Impact of Subretinal Drusenoid Deposits on Ellipsoid Zone-Related Thickness Metrics.

PURPOSE: Ellipsoid zone (EZ) attenuation is a widely used endpoint in retinal disease trials and is quantified as the distance between the EZ and retinal pigment epithelium (RPE). This study assessed the impact of subretinal drusenoid deposits (SDDs) on EZ-based quantitative metrics in nonneovascular age-related macular degeneration (AMD). METHODS: Spectral-domain optical coherence tomography volumes from 83 eyes (44 patients) with SDDs from the Amish Eye Study were analyzed. A semi-automated deep learning-based segmentation with manual correction delineated the inner EZ, inner RPE, and inner

Biomimetic hydrogel platform reveals active force transduction from retinal pigment epithelium to photoreceptors.

In the eye, the retinal pigment epithelium (RPE) maintains the functionality and welfare of retinal photoreceptors and forms a tight, interlocked structure with photoreceptor outer segments (POSs). The RPE-retina interaction is difficult to recapitulate in vitro, limiting the studies addressing the retinal maintenance functions of the RPE. To overcome this challenge, we constructed a retina-mimicking structure using a soft polyacrylamide hydrogel coated with Matrigel. This structure was introduced to RPE cells' apical side to model the RPE-retina interface in vitro. As a result, RPE cells atta

The retina of Spalax ehrenbergi: novel histologic features supportive of a modified photosensory role.

PURPOSE: The retina of the blind mole rat Spalax ehrenbergi was compared with other vertebrate photosensitive organs in an attempt to correlate its histologic organization with a presumptive nonvisual photoreceptor role. METHODS: The eyes of eight adult animals were analyzed by light and electron microscopy, using conventional staining and immunolabeling with antibodies against phototransduction proteins and calretinin. RESULTS: Rods accounted for most of the photoreceptor cells in the Spalax retina, although their morphology is dissimilar to that of sighted mammals, in that they contained o

Diabetes Renders Photoreceptors Susceptible to Retinal Ischemia-Reperfusion Injury.

PURPOSE: Studies have suggested that photoreceptors (PR) are altered by diabetes, contributing to diabetic retinopathy (DR) pathology. Here, we explored the effect of diabetes on retinal ischemic injury. METHODS: Retinal ischemia-reperfusion (IR) injury was caused by elevation of intraocular pressure in 10-week-old BKS db/db type 2 diabetes mellitus (T2DM) mice or C57BL/6J mice at 4 or 12 weeks after streptozotocin (STZ)-induced type 1 diabetes mellitus (T1DM), and respective nondiabetic controls. Retinal neurodegeneration was evaluated by retinal layer thinning, TUNEL staining, and neuron lo

Degenerated Cones in Cultured Human Retinas Can Successfully Be Optogenetically Reactivated.

Biblical references aside, restoring vision to the blind has proven to be a major technical challenge. In recent years, considerable advances have been made towards this end, especially when retinal degeneration underlies the vision loss such as occurs with retinitis pigmentosa. Under these conditions, optogenetic therapies are a particularly promising line of inquiry where remaining retinal cells are made into "artificial photoreceptors". However, this strategy is not without its challenges and a model system using human retinal explants would aid its continued development and refinement. Her

Vitamin E deficiency and the retina: photoreceptor and pigment epithelial changes.

To investigate the role of normal vitamin E levels and the interrelationships between vitamin E and A in maintaining the visual cells of the retina, weanling female Sprague-Dawley rats were fed vitamin E-free diets differing tenfold in their vitamin A content (0.8 and 8.0 mg of retinol per kilogram of diet). Rats on vitamin E-free diets with the higher vitamin A level exhibited marked disruption of photoreceptor outer segment membranes and a fivefold increase in the number of lipofuscin granules in the pigment epithelial cells which ingest these membranes. Rats on vitamin E-free diets with the

Potential role of myeloid bodies in protection against photo-oxidative damage of the retinal pigment epithelium.

Myeloid bodies are organized forms of the smooth endoplasmic reticulum that occur in the retinal pigment epithelium of vertebrates. The latter is a monolayer of cells, situated between the neural retina and the chroroid vasculature, that is essential for photoreceptor homeostasis and function. Myeloid bodies operate in an environment of high oxygen tension and chronic light exposure where phagocytosis of photoreceptor outer segments takes place, resulting in photo-oxidative stress. Over time, such damage can result in pigment epithelium cell death, retinal degeneration and blindness. The purpo

Intraperitoneal chromophore injections delay early-onset and rapid retinal cone degeneration in a mouse model of Leber congenital amaurosis.

Highly expressed in the retinal pigment epithelium (RPE), the RPE-specific 65-kDa (RPE65) enzyme is indispensable to generate 11-cis-retinal (11cRAL), a chromophore for rhodopsin and cone photopigments. RPE65 deficiency can lead to Leber congenital amaurosis type 2 (LCA2), in which the isomerization of photobleached all-trans-retinal into photosensitive 11cRAL is blocked, ultimately causing severe retinal dysfunction and degeneration. The related mouse models, which are constructed through gene knockout or caused by spontaneous mutations, morphologically present with early-onset and rapid reti

[The effect of cyclooxygenase inhibitors on the course of hereditary retinal dystrophy in RCS rats].

