Maternal epigenetic signatures are associated with small for gestational age births among black women.

Approximately 10% US infants are born small for gestational age (SGA), a condition linked to increased morbidity and mortality. Infants born to Black women are twice as likely to be SGA compared with those born to White women. Although maternal factors, including epigenetic modifications, likely contribute to SGA, the underlying biological mechanisms remain poorly understood. We evaluated whether epigenetic modifications in early pregnancy were associated with SGA among pregnant Black women. We analyzed data from 931 pregnant non-Hispanic Black women (6-13 weeks of gestation) enrolled in the N

Oleate attenuates palmitate-induced endoplasmic reticulum stress and apoptosis in placental trophoblasts.

Pre-pregnancy obesity is increasingly common and predisposes pregnant women and offspring to gestational diabetes, pre-eclampsia, fetal growth abnormalities and stillbirth. Obese women exhibit elevated levels of the two most common dietary fatty acids, palmitate and oleate, and the maternal blood containing these nutrients bathes the surface of trophoblasts of placental villi in vivo We test the hypothesis that the composition and concentration of free fatty acids modulate viability and function of primary human villous trophoblasts in culture. We found that palmitate increases syncytiotrophob

Melatonin Partially Attenuates Oxycodone-Induced Placental Stress Signaling and Fetal Brain Apoptosis in a Sex-Specific Manner.

BACKGROUND: Maternal oxycodone (oxy) exposure can disrupt placental function and fetal neurodevelopment, but the molecular mechanisms remain unclear. We investigated whether prenatal oxy exposure activates inflammation and stress response pathways in the placenta and fetal brain, and if maternal melatonin supplementation attenuates these effects. METHODS: Female Sprague-Dawley rats received either saline or oxy via oral gavage for 15 days before mating (10-15mg/kg/day dose escalation) and throughout pregnancy (15mg/kg/day). From gestational day (GD) 12.5, half of the dams received melatonin (

Viscoelasticity quantification by ultrasound for placental assessment in healthy versus gestational diabetes mellitus pregnancies.

INTRODUCTION: Viscosity is a novel parameter, recently introduced in the use of elastographic techniques. Ultrasound viscoelastic imaging quantifies tissue stiffness and viscosity. This study investigated its application for quantitatively assessing placental viscoelastic parameters and identifying functional alterations in healthy versus gestational diabetes mellitus (GDM) pregnancies. METHODS: Pregnant women undergoing mid-trimester checkups (Jan-Apr 2025) at Zhongda Hospital were prospectively enrolled. Based on Oral Glucose Tolerance Test (OGTT) results, participants were categorized into

Plasma unconjugated estriol values in high-risk pregnancy.

The usefulness of plasma estriol concentrations in the management of high-risk pregnancy is controversial. In this study, serial plasma unconjugated estriol (E3) concentrations determined by radioimmunoassay were evaluated in 321 patients with high-risk pregnancy for correlation with fetal and neonatal performance. A plasma E3 concentration of 4 ng. per milliliter or less in late pregnancy correlated significantly with low one-and five-minute Apgar scores and selected neonatal problems in pregnancies complicated by chronic hypertension and intrauterine growth retardation, with a similar trend

Malaria in pregnancy: baby steps.

PURPOSE OF REVIEW: Malaria threatens pregnant women and their babies, particularly in Africa. RECENT FINDINGS: This century, the number of women at risk of malaria in pregnancy has decreased globally, apart from in Africa, where it has increased. Low and sub microscopic infections are increasingly documented but remain hard to diagnose with current point-of-care tests, and their contribution to morbidity and transmission are unclear. Artemether-lumefantrine has been endorsed for treatment in first trimester, but many women attend antenatal clinics later in pregnancy, and reaching high-risk yo

Mechanical characterization of arteries affected by fetal growth restriction in guinea pigs (Cavia porcellus).

