Tiger nut and walnut extracts modulate extracellular metabolism of ATP and adenosine through the NOS/cGMP/PKG signalling pathway in kidney slices.

BACKGROUND: Tiger nut (Cyperus esculentus L.) and walnut (Tetracarpidium conophorum Müll. Arg.) have been reportedly used in the treatment of inflammatory diseases such as atherosclerosis, prevent heart attack and improve blood circulation, reduce serum cholesterol level as well as inhibit oxidation reactions. PURPOSE: This study investigated the effect of tiger nut and walnut hydro-alcoholic extracts on extracellular metabolism of ATP through the NOS/cGMP/PKG signaling pathway induced by Nω-nitro-L-arginine methyl ester hydrochloride (L-NAME) in kidney slices. METHODS: The plants were extra

Catch a tiger snake by its tail: Differential toxicity, co-factor dependence and antivenom efficacy in a procoagulant clade of Australian venomous snakes.

A paradigm of venom research is adaptive evolution of toxins as part of a predator-prey chemical arms race. This study examined differential co-factor dependence, variations relative to dietary preference, and the impact upon relative neutralisation by antivenom of the procoagulant toxins in the venoms of a clade of Australian snakes. All genera were characterised by venoms rich in factor Xa which act upon endogenous prothrombin. Examination of toxin sequences revealed an extraordinary level of conservation, which indicates that adaptive evolution is not a feature of this toxin type. Consisten

Dissociation of lethal toxicity and enzymic activity of notexin from Notechis scutatus scutatus (Australian-tiger-snake) venom by modification of tyrosine residues.

Notexin from Notechis scutatus scutatus snake venom was subjected to tyrosine modification with p-nitrobenzenesulphonyl fluoride (NBSF), and four modified derivatives were separated by h.p.l.c. The results of amino acid analysis and sequence determination revealed that only Tyr-7, Tyr-70 and Tyr-77 were modified in notexin. Modification of Tyr-7 resulted in decreases in lethal toxicity and enzymic activity by 70.2% and 22.7% respectively. Conversely, modification of Tyr-77 caused a 1.8-fold increase in enzymic activity, in contrast with the loss of 52.5% of lethality. A drastic decrease in let

Fear of humans mediates spatiotemporal interactions between endangered large Asian felids.

Species competitive interactions influence carnivore demography. Humans, as global 'super predators,' may mediate interactions among sympatric carnivores, although the effects of ecological fear on these interactions remain less understood. We used two years of camera trap data from more than 300 sites in northeast China to fit two-species, dynamic occupancy models. We then assessed how direct human passage affects spatial and temporal interactions between tigers and leopards, and the consequences for predator‒prey overlap in a temperate forest. We found evidence that median site occupancy, co

Larvicidal and Nematicidal Activities of 3-Acylbarbituric Acid Analogues against Asian Tiger Mosquito, Aedes albopictus, and Pine Wood Nematode, Bursaphelenchus xylophilus.

Widespread concern for the occurrence of resistant strains, along with the avoidance of the use of highly toxic insecticides and their wide environmental dispersal, highlights the need for the development of new and safer pest control agents. Natural products provide inspiration for new chemical entities with biological activities, and their analogues are good lead compounds for the development of new pest control agents. For this purpose, we evaluated the larvicidal and nematicidal activities of 48 3-acylbarbituric acid analogues against the Asian tiger mosquito, Aedes albopictus and the pine

A novel tool to identify the relative contribution of lymphoid cell types that contribute to IL-10 production during the infection with Schistosoma mansoni: the TIGER index.

INTRODUCTION: Infection with the trematode helminth Schistosoma mansoni affects more than 200 million people worldwide. Infected patients are thought to show a decreased incidence of asthma and autoimmune diseases, which is, among others, considered a result of an increased production of the immunoregulatory cytokine IL-10. However, the location and the type of cell that is responsible for the highest production of IL-10 in vivo are still unknown. AIM: Identification of the hierarchy of IL-10 producing cell types in the mesenteric lymph node and spleen during the course of the murine infectio

The action of notexin from tiger snake venom (Notechis scutatus scutatus) on acetylcholine release and compartmentation in synaptosomes from electric organ of Torpedo marmorata.

At rest, in the presence of calcium, notexin induced a rapid and concentration-dependent leakage of acetylcholine from nerve endings. In the presence of 20 nM notexin (5 min), synaptosomes were well-preserved structurally and they responded to addition of A23187 ionophore by a normal calcium-dependent acetylcholine release. When stimulated by high-K+ depolarization, evoked acetylcholine release was increased when notexin was present. These findings demonstrate that notexin (up to 100 nM) does not inhibit the acetylcholine release process itself. Further studies on intracellular acetylcholine c

Assessment of a Tiger Milk Medicinal Mushroom (Lignosus rhinocerus TM02®)-Based Oral Gel for Pain Reduction and Healing in Minor Aphthous Stomatitis.

