Animal models in research on retinal degenerations: past progress and future hope.

The retinal degenerations (RDs) are a family of inherited retinal degenerative diseases (dystrophies) that lead to vision loss. Although phenotypically very different, the RDs have several characteristics in common. They all are caused by gene mutations or at least have a genetic component in the etiology. They all lead to photoreceptor dysfunction, many leading to the death of both rod and cone photoreceptors. The mechanism of cell death in most of the RDs seems to be through the process of apoptosis. It is estimated that more than fifteen million people around the world have vision loss due

Outer retinal tubulation associated with photoreceptor degeneration.

Outer retinal tubulation (ORT) is a distinct structural manifestation of chronic photoreceptor degeneration, observed across a broad spectrum of retinal diseases. Initially described histologically as rosette-like formations, ORT has gained clinical relevance with the advent of high-resolution imaging modalities such as spectral-domain optical coherence tomography (SD-OCT) and adaptive optics scanning laser ophthalmoscopy (AO-SLO), which enable in vivo visualization of its tubular architecture. ORT arises from sustained photoreceptor and retinal pigment epithelium (RPE) injury, leading to the

Bmal1 deletion alters mitochondrial microstructure and function in mouse cone photoreceptors.

The mammalian retina contains an autonomous circadian system that regulates ocular physiology. The deletion of the core clock gene Bmal1 in the mouse retina disrupts retinal circuitry, alters cone spectral identity, and reduces cone viability. Cone photoreceptors have the highest energy demand among retinal neurons and are continuously exposed to high levels of oxidative stress, making them susceptible to mitochondrial dysfunction. To investigate the role of Bmal1 in mitochondrial biology, we analyzed mitochondrial function and ultrastructure in 661W cells and mouse retinas lacking Bmal1. Loss

Chronic exposure to aerosolized Arizona test dust reduces visual acuity in mice.

Air pollution is associated with increased incidence of age-related macular degeneration (AMD), glaucoma and other retinal diseases. The time course of molecular and cellular changes induced by chronic air pollution exposure that lead to retinal pathology are unknown. In this study, we investigated the effects of moderate levels of air pollution on visual acuity and retinal phenotypes in mice using Arizona test dust (ATD) as a surrogate for ambient air pollution. Mice were exposed to aerosolized standardized test dust for three hours a day, four days a week, for up to four months in a custom-b

GCAP neuronal calcium sensor proteins mediate photoreceptor cell death in the rd3 mouse model of LCA12 congenital blindness by involving endoplasmic reticulum stress.

Loss-of-function mutations in the retinal degeneration 3 (RD3) gene cause inherited retinopathy with impaired rod and cone function and fast retinal degeneration in patients and in the natural strain of rd3 mice. The underlying physiopathology mechanisms are not well understood. We previously proposed that guanylate cyclase-activating proteins (GCAPs) might be key Ca2+-sensors mediating the physiopathology of this disorder, based on the demonstrated toxicity of GCAP2 when blocked in its Ca2+-free form at photoreceptor inner segments. We here show that the retinal degeneration in rd3 mice is su

Retinitis pigmentosa, pigmentary retinopathies, and neurologic diseases.

Retinitis pigmentosa (RP) refers to a group of inherited retinal diseases with phenotypic and genetic heterogeneity. The pathophysiologic basis of the progressive visual loss in patients with RP is not completely understood but is felt to be due to a primary retinal photoreceptor cell degenerative process mainly affecting the rods of the peripheral retina. In most cases RP is seen in isolation (nonsyndromic), but in some other cases it may be a part of a genetic, metabolic, or neurologic syndrome or disorder. Nyctalopia, or night blindness, is the most common symptom of RP. The classic fundus

Overproduction of bioactive retinoic acid in cells expressing disease-associated mutants of retinol dehydrogenase 12.