Fetal growth restriction (FGR) is a perinatal condition associated with a low birth weight that results mainly from maternal and placental constrains. Newborns affected by this condition are more likely to develop in the long term cardiovascular diseases whose origins would be in an altered vascular structure and function defined during fetal development. Thus, this study presents the modeling and numerical simulation of systemic vessels from guinea pig fetuses affected by FGR. We aimed to characterize the biomechanical properties of the arterial wall of FGR-derived the aorta, carotid, and fem

An integrated model of preeclampsia: a multifaceted syndrome of the maternal cardiovascular-placental-fetal array.

Maternal tolerance of the semiallogenic fetus necessitates conciliation of competing interests. Viviparity evolved with a placenta to mediate the needs of the fetus and maternal adaptation to the demands of pregnancy and to ensure optimal survival for both entities. The maternal-fetal interface is imagined as a 2-dimensional porous barrier between the mother and fetus, when in fact it is an intricate multidimensional array of tissues and resident and circulating factors at play, encompassing the developing fetus, the growing placenta, the changing decidua, and the dynamic maternal cardiovascul

Discrete placental gene expression signatures accompany diabetic disease classifications during pregnancy.

BACKGROUND: Gestational diabetes mellitus affects up to 10% of pregnancies and is classified into subtypes gestational diabetes subtype A1 (GDMA1) (managed by lifestyle modifications) and gestational diabetes subtype A2 (GDMA2) (requiring medication). However, whether these subtypes are distinct clinical entities or more reflective of an extended spectrum of normal pregnancy endocrine physiology remains unclear. OBJECTIVE: Integrated bulk RNA-sequencing (RNA-seq), single-cell RNA-sequencing (scRNA-seq), and spatial transcriptomics harbors the potential to reveal disease gene signatures in sub

Birthweight differences, the transfusion syndrome and the cognitive development of monozygotic twins.

Monochorionic twins may differ from dichorionic monozygotic (MZ) twins because of the transfusion syndrome and the timing of the cleavage of the zygote. Intrapair birthweight differences may be an indicator of these intrauterine variables. Previous evidence concerning weight differences and intellectual ability in MZ twins is reviewed with recommendations that future research also incorporates full placental data. Poorer scores on a nonverbal test are found for the lighter male twin of pairs with large intrapair differences in birthweight. Co-twin concordance rates for test scores are also hig

Maternal Licorice Consumption During Pregnancy and Pubertal, Cognitive, and Psychiatric Outcomes in Children.

Earlier puberty, especially in girls, is associated with physical and mental disorders. Prenatal glucocorticoid exposure influences the timing of puberty in animal models, but the human relevance of those findings is unknown. We studied whether voluntary consumption of licorice, which contains glycyrrhizin (a potent inhibitor of placental 11β-hydroxysteroid dehydrogenase type 2, the "barrier" to maternal glucocorticoids), by pregnant women was associated with pubertal maturation (height, weight, body mass index for age, difference between current and expected adult height, Tanner staging, scor

Stem Cell-Based Trophoblast Models to Unravel the Genetic Causes of Human Miscarriages.

Miscarriage affects approximately 15% of clinically recognized pregnancies, and 1-3% of couples experience pregnancy loss recurrently. Approximately 50-60% of miscarriages result from chromosomal abnormalities, whereas up to 60% of euploid recurrent abortions harbor variants in candidate genes. The growing number of detected genetic variants requires an investigation into their role in adverse pregnancy outcomes. Since placental defects are the main cause of first-trimester miscarriages, the purpose of this review is to provide a survey of state-of-the-art human in vitro trophoblast models tha

MMP8 Promotes NETosis in Gestational Diabetes Mellitus.

