The activity and distribution of 10 enzymes was determined in the ruptured knee meniscus of 23 patients, when meniscus was operatively removed. The activities of NADH and SDH indicating oxydative energy metabolism were low in the ruptured meniscus as well as in the synovium close to it. On the contrary, NADPH and LDH, indicating anaerobic energy metabolism and G-6-PDH as an indicator of pentose-phosphate shunt, showed moderate or high activity. The activities of GLDH, ATPase, AcPase, AlPase, and LAPase were low in the meniscus tissue, but moderate and sometimes high in synovial tissue and fibr

Pulmonary hypertension (PH) is a disease of hyperplasia of pulmonary vascular cells. The pentose phosphate pathway (PPP)-a fundamental glucose metabolism pathway-is vital for cell growth. Because treatment of PH is inadequate, our goal was to determine whether inhibition of glucose-6-phosphate dehydrogenase (G6PD), the rate-limiting enzyme of the PPP, prevents maladaptive gene expression that promotes smooth muscle cell (SMC) growth, reduces pulmonary artery remodeling, and normalizes hemodynamics in experimental models of PH. PH was induced in mice by exposure to 10% oxygen (Hx) or weekly inj

Three strains of Gram-negative, aerobic, yellow-pigmented, chemo-organotrophic bacteria, motile by a polar flagellum, were isolated from the rhizosphere of spring barley (Hordeum vulgare L.) at a research field near Copenhagen, Denmark. The three strains, LJ79, LJ96T and LJ99, formed visible colonies on one-tenth-strength tryptic soy broth supplemented with agar (1/10 TSBA) after incubation for 6 days at 15 degrees C. The strains hydrolysed starch, casein (skimmed milk), gelatin and various pentoses and hexoses and grew on MacConkey agar and full-strength TSBA. Growth on 1/10 TSBA occurred at …

Previously, we demonstrated that disruption of ribose-5-phosphate biosynthesis in the pentose phosphate pathway increases the sensitivity of E. coli bacteria to a broad spectrum of antibiotics, and this sensitivity can be suppressed by deletion of the deoB gene. In this study, we showed that antibiotic sensitivity can also be suppressed by the deletion of the apt gene, which, along with the deoBD genes, constitutes the salvage pathway for purine base uptake. We propose that in the rpiAB mutant, these genes, together with the prs gene responsible for phosphoribosyl pyrophosphate (PRPP) synthesi

Acute lung injury (ALI) is a disease with high morbidity and mortality caused by multiple heterogeneous aetiological factors. Neutrophils play an important role in the development and progression of ALI. Neutrophil metabolic reprogramming, including sugars, pentose phosphates, lipids, amino acids and others, is thought to be involved in the regulation of neutrophil functions such as chemotaxis, phagocytosis and bacteriostasis. These functions contribute to inflammatory activation in ALI, and excessive inflammation may further exacerbate lung injury. Previous studies have focused on neutrophil

On the basis of clinico-laboratory examination of 500 patients suffering from diabetes mellitus a comparison was made of the influence of treatment with insulin and sulfanilamides on disturbed metabolic processes promoting development of vascular affections. There were found no differences in the action of insulin and oral agents on the indices of lipid, protein and mucopolysaccharide metabolism and on activity of the enzymes of the pentose-phosphate cycle. Corticosteroid excretion normalized with the compensation of diabetes, irrespective of the type of the sugar-reducing preparations. A more

BACKGROUND: Diabetic cardiomyopathy (DCM) is a major myocardial complication of type 2 diabetes and a contributor to heart failure. However, coordinated remodeling across the cardiac transcriptome, proteome and metabolome in DCM remains incompletely defined.
METHODS: Type 2 diabetes was induced in mice by a high-fat diet combined with single-dose streptozotocin. DCM was characterized by echocardiography, histology, transmission electron microscopy, and systemic metabolic and inflammatory assessments. Left ventricular myocardium underwent RNA sequencing, quantitative proteomics and untargeted