Daily administration of acetyl salicylic acid (ASA) and ibuprofen leads to an appreciable retardation in the process of retinal degeneration in the RCS rat which is dependent on the dosage given. The photoreceptor cell nuclei and inner segments are relatively well preserved in all regions of the retina. While the outer nuclear layer of 32 day old RCS rats is usually composed of only 3-4 rows, we found 8-9 nuclear rows exhibiting minimal pyknotic change in animals which had been treated with higher doses of ASA. These differences in layer thickness could be confirmed using morphometric analysis

Novel Porcine Retina Cultivation Techniques Provide Improved Photoreceptor Preservation.

Age-related macular degeneration (AMD) is the leading cause of blindness in industrialized countries among people over 60 years. It has multiple triggers and risk factors, but despite intense research efforts, its pathomechanisms are currently not completely understood. AMD pathogenesis is characterized by soft drusen in Bruch's membrane and involves the retinal pigment epithelium-Bruch's membrane-choroid complex and adjacent structures, like photoreceptors. This study explores the potential of novel cultivation techniques to preserve photoreceptors in retinal explants to gain better insights

Fine structure of the retinal epithelium and tapetum lucidum of the opossum (Didelphis virginiana).

The fine structure of the retinal epithelium has been studied by electron microscopy in the opossum (Didelphis virginiana). The retinal epithelium, over most of the retina, is typical of that in other vertebrates and consists of a single layer of heavily pigmented, cuboidal cells. These cells display extensive basal (scleral) infoldings and numerous apical (vitreal) processes which enclose photoreceptor outer segments. A semicircular area of the retinal epithelium in the superior fundus is further specialized as a tapetum lucidum. The reflecting material consists of a large quantity of lipoida

Specialised features of melanosomes in health and disease in the retinal pigment epithelium.

This mini-review focuses on melanosome biogenesis, positioning and function in the retinal pigment epithelium (RPE) where melanosomes absorb light scatter and protect against the harmful effects of photo-oxidation. RPE melanosomes share a common biogenesis pathway with those of skin melanocytes but are made primarily embryonically and are retained by the RPE throughout life. They do however move from the cell body into the apical processes which, in mammalian RPE, is regulated by a machinery related to that regulating melanosome distribution in skin melanocytes. Melanosomes in the RPE make ext

A 3D iPSC retina model reveals non-cell-autonomous and non-neuronal mechanism of photoreceptor degeneration in a lysosomal storage disorder.

UNLABELLED: Disruption of photoreceptor-retinal pigment epithelium (RPE) interface with loss of photoreceptor outer segments (POSs) in the retina is a pathological hallmark of several neurodegenerative and retinal diseases including lysosomal storage disorder's like CLN3 disease. However, the retina is a functional composite in vivo; and in vitro stem cell models of retina that enable investigation of the photoreceptor-RPE interface in healthy and diseased retina are lacking. Here, we developed a 3D human pluripotent stem cell (hPSC)-derived retina model to investigate the photoreceptor-RPE in

Slowly progressive changes of the retina and retinal pigment epithelium in Briard dogs with hereditary retinal dystrophy. A morphological study.

Seven eyes from 2 generations of Briard dogs (5 weeks--7 years old) with congenital night blindness and (in the second generation) impairment of day vision to varying degrees, were examined by light and electron microscopy. Specimens from 4 locations were studied: the central area, the midperiphery of the tapetal area, the upper periphery and the lower periphery. Disorientation of rod outer segment disc membranes was seen in the 5-week-old dog. Large electron-lucent inclusions were found in the RPE at 3.5 months of age. These inclusions occurred most frequently in the central and midperipheral

Retinal degeneration in cats fed casein. IV. The early receptor potential.

Electroretinographic studies of casein-fed cats with retinal taurine deficiency revealed that the early receptor potential (ERP) was initially normal in amplitude at a time when the a-wave and b-wave of the electroretinogram were substantially reduced or even nondetectable. The preserved ERP's in these taurine-deficient cats could be correlated with the histologic finding that their outer segments were relatively intact over 90% of the retinal area subtended by the test flash. The sequence of electroretinographic changes in these taurine-deficient cats was also consistent with previous biochem

Progesterone administration fails to protect albino male rats against photostress-induced retinal degeneration.

PURPOSE: Female patients show better recovery after brain injury and lower incidence of vascular diseases before menopause. The aim of this study was to test the protective effect of female sexual hormones against photostress-induced photoreceptor apoptosis. METHODS: Five week old male albino Sprague-Dawley rats were injected intraperitoneally with progesterone (60 mg/kg body weight) for 4 days. The control group was injected with the vehicle only (benzyl alcohol). Both groups were halved and one was stressed with light (2700 lux for 24 hours) and the other remained under the original dim cyc