Background: Neutrophil extracellular traps (NETs) have been known to be involved in the gestational diabetes mellitus (GDM), but the underlying role remains poorly understood. Methods: We conducted an integrated analysis of bulk RNA-seq data, single-cell transcriptomic sequencing (scRNA-seq) data, clinical laboratory findings, and cell models to identify hub genes linked to NET formation (NETosis) and to elucidate how NETs contribute to placental injury in GDM. Results: We found that circulating NET levels were increased in pregnant women with GDM both under fasting conditions and following an

Functional magnetic resonance imaging (magnetization transfer) and stereological analysis of human placentae in normal pregnancy and in pre-eclampsia and intrauterine growth restriction.

Magnetic resonance imaging provides a non-invasive method for investigating functional changes in the human placenta in vivo. In this study, we combine a magnetic resonance imaging technique called magnetization transfer with established stereological methods in order to analyse and compare placentae from normal (16-36 weeks of gestation) and complicated (pre-eclampsia, intrauterine growth restriction) pregnancies. Magnetization transfer provided an in vivo measure of the ratio of bound protons:total protons and stereological analysis of histological sections was used to estimate a residual:to

Diagnostic Approach to Abnormal Alkaline Phosphatase Value.

Alkaline phosphatase (ALP) is abundantly represented in nature, being fundamental for a number of processes. In addition to its fundamental function in skeletal mineralization, its roles in the pathogenesis of other diseases are being explored. The measurement of total ALP activity in serum or in plasma is a useful biomarker in clinical practice. Indeed, routine measurement of serum total ALP is a long-standing established part of initial biochemical evaluation of patients both in the hospital setting and on an ambulatory basis. Raised or reduced values of this enzyme activity are indicative o

Invited review. A contemporary approach to placental pathology.

A brief description is given of the significance of gross and histological abnormalities of the placenta. The question of placental ageing is discussed and the possible ill-effects of inflammation and immune-mediated damage to the placenta considered. Finally, the pathology of the utero-placental vasculature is outlined, and the view is expressed that most cases of apparent 'placental insufficiency' are, in reality, due to maternal vascular insufficiency.

Single-Cell Transcriptional Response of the Placenta to the Ablation of Caveolin-1: Insights into the Adaptive Regulation of Brain-Placental Axis in Mice.

Caveolin-1 (Cav1) is a major plasma membrane protein that plays important functions in cellular metabolism, proliferation, and senescence. Mice lacking Cav1 show abnormal gene expression in the fetal brain. Though evidence for placental influence on brain development is emerging, whether the ablation of Cav1 affects the regulation of the brain-placental axis remains unexamined. The current study tests the hypothesis that gene expression changes in specific cells of the placenta and the fetal brain are linked to the deregulation of the brain-placental axis in Cav1-null mice. By performing singl

SLC20a1/PiT-1 is required for chorioallantoic placental morphogenesis.

The placenta mediates the transport of nutrients, such as inorganic phosphate (Pi), between the maternal and fetal circulatory systems. The placenta itself also requires high levels of nutrient uptake as it develops to provide critical support for fetal development. This study aimed to determine placental Pi transport mechanisms using in vitro and in vivo models. We observed that Pi (P33) uptake in BeWo cells is sodium dependent and that SLC20A1/Slc20a1 is the most highly expressed placental sodium-dependent transporter in mouse (microarray), human cell line (RT-PCR) and term placenta (RNA-seq

Human placental extract ameliorates methotrexate-induced hepatotoxicity in rats via regulating antioxidative and anti-inflammatory responses.

PURPOSE: Methotrexate (MTX) induces hepatotoxicity, limiting its clinical efficacy as a widely known chemotherapy drug. In the current study, we examined the protective effect of human placenta extract (HPE) against MTX-induced liver damage in rats, as well as its ability to regulate antioxidative and anti-inflammatory liver responses. METHODS: Male rats were orally administered MTX at a daily dose of 5 mg/kg-body-weight in the presence or absence of HPE (10.08 mg/kg) for 2 weeks. We measured the biological effects of MTX and HPE on the levels of liver enzymes, lipid profile, lipid peroxidati
