium, whereas the curve of PDE activity dependence on pH has a broad maximum. PDE is inhibited by methylxanthi

Yeast single-cell proteins from CO2 with tandem biological systems.

Production of customized yeast single-cell protein (SCP) from CO2 offers a sustainable avenue to convert a major carbon emission process into a carbon-conserving production route of food and feed. Here, we highlight a specific approach for SCP production from CO2 using sequentially connected yet spatially separated tandem biological systems, which consist of a first unit for CO2 fixation followed by a second unit for precision fermentation toward customized SCPs. We discuss the design principles of the system, review current efforts to convert CO2 using natural and engineered autotrophs, and o

Nutrient limitations on photosynthesis: from individual to combinational stresses.

Liebig's law of the minimum states that increasing photosynthetic productivity on nutrient-impoverished soils depends on addressing the most limiting nutrient. Research has identified the roles of different mineral nutrients in photosynthetic processes. However, diffusional and biochemical regulation of photosynthesis both feature patterns of cumulative effects that jointly determine photosynthetic capacity. More importantly, responses to multiple nutrient stresses are not simply additive and require a comprehensive understanding of how these stresses interact and impact photosynthetic perform

The respiratory enzyme complex Rnf is vital for metabolic adaptation and virulence in Fusobacterium nucleatum.

This paper illuminates the significant question of how the oral commensal Fusobacterium nucleatum adapts to the metabolically changing environments of several extra-oral sites such as placenta and colon to promote various diseases as an opportunistic pathogen. We demonstrate here that the highly conserved Rhodobacter nitrogen-fixation complex, commonly known as Rnf complex, is key to fusobacterial metabolic adaptation and virulence. Genetic disruption of this Rnf complex causes global defects in polymicrobial interaction, biofilm formation, cell growth and morphology, hydrogen sulfide producti

Phosphite utilization by the globally important marine diazotroph Trichodesmium.

Species belonging to the filamentous cyanobacterial genus Trichodesmium are responsible for a significant fraction of oceanic nitrogen fixation. The availability of phosphorus (P) likely constrains the growth of Trichodesmium in certain regions of the ocean. Moreover, Trichodesmium species have recently been shown to play a role in an emerging oceanic phosphorus redox cycle, further highlighting the key role these microbes play in many biogeochemical processes in the contemporary ocean. Here, we show that Trichodesmium erythraeum IMS101 can grow on the reduced inorganic compound phosphite as i…

Fate of nitrogen-fixing bacteria in crude oil contaminated wetland ultisol.

The effect of crude oil on the growth of legumes (Calopogonium muconoides and Centrosema pubescens) and fate of nitrogen-fixing bacteria in wetland ultisol was investigated using standard cultural techniques. The results revealed observable effects of oil on soil physico-chemistry, plant growth and nodulation as well as on densities of heterotrophic, hydrocarbonoclastic and nitrogen fixing bacteria. The effects however varied with different levels (0.5%, 1%, 5%, 10%, 15% and 20%) of pollution. Ammonium and nitrate levels were high in the unpolluted soil but decreased with increase in pollution

Energy-Efficient Cascade Synthesis of Ammonia from Air via Spark Plasma-Mediated Nitrite Generation and CuRu Nanoalloy Electrocatalysis.

Plasma-electrocatalytic cascade ammonia synthesis offers a promising route for decentralized nitrogen fixation, yet remains hindered by excessive NO3 - formation and poor energy matching between the two stages. Here, we propose a strategy that breaks this bottleneck by selectively generating NO2 - via spark-discharge plasma and efficiently reducing it over a CuRu0.5 nanoalloy. Systematic optimization of the plasma discharge and electrocatalytic interface enables synergistic energy matching, delivering a record-low specific energy consumption of 273.41 kWh/kg NH3. Mechanistic studies reveal tha…

Nitrogen budgets in Japan from 2000 to 2015: Decreasing trend of nitrogen loss to the environment and the challenge to further reduce nitrogen waste.

The benefits of the artificial fixation of reactive nitrogen (Nr, nitrogen [N] compounds other than dinitrogen), in the form of N fertilizers and materials are huge, while at the same time posing substantial threats to human and ecosystem health by the release of Nr to the environment. To achieve sustainable N use, Nr loss to the environment must be reduced. An N-budget approach at the national level would allow us to fully grasp the whole picture of Nr loss to the environment through the quantification of important N flows in the country. In this study, the N budgets in Japan were estimated f
