A mechanism of target mRNA selection and activity regulation in meiosis-related RBM46-MEIOC-YTHDC2 complex.

MEIOC and its two partners, RBM46 and YTHDC2, function as posttranscriptional regulators to promote mitotic-to-meiotic transition in mammalian germ cells. However, the molecular mechanism underlying target mRNA selection and degradation by MEIOC complex is largely unknown. Here, we demonstrate that exogenously expressed RBM46/MEIOC/YTHDC2 binds to Rad21 and Meioc 3'UTR and targets these transcripts for degradation in cultured somatic cells. YTHDC2 stabilizes MEIOC through inhibiting its ubiquitination, and the coiled-coil domain of MEIOC mediates its association with YTHDC2 and RBM46. The targ

Familial cartilage-hair hypoplasia: prenatal ultrasound features and clinical outcomes in three siblings with identical RMRP variants.

Cartilage-hair hypoplasia (CHH) is a rare autosomal recessive skeletal dysplasia. It is often associated with short stature, metaphyseal abnormalities, hair hypoplasia and immune dysfunction. This case describes a pregnant woman in her mid-30s with two previous children diagnosed with CHH with molecular confirmation of two pathogenic variants of the ribonuclease mitochondrial RNA processing gene. The couple is healthy and non-consanguineous. Routine ultrasound examination of the current pregnancy suggested this fetus was also likely to be affected. The couple opted not to perform invasive pren

Structural analysis of the active site architecture of the VapC toxin from Shigella flexneri.

The VapC toxin from the Shigella flexneri 2a virulence plasmid pMYSH6000 belongs to the PIN domain protein family, which is characterized by a conserved fold with low amino acid sequence conservation. The toxin is a bona fide Mg(2+) -dependent ribonuclease and has been shown to target initiator tRNA(fMet) in vivo. Here, we present crystal structures of active site catalytic triad mutants D7A, D7N, and D98N of the VapC toxin in absence of antitoxin. In all structures, as well as in solution, VapC forms a dimer. In the D98N structure, a Hepes molecule occupies both active sites of the dimer and

RNASET2 is required for ROS propagation during oxidative stress-mediated cell death.

RNASET2 is a ubiquitously expressed acidic ribonuclease that has been implicated in diverse pathophysiological processes including tumorigeneis, vitiligo, asthenozoospermia, and neurodegeneration. Prior studies indicate that RNASET2 is induced in response to oxidative stress and that overexpression of RNASET2 sensitizes cells to reactive oxygen species (ROS)-induced cell death through a mechanism that is independent of catalytic activity. Herein, we report a loss-of-function genetic screen that identified RNASET2 as an essential gene for lipotoxic cell death. Haploinsufficiency of RNASET2 conf

Androgens down-regulate myosin light chain kinase in human prostate cancer cells.

Androgens play a major role in the growth and survival of primary prostate tumors. The molecular mechanisms involved in prostate cancer progression are not fully understood but genes that are regulated by androgens clearly influence this process. We searched for new androgen-regulated genes using the Affymetrix GeneChip Human Genome U95 Set in the androgen-sensitive LNCaP prostate cancer cell line. Analysis of gene expression profiles revealed that myosin light chain kinase (MLCK) mRNA levels were markedly down-regulated by the synthetic androgen R1881. The microarray data were confirmed by ri

Local renin-angiotensin system in the pineal gland.

Besides the classical endocrine renin-angiotensin system (RAS), a local RAS has been described also in the brain. We attempted to clarify the existence of a local RAS in the pineal gland. Through the use of a ribonuclease protection assay, it proved possible to detect the mRNA for angiotensinogen (AOGEN), for the angiotensin receptor type 1A (AT1a) and 1B (AT1b) and for the angiotensin-converting enzyme (ACE) in pineal glands from rats. Renin mRNA, however, could not be found by this method. By in situ hybridization and immunocytochemistry, AOGEN mRNA was co-localized with the astrocyte marker

The molecular biology of hair cell regeneration in the avian cochlea.

