Rapid generation of a tomato male sterility system and its feasible application in hybrid seed production.

A practical approach for the rapid generation and feasible application of green hypocotyl male-sterile (GHMS) tm6 dfr lines in tomato hybrid breeding was established. Male sterility enables reduced cost and high seed purity during hybrid seed production. However, progress toward its commercial application has been slow in tomato due to the disadvantages of most natural male-sterile mutants. Here, we developed a practical method for efficient tomato hybrid seed production using a male-sterile system with visible marker, which was rapidly generated by CRISPR/Cas9-mediated gene editing. Fine mapping and candidate gene analysis of qSB12YSB, a gene conferring major quantitative resistance to rice sheath blight.

qSB12YSB, a major quantitative sheath blight resistance gene originated from rice variety YSBR1 with good breeding potential, was mapped to a 289-Kb region on chromosome 12. Sheath blight (ShB), caused by Rhizoctonia solani kühn, is one of the most serious global rice diseases. Rice resistance to ShB is a typical of quantitative trait controlled by multiple quantitative trait loci (QTLs). Many QTLs for ShB resistance have been reported while only few of them were fine-mapped. In this study, we identified a QTL on chromosome 12, in which the qSB12YSB resistant allele shows significant ShB resis

Minimum sperm dose for optimal fertility after artificial insemination in ostriches.

Artificial insemination (AI) in ostriches may present potential solutions to high proportions of infertile eggs commonly recorded on commercial farms and assist in reducing the number of males for breeding purposes thereby leading to a more economical and efficient farming system. Although non-invasive methods to collect semen and to artificially inseminate female ostriches have been developed, the insemination dose for maximum fertility of eggs remains unknown. This study was thus conducted to determine the minimum sperm dose that would promote fertility of eggs following AI in female ostrich

Development and characterization of GR2E Golden rice introgression lines.

Golden Rice with β-carotene in the grain helps to address the problem of vitamin A deficiency. Prior to commercialize Golden Rice, several performance and regulatory checkpoints must be achieved. We report results of marker assisted backcross breeding of the GR2E trait into three popular rice varieties followed by a series of confined field tests of event GR2E introgression lines to assess their agronomic performance and carotenoid expression. Results from confined tests in the Philippines and Bangladesh have shown that GR2E introgression lines matched the performance of the recurrent parents

The knockout of ClaCSLH1 induced dwarfing in watermelon.

In agriculture, selecting ideal plant types with desirable traits, such as dwarfing and upright stem structures, significantly enhances crop yield and quality by optimizing light absorption, spatial efficiency, and nutrient utilization. Developing new varieties of dwarf watermelon is a crucial objective in watermelon breeding. In this study, we constructed an F2 population using the wild-type V063 as the paternal parent and the dwarf variety dw-n as the maternal parent. The dwarfing trait was found to be governed by a pair of recessive alleles. Through bulk segregant analysis sequencing (BSA-s

Nucleoredoxin gene SINRX1 negatively regulates tomato immunity by activating SA signaling pathway.

The tomato (Solanum lycopersicum) is widely consumed globally and renowned for its health benefits, including the reduction of cardiovascular disease and prostate cancer risk. However, tomato production faces significant challenges, particularly due to various biotic stresses such as fungi, bacteria, and viruses. To address this challenges, we employed the CRISPR/Cas9 system to modify the tomato NUCLEOREDOXIN (SlNRX) genes (SlNRX1 and SlNRX2) belonging to the nucleocytoplasmic THIOREDOXIN subfamily. CRISPR/Cas9-mediated mutations in SlNRX1 (slnrx1) plants exhibited resistance against bacterial

Artificial Selection on the GA2ox Gene Family Contributes to Plant Architecture Improvement in Upland Cotton.

Gibberellins (GAs) play a crucial regulatory role in the growth and development of cotton (Gossypium hirsutum L.). Through bioinformatics analyses, we identified a total of 39 GA2ox genes (encoding gibberellin 2-oxidases) in the cotton genome, designated GhGA2ox1 to GhGA2ox39. Based on phylogenetic analysis, these genes were classified into five groups. We further examined their gene structures, conserved motifs, and chromosomal distributions, revealing that members within the same group shared similar structural and motif organizations. Collinearity and cis-element analyses provided important

Simultaneous effects of leaf irradiance and soil moisture on growth and root system architecture of novel wheat genotypes: implications for phenotyping.

Plants in the field are exposed to varying light and moisture. Agronomic improvement requires knowledge of whole-plant phenotypes expressed in response to simultaneous variation in these essential resources. Most phenotypes, however, have been described from experiments where resources are varied singularly. To test the importance of varying shoot and root resources for phenotyping studies, sister pre-breeding lines of wheat were phenotyped in response to independent or simultaneous exposure to two light levels and soil moisture profiles. The distribution and architecture of the root systems d

Development of a low-coverage whole genome sequencing screen for apomixis using a diverse set of Malus germplasm.

