Induction of a heat-shock-type response in Saccharomyces cerevisiae following glucose limitation.

The protein pattern of yeast cells which have arrested proliferation in response to glucose exhaustion is drastically different from that of exponentially growing cells (Boucherie, 1985). In this study, we used two-dimensional gel electrophoresis to characterize the protein events responsible for these alterations. We found that the induction of heat-shock proteins is one of the major events responsible for these changes. This induction accounts for the synthesis of 18 of the 35 novel polypeptides observed in glucose-limited cells. It was shown to occur in combination with two other protein ev

Enzyme-substitution therapy with the yeast Saccharomyces cerevisiae in congenital sucrase-isomaltase deficiency.

Sucrase-isomaltase deficiency is an inherited disaccharidase deficiency that leads to malabsorption of sucrose, with resulting diarrhea and abdominal distention and cramps. We investigated the sucrose-splitting effect of viable yeast cells in eight children with congenital sucrase-isomaltase deficiency, by means of the sucrose hydrogen breath test. This test is based on the fact that hydrogen is released from the malabsorbed sucrose by the colonic microflora. We found that 0.3 g of lyophilized Saccharomyces cerevisiae, given after loading with 2 g of sucrose per kilogram of body weight, reduce…

Protein synthesis during transition and stationary phases under glucose limitation in Saccharomyces cerevisiae.

Metabolic changes have been investigated during continuous growth of yeast cells inoculated in glucose-containing medium until the cells entered the stationary phase in response to glucose exhaustion. Well in advance of glucose exhaustion, a transition phase was observed, characterized by a decrease in the growth rate and a progressive reduction of protein and RNA accumulation. Two-dimensional gel analysis of the proteins synthesized during this stage showed that the pattern of proteins remained similar to that of log-phase cells. When the cells entered the stationary phase, protein accumulati

Adenosine 3',5'-phosphate phosphodiesterase and pheromone response in the yeast Saccharomyces cerevisiae.

Theophylline, aminophylline, and isobutylmethylxanthine, compounds reported to be inhibitors of adenosine 3',5'-phosphate (cAMP) phosphodiesterase, prevented the alpha-factor-induced cell cycle arrest of Saccharomyces cerevisiae a cells. To determine whether the in vivo effect of these methylxanthines on yeast pheromone response was related to their known biochemical mode of action, two assays for cAMP phosphodiesterase based on affinity of the product of the reaction (5'-AMP) for boronate groups were developed and were used to monitor the activity of the low Km cAMP phosphodiesterase present …

Ultra-high-field solid-state NMR of intact Saccharomyces cerevisiae cells at 1.2 GHz.

The increasing availability of ultra-high-field NMR spectrometers is opening new opportunities for the characterization of complex and structurally heterogeneous biological assemblies by solid-state NMR, including lipid membranes, cell walls, and even intact cells. Here, we investigate the use of a 1.2 GHz (28.2 T) spectrometer, the highest magnetic field currently commercially available, for the analysis of intact Saccharomyces cerevisiae cells. Budding yeast is an experimentally tractable eukaryotic model organism that is widely used in biotechnology and industrial fermentation. Characteriza…

Adaptive laboratory evolution of maltose utilization in wild Irish isolates of Saccharomyces eubayanus.

The first European isolates of Saccharomyces eubayanus were discovered in Ireland in 2022 and belong to the same clade as the S. eubayanus parent of the hybrid lager yeast Saccharomyces pastorianus. Here, we report the isolation of 10 additional Irish strains and we explore maltose metabolism. Maltose metabolism genes are clustered in subtelomeric MAL loci including maltase (MALS), maltose transporter (MALT), and regulator (MALR). Despite having intact MAL loci, the Irish strains do not grow on maltose. Two Irish S. eubayanus strains (UCD650 and UCD926) were passaged in medium containing malto…

Bacteriocin-producing Saccharomyces cerevisiae inhibits bacterial contamination in raw starch-to-ethanol fermentations.

Contaminating lactic acid bacteria (LAB) compete with yeast cells for nutrients during starch-to-ethanol fermentations, affecting yeast cell viability and ethanol yields, with significant economic implications. Antibiotics are typically used to control LAB contaminants in the bioethanol industry; however, the global rise in antibiotic resistance necessitates the use of alternative antimicrobials, such as recombinant bacteriocins. In this study, 31 LAB were isolated from red sorghum flour fermentations, including Enterococcus (80.65%), Pediococcus (16.13%) and Lactococcus (3.22%) species, with

Transcriptomics-guided identification of engineering targets for improving salt tolerance in Saccharomyces cerevisiae.

BACKGROUND: High-salinity conditions frequently impair the fermentation performance of Saccharomyces cerevisiae in industrial processes involving high-osmolarity substrates. Identifying genetic determinants that enhance salt tolerance is therefore essential for the development of robust yeast cell factories. In this study, a comparative transcriptomic analysis was performed to investigate the transcriptional responses of a salt-tolerant strain, E-158, and its parental strain, KF-7, under 1.25 M NaCl stress, with the aim of identifying potential targets for strain engineering. RESULTS: Compara…

ENA1 deficiency attenuates Saccharomyces 'boulardii' probiotic yeast virulence in immunosuppressed mouse fungaemia model.

