Hub genes for early diagnosis and therapy of adamantinomatous craniopharyngioma.

BACKGROUND: Adamantinomatous craniopharyngioma (ACP) is a subtype of craniopharyngioma, a neoplastic disease with a benign pathological phenotype but a poor prognosis in the sellar region. The disease has been considered the most common congenital tumor in the skull. Therefore, this article aims to identify hub genes that might serve as genetic markers of diagnosis, treatment, and prognosis of ACP. METHODS: The procedure of this research includes the acquisition of public data, identification and functional annotation of differentially expressed genes (DEGs), construction and analysis of prot

Changes in the chemical composition of mineralised teeth in children after antineoplastic treatment.

INTRODUCTION: Chemotherapy, neoplasms, and their complications linked to malabsorption, malnutrition, and metabolic disorders may lead to improper tooth development and frequent severe caries in patients during/after antineoplastic treatment and to a more frequent improper tooth development in patients undergoing chemotherapy during odontogenesis. However, the causes of these abnormalities remain unknown; there are no studies on the impact of antineoplastic treatment and its complications on the chemical composition of mineralised teeth. AIM OF THE STUDY: To compare the chemical composition o

Accelerated enamel mineralization in Dspp mutant mice.

Dentin sialophosphoprotein (DSPP) is one of the major non-collagenous proteins present in dentin, cementum and alveolar bone; it is also transiently expressed by ameloblasts. In humans many mutations have been found in DSPP and are associated with two autosomal-dominant genetic diseases - dentinogenesis imperfecta II (DGI-II) and dentin dysplasia (DD). Both disorders result in the development of hypomineralized and mechanically compromised teeth. The erupted mature molars of Dspp(-/-) mice have a severe hypomineralized dentin phenotype. Since dentin and enamel formations are interdependent, we

The effect of fluoride on the developing tooth.

This review aims to outline the effects of fluoride on the biological processes involved in the formation of tooth tissues, particularly dental enamel. Attention has been focused on mechanisms which, if compromised, could give rise to dental fluorosis. The literature is extensive and often confusing but a much clearer picture is emerging based on recent more detailed knowledge of odontogenesis. Opacity, characteristic of fluorotic enamel, results from incomplete apatite crystal growth. How this occurs is suggested by other changes brought about by fluoride. Matrix proteins, associated with the

Evaluating dental developmental abnormalities in osteogenesis imperfecta: associations with timing and cumulative exposure to intravenous pamidronate- a pilot study.

OBJECTIVES: Bisphosphonates are widely used in pediatric Osteogenesis Imperfecta (OI), but their potential impact on odontogenesis is not well characterized. We examined whether intravenous pamidronate exposure is associated with specific developmental dental abnormalities in OI, with particular attention to treatment timing, duration, cumulative exposure, and gender. MATERIALS AND METHODS: Fifty-eight individuals with OI (38 treated and 20 untreated) underwent standardized clinical examination and panoramic radiography. Outcomes included enamel hypoplasia, dentinogenesis imperfecta (DI), den

Molecular Profiling of Odontoclasts during Physiological Tooth Replacement.

The odontoclast is a rarely studied cell type that is overly active in many dental pathologies, leading to tooth loss. It is difficult to find diphyodont mammals in which either physiological or pathological root resorption can be studied. Here we use the adult leopard gecko, which has repeated cycles of physiological tooth resorption and shedding. RNA-seq was carried out to compare gene expression profiles of functional teeth to developing teeth. Genes more highly expressed in bell-stage developing teeth were related to morphogenesis (PTHLH, SFRP2, SHH, EDAR). Some genes expressed in osteocla

Abnormalities in Tooth Formation after Early Bisphosphonate Treatment in Children with Osteogenesis Imperfecta.

Treatment with intravenous bisphosphonate (BP) in children and adolescents with osteogenesis imperfecta (OI) started in Sweden in 1991. No human studies on the role of BP therapy in development of disturbances in tooth mineralization or tooth morphology have been published. The study cohort comprised 219 individuals who were divided into four groups: group 1, BP treatment onset before 2 years of age (n = 22); group 2, BP treatment onset between 2 and 6 years of age (n = 20); group 3, BP treatment onset between 6 and 10 years of age (n = 13); and a control group of patients with OI who had not

Fate of Worn-Out Functional Teeth in the Upper Jaw Dentition of Sicyopterus japonicus (Gobioidei: Sicydiinae) During Tooth Replacement.