Retinol dehydrogenase 12 (RDH12) is an NADP(+)-dependent oxidoreductase that in vitro catalyzes the reduction of all-trans-retinaldehyde to all-trans-retinol or the oxidation of retinol to retinaldehyde depending on substrate and cofactor availability. Recent studies have linked the mutations in RDH12 to severe early-onset autosomal recessive retinal dystrophy. The biochemical basis of photoreceptor cell death caused by mutations in RDH12 is not clear because the physiological role of RDH12 is not yet fully understood. Here we demonstrate that, although bi-directional in vitro, in living cells

Gain-of-function nature of Cav1.4 L-type calcium channels alters firing properties of mouse retinal ganglion cells.

Proper function of Cav1.4 L-type calcium channels is crucial for neurotransmitter release in the retina. Our understanding about how different levels of Cav1.4 channel activity affect retinal function is still limited. In the gain-of-function mouse model Cav1.4-IT we expected a reduction in the photoreceptor dynamic range but still transmission toward retinal ganglion cells. A fraction of Cav1.4-IT ganglion cells responded to light stimulation in multielectrode array recordings from whole-mounted retinas, but showed a significantly delayed response onset. Another significant number of cells sh

Rod/Cone Gap Junctions: Plastic Electrical Synapses at the First Visual Synapse.

Electrical coupling mediated by gap junctions is a defining feature of almost every neuron in the retina, as it is throughout the central nervous system. By allowing direct current flow between neurons, electrical coupling helps establish functional circuits, allows signals to be shared, and improves the signal-to-noise ratio, thereby regulating neuronal activity at the dendritic, single-neuron, and network levels. Remarkably, electrical coupling is not static or constitutive. It shows a high degree of plasticity, changing in strength on time scales ranging from milliseconds to days, thereby p

Native components analysis of the spectral electroretinogram.

The electroretinogram (ERG) is a mass electrical response from all electrically activated components of the retina, recorded with the goal of identifying the individual contributions of relevant components for the purposes of electrodiagnosis of eye diseases and other systemic medical conditions. The primary hypothesis being tested was that the ERGs across the spectrum in the mesopic range of intensities could be fully accounted with a duplex (two-component) model of linear combinations of rod- and cone-pathway responses. Full-field square-wave ERGs were measured with the RETeval device at 2 H

The effect of TIMP-1 on the cone mosaic in the retina of the rat model of retinitis pigmentosa.

PURPOSE: The array of photoreceptors found in normal retinas provides uniform and regular sampling of the visual space. In contrast, cones in retinas of the S334ter-line-3 rat model for RP migrate to form a mosaic of rings, leaving large holes with few or no photoreceptors. Similar mosaics appear in human patients with other forms of retinal dystrophy. In the current study, we aimed to investigate the effect of tissue inhibitor of metalloproteinase-1 (TIMP-1) on the mosaic of cones in S334ter-line-3 rat retinas. We focused on TIMP-1 because it is one of the regulators of the extracellular matr

Circadian photoreception in humans and mice.

Circadian rhythms are the endogenous oscillations, occurring with a periodicity of approximately twenty-four hours, in the biochemical and behavioral functions of organisms. In mammals, the phase and period of the rhythm are synchronized to the daily light-dark cycle by light input through the eye. Certain retinal degenerative diseases affecting the photoreceptor cells, both rods and cones, in the outer retina reveal that classical opsins (i.e., rhodopsin and color opsins located in these cells) are essential for vision, but are not required for circadian photoreception. The mammalian cryptoch

Normal S cone electroretinogram b-wave in Oguchi's disease.

AIM: The short wavelength sensitive (S) cone electroretinograms (ERGs) were examined in two patients with Oguchi's disease to study S cone function. METHODS: Ganzfeld colour flashes under bright white background illumination were used to elicit S cone, and mixed long (L) and middle (M) wavelength sensitive cone ERGs. RESULTS: The S cone ERG b-wave was normal with short wavelength stimuli with normal L and M cone responses in both patients. CONCLUSIONS: These ERG results indicate that the S cone system as well as the L and M cone system is not defective in Oguchi's disease, while the S cone

Transcriptional Corepressors Panky and Panky-Like Modulate Ganglioside Levels and Are Essential for Retinal Cone Photoreceptor Structure and Survival.