Recurrent aphthous stomatitis (RAS) is a common oral mucosal condition characterized by painful ulcerations with no consistently effective treatment. Lignosus rhinocerus (tiger milk mushroom) has been traditionally used in Southeast Asia for its wound-healing and anti-inflammatory properties, yet its clinical application for RAS remains insufficiently studied. This double-blind, randomized clinical trial evaluated the efficacy and safety of an oral gel containing L. rhinocerus TM02® (xLr®) extract in adults with minor RAS. Twenty-one participants aged 20 to 50 years were allocated to placebo

Genetic Diversity and Biological Characteristics of a Novel Feline Calicivirus Strain Isolated From Zoo-Captive South China Tiger (Panthera tigris amoyensis).

Feline calicivirus (FCV) is a common pathogen causing upper respiratory tract disease in domestic cats. Nonetheless, its threat and genetic features in endangered wild felids remain poorly understood. In this study, a novel FCV strain from a captive South China tiger (Panthera tigris amoyensis) was isolated, designated as FCV LYH1224. Viral RNA was detected in multiple tissues of the deceased tiger, including the heart, liver, spleen, lungs, and kidneys, suggesting broad tissue tropism and FCV contributed substantially to the fatal outcome. In vitro growth kinetics showed that the virus replic

Trophic Ecology of the Tiger Shark (Galeocerdo cuvier) and Bengal Whipray (Brevitrygon imbricata) Harvested by Sri Lankan Fisheries Based on Stable Isotope Analysis.

Sri Lankan fisheries have substantial elasmobranch catches, but the local ecology of individual species is not well characterized. We examine the tiger shark (Galeocerdo cuvier) and Bengal whipray (Brevitrygon imbricata), two elasmobranch species with variable life history and feeding ecology that represent differing trophic guilds. Tiger sharks have a global distribution and are well-studied in some regions, but there is a lack of ecological information specific to the Indian Ocean. In contrast, Bengal whiprays are often misidentified at the species level and are thought to largely feed on be

A randomized non-inferiority study of low-dose and standard-dose ticagrelor after intervention for acute coronary syndrome: study protocol for the TIGER STUDY.

BACKGROUND: Current guidelines recommend that patients with acute coronary syndrome (ACS) who have successfully undergone percutaneous coronary intervention (PCI) should continue to use dual antiplatelet therapy (DAPT) for 12 months. The long-term use of standard-dose dual antiplatelet therapy will increase the risk of bleeding. An optimized antiplatelet strategy that can prevent ischemic events and reduce the risk of bleeding remains to be explored. METHODS: The study is a prospective, multicenter, randomized, open-label, controlled study involving 2090 patients from six clinical centers in

Coagulation factor activity patterns of venom-induced consumption coagulopathy in naturally occurring tiger snake (Notechis scutatus) envenomed dogs treated with antivenom.

BACKGROUND: Venom-induced consumption coagulopathy (VICC) from tiger snake (Notechis scutatus) envenomation results in a dose-dependent coagulopathy that is detectable on coagulometry. However, individual coagulation factor activities in dogs with tiger snake envenomation have not been determined. This study aimed to characterise VICC and the time course of recovery in tiger snake envenomed dogs and to investigate an association between tiger snake venom (TSV) concentrations and factor activity. METHODS: This was a prospective, observational, cohort study. The study cohort was 11 dogs of any

Role of Clathrin Assembly Protein-2 Beta Subunit during White Spot Syndrome Virus Infection in Black Tiger Shrimp Penaeus monodon.

White spot syndrome virus (WSSV) is one of the most lethal viruses severely affecting shrimp industry. This disease can cause 100% mortality of farmed shrimp within a week. This work aims to characterize clathrin assembly proteins in Penaeus monodon and investigate their roles in WSSV entry. In general, clathrin assembly proteins form complexes with specific receptors and clathrins, leading to clathrin-mediated endocytosis. Adaptor protein 2 (AP-2), which is responsible for endocytosis at plasma membrane, consists of four subunits including α, β2, μ2 and σ2. Knockdown of clathrin coat AP17, or

Genome Sequencing and Genome-Wide Analysis Uncover Candidate Genes and Loci Linked to Key Production Traits in the Philippine Black Tiger Shrimp (Penaeus monodon).

Penaeus monodon (black tiger shrimp) is a key species in aquaculture globally and in the Philippines. The selection and development of stocks with desirable production traits, such as enhanced growth and disease resistance, are critical for improving production. In this study, we leveraged a newly assembled, high-quality genome sequence from a Philippine population to perform genome-wide scans for SNPs associated with key production traits. Using cohort-based extreme phenotype GWAS (XP-GWAS), we identified variants linked to resistance against white spot syndrome virus (WSSV), body weight, and