In the past decade, plant biologists have made several major discoveries pertaining to the genetic basis of apomixis (clonal propagation by seed) that have shown promise in preserving high-value hybrid rice and sorghum genotypes. This progress was made possible by foundational gene discovery efforts in model species and natural apomicts, but pleiotropic obstacles still limit its broad agricultural adoption, especially in eudicots. Thus, it follows that investigations of novel apomicts should lead to the development of new molecular tools for plant breeding. The two most common ways to identify…

Magnesium Nutrient Application Induces Metabolomics and Physiological Responses in Mulberry (Morus alba) Plants.

Mulberry (Morus alba) is a significant plant with numerous economic benefits; however, its growth and development are affected by nutrient levels. A high level of magnesium (Mg) or magnesium nutrient starvation are two of the significant Mg factors affecting plant growth and development. Nevertheless, M. alba's metabolic response to different Mg concentrations is unclear. In this study, different Mg concentrations, optimal (3 mmol/L), high (6 mmol/L and 9 mmol/L), or low (1 and 2 mmol/L) and deficient (0 mmol/L), were applied to M. alba for three weeks to evaluate their effects via physiologic

Creation of fertility-restored materials for Ogura CMS in Brassica oleracea by introducing Rfo gene from Brassica napus via an allotriploid strategy.

Ogura CMS fertility-restored materials, with 18 chromosomes, normal seed setting, stable fertility and closer genetic background to the parent Chinese kale, were successfully developed in B. oleracea via a triploid strategy for the first time. Ogura cytoplasmic male sterility (CMS) is the most widely used sterile type in seed production for commercial hybrids of Brassica oleracea vegetables. However, the natural Ogura CMS restorer line has not been found in B. oleracea crops. In this study, the triploid strategy was used with the aim to create euploid B. oleracea progenies with the Rfo gene. T

An improved method for high-temperature induced embryo sac chromosome doubling in Populus simonii Carr. × P. nigra var. italica (Moench.) Kochne, an interspecific hybrid of Tacamahaca and Aigeiros poplars.

Chromosome doubling is considered an important technique in poplar breeding, with many triploid clones being artificially induced and selected for promotion in the north and northeast of China because of their outstanding traits in vegetative growth and environmental adaption. In this study, the triploid yield of Populus simonii Carr × P. nigra var. italica (Moench.) Kochne was 23.41%, which exceeded the yield attained in our previous studies due to the use of an optimized method of chromosome doubling in the embryo sac at a high temperature. StCYP90C1 modulates plant architecture via regulating brassinosteroid biosynthesis in potato.

Brassinosteroid (BR) exert a crucial regulatory role in shaping plant architecture. However, the BR biosynthesis and underlying genetic variations governing potato plant architecture remain poorly understood. Here, we identified a single recessive locus, ap, which controls the abnormal architectural plant (AP) with dark-green curled leaves, shortened internodes, and delayed flowering, using a BC4S1 population. The functional gene, StCYP90C1, encoding 3-epi-6-deoxocathasterone 23-monooxygenase, was identified at this locus. StCYP90C1 expression was significantly lower in the leaves of APs than

Chemical Signaling and Integrated Strategies for Biological Control in Crop Agroecosystems.

Food security faces significant challenges due to the growing threat of insect pests and global climate change, making it essential to adopt sustainable agricultural strategies. Plants possess intrinsic defense systems that are activated in response to herbivore attack, including the emission of specialized volatile organic compounds (VOC) known as herbivore-induced plant volatiles (HIPVs). These volatiles aid the foraging of natural enemies (predators and parasitoids), thereby contributing to biological control of pest populations. Harnessing these chemical defense mechanisms offers a promisi

Molecular dissection and functional characterization of the liguleless1 gene for manipulation of leaf angle in maize.

Recessive liguleless1 (lg1) gene significantly reduces leaf angle in maize and has become the choice in breeding for high plant density. Here, we sequenced the entire lg1 gene (5560 bp) among seven wild-type (Lg1) and one mutant (lg1) inbreds. The analysis revealed a total of 229 SNPs and 155 InDels within the Lg1 gene. The study also revealed the existence of three exons, with lg1-mutant having two exons. The lg1-mutant harboured an insertion of 130 bp Tourist MITE transposable element in exon-2 at 1663rd base, which deleted 157 amino acids of C-terminal region of the mutant LG1 protein. The

Genome-wide association study of diverse soybean [Glycine max (L.) Merrill] accessions for agronomic and seed composition traits in Rwanda.

This research explored the genetic basis of adaptation for soybean to the low-latitude but high-elevation production regions in Rwanda. Soybean [Glycine max (L.) Merrill] is an increasingly important crop in Rwanda; however, production remains constrained by limited availability of high-yielding, locally adapted cultivars. The varieties introduced through Rwanda's Crop Intensification Program were developed for other regions of sub-Saharan Africa and exhibit poor adaptation, delayed maturity and low productivity in Rwanda. This study evaluated diverse germplasm to support development of a nati