The biological functions and mechanisms of many cell type-specific transcriptional cofactors remain unclear. We previously identified Panky (Ankrd33) as a repressive transcriptional cofactor that suppresses Crx-mediated transactivation of photoreceptor genes; however, the biological function of Panky remains unclarified. Here, we investigated the functions of Panky and its paralog, Panky-like (Ankrd33b), in mice of either sex. Single knock-out (KO) mouse retinas of Panky or Panky-like did not show significant alterations at both histological and functional levels compared with those of control

Retinal Pigment Epithelium Remodeling in Mouse Models of Retinitis Pigmentosa.

In retinitis pigmentosa (RP), one of many possible genetic mutations causes rod degeneration, followed by cone secondary death leading to blindness. Accumulating evidence indicates that rod death triggers multiple, non-cell-autonomous processes, which include oxidative stress and inflammation/immune responses, all contributing to cone demise. Inflammation relies on local microglia and recruitment of immune cells, reaching the retina through breakdowns of the inner blood retinal barrier (iBRB). Leakage in the inner retina vasculature suggests similarly altered outer BRB, formed by junctions bet

Predictive Mathematical Models for the Spread and Treatment of Hyperoxia-induced Photoreceptor Degeneration in Retinitis Pigmentosa.

PURPOSE: To determine whether the oxygen toxicity hypothesis can explain the distinctive spatio-temporal patterns of retinal degeneration associated with human retinitis pigmentosa (RP) and to predict the effects of antioxidant and trophic factor treatments under this hypothesis. METHODS: Three mathematical models were derived to describe the evolution of the retinal oxygen concentration and photoreceptor density over time. The first model considers only hyperoxia-induced degeneration, while the second and third models include mutation-induced rod and cone loss respectively. The models were f

Retinal structure and function in monkeys with fetal alcohol exposure.

Exposure to ethanol in utero leads to several brain development disorders including retinal abnormalities whose underlying cellular pathogenesis remains elusive. We recently reported that fetal alcohol exposure (FAE) in vervet monkeys induces anomalies of full-field electroretinogram (ERG) waveforms that suggest premature aging of the retina. The goal of this study is to characterize the anatomo-functional mechanisms underlying the retinal changes observed in fetal alcohol exposed (FAE) monkeys, and age- and sex-matched normals. First, we examined in vivo the fundus of the eyes, measured intra

Proton-mediated feedback inhibition of presynaptic calcium channels at the cone photoreceptor synapse.

Generation of center-surround antagonistic receptive fields in the outer retina occurs via inhibitory feedback modulation of presynaptic voltage-gated calcium channels in cone photoreceptor synaptic terminals. Both conventional and unconventional neurotransmitters, as well as an ephaptic effect, have been proposed, but the intercellular messaging that mediates the inhibitory feedback signal from postsynaptic horizontal cells (HCs) to cones remains unknown. We examined the possibility that proton concentration in the synaptic cleft is regulated by HCs and that it carries the feedback signal to

Multimodal Imaging in Choroideremia.

Choroideremia (CHM) is associated with progressive degeneration of the retinal pigment epithelium (RPE), choriocapillaris (CC), and photoreceptors. As animal models of CHM are lacking, most information about cell survival has come from imaging affected patients. This chapter discusses a combination of imaging techniques, including fundus-guided microperimetry, confocal and non-confocal adaptive optics scanning laser ophthalmoscopy (AOSLO), fundus autofluorescence (FAF), and swept-source optical coherence tomography angiography (SS-OCTA) to analyze macular sensitivity, cone photoreceptor outer