The sensory cells of the ear, the hair cells, are damaged by loud noise or certain types of drugs. In the bird cochlea, new hair cells are produced to replace those that are lost. Regeneration also occurs in the vestibular epithelia of birds, fish, and mammals but does not occur in the mammalian cochlea. In order to further our understanding of the regeneration process in the bird cochlea, we have begun to identify the genes that are involved. However, the small size of this organ has made it difficult to use traditional molecular biology methods to address these problems. Recently, many molec

Targeted disruption of the estrogen receptor-alpha gene in female mice: characterization of ovarian responses and phenotype in the adult.

Targeted disruption of the mouse estrogen receptor-alpha gene (estrogen receptor-alpha knockout; ERKO) results in a highly novel ovarian phenotype in the adult. The ERKO mouse model was used to characterize ER alpha-dependent processes in the ovary. Visualization of the ovaries of 10-, 20-, and 50-day-old wild-type (WT) and ERKO mice showed that the ERKO phenotype developed between 20 and 50 days of age. Developmental progression through the primordial, primary, and antral follicle stages appeared normal, but functional maturation of preovulatory follicles was arrested resulting in atresia or

The role of angiogenin in bladder cancer: where are we in 2025?

PURPOSE: The diagnosis of bladder cancer (BCa), a condition that represents approximately 4% of new cancer cases in the United States, could be greatly improved by the development of non-invasive biomarker-based testing. Current diagnostic methods include cystoscopy and cytology; however, these techniques are overly invasive and may cause patient discomfort. Angiogenin (ANG), a member of the RNase A superfamily, was implicated in tumor growth due to its role in angiogenesis. ANG pathophysiology and potential diagnostic and therapeutic role in BCa are discussed. METHODS: A narrative review was

Synchronous detection of Burkholderia pseudomallei and its ceftazidime resistance mutation based on RNase-HII hydrolysis combined with lateral flow strip assay.

This study focused on the development of a reaction system using rhPCR to amplify a specific gene, ORF2, of B. pseudomallei and to identify the P174L mutation associated with increased drug resistance to ceftazidime (CAZ). The system incorporated universal primer probes and a simple temperature cycle reaction. The amplified products were then analyzed using lateral flow strip assay (LFSA) for strain identification and mutation interpretation. The developed system provides a reliable basis for diagnosing melioidosis and selecting appropriate drugs. Its potential impact is particularly significa

Simple, inexpensive and RNase-free purification of plasmid DNA by fractional precipitation with isopropanol.

We present a modified alkaline lysis method for purification of plasmid DNA (pDNA) from bacterial extract using fractional precipitation with isopropanol (FPI). This method includes two successive precipitations with 0.33 and 0.36 volumes of isopropanol and separates pDNA from total RNA and most of the lipopolysaccharides. Using different quality control tests, we demonstrate that plasmids purified with FPI show superior quality compared to plasmids prepared with commercial kits based on spin-column chromatography.

Non-meningothelial mesenchymal tumours of the CNS in the diagnostic practice of the pathologist.

INTRODUCTION: Primary mesenchymal tumours of the central nervous system with uncertain differentiation are rare. The 5th edition of the WHO classification groups three of these entities according to their molecular profiles into: intracranial mesenchymal tumours with FET-CREB fusion, sarcomas with CIC rearrangement, and primary intracranial sarcomas with DICER1 mutations. The objectives of this study were to carry out a systematic review of the literature and to determine whether a specific morphology predominates in relation to the proposed molecular types. MATERIALS AND METHODS: A systemati

Nuclease resistance and RNase H sensitivity of oligonucleotides bridged by oligomethylenediol and oligoethylene glycol linkers.