The efficient fermentation of D-xylose by yeasts remains a major bottleneck for the sustainable production of second-generation bioethanol, as this pentose represents a substantial fraction of hemicellulosic sugars in lignocellulosic biomass. While conventional industrial yeasts show limited native capacity for D-xylose utilization, numerous autochthonous (native or wild-derived) yeasts isolated from natural environments harbor metabolic traits that have been naturally selected and enable effective pentose assimilation under challenging conditions. Understanding and harnessing this diversity i

BACKGROUND: Emerging perspectives on tumor metabolism reveal its heterogeneity, a characteristic yet to be fully explored in gliomas. To advance therapies targeting metabolic processes, it is crucial to uncover metabolic differences and identify distinct metabolic subtypes. Therefore, we aimed to develop a classification system for gliomas based on the enrichment levels of four key metabolic pathways: glutaminolysis, glycolysis, the pentose phosphate pathway, and fatty acid oxidation.
METHODS: Energy-related features of glioma were characterized through integrative analyses of multiple datase

This review article delves into the complexities of granuloma formation, focusing on the metabolic reprogramming within these immune structures, especially in tuberculosis and sarcoidosis. It underscores the role of the monocyte-macrophage lineage in granuloma formation and maintenance, emphasizing the adaptability of these cells to environmental cues and inflammatory stimuli. Key to the discussion is the macrophage polarization influenced by various cytokines, with a detailed exploration of the metabolic shifts towards glycolysis under hypoxic conditions and the utilization of the pentose pho

Mature human erythrocytes are dependent on anerobic glycolysis, i.e. catabolism (oxidation) of one glucose molecule to produce two ATP and two lactate molecules. Proliferating tumor cells mimick mature human erythrocytes to glycolytically generate two ATP molecules. They deliberately avoid or switch off their respiration, i.e. tricarboxylic acid (TCA) cycle and oxidative phosphorylation (OXPHOS) machinery and consequently dispense with the production of additional 36 ATP molecules from one glucose molecule. This phenomenon is named aerobic glycolysis or Warburg effect. The present review deals

Tumor-associated macrophages (TAMs) possess both tumor-promoting and tumor-inhibiting roles. Here, we explore TAMs' anti-tumor functions, focusing on the immune responsive gene 1 (IRG1) and its product, itaconate, in lung cancer development. Spatial metabolomics reveals that endogenous itaconate is markedly depleted within lung tumor regions compared with adjacent non-tumor tissue. Single-cell RNA sequencing shows that macrophages are the primary cells expressing IRG1 in human and mouse lung tumors. Both IRG1 knockout and transplantation of IRG1-depleted bone marrow leads to increased lung tum

Doxorubicin (DOX), although effective in treating cancer, has significant cardiac side effects, which limit its clinical utility. In this study, we collected time-course transcriptomics and metabolomics data from the human cardiomyocyte cell line AC16, which we analyzed along with curated public transcriptomics data on DOX-induced toxicity. We developed a multiomics analysis workflow and a computational toolbox, pipeGEM, to integrate RNA-seq data with metabolic models, enabling the simulation of DOX-induced metabolic perturbations at a sample-specific level. Our results revealed that DOX affec

Glycogen storage disease type Ia (GSD-Ia) is caused by a deficiency in glucose-6-phosphatase-α (G6Pase-α or G6PC), a key enzyme in endogenous glucose production. This autosomal recessive disorder is characterized by impaired glucose homeostasis and long-term complications of hepatocellular adenoma/carcinoma (HCA/HCC). We have shown that hepatic G6Pase-α deficiency-mediated steatosis leads to defective autophagy that is frequently associated with carcinogenesis. We now show that hepatic G6Pase-α deficiency also leads to enhancement of hepatic glycolysis and hexose monophosphate shunt (HMS) that

Coffee extracts contain numerous bioactive compounds. Given the dietary link between coffee consumption and colorectal cancer, this study compared the effects of roasted and green (unroasted) coffee extracts on human colorectal cancer cells (HCT116) and non-cancerous fibroblasts (BJ-5ta) to evaluate how processing influences proliferation and molecular signaling. Real-time cell analysis (RTCA), qRT-PCR, and label-free quantitative proteomic analysis were performed to assess cellular responses. MTS and RTCA showed that in BJ-5Ta fibroblasts, coffee extracts increased proliferation in the order