Recently, fungal infections originating from the probiotic Saccharomyces 'boulardii' yeast are increasingly reported. Here, we aimed to reveal the background of and to diminish the virulence of this yeast, mitigating infection risks in vulnerable patient groups. Product and human isolates of S. 'boulardii' were subjected to in-host selection and their subclone lineages were compared phenotypically to identify target phenotypes and associated genes. More virulent isolates showed signs of selection for high osmotic tolerance in immunosuppressed mouse model, hence the genes NHA1 and ENA1 were del…

Metabolic engineering of Saccharomyces cerevisiae for redox-assisted co-fermentation of l-rhamnose.

Rapid proliferation of Ulva species, commonly referred to as green tides, significantly impacts coastal regions globally, causing environmental degradation and economic disruption. Efficient utilization of Ulva biomass as a sustainable feedstock requires microbial strains capable of metabolizing its diverse sugars, including l-rhamnose, a major ulvan-derived carbohydrate constituent that industrial yeast Saccharomyces cerevisiae cannot naturally utilize. Here, we engineered S. cerevisiae for l-rhamnose utilization by integrating a heterologous l-rhamnose transporter and a complete fungal l-rha…

De novo coassembly of functional chlorophyll- and carotenoid-protein complexes in Saccharomyces cerevisiae.

Chlorophylls are essential for photosynthesis, yet their heterologous production is a major challenge in synthetic biology due to pathway complexity and the diversion of resources away from essential cofactor biosynthesis. Here, we reconstruct the complete chlorophyll biosynthetic pathway in Saccharomyces cerevisiae. By integrating 13 heterologous genes and deleting 3 endogenous genes, we engineered a yeast chassis capable of producing chlorophyll a at titers of 26 micrograms per gram of dry cell weight. This platform was further expanded with a ketocarotenoid biosynthetic module to produce ec…

Increased replication-associated single-stranded DNA promotes formaldehyde-induced mutagenesis in Saccharomyces cerevisiae.

Formaldehyde (FA) is an environmentally abundant and endogenously produced aldehyde that has been shown to cause DNA damage, mutagenesis, and carcinogenesis. Several studies have demonstrated that FA induces guanine mutations resulting in a mutation signature like SBS40. In this work, we use Saccharomyces cerevisiae to demonstrate that replication defects generating single-stranded DNA (ssDNA) caused by the downregulation of the major replicative polymerases results in elevated FA mutagenesis. We found that loss of Mrc1 (CLASPIN) resulted in a high accumulation of ssDNA and FA mutagenesis, and…

Genetic pre-adaptations in Saccharomyces cerevisiae Andean chicha isolates facilitate industrial brewery application.

Strains of Saccharomyces cerevisiae were isolated from a traditionally produced Andean maize-based chicha from Ecuador and characterised with respect to their potential use in industrial beer brewing. Whole-genome sequencing revealed that the strains were related to the 'French Guiana' and 'Mexican Agave' S. cerevisiae clades, though the available evidence indicates that they belong to a previously undescribed population, and are thus unrelated to traditional European brewing strains. Small-scale screening for wort fermentation revealed two strains with brewing potential. These outperformed co…

Co-culture dynamics between Lachancea cidri and Saccharomyces cerevisiae impact fermentative aroma profile.

The growing demand for more aromatized and complex fermented beverages has encouraged research into non-conventional yeasts that combine bio-flavor and reproducible fermentation profiles. In this study, we explore the use of the non-conventional yeast Lachancea cidri strain CBS2950 to ferment Synthetic Wine Must in single (SWM 60 and 180 mg/L YAN) and mixed (SWM 180 mg/L YAN) fermentations with the commercial Saccharomyces cerevisiae strain EC1118. We identified that L. cidri can tolerate high ethanol concentrations (10-12 % v/v) and antimicrobial compounds commonly used in wine, such as coppe…

Inhibition of diastatic yeasts by Saccharomyces killer toxins to prevent hyperattenuation during brewing.

Secondary fermentation in beer can result in undesirable consequences, such as off-flavors, increased alcohol content, hyperattenuation, gushing, and the spontaneous explosion of packaging. Strains of Saccharomyces cerevisiae var. diastaticus are a major contributor to such spoilage due to their production of extracellular glucoamylase enzyme encoded by the STA1 gene. Saccharomyces yeasts can naturally produce antifungal proteins named "killer" toxins that inhibit the growth of competing yeasts. Challenging diastatic yeasts with killer toxins revealed that 91% of strains are susceptible to the…

Dehydrozingerone enhances the fungicidal activity of glabridin against Saccharomyces cerevisiae and Candida albicans.

Drug resistance commonly occurs when treating immunocompromized patients with fungal infections. Dehydrozingerone-a phenolic compound isolated from the rhizome of Zingiber officinale-inhibits drug efflux in Saccharomyces cerevisiae by overexpression of the ATP-binding cassette (ABC) transporter Pdr5p. We aimed to investigate whether dehydrozingerone enhances the antifungal activity of glabridin-an isoflavan isolated from the roots of Glycyrrhiza glabra L.-by attenuating multidrug resistance through the intrinsic expression system of multidrug-efflux-related genes in a wild-type strain of the m…

Heterologous Expression of Candida Antifungal Target Genes in the Model Organism Saccharomyces cerevisiae.

Understanding how a gene variant may influence antifungal resistance, or other phenotypic characteristics, is an important step in determining or dissecting resistance mechanisms. The influence of specific genes or gene alleles on a phenotype can initially be assessed within the model organism, Saccharomyces cerevisiae. S. cerevisiae exhibits efficient rates of homologous recombination making it amendable for heterologous expression and represents a susceptible organism that can be used to determine changes in antifungal susceptibilities. Many groups have developed different methodologies for …