The properties of new chimeric oligodeoxynucleotides made of short sequences (tetramers, pentamers, octamers, and decamers) bridged by hexamethylenediol and hexaethylene glycol linkers have been investigated. These chimeric oligonucleotides showed an improved resistance toward snake venom 3'-phosphodiesterase, with an increased stability when a terminal 3'-3'-internucleotide phosphodiester bond is present. It also has been demonstrated that the hybrid complexes formed by bridged oligonucleotides and a complementary 20-mer RNA are able to elicit the activity of ribonuclease H (RNase H) from Esc

Purification of C/D-Box snoRNP and Fibrillarin-Associated Factors from Arabidopsis thaliana Plants Using GFP-Tagged Proteins.

Fibrillarin is an evolutionarily conserved methyltransferase found in Archaea, algae, yeast, animals, and plants. It catalyzes the 2'-O-methylation (2'-O-Me) of ribosomal RNAs (rRNAs), small nucleolar RNAs (snoRNAs), small nuclear RNAs (snRNAs), and transfer RNAs (tRNAs) as well as the glutamine methylation (Qme) in histone H2A. Fibrillarin interacts with specific accessory proteins and snoRNA guides to perform 2'-O-Me, although it can also act independently in the case of Qme. Additionally, fibrillarin has been reported to exhibit ribonuclease activity and interact with long non-coding RNAs t

In vitro cleavage requirements and specificities of mycobacterial RNase E.

Regulation of RNA pools allows for adaptation to stress, which is important in pathogenic bacteria such as Mycobacterium tuberculosis (Mtb). RNA degradation is a significant contributor to RNA abundance, and Ribonuclease (RNase) E has a rate-limiting role in degradation of most mycobacterial transcripts. Many open questions remain about the RNA substrate requirements and specificities for efficient cleavage by mycobacterial RNase E. Here, we demonstrate that RNase E is only active on substrates with a minimum length of approximately 27 nt using both Mtb and Mycolicibacterium smegmatis (Msm) en

Isolation and Analysis of Double-Stranded RNA from Virus-Infected Plant and Fungal Tissue.

A simple, rapid method for the isolation of double-stranded RNA (dsRNA) from virus-infected plant and fungal tissues provides a new approach to virus detection and identification. Diseased tissue was phenol-extracted to isolate cellular nucleic acids, and viral dsRNA was selectively purified from other nucleic acids by binding to cellulose powder in 15% ethanol either in small columns or by a batch procedure. The product was analyzed first by gel electrophoresis and then by ribonuclease treatment to identify dsRNA. The method permits rapid and efficient isolation and analysis of dsRNA from sma

CRISPR-Mediated Genome Engineering in Aedes aegypti.

CRISPR-mediated genome engineering technologies have been adapted to a wide variety of organisms with high efficiency and specificity. The yellow fever mosquito, Aedes aegypti , is one such organism. It is also responsible for transmitting a wide variety of deadly viruses including Dengue, Zika, Yellow fever, and Chikungunya. The key to successful CRISPR-mediated gene editing applications is the delivery of both Cas9 ribonuclease and single-guide RNA (sgRNA ) to the nucleus of desired cells. Various methods have been developed for supplying the Cas9 endonuclease, sgRNA , and donor DNA to Ae. a

Anti-gliomas Effect of Chlorotoxin-Conjugated Onconase at High Dose.

Malignant gliomas are rarely curable malignant tumors in the central nervous system. Chlorotoxin (CTX) is a peptide derived from scorpion venom, which can selectively target malignant gliomas. Onconase (Onc) is a small cytotoxic ribonuclease derived from frogspawn that exhibits cytotoxicity against some tumor cells. In the present study, we found that CTX-conjugated Onc (CTX-Onc) shows better anti-tumor effect than the physical mixture of CTX and Onc (CTX + Onc) on the nude mice carrying subcutaneous glioblastoma cell-derived tumor. However, CTX-Onc does not show dose-dependent anti-tumor effe