Mochizuki and Fukui (Jpn J Ichthyol 30 () 27-36) studied the development and replacement of the upper jaw teeth in a Japanese fish species, Sicyopterus japonicus (Gobioidei: Sicydiinae), and they reported that worn-out functional teeth in the upper jaw were not shed outside the skin but were taken into the soft tissue of the upper jaw and completely resorbed there. To date, however, this phenomenon appears poorly documented. Furthermore, the mechanism for the resorption of these teeth remains to be determined. In this study, we examined this phenomenon by using 3D microcomputed tomography (m-C

Permanent maxillary canines - review of eruption pattern and local etiological factors leading to impaction.

The position of the permanent maxillary canine at the angle of the mouth is strategically significant in maintaining the harmony and symmetry of the occlusal relationship. However, the maxillary canine is the second most frequently impacted tooth, with prevalence reported to be between 1% and 2%. Moreover, treatment of this condition is often complex and involves substantial time and financial cost. Hence, it is only prudent to monitor the eruption and identify the etiological factors that lead to impaction of the maxillary canine. Numerous researchers have tried to identify specific and nonsp

The genetics of human tooth agenesis: new discoveries for understanding dental anomalies.

The important role of genetics has been increasingly recognized in recent years with respect to the understanding of dental anomalies, such as tooth agenesis. The lack of any real insight into the cause of this condition has led us to use a human molecular genetics approach to identify the genes perturbing normal dental development. We are reporting a strategy that can be applied to investigate the underlying cause of human tooth agenesis. Starting with a single large family presenting a clearly recognizable and well-defined form of tooth agenesis, we have identified a defective gene that affe

Chondroitin sulfates in developing mouse tooth germs. An immunohistochemical study with monoclonal antibodies against chondroitin-4 and chondroitin-6 sulfates.

The role of glycosaminoglycans and proteoglycans during ontogenesis is not known. The developing tooth offers a potentially important model for studies of structure-function relationships. In this study, we have analysed the temproal and spatial expression of chondroitins of differing sulfation patterns in embryonic molars and incisors. For this purpose, we have used monoclonal antibodies (Mabs) specific for unsulfated, 4-sulfated, and 6-sulfated forms of chondroitin in conjunction with indirect immunofluorescence or immunoperoxidase labeling. Unsulfated chondroitin was not detected in embryon

Hypodontia in the beagle after perinatal whole-body 60Co gamma irradiation.

As part of a long-term study to evaluate health effects of pre- and postnatal irradiation, dental development was examined. Beagles were irradiated in utero at 8, 28, or 55 days postcoitus or postnatally at 2, 70, or 365 days postpartum. Whole-body 60Co gamma radiation doses ranged from 0 to 3.8 Gy. There was an age-dependent dose-related increase in premolar hypodontia for animals irradiated at 55 days postcoitus or 2 days postpartum with doses of 0.83 Gy or higher and for those irradiated at 28 days postcoitus with 1.2 Gy or higher.

Longitudinal growth and dental development in osteosarcoma patients.

The body height at 8.0 years and at the time of diagnosis, as well as the dental maturity, were analysed in a sample of patients with classical and primary osteosarcoma, registered in Sweden between 1972 and 1974, in order to investigate whether these patients have an accelerated body height growth. In contrast to previous reports the height development was found to be normal. The dental maturity was also normal.

The role of ascorbic acid on the structural integrity of developing tooth germs.

Tooth germs grown in ascorbate deficient medium for up to 20 days underwent progressive and widespread changes. Proliferation and differentiation of preameloblasts and preodontoblasts progressed normally. Newly differentiated odontoblasts, however, became vacuolated when they began secreting: this suggested a metabolic disturbance. Failure to maintain differentiated odontoblasts, ameloblasts and pulpal cells resulted in aberrant dentin matrix, cessation of dentin production, and finally overall structural collapse with loss of normal morphology. Biochemical studies then were undertaken to defi

Deciduous tooth chronology in the mandible of the domestic pig.

Fetuses of known age, collected from 20 days' gestation to term, were used to characterize the chronology of deciduous tooth development within the right mandible of swine. Tooth development was first observed at 32 days' embryonic development, with the differentiation of the deciduous third molar. Bud stages for the remaining deciduous teeth differentiated within the period of 32 to 38 days of embryonic development. Although the initial histological appearance of these teeth covered a short period of time, it was apparent that each tooth continued to develop at its own rate. The deciduous sec
